ABSTRACT

The walls of venules of paracortical zone of popliteal lymph nodes were studied in 127 2-5 Ys old male dogs (15-20 kg of body weight) after subcutaneous injection of BCG vaccine 0.2 mg/kg in dose. 6 hrs after the administration of BCG vaccine the venule diameter was found to reduce. Then it gradually increased and returned to initial parameters after 3 months. Lymphocyte number was increased in venular lumen and walls following antigenic treatment. Microvilli number growth was observed in endotheliocytes. Basement membrane in the venules was loosened. Lymphocytes through endotheliocytes. Maximal lymphocyte activity and modifications of venular walls was observed on d 7 following the BCG vaccine administration.

ABSTRACT

In the experiment performed on 127 dogs by means of cytospectrofluorometric analysis, using fluorochrome acridine orange in dynamics up to 1 year, changes in the level of chromatin activation and RNA content have been studied in lymphocytes of the germinative centers and the crown of lymphoid nodules, in the paracortical zone and medullary cords of the regional and contralateral popliteal lymph nodes, after subcutaneous injection of antigen (BCG vaccine, 0.2 mg/kg) into the lateral area of the foot of the left pelvic extremity. The immune response is accompanied with a periodical increase in the level of chromatin activation and RNA content in populations of lymphocytes in the regional and contralateral popliteal lymph nodes with maximum in 6 h, 3-7 days, 1-3 months after the antigen injection. The intensity of these processes has an unequal level in lymphoid cells of various structural components; it is higher in lymphocytes of the contralateral lymph node.

ABSTRACT

By means of morphometry, light and electron microscopy methods peculiarities in distribution of small, middle and large lymphocytes, as well as plasmocytes in various zones of the popliteal lymph nodes have been studied in normal and in dynamics up to one year after subcutaneous injection of BCG vaccine into the left hind paw. The antigen produces certain changes in density and morphological parameters of lymphoid cells both in the regional and in the contralateral lymph nodes. For them 3 periods are specific. During the first 3 days they are not antigen-dependent (stipulated by the stress reaction), during 7-24 days antigen-dependent processes of proliferation and differentiation of lymphocytes get into action. In 3 months a new wave of the immune response is observed.

ABSTRACT

Investigation of the character of the luminiscent spectrum of nuclear structures in periferal blood leucocytes stained with acridine orange showed that a specific alteration of leucocytes of an animal sensitized by Klebsiella rhinoscleromatis causes an increase in the values of the luminescence relative intensity Jrel=J640 divided by J530. This indicates to DNA denaturation, occuring in the process of alteration.

ABSTRACT

Neutrophil segmentonuclear leukocytes and lymphocytes of the human peripheral blood vitally stained with Acridine Orange (AO) in concentrations of 250 and 330 mcg/ml show different fluorescence dynamics. The number of neutrophil segmentonuclear granulocytes with green fluorescence of nuclei decreases, whereas the number of cells with red fluorescence of nuclei increases. As a criterion of this process, time T1/2 is taken during which the number of green-fluorescent cells decreases twofold. With AO concentrations of 250 and 330 mcg/ml, T1/2 is equal to 40 or 5 minutes, resp. The nuclei of lymphocytes within a 60 minutes observation show green fluorescence. This effect is likely to be due to structural-functional peculiarities of neutrophil segmentonuclear granulocytes and lymphocytes.
