ABSTRACT

Soybean is the main oilseed cultivated worldwide. Even though Brazil is the world's largest producer and exporter of soybean, its production is severely limited by biotic factors. Soil borne diseases are the most damaging biotic stressors since they significantly reduce yield and are challenging to manage. In this context, the present study aimed to evaluate the potential of a bacterial strain (Ag109) as a biocontrol agent for different soil pathogens (nematodes and fungi) of soybean. In addition, the genome of Ag109 was wholly sequenced and genes related to secondary metabolite production and plant growth promotion were mined. Ag109 showed nematode control in soybean and controlled 69 and 45% of the populations of Meloidogyne javanica and Pratylenchus brachyurus, respectively. Regarding antifungal activity, these strains showed activity against Macrophomia phaseolina, Rhizoctonia solani, and Sclerotinia sclerotiorum. For S. sclerotiorum, this strain increased the number of healthy plants and root dry mass compared to the control (with inoculation). Based on the average nucleotide identity and digital DNA-DNA hybridization, this strain was identified as Bacillus velezensis. Diverse clusters of specific genes related to secondary metabolite biosynthesis and root growth promotion were identified, highlighting the potential of this strain to be used as a multifunctional microbial inoculant that acts as a biological control agent while promoting plant growth in soybean.

ABSTRACT

Chili peppers (Solanaceae family) have great commercial value. They are commercialized in natura and used as spices and for ornamental and medicinal purposes. Although three whole genomes have been published, limited information about satellite DNA sequences, their composition, and genomic distribution has been provided. Here, we exploited the noncoding repetitive fraction, represented by satellite sequences, that tends to accumulate in blocks along chromosomes, especially near the chromosome ends of peppers. Two satellite DNA sequences were identified (CDR-1 and CDR-2), characterized and mapped in silico in three Capsicum genomes (C. annuum, C. chinense, and C. baccatum) using data from the published high-coverage sequencing and repeats finding bioinformatic tools. Localization using FISH in the chromosomes of these species and in two others (C. frutescens and C. chacoense), totaling five species, showed signals adjacent to the rDNA sites. A sequence comparison with existing Solanaceae repeats showed that CDR-1 and CDR-2 have different origins but without homology to rDNA sequences. Satellites occupied subterminal chromosomal regions, sometimes collocated with or adjacent to 35S rDNA sequences. Our results expand knowledge about the diversity of subterminal regions of Capsicum chromosomes, showing different amounts and distributions within and between karyotypes. In addition, these sequences may be useful for future phylogenetic studies.

ABSTRACT

Soybean and maize are some of the main drivers of Brazilian agribusiness. However, biotic and abiotic factors are of great concern, causing huge grain yield and quality losses. Phosphorus (P) deficiency is important among the abiotic factors because most Brazilian soils have a highly P-fixing nature. Thus, large amounts of phosphate fertilizers are regularly applied to overcome the rapid precipitation of P. Searching for alternatives to improve the use of P by crops is essential to reduce the demand for P input. The use of multifunctional rhizobacteria can be considered one of these alternatives. In this sense, the objective of the present work was to select and validate bacterial strains with triple action (plant growth promoter, phosphate solubilizer, and biocontrol agent) in maize and soybean, aiming to develop a multifunctional microbial inoculant for Brazilian agriculture. Bacterial strains with high indole acetic acid (IAA) production, phosphate solubilization, and antifungal activity against soil pathogenic fungi (Rhizoctonia solani, Macrophomina phaseolina, and Fusarium solani) were selected from the maize rhizosphere. Then, they were evaluated as growth promoters in maize under greenhouse conditions. Based on this study, strain 03 (Ag75) was selected due to its high potential for increasing biomass (root and shoot) and shoot P content in maize. This strain was identified through genomic sequencing as Bacillus velezensis. In field experiments, the inoculation of this bacterium increased maize and soybean yields by 17.8 and 26.5%, respectively, compared to the control (25 kg P2O5). In addition, the inoculation results did not differ from the control with 84 kg P2O5, indicating that it is possible to reduce the application of phosphate in these crops. Thus, the Ag75 strain has great potential for developing a multifunctional microbial inoculant that combines the ability to solubilize phosphate, promote plant growth, and be a biocontrol agent for several phytopathogenic fungi.

ABSTRACT

Increasing phosphorus (P) use efficiency in agricultural systems is urgent and essential to significantly reduce the global demand for this nutrient. Applying phosphate-solubilizing and plant growth-promoting bacteria in the rhizosphere represents a strategy worthy of attention. In this context, the present work aimed to select and validate bacterial strains capable of solubilizing phosphorous and promoting maize growth, aiming to develop a microbial inoculant to be used in Brazilian agriculture. Bacterial strains from the maize rhizosphere were evaluated based on their ability to solubilize phosphate and produce indole acetic acid. Based on these characteristics, 24 strains were selected to be further evaluated under laboratory, greenhouse, and field conditions. Among the selected strains, four (I04, I12, I13, and I17) showed a high potential to increase maize root growth and shoot P content. Strains I13 (Ag87) and I17 (Ag94) were identified by genomic sequencing as Bacillus megaterium and Lysinibacillus sp., respectively. These strains presented superior yield increments relative to the control treatment with 30% P. In addition, combining Ag87 and Ag94 resulted in even higher yield gains, indicating a synergistic effect that could be harnessed in a commercial inoculant for Brazilian agriculture.

ABSTRACT

The population growth trend in recent decades has resulted in continuing efforts to guarantee food security in which leguminous plants, such as the common bean (Phaseolus vulgaris L.), play a particularly important role as they are relatively cheap and have high nutritional value. To meet this demand for food, the main target for genetic improvement programs is to increase productivity, which is a complex quantitative trait influenced by many component traits. This research aims to identify Quantitative Trait Nucleotides (QTNs) associated with productivity and its components using multi-locus genome-wide association studies. Ten morpho-agronomic traits [plant height (PH), first pod insertion height (FPIH), number of nodules (NN), pod length (PL), total number of pods per plant (NPP), number of locules per pod (LP), number of seeds per pod (SP), total seed weight per plant (TSW), 100-seed weight (W100), and grain yield (YLD)] were evaluated in four environments for 178 Mesoamerican common bean domesticated accessions belonging to the Brazilian Diversity Panel. In order to identify stable QTNs, only those identified by multiple methods (mrMLM, FASTmrMLM, pLARmEB, and ISIS EM-BLASSO) or in multiple environments were selected. Among the identified QTNs, 64 were detected at least thrice by different methods or in different environments, and 39 showed significant phenotypic differences between their corresponding alleles. The alleles that positively increased the corresponding traits, except PH (for which lower values are desired), were considered favorable alleles. The most influenced trait by the accumulation of favorable alleles was PH, showing a 51.7% reduction, while NN, TSW, YLD, FPIH, and NPP increased between 18 and 34%. Identifying QTNs in several environments (four environments and overall adjusted mean) and by multiple methods reinforces the reliability of the associations obtained and the importance of conducting these studies in multiple environments. Using these QTNs through molecular techniques for genetic improvement, such as marker-assisted selection or genomic selection, can be a strategy to increase common bean production.

ABSTRACT

KEY MESSAGE: QTNs significantly associated to nine mineral content in grains of common bean were identified. The accumulation of favorable alleles was associated with a gradually increasing nutrient content in the grain. Biofortification is one of the strategies developed to address malnutrition in developing countries, the aim of which is to improve the nutritional content of crops. The common bean (Phaseolus vulgaris L.), a staple food in several African and Latin American countries, has excellent nutritional attributes and is considered a strong candidate for biofortification. The objective of this study was to identify genomic regions associated with nutritional content in common bean grains using 178 Mesoamerican accessions belonging to a Brazilian Diversity Panel (BDP) and 25,011 good-quality single nucleotide polymorphisms. The BDP was phenotyped in three environments for nine nutrients (phosphorus, potassium, calcium, magnesium, copper, manganese, sulfur, zinc, and iron) using four genome-wide association multi-locus methods. To obtain more accurate results, only quantitative trait nucleotides (QTNs) that showed repeatability (i.e., those detected at least twice using different methods or environments) were considered. Forty-eight QTNs detected for the nine minerals showed repeatability and were considered reliable. Pleiotropic QTNs and overlapping genomic regions surrounding the QTNs were identified, demonstrating the possible association between the deposition mechanisms of different nutrients in grains. The accumulation of favorable alleles in the same accession was associated with a gradually increasing nutrient content in the grain. The BDP proved to be a valuable source for association studies. The investigation of different methods and environments showed the reliability of markers associated with minerals. The loci identified in this study will potentially contribute to the improvement of Mesoamerican common beans, particularly carioca and black beans, the main groups consumed in Brazil.

ABSTRACT

Brazil is the world's largest producer of common bean. Knowledge of the genetic diversity and relatedness of accessions adapted to Brazilian conditions is of great importance for the conservation of germplasm and for directing breeding programs aimed at the development of new cultivars. In this context, the objective of this study was to analyze the genetic diversity, population structure, and linkage disequilibrium (LD) of a diversity panel consisting of 219 common bean accessions, most of which belonging to the Mesoamerican gene pool. Genotyping by sequencing (GBS) of these accessions allowed the identification of 49,817 SNPs with minor allele frequency > 0.05. Of these, 17,149 and 12,876 were exclusive to the Mesoamerican and Andean pools, respectively, and 11,805 SNPs could differentiate the two gene pools. Further the separation according to the gene pool, bayesian analysis of the population structure showed a subdivision of the Mesoamerican accessions based on the origin and color of the seed tegument. LD analysis revealed the occurrence of long linkage blocks and low LD decay with physical distance between SNPs (LD half decay in 249 kb, corrected for population structure and relatedness). The GBS technique could effectively characterize the Brazilian common bean germplasms, and the diversity panel used in this study may be of great use in future genome-wide association studies.

ABSTRACT

Bacterial leaf streak (BLS), caused Xanthomonas vasicola pv. vasculorum (Xvv), has become a major concern for maize production, mainly in the United States and South America. Therefore, this study aimed to establish a protocol for Xvv inoculation in young maize plants under controlled conditions and to develop and validate a diagrammatic scale for evaluation of maize hybrids in regard to BLS resistance. The study was carried out in three steps: the establishment of a protocol for inoculation of Xvv in young maize plants under controlled conditions; the development and validation of a diagrammatic scale for BLS severity evaluation; and the screening for BLS resistance of 45 hybrids using the proposed protocol for bacterial inoculation and the diagrammatic scale developed in this study. Besides reproducing a more natural Xvv infection, the bacterial suspension spraying without injury inoculation method induced higher disease incidence and severity, as well as reproducibility of results under the experimental conditions established in this study. The proposed diagrammatic scale allowed evaluating BLS severity with up to 97.49% of the leaf area affected by the disease. Further, the use of the diagrammatic scale resulted in an increase of accuracy from 0.909 up to 0.992. The reaction of 45 maize hybrids to BLS allowed establishing six major groups of susceptibility to the disease. The most resistant maize hybrids to BLS formed a group of 13 hybrids, with disease severity below 5%.

ABSTRACT

Common bean (Phaseolus vulgaris L.) is one of the most important crops worldwide and is considered an essential source of proteins, fibers, and minerals in the daily diet of several countries. Nitrogen (N) is considered the most important nutrient for common bean crop. On the other hand, the reduction of chemical fertilizers is a global challenge, and the development of cultivars with more N use efficiency (NUsE) is considered one of the main strategies to reduce the amount of N fertilizers. Genetic progress of NUsE has been reported in several crops; however, there was still no quantity in common bean. In this study, our goal was to analyze the genetic progress of seed yield (SY) and NUsE-related traits of 40 carioca common bean cultivars release from 1970 to 2017 in eight environments under low (zero) or high N (40 kg ha-1) in top-dressing. Genetic progress, principal component analysis, correlations among traits, and cultivar stability were analyzed using Bayesian approaches. The lowest values of the deviance information criterion (DIC) for the full model tested indicated the presence of the genotype × N × environment interaction for all evaluated traits. Nitrogen utilization efficiency (NUtE) and nitrogen uptake efficiency (NUpE) were the traits that most contributed to discriminate cultivars. The genetic progress of SY under high N (0.53% year-1, 95% HPD = 0.39; 0.65% year-1) was similar to that obtained in low N conditions (0.48% year-1, 95% HPD = 0.31; 0.64% year-1). These results indicate that modern cultivars do not demand more N fertilizers to be more productive. In addition, we observed a high genetic variability for NUsE-related traits, but there was no genetic progress for these variables. SY showed negative correlation with seed protein content (Prot) in both N conditions, and there was no reduction in Prot in modern cultivars. Both modern and old cultivars showed adaptability and stability under contrasting N conditions. Our study contributed to improve our knowledge about the genetic progress of common bean breeding program in Brazil in the last 47 years, and our data will help researchers to face the challenge of increase NUsE and Prot in the next few years.

ABSTRACT

Water deficit is one of the major limitations to soybean production worldwide, yet the genetic basis of drought-responsive mechanisms in crops remains poorly understood. In order to study the gene expression patterns in leaves and roots of soybean, two contrasting genotypes, Embrapa 48 (drought-tolerant) and BR 16 (drought-sensitive), were evaluated under moderate and severe water deficit. Transcription factors from the AP2/EREBP and WRKY families were investigated. Embrapa 48 showed 770 more up-regulated genes than BR 16, in eight categories. In general, leaves presented more differentially expressed genes (DEGs) than roots. Embrapa 48 responded to water deficit faster than BR 16, presenting a greater number of DEGs since the first signs of drought. Embrapa 48 exhibited initial modulation of genes associated with stress, while maintaining the level of the ones related to basic functions. The genes expressed exclusively in the drought-tolerant cultivar, belonging to the category of dehydration responsive genes, and the ones with a contrasting expression pattern between the genotypes are examples of important candidates to confer tolerance to plants. Finally, this study identified genes of the AP2/EREBP and WRKY families related to drought tolerance.

ABSTRACT

MAIN CONCLUSION: Plant growth-promoting bacteria association improved the enzymatic and non-enzymatic antioxidant pathways in Neotropical trees under drought, which led to lower oxidative damage and enhanced drought tolerance in these trees. Water deficit is associated with oxidative stress in plant cells and may, thus, negatively affect the establishment of tree seedlings in reforestation areas. The association with plant growth-promoting bacteria (PGPB) is known to enhance the antioxidant response of crops, but this strategy has not been tested in seedlings of Neotropical trees. We evaluated the effects of inoculation with two PGPB (Azospirillum brasilense and Bacillus sp.) on the antioxidant metabolism of Cecropia pachystachya and Cariniana estrellensis seedlings submitted to drought. We measured the activity of antioxidant enzymes and the content of non-enzymatic antioxidants in leaves, and biometrical parameters of the seedlings. In both tree species, drought decreased the activity of antioxidant enzymes and the content of non-enzymatic antioxidant compounds. For C. pachystachya, the enzymatic and non-enzymatic pathways were mostly influenced by A. brasilense inoculation, which enhanced ascorbate peroxidase (APX) and superoxide dismutase activities and positively affected the level of non-enzymatic antioxidant compounds. In C. estrellensis, A. brasilense inoculation enhanced APX activity. However, A. brasilense and Bacillus sp. inoculation had more influence on the non-enzymatic pathway, as both bacteria induced a greater accumulation of secondary compounds (such as chlorogenic acid, gallic acid, rutin and synapic acid) compared to that in non-inoculated plants under drought. For both species, PGPB improved biometrical parameters related to drought tolerance, as specific leaf area and leaf-area ratio. Our results demonstrate that PGPB induced antioxidant mechanisms in drought-stressed Neotropical trees, increasing drought tolerance. Thus, PGPB inoculation provides a biotechnological alternative to improve the success of reforestation programmes.

ABSTRACT

BACKGROUND: Plant genomes are rich in repetitive sequences, and transposable elements (TEs) are the most accumulated of them. This mobile fraction can be distinguished as Class I (retrotransposons) and Class II (transposons). Retrotransposons that are transposed using an intermediate RNA and that accumulate in a "copy-and-paste" manner were screened in three genomes of peppers (Solanaceae). The present study aimed to understand the genome relationships among Capsicum annuum, C. chinense, and C. baccatum, based on a comparative analysis of the function, diversity and chromosome distribution of TE lineages in the Capsicum karyotypes. Due to the great commercial importance of pepper in natura, as a spice or as an ornamental plant, these genomes have been widely sequenced, and all of the assemblies are available in the SolGenomics group. These sequences were used to compare all repetitive fractions from a cytogenomic point of view. RESULTS: The qualification and quantification of LTR-retrotransposons (LTR-RT) families were contrasted with molecular cytogenetic data, and the results showed a strong genome similarity between C. annuum and C. chinense as compared to C. baccatum. The Gypsy superfamily is more abundant than Copia, especially for Tekay/Del lineage members, including a high representation in C. annuum and C. chinense. On the other hand, C. baccatum accumulates more Athila/Tat sequences. The FISH results showed retrotransposons differentially scattered along chromosomes, except for CRM lineage sequences, which mainly have a proximal accumulation associated with heterochromatin bands. CONCLUSIONS: The results confirm a close genomic relationship between C. annuum and C. chinense in comparison to C. baccatum. Centromeric GC-rich bands may be associated with the accumulation regions of CRM elements, whereas terminal and subterminal AT- and GC-rich bands do not correspond to the accumulation of the retrotransposons in the three Capsicum species tested.

ABSTRACT

The main objective of this study was to evaluate Bacillus velezensis strain CMRP 4490 regarding its ability to inhibit soil-borne plant pathogens and to increase plant growth. The study included evaluation of in vitro antifungal control, sequencing the bacterial genome, mining genes responsible for the synthesis of secondary metabolites, root colonization ability, and greenhouse studies for the assessment of plant growth-promoting ability. The strain was obtained from soil samples in the north of Paraná in Brazil and was classified as a B. velezensis, which is considered a promising biological control agent. In vitro assay showed that B. velezensis CMRP 4490 presented antagonistic activity against Sclerotinia sclerotiorum, Macrophomina phaseolina, Botrytis cinerea, and Rhizoctonia solani with a mycelial growth inhibition of approximately 60%, without any significant difference among them. To well understand this strain and to validate its effect on growth-promoting rhizobacteria, it was decided to explore its genetic content through genome sequencing, in vitro, and greenhouse studies. The genome of CMRP 4490 was estimated at 3,996,396 bp with a GC content of 46.4% and presents 4,042 coding DNA sequences. Biosynthetic gene clusters related to the synthesis of molecules with antifungal activity were found in the genome. Genes linked to the regulation/formation of biofilms, motility, and important properties for rhizospheric colonization were also found in the genome. Application of CMRP 4490 as a coating film on soybean increased from 55.5 to 64% on germination rates when compared to the control; no differences were observed among treatments for the maize germination. The results indicated that B. velezensis CMRP 4490 could be a potential biocontrol agent with plant growth-promoting ability.

ABSTRACT

Few works have reported the relationship among genotype, temperature, rainfall and the chemical compounds of soybean. Therefore, the aim of this study was to investigate the interaction effect between soybean food-type inbred lines sowed in two different dates and the contents of protein, oil and isoflavones. Eight lines with null lipoxygenase seeds classified as food-type soybean were sowed in October 7 (early sowing) and October 29 (late sowing) at 2013/2014 crop year. The oil, protein and isoflavones contents were determined and the data were analyzed by variance analysis (ANOVA), principal component analysis (PCA) and UPGMA hierarchical. The genetic variability, sowing date and interactions between inbred lines and sowing date showed differences for all characteristics, except for oil content that did not showed a significant effect to the interaction. The greater participation of complex interaction was attributed to protein content with 87.82 %. According to PCA and UPGMA results, the food-type soybean lines were separated into three groups and were consistent in both sowing dates. The UEL 131 and UEL 153 lines showed the highest isoflavones content for the two sowing dates, indicating these genotypes as promising for breeding programs.

ABSTRACT

Water deficit is one of the main factors that reduce grain yield. A better understanding of the mechanisms related to this abiotic stress is a key aspect to design and act upon drought tolerance improvement in crop plants. Therefore, the major objective of this study was to investigate four common bean genotypes for drought tolerance and to establish their tolerance mechanisms. The experiment was carried out in a greenhouse, using the completely randomized design in a factorial arrangement (2 × 4), composed by 2 water conditions (well-watered and water deficit) and 4 cultivars, with six replicates per treatment. The four cultivars, two drought-sensitive (IAC Tybatã and BRS Pontal) and two drought-tolerant (IAPAR 81 and BAT 477), were evaluated for some physiological, biochemical and morphoagronomic traits. Drought promoted physiological and metabolic changes in the plants, reflecting on the morphoagronomic traits. Under water deficit, the genotype IAPAR 81 stood out from the others in terms of physiological characters, however, it presented a low efficiency concerning biochemical activities and a significant reduction in the morphoagronomic characters. The cultivar BAT 477 demonstrated to be drought-adapted presenting more efficient biochemical and morphoagronomic adaptions and the genotype BRS Pontal obtained morphoagronomic values similar to BAT 477, thus it may be classified as moderately tolerant to drought.

ABSTRACT

Heterosis has been exploited in Capsicum annuum commercial hybrids; however, the use of heterosis in C. baccatum still remains to be explored, and studies related to the genetics and breeding of this species are scarce. The present study aimed to estimate the combining ability of five parents of C. baccatum var. pendulum , representatives of two distinct types of fruits (namely, lady's finger and cambuci), to calculate heterosis and to evaluate the agronomic potential of the hybrids for yield and fruit quality. The hybrids were produced from a complete diallel without reciprocals. The parents and hybrids were evaluated under field conditions in a randomized block design with three replications, and the following traits were assessed: number of fruits per plant, fruit weight, yield per plant, fruit length, fruit diameter and soluble solids. All traits were significant for general and specific combining ability, indicating that additive and non-additive effects are involved in the genetic control of these traits. The hybrid combinations between the types lady's finger and cambuci provided elongated fruits with smaller diameters and greater weight compared with the parents of the cambuci type. However, these factors did not lead to a significant increase in the yield per plant due to the decreased number of fruits except in hybrid UENF 1616 x UENF 1732. Considering only the parents and hybrids within each type of fruit, the genitor UENF 1624 (lady's finger) and the hybrid UENF 1639 x UENF 1732 (cambuci x cambuci) stood out for achieving a high yield per plant.

The significant increase in maize production in the second season crop in Brazil, mainly in central-south of the country have stimulated breeding programs to select genotypes adapted to the climatic conditions of different sowing times. The objective of this study was to quantify the progenies x sowing times interaction and check its reflexes on the genetic progress with the use of multivariate selection for selection progenies of the maize Composite Isanão VF-1. Sowings were done in the second growing season, in 2004 and in first growing season of the 2004/05 crop year. A total of 71 half-sib progenies were evaluated in a randomized block design with three replications. The traits evaluated were: plant height, ear height, lodged and broken plants, prolificacy and grain yield. There was decomposition of progenies x sowing times interaction and gains from selection index Mulamba and Mock were estimated. There was a predominance of the simple interaction for most traits, except prolificacy, which revealed 86 percent of complex type interaction. For the index Mulamba and Mock, proportional gains most appropriate for the traits set was obtained by the economic weights assigned by attempts. The predicted gains were 1.41, 0.86, -13.03, 9.54 and 16.12 percent for plant height, ear height, lodged and broken plants, prolificacy and grain yield, respectively.

ABSTRACT

The genus Dimorphandra has distinguish relevance considering either medicinal or biodiversity aspects because it includes two species that are economically important flavonoids sources for pharmachemical industry (D. mollis Benth. and D. gardneriana Tul.), and species endemic to Brazil, such as D. jorgei Silva and D. wilsonii Rizz., threatened by extinction. In order to evaluate variability among accessions of D. mollis (fava-d'anta), D. gardneriana and D. wilsonii, it was collected fruit from individual plants from three Brazilian states in a total of 57 accessions, which were analyzed with RAPD markers. It was used 20 seeds per progeny; the DNA was extracted from fully-formed young leaves, which were collected in bulk. The data were analyzed using a binary matrix, in which the score one represented presence of a band and zero, absence. The similarity matrix was developed by using the arithmetic complement of the Jaccard index, later grouped based on the Neighbor Joining algorithm. It was found considerable intra and inter specific variability in Dimorphandra spp., which were separated into four groups. Though genetic variability was found, the collecting trips showed that most of these areas are subject to loss of genetic resources of fava-d'anta due to the following factors: continuous anthropic activity, propensity for natural fires, and loss of natural seed dispersers (large fruit-eating mammals). Therefore, protections of these areas and ex situ conservation are essentials for the maintenance of genetic variability of these species.

ABSTRACT

The economic importance of Spondias umbuzeiro has been increased in international market due to its exotic flavor and the ability to be used in juices, ice creams and other food preparation. For this reason, the knowledge of many aspects of this species is has been investigated by many researchers and also attracted the farmers interest. The aim of this research was to evaluate the development of umbu seedlings and to indicate the best stage of the seedling and also the more appropriate stage of the fork for grafting. Seeds were sown in wood beds with 1.5m wide10.0m long0.15m high, washed sand, and placed together and aligned in rows spaced 5.0cm apart. The following characteristics were evaluated: seedling size, leaves number, stem diameter at 5, 10 and 15cm from the soil, canopy and root fresh weight in the transplant date and also after 60, 120, 180 and 240 days after transplanting. The grafting technique studied consisted in a top bench grafting with seedlings from different origins and development stages. The efficiency of grafting technique was evaluated after 180 days after transplanting by observing the surviving plants. The experiments were carried out in a completely casualized design and the replicates made up of six plants per plot. It was concluded that from six months after transplanting the umbuzeiro seedlings can be grafted by using top cleft grafting. The efficiency of grafting technique reached 80 percent of all genotypes tested. The same results were observed considering plants after two months when the plants were already planting in a definitive place. The survival index decreased gradually when forks from plants older than six years were used.

ABSTRACT

Cherry tomato is becoming an important crop alternative mainly for small farmers that use family hand labor and use organic management to differentiate their product. The introduction of new genotypes in this market should be supported by germplasm characterization and evaluation. This germplasm could be recommended for farmers or introduced in breeding programs. The aim of this research was to test 40 cherry tomatoes accessions from different sources, being 36 accessions from germplasm collection of Department de Fitotecnia of UFRRJ and four cultivars, namely 'Joanna', 'Perinha Água Branca', 'Samambaia' and Super Sweet. The plants were grown in field conditions, under organic management, in a randomized block design with four replications and five plants per plot, from June to December 2004. The following variables were studied: fruit total yield; commercial and non-commercial fruit yield; longitudinal and equatorial fruit diameter; pulp thickness; minimum, medium and maximum fruit mass variation per plant. The data were analyzed considering Mahalanobis distance and the cluster analysis was performed using UPGMA (Unweighted Pair-Group Method Using an Arithmetic Average) and canonic variables. The adjustment between the similarity matrix and dendrograma was performed using the cophenetic correlation. The characters relative importance was determined by Singh´s method. The accessions were clustered in two groups, one of them with only one accession (ENAS 1021) and the second group, with 39 accessions, was subdivided generating seven subgroups. The cophenetic correlation was 0.9 meaning a very good adjustment between the distance matrix and the dendrogram. The first three canonic variables explained 78.58 percent of observed variation, forming seven groups. 'Perinha Agua Branca' was the one that produced lower proportion of non-commercial fruit yield (11.11 percent) while Super Sweet produced 48.05 percent of damaged fruit.
