ABSTRACT

"Iron prawn" is a condition of severe growth retardation that fishers call. The giant river prawn (Macrobrachium rosenbergii) is a commercially important species contains high protein content and functional nutrients. However, no proteomic information is available for this species. We performed the shotgun 2DLC-MS/MS proteomic analysis of the total protein from "iron prawn". Total 19,758 peptides corresponding to 2613 high-confidence proteins were identified. These proteins range in size from 40 to 70 kDa. KEGG analysis revealed that the largest group consisting total 102 KEGG pathway proteins comparing the "iron prawn" with the normal prawn. Additionally, 7, 11, 1, 6, and 5 commercially important enzymes were found in the eyestalk, liver, muscle, ovary, and testis, respectively. The functions of these differently expressed enzymes include immune system action against pathogens, muscle contraction, digestive system metabolism, cell differentiation, migration, and apoptosis in the severe growth retardation of "iron prawn". Our work provides insight into the understanding of the formation mechanism of "iron prawn".

ABSTRACT

The present study was undertaken to investigate the antiparasitic activity of extracellular products of Streptomyces albus. Bioactivity-guided isolation of chloroform extracts affording a compound showing potent activity. The structure of the compound was elucidated as salinomycin (SAL) by EI-MS, 1H NMR and 13C NMR. In vitro test showed that SAL has potent anti-parasitic efficacy against theronts of Ichthyophthirius multifiliis with 10 min, 1, 2, 3 and 4 h (effective concentration) EC50 (95% confidence intervals) of 2.12 (2.22-2.02), 1.93 (1.98-1.88), 1.42 (1.47-1.37), 1.35 (1.41-1.31) and 1.11 (1.21-1.01) mg L-1. In vitro antiparasitic assays revealed that SAL could be 100% effective against I. multifiliis encysted tomonts at a concentration of 8.0 mg L-1. In vivo test demonstrated that the number of I. multifiliis trophonts on Erythroculter ilishaeformis treated with SAL was markedly lower than that of control group at 10 days after exposed to theronts (P < 0.05). In the control group, 80% mortality was observed owing to heavy I. multifiliis infection at 10 days. On the other hand, only 30.0% mortality was recorded in the group treated with 8.0 mg L-1 SAL. The median lethal dose (LD50) of SAL for E. ilishaeformis was 32.9 mg L-1.

ABSTRACT

AIM:To observe the tear film and corneal endothelial cell density in cataract patients with high myopia.METHODS:From January 2016 to December 2016,38 cases (38 eyes) with high myopia and cataract were selected as study group,24 males (24 eyes) and 14 females (14 eyes),average 65.2±2.37(60-72) years old.Age-related cataract patients without high myopia were as control group,22 males (22 eyes) and 16 females (16 eyes),average 64.4±2.43 (61-70) years old.The tear film and corneal endothelial cell density of the two groups were observed at 3,7,14d and 1mo after operations.RESULTS:Between the two groups of preoperative S I t,BUT,FL,subjective rating,corneal endothelial cell density comparison,there were no statistically significant difference (P>0.05).In the two groups at 3,7,14d and 1mo after operations,BUT,FL,corneal endothelial cell density,subjective score comparison,the difference had statistical significance (P<0.01).Two groups after 3,7,14d comparative differences of S Ⅰ t were statistically significant (P < 0.01),not statistical significant at postoperative 1mo (P>0.05).At postoperative 3,7,14d,1mo,FL,subjective rating,corneal endothelial cell density of the two groups were compared with preoperative,the difference was statistically significant (P<0.01).In the two groups at 3,7,14d after operation,S I t compared with the same group preoperative difference was statistically significant (P < 0.01),no statistical significance when postoperative 1mo compared with preoperative (P>0.05).BUT of high myopia patients with age-related cataract surgery,at 3,7,14d and 1mo after operations decreased than preoperative,the difference was statistically significant (P< 0.01).Age-related cataract patients without high myopia at 3,7,14d after operation decreased than preoperative,the difference was statistically significant (P<0.01),there was no statistically significant difference between preoperative and postoperative 1 mo (P>0.05).CONCLUSION:Phacoemulsification cataract surgery in the treatment of high myopia cataract patients is safe and reliable,and less influence on tear film and corneal endothelial cell density.

ABSTRACT

OBJECTIVE: To investigate the cognitive impairment in patients with age-related macular degeneration (AMD). METHODS: Relevant articles were identified through a search of the following electronic databases through October 2015, without language restriction: 1) PubMed; 2) the Cochrane Library; 3) EMBASE; 4) ScienceDirect. Meta-analysis was conducted using STATA 12.0 software. Standardized mean differences with corresponding 95% confidence intervals were calculated. All of the included studies met the following four criteria: 1) the study design was a case-control or randomized controlled trial (RCT) study; 2) the study investigated cognitive function in the patient with AMD; 3) the diagnoses of AMD must be provided; 4) there were sufficient scores data to extract for evaluating cognitive function between cases and controls. The Newcastle-Ottawa Scale criteria were used to assess the methodological quality of the studies. RESULTS: Of the initial 278 literatures, only six case-control and one RCT studies met all of the inclusion criteria. A total of 794 AMD patients and 1,227 controls were included in this study. Five studies were performed with mini-mental state examination (MMSE), two studies with animal fluency, two studies with trail making test (TMT)-A and -B, one study with Mini-Cog. Results of the meta-analysis revealed lower cognitive function test scores in patients with AMD, especially with MMSE and Mini-Cog test (P≤0.001 for all). The results also showed that differences in the TMT-A (except AMD [total] vs controls) and TMT-B test had no statistical significance (P>0.01). The Newcastle-Ottawa Scale score was ≥5 for all of the included studies. Based on the sensitivity analysis, no single study influenced the overall pooled estimates. CONCLUSION: This meta-analysis suggests lower cognitive function test scores in patients with AMD, especially with MMSE and Mini-Cog test. The other cognitive impairment screening tests, such as animal fluency test and TMT, need more studies to assess.

ABSTRACT

A monthly investigation was conducted on the macrozoobenthos at 7 stations of Fenshuijiang Reservoir in Zhejiang Province of East China from November 2008 to October 2009. A total of 37 taxa were collected, most of which belonged to Oligocheata and Chironomidae. Limnodrilus hoffmeisteri was dominant in spring, summer and autumn, and Chironomus gr. plumosus was prevailing in winter. The collector-gatherer was the predominant group in species number, density and biomass. The mean annual density and biomass of the community were (488.0 +/- 48.8) ind X m(-2) and (1.86 +/- 0.49) g X m(-2), respectively. There were no significant differences in the macrozoobenthic density among the stations and in the macrozoobenthic biomass among the stations and among the seasons, but the macrozoobenthic density had a significant seasonal variation, with the sequence of spring > summer > winter > autumn. Water temperature and water depth were the main factors affecting the spatial distribution of the macrozoobenthos. Shannon index and Goodnight-Whitley index were not suitable for the assessment, whereas the Wright index, Carlander index, Pielou index and trophic level index showed that this reservoir was slightly polluted.

ABSTRACT

Based on the 2008-2009 survey data of fishery resources and eco-environment of Fenshuijiang Reservoir, a mass balance model for the Reservoir ecosystem was constructed by Ecopath with Ecosim software. The model was composed of 14 functional groups, including silver carp, bighead carp, Hemibarbus maculates, Cutler alburnus, Microlepis and other fishes, Oligochaeta, aquatic insect, zooplankton, phytoplankton, and organic detritus, etc. , being able to better simulate Fenshuijiang Reservoir ecosystem. In this ecosystem, there were five trophic levels (TLs), and the nutrient flow mainly occurred in the first three TLs. Grazing and detritus food chains were the main energy flows in the ecosystem, but the food web was simpler and susceptible to be disturbed by outer environment. The transfer efficiency at lower TLs was relatively low, indicating that the ecosystem had a lower capability in energy utilization, and the excessive stock of nutrients in the ecosystem could lead to eutrophication. The lower connectance index, system omnivory index, Finn' s cycled index, and Finn's mean path length demonstrated that the ecosystem was unstable, while the high ecosystem property indices such as Pp/R and Pp/B showed that the ecosystem was immature and highly productive. It was suggested that Fenshuijiang Reservoir was still a developing new reservoir ecosystem, with a very short history and comparatively high primary productivity.

ABSTRACT

Ikaros is an important transcription factor involved in the development and differentiation of hematopoietic cells. In this work, we found that chemotherapeutic drugs or ultraviolet radiation (UV) treatment could reduce the expression of full-length Ikaros (IK1) protein in less than 3h in leukemic NB4, Kasumi-1 and Jurkat cells, prior to the activation of caspase-3. Etoposide treatment could not alter the mRNA level of IK1 but it could shorten the half-life of IK1. Co-treatment with the proteasome inhibitor MG132 or epoxomicin but not calpain inhibitor calpeptin inhibited etoposide-induced Ikaros downregulation. Overexpression of IK1 could accelerate etoposide-induced apoptosis in NB4 cells, as evidenced by the increase of Annexin V positive cells and the more early activation of caspase 3. To our knowledge, this is the first report to show that upon chemotherapy drugs or UV treatment, IK1 could be degraded via the proteasome system in the early phase of apoptosis induction. These data might shed new insight on the role of IK1 in apoptosis and the post-translational regulation of IK1.

ABSTRACT

RIG-G is a retinoic acid- or interferon-induced gene with potential anti-proliferation function. However, the mechanism underlying ATRA-induced RIG-G induction is not completely understood. Here, we demonstrate that ATRA up-regulates the expression of PU.1, which in turn directly binds to the promoter and increases the expression of RIG-G gene. Luciferase reporter assay and electrophoretic mobility shift assay reveal that PU.1 preferentially binds to one of the two putative binding sites on the RIG-G promoter. Moreover, silencing of PU.1 by shRNA markedly inhibited ATRA- but not IFNα-induced expression of RIG-G. These data provide new insight into the mechanism of ATRA-induced RIG-G expression.

ABSTRACT

All-trans retinoic acid (ATRA), a natural ligand for the retinoic acid receptors (RARs), induces clinical remission in most acute promyelocytic leukemia (APL) patients through the induction of differentiation and/or eradication of leukemia-initiating cells. Here, we identify a novel natural ent-kaurene diterpenoid derived from Isodon pharicus leaves, called pharicin B, that can rapidly stabilize RAR-α protein in various acute myeloid leukemic (AML) cell lines and primary leukemic cells from AML patients, even in the presence of ATRA, which is known to induce the loss of RAR-α protein. Pharicin B also enhances ATRA-dependent the transcriptional activity of RAR-α protein in the promyelocytic leukemia-RARα-positive APL cell line NB4 cells. We also showed that pharicin B presents a synergistic or additive differentiation-enhancing effect when used in combination with ATRA in several AML cell lines and, especially, some primary leukemic cells from APL patients. In addition, pharicin B can overcome retinoid resistance in 2 of 3 NB4-derived ATRA-resistant subclones. These findings provide a good example for chemical biology-based investigations of pathophysiological and therapeutic significances of RAR-α and PML-RAR-α proteins. The effectiveness of the ATRA/pharicin B combination warrants further investigation on their use as a therapeutic strategy for AML patients.

ABSTRACT

Using the cDNA of rye cytoplasmic ribosomal protein ScRPS7 as a query probe, a highly homologous rice genomic contig was obtained from Huada rice genome database. The full-length cDNA sequence of rice cytoplasmic ribosomal protein S7 was assembled by informatics based on the contig. Furthermore, with the two primers designed according to this assembled cDNA, the full-length cDNA of rice ribosomal protein was cloned by RT-PCR and named as OsRPS7. The cDNA was 919bp in length and contained a complete Open Reading Frame (ORF) of 576bp, encoding a protein of 192 amino acid residues. The deduced amino acids of OsRPS7 showed 88%,72% and 72% identity with those from Secale cereale, Arabidopsis thaliana and Brassica oleracea, respectively. The genome structure of OsRPS7 was analyzed, and its function was predicted in this paper.
