ABSTRACT

The expansion of the area of harmfulness of the wheat yellow rust pathogen (Puccinia striiformis) (Pst) has be observed all over the world in the 2000s. This is due to the emergence of new highly aggressive invasive groups of races PstS1 and PstS2, adopted to the high temperatures, and also as a result of virulence mutations of regional pathogen populations. SCAR-markers were developed for identification of invasive races, and pathogen collections from many countries were studied. In these studies in first in Russia, the analysis of regional populations of P. striiformis for the presence of invasive races PstS1 and PstS2 was carried out. Single pustule isolates were obtained from urediosamples collected from common and durum wheat, triticale and wild grasses in seven regions of the Russian Federation (North Caucasian, Northwestern, Central Black Earth, Lower Volga, Middle Volga, Volga-Vyatka, West Siberian) in 2019-2020. In total 82 isolates were studied. Using SCAR markers, three genotypes were identified in the studied collection of P. striiformis, and one of which belongs to the invasive group PstS2. The other two genotypes had a different origin (other). Isolates of PstS2 group were received from pathogen population samples collected in the Russian Northwest in 2020. Virulence analysis revealed two phenotypes among them: PstS2_R1 (3 isolates) and PstS2_R2 (1 isolate). According to information from the Global Rust Reference Center ( http://www.wheatrust.org/ ), the main characteristic of isolates from invasive PstS2 group is virulence to wheat lines with resistance genes Yr2, Yr6, Yr7, Yr8, Yr9, and Yr25. The Russian R1 phenotype PstS2 was also characterized by virulence to these genes, as well as to Yr1, Yr32, and YrSp. The PstS2_R2 phenotype differed from PstS2_R1 for avirulence to Yr25 and virulence to Yr3 and Yr4. The main difference of Russian PstS2 isolates with detected in other countries is virulence to wheat lines with genes Yr4, Yr32, and YrSp. The first detection of invasive races in the Northwest of Russia indicates the relevance of annual monitoring of regional populations of P. striiformis.

ABSTRACT

The 7DL-7Ae#1L·7Ae#1S translocation with the Lr29 gene attracts the attention of bread wheat breeders by its effectiveness against Puccinia triticina. However, its impact on useful agronomic traits has been little studied. In this report, the prebreeding value of 7DL-7Ae#1L·7Ae#1S was studied in analogue lines (ALs) of spring bread wheat cultivars Saratovskaya 68 and Saratovskaya 70 during 2019-2021. The presence of the Lr29 gene was conf irmed by using molecular marker Lr29F24. The ALs with the Lr29 gene were highly resistant to P. triticina against a natural epiphytotics background and in laboratory conditions. 7DL-7Ae#1L·7Ae#1S in Saratovskaya 68 ALs reduced grain productivity in all years of research. On average, the decrease was 35 and 42 %, or in absolute f igures 1163 and 1039 against 1802 kg/ha in the cultivar-recipient. In Saratovskaya 70 ALs, there was a decrease in grain yield in 2019 and 2020, and there were no differences in 2021. On average, the decrease was 18 and 32 %, or in absolute f igures 1101 and 912 against 1342 kg/ha in the cultivar-recipient. The analogues of both cultivars showed a signif icant decrease in the weight of 1000 grains, which ranged from 14 to 20 % for Saratovskaya 68 and 17-18 % for Saratovskaya 70. An increase in the period of germination-earing was noted only in Saratovskaya 68 lines, which averaged 1.3 days. ALs of Saratovskaya 70 had no differences in this trait. 7DL-7Ae#1L·7Ae#1S did not affect plant height and lodging resistance in all ALs. Studies of the bread-making quality in lines with 7DL-7Ae#1L·7Ae#1S revealed a signif icant increase in grain protein and gluten content. As for the effect on the alveograph indicators, there were differences between ALs of both cultivars. While Saratovskaya 68 ALs had a decrease in elasticity and in the ratio of dough tenacity to the extensibility, Saratovskaya 70 lines had an increase in these indicators. All lines increased the f lour strength and the loaves volume, but while Saratovskaya 68 ALs had an increased porosity rating, Saratovskaya 70 ALs had the same rating as the recipient.

ABSTRACT

Aim    To study changes in clinical and echocardiographic parameters in patients after documented COVID-19 pneumonia at 3 months and one year following discharge from the hospital. Material and methods    The study included 116 patients who have had documented COVID-19 pneumonia. Patients underwent a comprehensive clinical evaluation at 3 months ± 2 weeks (visit 1) and at one year ± 3 weeks after discharge from the hospital (visit 2). Mean age of the patients was 49.0±14.4 years (from 19 to 84 years); 49.6 % were women. Parameters of global and segmentary longitudinal left ventricular (LV) myocardial strain were studied with the optimal quality of visualization during visit 1 in 99 patients and during visit 2 in 80 patients.Results    During the follow-up period, the incidence rate of cardiovascular diseases (CVD) increased primarily due to development of arterial hypertension (AH) (58.6 vs. 64.7 %, р=0.039) and chronic heart failure (CHF) (35.3% vs. 40.5 %, р=0.031). Echocardiography (EchoCG) showed decreases in values of end-diastolic dimension and volume, LV end-systolic and stroke volumes (25.1±2.6 vs. 24.5±2.2 mm /m2, p<0.001; 49.3±11.3 vs. 46.9±9.9 ml /m2, p=0.008; 16.0±5.6 vs. 14.4±4.1 ml /m2, p=0.001; 36.7±12.8 vs. 30.8±8.1 ml /m2, p<0.001, respectively). LV external short-axis area (37.1 [36.6-42.0] vs. 38.7 [35.2-43.1] cm2, р=0.001) and LV myocardial mass index calculated with the area-length formula (70.0 [60.8-84.0] vs. 75.4 [68.2-84.9] g /m², р=0.024) increased. LV early diastolic filling velocity (76.7±17.9 vs. 72.3±16.0 cm /sec, р=0.001) and lateral and septal early diastolic mitral annular velocities decreased (12,10±3,9 vs. 11.5±4.1 cm /sec, р=0.004 and 9.9±3.3 vs. 8.6±3.0 cm /sec, р<0.001, respectively). The following parameters of LV global longitudinal (-20.3±2.2 vs. -19.4±2.7 %, р=0.001) and segmental strain were impaired: apical segments (anterior, from -22.3±5.0 to -20.8±5.2 %, р=0.006; inferior, from -24.6±4.9 to -22.7±4.6, р=0.003; lateral, from -22.7±4.5 to -20.4±4.8 %, р<0.001; septal, from -25.3±4.2 to -23.1±4.4 %, р<0.001; apical, from -23.7±4.1 to -21.8±4.1 %, р<0.001), mid-cavity (anteroseptal, from -21.1±3.3 to -20.4±4.1 %, р=0.039; inferior, from -21.0±2.7 to -20.0±2.9 %, р=0.039; lateral, from -18.4±3.7 to -17.6±4.4 %, р=0.021). RV basal and mid-cavity sphericity indexes increased (0.44±0.07 vs. 0.49±0.07 and 0.37±0.07 vs. 0.41±0.07, respectively, р<0.001 for both). A tendency for increased calculated pulmonary arterial systolic pressure (22.5±7.1 and 23.3±6.3 mm Hg, р=0.076) was observed. Right ventricular outflow tract velocity integral decreased (18.1±4.0 vs. 16.4±3.7 cm, р<0.001).Conclusion    Patients after COVID-19 pneumonia one year after discharge from the hospital, compared to the follow-up data 3 months after the discharge, had an increased incidence of CVD, primarily due to the development of AH and CHF. EchoCG revealed changes in ventricular geometry associated with impairment of LV diastolic and systolic function evident as decreases in LV global longitudinal strain and LV myocardial apical and partially mid-cavity strain.

ABSTRACT

Common winter wheat is the main grain crop cultivated in the North Caucasus. Rust disease damage is one of the factors limiting wheat productivity. There are three species of rust in the region: leaf (Puccinia triticina), stem (P. graminis) and stripe rust (P. striiformis), and their signif icance varies from year to year. The most common is leaf rust, but in the last decade the frequency of its epiphytotic development has signif icantly decreased. At the same time, an increase in the harmfulness of stripe rust (P. striiformis) is noted. Stem rust in the region is mainly absent or observed at the end of the wheat growing season to a weak degree. Only in some years with favorable weather conditions its mass development is noted on susceptible cultivars. It is believed that the sources of infection with rust species in the North Caucasus are infested soft wheat crops, wild-growing cereals and exodemic infection carried by air currents from adjacent territories. In the North Caucasus, forage and wild grasses are affected by Puccinia species almost every year. Depending on weather conditions, the symptom expression is noted from late September to December and then from late February to May-June. Potentially, an autumn infection on grasses can serve as a source for infection of winter soft wheat cultivars sown in October. The purpose of these studies is to characterize the virulence of P. triticina, P. graminis, P. striiformis on wild cereals and to assess the specialization of causative agents to winter wheat in the North Caucasus. Infectious material represented by leaves with urediniopustules of leaf, stem and stripe rusts was collected from wild cereals (Poa spp., Bromus spp.) in the Krasnodar Territory in October-November 2019. Uredinium material from P. triticina, P. striiformis, and P. graminis was propagated and cloned. Monopustular Puccinia spp. isolates were used for virulence genetics analysis. In experiments to study the specialization of rust species from wild-growing cereals on common wheat, 12 winter cultivars were used (Grom, Tanya, Yuka, Tabor, Bezostaya 100, Yubileynaya 100, Vekha, Vassa, Alekseich, Stan, Gurt, Bagrat). These cultivars are widely cultivated in the North Caucasus region and are characterized by varying degrees of resistance to rust. Additionally, wheat material was inoculated with Krasnodar populations of P. triticina, P. striiformis, P. graminis from common wheat. In the virulence analysis of P. triticina on cereal grasses, four phenotypes (races) were identif ied: MCTKH (30 %), TCTTR (30 %), TNTTR (25 %), MHTKH (15 %), and f ive were identif ied in P. graminis (RKMTF (60 %), TKTTF, RKLTF, QKLTF, LHLPF (10 % each). Among P. striiformis isolates, three phenotypes were identif ied using the International and European sets of differentiating cultivars - 111E231 (88 %), 111E247 (6 %) and 78E199 (6 %). Using isogenic Avocet lines, 3 races were also identif ied, which differed among themselves in virulence to the Yr1, Yr11, Yr18 genes (with the prevalence of virulent ones (94 %)). Composite urediniums' samples (a mixture of all identif ied races) of grass rust of each species were used to inoculate winter wheat cultivars. The most common winter wheat cultivars (75 %) were characterized by a resistant response when infected with P. graminis populations from common wheat and cereal grasses. All these cultivars were developed using donors of the rye translocation 1BL.1RS, in which the Lr26, Sr31, and Yr9 genes are localized. The number of winter wheat cultivars resistant to leaf rust in the seedling phase was lower (58 %). At the same time, all the studied cultivars in the seedling phase were susceptible to P. striiformis to varying degrees. The virulence analysis of the leaf, stem and stripe rust populations did not reveal signif icant differences in the virulence of the pathogens between wild-growing cereals and soft wheat. Urediniomaterial of all studied rust species successfully infested soft wheat cultivars. The results obtained indicate that grasses are rust infection reservoirs for common wheat crops in the North Caucasus.

ABSTRACT

Aim      To study long-term results and to identify predictors of death in patients with ST-segment elevation acute myocardial infarction (STEMI) who underwent endovascular revascularization.Materials and methods This study included 283 patients registered in the hospital registry of percutaneous coronary interventions (PCI) for STEMI from 2006 through 2009. Analysis of 10-year results included all-cause and cardiovascular death rate, incidence of recurrent myocardial infarction (MI), repeated revascularization, stroke, stent restenosis and thrombosis. Also, a composite endpoint МАССЕ (Major Adverse Cardiovascular and Cerebrovascular Events) was evaluated, which included death, recurrent MI, repeated PCI, stent restenosis and thrombosis, coronary bypass, and stroke.Results Information about the health condition was provided by 204 (72.1 %) patients. Mean follow-up period was 120.1±9.5 months. All-cause mortality was 25.5 % with cardiovascular death determined in 19.1 % of cases. Recurrent MI developed in 21.6 % of patients; in 1.5 % of cases, recurrent MI resulted from thrombosis of previously implanted stents. Repeated PCI was performed for 31.9 % of patients; in 13.7 % of cases, the PCI was performed for stent restenosis. Coronary bypass was performed for 5.4 % of patients. Incidence of stroke was 10.3 %. Major cardiovascular and cerebrovascular complications (МАССЕ) during the follow-up period were determined in 60.3 % patients. According to the Cox proportional hazards regression model, age ≥65 years (odds ratio (OR), 3.75 at 95 % confidence interval (CI) from 1.75 to 8.03; р=0.001) and incomplete coronary revascularization (OR, 3.09 at 95 % CI from 1.52 to 6.30; р=0.002) were independent predictors of death based on data of the 10-year observation.Conclusion      Therefore, at 10 years following endovascular revascularization, STEMI patients showed a moderate death rate with a high incidence of major cardiovascular and cerebrovascular complications. The leading causes for fatal outcomes were recurrent cardiovascular complications. The major predictors of death for the coming 10-year period included age ≥65 years and incomplete myocardial revascularization.

ABSTRACT

Classical virulence analysis is based on discovering virulence phenotypes of isolates with regard to a composition of resistance genes in a differential set of host genotypes. With such a vision, virulence phenotypes are usually treated in a genetic manner as one of two possible alleles, either virulence or avirulence in a binary locus. Therefore, population genetics metrics and methods have become prevailing tools for analyzing virulence data at multiple loci. However, a basis for resolving binary virulence phenotypes is infection type (IT) data of host-pathogen interaction that express functional traits of each specific isolate in a given situation (particular host, environmental conditions, cultivation practice, and so on). IT is determined by symptoms and signs observed (e.g., lesion type, lesion size, coverage of leaf or leaf segments by mycelium, spore production and so on), and assessed by IT scores at a generally accepted scale for each plant-pathogen system. Thus, multiple IT profiles of isolates are obtained and can be subjected to analysis of functional variation within and among operational units of a pathogen. Such an approach may allow better utilization of the information available in the raw data, and reveal a functional (e.g., environmental) component of pathogen variation in addition to the genetic one. New methods for measuring functional variation of plant-pathogen interaction with IT data were developed. The methods need an appropriate assessment scale and expert estimations of dissimilarity between IT scores for each plant-pathogen system (an example is presented). Analyses of a few data sets at different hierarchical levels demonstrated discrepancies in results obtained with IT phenotypes versus binary virulence phenotypes. The ability to measure functional IT-based variation offers promise as an effective tool in the study of epidemics caused by plant pathogens.
