ABSTRACT

The Hessian fly, Mayetiola destructor (Say) (Diptera: Cecidomyiidae), is a destructive wheat pest worldwide and an important alien species in China. Based on 258 distribution records and nine environmental factors of the Hessian fly, we predicted the potential distribution area in China under three current and future (2050s and 2070s) climate change scenarios (RCP2.6, RCP4.5, and RCP8.5) via the optimized MaxEnt model. Under the current climate conditions, the suitable distribution areas of the Hessian fly in China were 25-48° N, 81-123° E, and the total highly suitable distribution area is approximately 9.63 × 105 km2, accounting for 9.99% of the total national area. The highly suitable areas are mainly located in northern Xinjiang and central and eastern China. With the rising global temperatures, except for the high-suitable areas under the RCP8.5 scenario, most potential geographic distribution areas would expand in the future. The minimum temperature in February (tmin-2), precipitation in March (prec-3), maximum temperature in November (tmax-11), and precipitation seasonality (bio-15) are important factors that affect the potential geographic distribution of the Hessian fly. This study provides an important reference and empirical basis for management of the Hessian fly in the future.

ABSTRACT

KEY MESSAGE: Two ß-1,3-glucanase genes from sugarcane were cloned and characterized. They were all located in apoplast and involves in different expression patterns in biotic and abiotic stress. Smut caused by Sporisorium scitamineum is a serious disease in the sugarcane industry. ß-1,3-Glucanase, a typical pathogenesis-related protein, has been shown to express during plant-pathogen interaction and involves in sugarcane defense response. In this study, ß-1,3-glucanase enzyme activity in the resistant variety increased faster and lasted longer than that of the susceptible one when inoculated with S. scitamineum, along with a positive correlation between the activity of the ß-1,3-glucanase and smut resistance. Furthermore, two ß-1,3-glucanase genes from S. scitamineum infected sugarcane, ScGluA1 (GenBank Accession No. KC848050) and ScGluD1 (GenBank Accession No. KC848051) were cloned and characterized. Phylogenetic analysis suggested that ScGluA1 and ScGluD1 clustered within subfamily A and subfamily D, respectively. Subcellular localization analysis demonstrated that both gene products were targeted to apoplast. Escherichia coli Rosetta (DE3) cells expressing ScGluA1 and ScGluD1 showed varying degrees of tolerance to NaCl, CdCl2, PEG, CuCl2 and ZnSO4. Q-PCR analysis showed up-regulation of ScGluA1 and slight down-regulation of ScGluD1 in response to S. scitamineum infection. It suggested that ScGluA1 may be involved in the defense reaction of the sugarcane to the smut, while it is likely that ScGluD1 was inhibited. The gene expression patterns of ScGluA1 and ScGluD1, in response to abiotic stresses, were similar to sugarcane response against smut infection. Together, ß-1,3-glucanase may function in sugarcane defense mechanism for S. scitamineum. The positive responses of ScGluA1 and the negative responses of ScGluD1 to biotic and abiotic stresses indicate they play different roles in interaction between sugarcane and biotic or abiotic stresses.

ABSTRACT

Qiemo was an ancient country on the south branch of the Silk Road. The Zagunluke tomb site is located at the Qiemo County of the Xinjiang Uygur Autonomous Region. Glass beads and only colourless glass cup were excavated from the 3rd cultural layer of the tomb site M133 and M49, dated between the 1st AD-6th AD. LA-ICP-AES was applied to analyse chemical composition of these glass finds with the corning glass as reference. According to the result, characteristics of chemical composition are very similar to typical soda-lime glass, which indicates the glasses were imported productions from the west. These soda-lime glasses were divided into two groups in terms of flux source: natron glass and plant ash glass. This analytical research indicates the history of glass trade and communication between the East and the West on the Silk Road.

ABSTRACT

OBJECTIVE: To study whether the porcine alpha1, 3 galactosyltransferase gene siRNA targeted heterozygous hepatocyte negatively expresses GT mRNA and resists to the cytotoxicity of nature antibody in human serum. METHODS: The porcine alpha1, 3 galactosyltransferase gene siRNA targeted vector (pPNTloxPGTsiRNA) were construct with pPNTloxPGT and pMXSV/U6 vector. Positive-negative selection was used to produce a heterozygous pPNTloxPGTsiRNA knockout (+/-) clone. The GT mRNA expressions were detected with northern blot. Complement-mediated NAb cytotoxicity after incubation of hepatocytes with NAbs and complement was determined using 3- (4, 5-dimethylthiazol-2-yl)-5-(3-carboxymethyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium(MTS, tetrazolium salt) colorimetric assay. RESULTS: The pPNTloxPGTsiRNA targeted porcine hepatocyte (+/-) negative express GT mRNA. Only 14% to 18% cytotoxicity can be detected at the highest serum concentration. The pPNTloxPGT targeted porcine hepatocyte (+/-) express GT mRNA just as the wild type porcine cells and the cytotoxicity are 77% to 83%. CONCLUSION: The porcine a1, 3 galactosyltransferase gene siRNA targeted heterozygous hepatocyte (+/-) negative express GT and resisted to nature antibody in human serum.

ABSTRACT

The capillary model was used to analyze the hydraulic conditions in the deep bed filtration process. The physicochemical interaction forces between the filter media and suspended particles and their influence on deep bed filtration process were also studied theoretically. Through the comparison of the hydraulic and physicochemical forces, the key influencing factors on the filtration process were proposed and investigated. Pilot study of the microflocculation deep bed filtration was carried out in the No. 9 Potable Water Treatment Plant of Beijing, and the experimental results of hydraulic head loss, particle distribution and entrapment were presented. The theoretical prediction was reasonably consistent with the experimental results under different conditions, which indicated that the regulation and control of micro-flocculation and deep bed filtration could be realized by the evaluation of the physicochemical interactions. Further theoretical and experimental research should be carried out to investigate the interaction mechanism and its application in the deep bed filtration and other cases.
