ABSTRACT

High temperature during grain filling considerably reduces yield and quality in rice, but its molecular mechanisms are not fully understood. We investigated the functions of a seed preferentially expressed Aux/IAA gene, OsIAA29, under high temperature-stress in grain filling using CRISPR/Cas9, RNAi, and overexpression. We observed that the osiaa29 had a higher percentage of shrunken and chalkiness seed, as well as lower 1000-grain weight than ZH11 under high temperature. Meanwhile, the expression of OsIAA29 was induced and the IAA content was remarkably reduced in the ZH11 seeds under high temperature. In addition, OsIAA29 may enhance the transcriptional activation activity of OsARF17 through competition with OsIAA21 binding to OsARF17. Finally, chromatin immunoprecipitation quantitative real-time PCR (ChIP-qPCR) results proved that OsARF17 regulated expression of several starch and protein synthesis related genes (like OsPDIL1-1, OsSS1, OsNAC20, OsSBE1, and OsC2H2). Therefore, OsIAA29 regulates seed development in high temperature through competition with OsIAA21 in the binding to OsARF17, mediating auxin signaling pathway in rice. This study provides a theoretical basis and gene resources for auxin signaling and effective molecular design breeding.

ABSTRACT

Lodging, a commonly occurring rice crop disaster, seriously reduces rice quality and production. Monitoring rice lodging after a typhoon event is essential for evaluating yield loss and formulating suitable remedial policies. The availability of Sentinel-1 and Sentinel-2 open-access remote sensing data provides large-scale information with a short revisit time to be freely accessed. Data from these sources have been previously shown to identify lodged crops. In this study, therefore, Sentinel-1 and Sentinel-2 data after a typhoon event were combined to enable monitoring of lodging rice to be quickly undertaken. In this context, the sensitivity of synthetic aperture radar (SAR) features (SF) and spectral indices (SI) extracted from Sentinel-1 and Sentinel-2 to lodged rice were analyzed, and a model was constructed for selecting optimal sensitive parameters for lodging rice (OSPL). OSPL has high sensitivity to lodged rice and strong ability to distinguish lodged rice from healthy rice. After screening, Band 11(SWIR-1) and Band 12(SWIR-2) were identified as optimal spectral indices (OSI), and VV, VV + VH and Shannon Entropy were optimal SAR features (OSF). Three classification results of lodging rice were acquired using the Random Forest classification (RFC) method based on OSI, OSF and integrated OSI-OSF stack images, respectively. Results indicate that an overall level of accuracy of 91.29% was achieved with the combination of SAR and optical optimal parameters. The result was 2.91% and 6.05% better than solely using optical or SAR processes, respectively.

ABSTRACT

BACKGROUND: Existing studies of PLK1 in cervical cancer had several flaws. The methods adopted by those studies of detecting PLK1 expression in cervical cancer were single and there lacks comprehensive evaluation of the clinico-pathological significance of PLK1 in cervical cancer. METHODS: A total of 303 cervical tissue samples were collected for in-house tissue microarrays. Immunohistochemistry was performed for evaluating PLK1 expression between cervical cancer (including cervical squamous cell carcinoma (CESC) and cervical adenocarcinoma) and non-cancer samples. The Expression Atlas database was searched for querying PLK1 expression in different cervical cancer cell lines and different tissues in the context of pan-cancer. Standard mean difference (SMD) was calculated and the summarized receiver's operating characteristics (SROC) curves were plotted for integrated tissue microarrays, exterior high-throughput microarrays and RNA sequencing data as further verification. The effect of PLK1 expression on the overall survival, disease-free survival and event-free survival of cervical cancer patients was analyzed through Kaplan Meier survival curves for cervical cancer patients from RNA-seq and GSE44001 datasets. The gene mutation and alteration status of PLK1 in cervical cancer was inspected in COSMIC and cBioPortal databases. Functional enrichment analysis was performed for genes correlated with PLK1 from aggregated RNA-seq and microarrays. RESULTS: A total of 963 cervical cancer samples and 178 non-cancer samples were collected from in-house tissue microarrays and exterior microarrays and RNA-seq datasets. The combined expression analysis supported overexpression of PLK1 in CESC, cervical adenocarcinoma and all types of cervical cancer (SMD = 1.59, 95%CI [0.56-2.63]; SMD = 2.99, 95%CI [0.75-5.24]; SMD = 1.57, 95% CI [0.85-2.29]) and the significant power of PLK1 expression in distinguishing CESC or all types of cervical cancer samples from non-cancer samples (AUC = 0.94, AUC = 0.92). Kaplan-Meier survival curves showed that the event-free survival rate of cervical cancer patients with higher expression of PLK1 was shorter than that of patients with lower PLK1 (HR = 2.020, P = 0.0197). Genetic alteration of PLK1 including missense mutation and mRNA low occurred in 6% of cervical cancer samples profiled in mRNA expression. Genes positively or negatively correlated with PLK1 were mainly assembled in pathways such as DNA replication, cell cycle, mismatch repair, Ras signaling pathway, melanoma, EGFR tyrosine kinase inhibitor resistance and homologous recombination (P < 0.05). CONCLUSIONS: Here, we provided sufficient evidence of PLK1 overexpression in cervical cancer. The overexpression of PLK1 in cervical cancer and the contributory effect of it on clinical progression indicated the hopeful prospect of PLK1 as a biomarker for cervical cancer.

ABSTRACT

Timely and accurate estimation of rice parameters plays a significant role in rice monitoring and yield forecasting for ensuring food security. Compact-polarimetric (CP) synthetic aperture radar (SAR), a good compromise between the dual- and quad-polarized SARs, is an important part of the new generation of Earth observation systems. In this paper, the ability of CP SAR data to retrieve rice biophysical parameters was explored using a modified water cloud model. The results showed that S1 was superior to other CP variables in rice height inversion with a coefficient of determination (R²) of 0.92 and a root-mean-square error (RMSE) of 5.81 cm. RL was the most suitable for inverting the volumetric water content of the rice canopy, with an R² of 0.95 and a RMSE of 0.31 kg/m³. The m-χ decomposition produced the highest accuracies for the ear biomass: R² was 0.89 and RMSE was 0.17 kg/m². The highest accuracy of leaf area index (LAI) retrieval was obtained for RH (right circular transmit and horizontal linear receive) with an R² of 0.79 and a RMSE of 0.33. This study illustrated the capability of CP SAR data with respect to retrieval of rice biophysical parameters, especially for height, volumetric water content of the rice canopy, and ear biomass, and this mode may offer the best option for rice-monitoring applications because of swath coverage.

ABSTRACT

BACKGROUND: This study was carried out to discover the underlying role that HOXA11 plays in lung squamous cancer (LUSC) and uncover the potential corresponding molecular mechanisms and functions of HOXA11-related genes. METHODS: Twenty-three clinical paired LUSC and non-LUSC samples were utilized to examine the level of HOXA11 using quantitative real-time polymerase chain reaction (qRT-PCR). The clinical significance of HOXA11 was systematically analyzed based on 475 LUSC and 18 non-cancerous adjacent tissues from The Cancer Genome Atlas (TCGA) database. A total of 102 LUSC tissues and 121 non-cancerous tissues were available from Oncomine to explore the expressing profiles of HOXA11 in LUSC. A meta-analysis was carried out to further assess the differential expression of HOXA11 in LUSC, including in-house qRT-PCR data, expressing data extracted from TCGA and Oncomine databases. Moreover, the enrichment analysis and potential pathway annotations of HOXA11 in LUSC were accomplished via Gene Oncology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG). The expression of hub genes and according correlations with HOXA11 were assessed to further explore the biological role of HOXA11 in LUSC. RESULTS: HOXA11 expression in LUSC had a tendency to be upregulated in comparison to adjacent non-cancerous tissues by qRT-PCR. TCGA data displayed that HOXA11 was remarkably over-expressed in LUSC compared with that in non-LUSC samples, and the area under curves (AUC) was 0.955 (P < 0.001). A total of 1523 co-expressed genes were sifted for further analysis. The most significant term enriched in the KEGG pathway was focal adhesion. Among the six hub genes of HOXA11, including PARVA, ILK, COL4A1, COL4A2, ITGB1, and ITGA5, five (with the exception of COL4A1) were significantly decreased compared with the normal lung tissues. Moreover, the expression of ILK was negatively related to HOXA11 (r = - 0.141, P = 0.002). CONCLUSION: High HOXA11 expression may lead to carcinogenesis and the development of LUSC. Furthermore, co-expressed genes might affect the prognosis of LUSC.
