ABSTRACT

Peptidyl arginine deiminase 4 (PAD4) is an important biocatalytic enzymes involved in the conversion of protein arginine to citrulline, its dysregulation has a great impact on many physiological processes. Recently, PAD4 has emerged as a potential therapeutic target for the treatment of various diseases including rheumatoid arthritis (RA). Traditional Chinese Medicines (TCMs), also known as herbal plants, have gained great attention by the scientific community due to their good therapeutic performance and far fewer side effects observed in the clinical treatment. However, limited researches have been reported to screen natural PAD4 inhibitors from herbal plants. The color developing reagent (COLDER) or fluorescence based methods have been widely used in PAD4 activity assay and inhibitor screening. However, both methods measure the overall absorbance or fluorescence in the reaction solution, which are easy to be affected by the background interference due to colorful extracts from herbal plants. In this study, a simple, and robust high-performance liquid chromatography ultraviolet-visible (HPLC-UV) based method was developed to determine PAD4 activity. The proposed strategy was established based on COLDER principle, while used hydrophilic l-arginine instead of hydrophobic N-benzoyl-l-arginine ethyl ester (BAEE) as a new substrate to determine PAD4 inhibition activity of herbal extracts. The herbal extracts and PAD4 generated hydrophobic l-citrulline were successfully separated by the HPLC, and the developed method was optimized and validated with a known PAD4 inhibitor (GSK484) in comparison with COLDER assay. The IC50 value of GSK484 measured by HPLC-UV method was 153 nM, and the detection limit of the citrulline was 0.5 nmol, respectively, with a linear range of 0.5 nmol to 20 nmol. The IC50 value of the HPLC-UV method was improved by nearly three times compared with COLDER assay (527 nM), and the results indicated the reliability of PAD4 inhibition via HPLC-UV method. The inhibitory effect against PAD4 were fast and accurately screened for the twenty-four extracts from eight herbs. Among them, Ephedra Herba extracts showed significant inhibitory activity against the PAD4 with the IC50 values of three extracts (ethanol, ethyl acetate and water) ranging from 29.11 µg/mL to 41.36 µg/mL, which may help researchers to discover novel natural compounds holding high PAD4 inhibition activity.

ABSTRACT

Only rarely have polyoxometalates been found to form core-shell nanoclusters. Here, we succeeded in isolating a series of rare giant and all-inorganic core-shell cobalt polyoxoniobates (Co-PONbs) with diverse shapes, nuclearities and original topologies, including 50-nuclearity {Co12 Nb38 O132 }, 54-nuclearity {Co20 Nb34 O128 }, 62-nuclearity {Co26 Nb36 O140 } and 87-nuclearity {Co33 Nb54 O188 }. They are the largest Co-PONbs and also the polyoxometalates containing the greatest number of Co ions and the largest cobalt clusters known thus far. These molecular Co-PONbs have intriguing and atomically precise core-shell architectures comprising unique cobalt oxide cores and niobate oxide shells. In particular, the encapsulated cobalt oxide cores with different nuclearities have identical compositions, structures and mixed-valence Co3+ /Co2+ states as the different sized Co-O moieties of the bulk cubic-spinel Co3 O4 , suggesting that they can serve as various molecular models of the cubic-spinel Co3 O4 . The successful construction of the series of the Co-PONbs reveals a feasible and versatile synthetic method for making rare core-shell heterometallic PONbs. Further, these new-type core-shell bimetal species are promising cluster molecular catalysts for visible-light-driven CO2 reduction.

ABSTRACT

This work reports the interesting and unique cation-exchange behaviors of the first indium-bridged purely inorganic 3D framework based on high-nuclearity polyoxoniobates as building units. Each nanoscale polyoxoniobate features a fascinating near-icosahedral core-shell structure with six pairs of unique inorganic "molecular tweezers" that have changeable openings for binding different metal cations via ion-exchanges and exhibit unusual selective metal-uptake behaviors. Further, the material has high chemical stability so that can undergo single-crystal-to-single-crystal metal-exchange processes to produce a dozen new crystals with high crystallinity. Based on these crystals and time-dependent metal-exchange experiments, we can visually reveal the detailed metal-exchange interactions and mechanisms of the material at the atomic precision level. This work demonstrates a rare systematic and atomic-level study on the ion-exchange properties of nanoclusters, which is of significance for the exploration of cluster-based ion-exchange materials that are still to be developed.

ABSTRACT

Neutrophils play a pivotal role in innate immunity by releasing neutrophils extracellular traps (NETs). Excessive NETs are detrimental to the local tissue and further exacerbate inflammation. Protein arginine deiminases (PAD) mediate histone citrullination and NET formation that, in turn, exacerbate endotoxin shock damages. In this study, we further investigated the molecular mechanism underlying PAD and NETs in endotoxic stress in mice. The control group mice were injected with solvent, the LPS endotoxic shock group mice were intraperitoneally injected with LPS at 35 mg/kg only, while the LPS and PAD inhibitor YW3-56 treatment group mice were injected with YW3-56 at 10 mg/kg prior to the LPS injection. YW3-56 significantly prolonged the survival time of the LPS-treated mice. NETs, cfDNA, and inflammatory factors were detected by ELISA in serum, paitoneal cavity, and lung at 24 h after LPS administration. Lung injuries were detected by immunostaining, and lung tissue transcriptomes were analyzed by RNA-seq at 24 h after LPS administration. We found that YW3-56 altered neutrophil tissue homeostasis, inhibited NET formation, and significantly decreased cytokines (IL-6, TNFα and IL-1ß) levels, cytokines gene expression, and lung tissue injury. In summary, NET formation inhibition offers a new avenue to manage inflammatory damages under endotoxic stress.

ABSTRACT

Peptidylarginine deiminases (PADs) are indispensable enzymes for post-translational modification of proteins, which can convert Arg residues on the surface of proteins to citrulline residues. The PAD family has five isozymes, PAD1, 2, 3, 4, and 6, which have been found in multiple tissues and organs. PAD2 and PAD4 were detected in cerebral cortex and hippocampus from human and rodent brain. In the central nervous system, abnormal expression and activation of PADs are involved in the pathological changes and pathogenesis of Alzheimer's disease (AD). This article reviews the classification, distribution, and function of PADs, with an emphasis on the relationship between the abnormal activation of PADs and AD pathogenesis, diagnosis, and the therapeutic potential of PADs as drug targets for AD.

ABSTRACT

A rare and novel Cr-substituted polyoxoniobate (PONb), [Cr2.5Nb27.5O66(OH)20(H2O)2]7-, has been synthesized by a combination of hydrothermal and conventional solution methods. The PONb shows an unknown tetrameric structure, which is the largest Cr-containing PONb to date. Interestingly, every two PONb tetramers can be joined together by alkali-metal cations to form a discrete cubelike ionic cluster. The obtained PONb not only enriches the very limited members of the Cr-substituted PONb family but also exhibits high ionic conductivity.

ABSTRACT

For accurate motion compensation (MOCO) in airborne synthetic aperture radar (SAR) imaging, a high-precision inertial navigation system (INS) is required. However, an INS is not always precise enough or is sometimes not even included in airborne SAR systems. In this paper, a new, raw, data-based range-invariant motion compensation approach, which can effectively extract the displacements in the line-of-sight (LOS) direction, is proposed for high-resolution sliding spotlight SAR mode. In this approach, the sub-aperture radial accelerations of the airborne platform are estimated via a well-developed weighted total least square (WTLS) method considering the time-varying beam direction. The effectiveness of the proposed approach is validated by two airborne sliding spotlight C band SAR raw datasets containing different types of terrain, with a high spatial resolution of about 0.15 m in azimuth.

ABSTRACT

A radical C-H arylation reaction of oxazoles with (hetero)aryl iodides using Cs2CO3 as base/electron donor and 1,1'-bis(diphenylphosphino) ferrocene (dppf) as a catalytic SET mediator is reported. The overall reaction likely follows the general base-promoted homolytic aromatic substitution mechanism through a radical-chain pathway. DFT calculations suggest that dppf forms a complex with CsCO3-, enhancing its SET reducing ability to generate an aryl radical from ArI.
