ABSTRACT

Within the context of maximizing the use of dietary phosphorus, a growing-finishing pig study was conducted to determine the optimal total dietary calcium (Ca) to digestible phosphorus (dP) ratio and to verify the possibility of mineral phosphate removal during the finishing period on growth performance and mineral status. The potential for replacing chemical and mechanical bone properties by dual energy X-ray absorptiometry (DXA) measures on non-dissected feet was also verified. Three Ca to dP ratios (2.2:1, 2.5:1 and 2.8:1) within two dP levels (P+, P-) were fed during 91 days to 84 pigs. The grower and finisher P+ diets contained 3.0 and 2.4 and P- diets contained 2.5 and 1.7 g dP/kg, respectively. Growth performance and blood serum mineral content were independent of treatments, except that 2.2:1 impaired finisher feed conversion ratio compared to 2.5:1 and 2.8:1. Urinary P concentration increased by 37% in 2.2:1 compared to 2.5:1 and 2.8:1. Maximal load on bone and DXA mineral density were reduced in 2.2:1 compared to 2.8:1. Bone ash and volumetric density were reduced in 2.2:1 and 2.5:1 compared to 2.8:1. Diet P- reduced bone ash, maximal load, volumetric density and DXA bone mineral content and density. No interaction was observed between Ca and dP level. Therefore, 2.2:1 was insufficient for an efficient metabolic use of P, 2.5:1 was sufficient to maximize growth performance and 2.8:1 further improved bone mineralization. Increasing dietary Ca did not impair bone zinc content. Diets P- without supplemented mineral phosphates during the finisher period resulted, per pig, in a decrease of its use by 65% and of the calculated P excretion by 41%, without impaired growth performance. Finally, DXA data responded to dietary treatments as did labor intensive chemical and mechanical bone properties.

ABSTRACT

Acute pain and peripheral sensitization development after cautery disbudding was investigated in 33 calves administered preventive multimodal analgesia. The animals were assigned randomly to three groups: 1) Group SH (Control), undergoing sham disbudding at 1 and 4weeks of age; 2) Group ED (Early Disbudding), undergoing disbudding at 1week of age and sham disbudding at 4weeks of age; 3) Group LD (Late Disbudding), undergoing sham disbudding at 1week of age and disbudding at 4weeks of age. Physiological parameters (heart rate, respiratory rate, rectal temperature, invasive blood pressure, cortisol, ß-endorphin, interleukin-1ß, interleukin-6, tumor necrosis factor-α and haptoglobin plasmatic concentration), local variables (tactile sensitivity score, pressure pain thresholds and horn temperature), behavior and pain scores [multidimensional pain scale and visual analogue scale (VAS)] were assessed at baseline and at several pre-determined time points until 24h after disbudding. Tactile sensitivity score significantly and equally increased in both groups ED and LD and pressure pain thresholds significantly decreased in group LD until 24h after disbudding compared to group SH. Pain and VAS scores significantly and equally increased in both groups ED and LD until 24h after disbudding compared to group SH. No significant differences in physiological parameters, behavior and horn temperature were detected among groups. The present study suggests that acute pain and peripheral sensitization develop and do not differ in calves disbudded at 1week and 4weeks of age. Moreover, the use of physiological and behavioral parameters as sole indicators of acute pain might lead to improper conclusions and should be reassessed.

ABSTRACT

BACKGROUND: Drugs for the treatment of groups of pigs receiving liquid feed are frequently mixed into the feed and administered via the pipelines of the feeding installations. In-feed antimicrobials may select antimicrobial resistant strains among the bacteria which form the biofilm of these pipelines and are shed into the liquid feed. OBJECTIVE AND METHODS: In order to evaluate the risk of selecting antimicrobial resistant bacteria in the biofilm of liquid feeding installations, the effect of the administration of antimicrobials via the pipelines on the occurrence of antimicrobial resistance in the feed was examined in a case-control study. A premix containing either sulphonamide plus trimethoprim or sulphonamide plus chlortetracycline plus tylosin or chlortetracycline was administered via the pipelines to each batch of bought-in fattening pigs in 7, 3 and 3 case farms respectively, whereas antimicrobials had not been administered via the liquid feeding installation for at least 2 years in the 14 control farms. Enterobacteriaceae and sulphonamide-trimethoprim resistant Enterobacteriaceae were counted in twelve and eight feed samples collected in each case and in each control farm respectively during one fattening period. The semiparametric Generalized Estimating Equations (GEE) method was used for the statistical data analysis. RESULTS: The ratio of sulphonamide and trimethoprim resistant to total Enterobacteriaceae was higher in the feed of the case farms compared to the control farms (P < 0.001) and did not decrease after treatment during the fattening period. CONCLUSION: The administration of antimicrobials via the liquid feeding installation selects antibiotic resistant bacteria in the biofilm lining the pipelines, which may contaminate the liquid feed for extended periods and transmit their resistance genes to the gastrointestinal flora of the pigs. Alternatives to the administration of antimicrobials via pipelines of liquid feeding installations for group treatment should be developed.

ABSTRACT

We investigated the nutritional effects on carcass traits, gene expression and DNA methylation in a three generation Large White pig feeding experiment. A group of experimental (E) F0 boars were fed a standard diet supplemented with high amounts of methylating micronutrients whereas a control group (C) of F0 boars received a standard diet. These differentially fed F0 boars sired F1 boars which then sired 60 F2 pigs. Carcass traits were compared between 36 F2 descendants of E F0 boars and 24 F2 descendants of C F0 boars. The two F2 offspring groups differed with respect to backfat percentage (P = 0.03) and tended to differ with respect to adipose tissue (P = 0.09), fat thickness at the 10(th) rib (P = 0.08) and at the croup (P = 0.09) as well as percentages of shoulder (P = 0.07). Offspring from the experimental F0 boars had a higher percentage of shoulder and were leaner compared to the control group. Gene expression profiles showed significant twofold differences in mRNA level between 8 C F2 offspring and 8 E F2 offspring for 79, 64 and 53 genes for muscle, liver and kidney RNA, respectively. We found that in liver and muscle respective pathways of lipid metabolism and metabolic pathway were over-represented for the differentially expressed genes between these groups. A DNA methylation analysis in promoters of differentially expressed genes indicated a significant difference in DNA methylation at the IYD gene. If these responses on carcass traits, gene expression and DNA methylation withstand verification and can indeed be attributed to transgenerational epigenetic inheritance, it would open up pioneering application in pork production and would have implications for human health.

ABSTRACT

Synthetic biology has been successfully used to program novel metabolic function in mammalian cells and to design the first-generation of prosthetic networks that have shown the potential for the treatment of obesity, hormone-related disorders and hyperuricemia in small-animal model systems. By functionally rewiring luteinizing hormone receptor signaling to CREB1 (cAMP-responsive element binding protein 1)-mediated transgene expression via the common cyclic adenosine monophosphate (cAMP) second messenger pool we have designed an artificial insemination device which enables lutropin-triggered in-utero release of sperms protected inside cellulose-based implants. Swiss dairy cows treated with such in-utero implants containing spermatozoa and mammalian cells transgenic for luteinizing hormone receptor and CREB1-inducible expression of an engineered cellulase showed ovulation-triggered implant degradation and sperm release leading to successful fertilization of the animals. Synthetic devices plugged into endogenous control circuitry enable the body to automatically control spatio-temporal metabolic activities that could improve the economics of cattle breeding and provide novel opportunities for future therapeutic interventions.

ABSTRACT

A diet contaminated with 2.8 mg deoxynivalenol (DON)/kg was fed at 6 kg per day to 32 mycotoxin-exposed pluriparous sows (M) during lactation. The 31 control sows (C) received 6 kg of an uncontaminated diet. Although more contaminated diet was refused (P = 0.05), DON exposure had no effect (P > 0.1) on body weight loss of the sows during lactation (M: 27.9 ± 12.3 kg; C: 29.7 ± 10.2 kg), the number of weaned piglets (M: 9.8 ± 1.4; C: 9.7 ± 1.6) and their daily weight gain (M: 266 ± 70 g; C: 272 ± 64 g). Several sows were culled after weaning for reasons unrelated to the experiment. Compared with the remaining 21 C sows, the remaining 26 M sows had an identical interval between weaning and the next farrowing (M: 120 ± 1 days; C: 120 ± 1 days) and a similar litter size (M: 14.5 ± 2.7; C: 14.9 ± 3.0; P > 0.10). The daily intake of 17 mg DON during lactation thus did not affect the reproductive performance of the sows.

ABSTRACT

Nine gilts weighing 80 kg at the beginning of the trial were fed a mycotoxin contaminated diet containing 2 mg deoxynivalenol (DON) and 0.4 mg zearalenone (ZON) per kg (Diet M). Their daily weight gain until 103 kg BW was reduced in comparison to the nine control animals fed an uncontaminated diet (Diet C) (763 vs. 912 g; p = 0.02). There was no treatment effect on the age at first observed oestrus. Seven and eight gilts receiving Diet M and C, respectively, became pregnant after being mated once or being again mated three weeks later. The examination of the uteri of gilts slaughtered 35-61 days after mating showed that the exposure to DON and ZON had no effect on the number of foetuses per gilt (p = 0.54), but increased their growth rate (p = 0.003). Thus, low dietary DON and ZON levels had no negative effects on the reproductive parameters examined. The hypothesis that the bulbourethral gland weight of barrows can be used for the bioassay of low dietary ZON levels was rejected since feeding Diet M from 80-103 kg BW did not increase the weight of that accessory sex gland (p = 0.51).

ABSTRACT

Three temperate forages, sainfoin, birdsfoot trefoil, and chicory, characterized by elevated contents of plant secondary compounds, were compared to a ryegrass-clover mixture (control) in dried (Experiment 1) and ensiled form (Experiment 2) in their palatability and nutritional value. Palatability was measured in adult wethers (n = 6) allowed to choose between the familiar control forage and one of the three test plants. Palatability index was calculated from differences in intake of control and test plants measured after given times. Generally at first contact, palatability of the unfamiliar plants was low. Lag time until palatability index approached or exceeded a value of 100 was 2-5 d, but could not be related to the content of condensed tannins. Sainfoin had a high palatability, the highest content of condensed tannins (77.4 +/- 10.23 g/kg DM), a high content of duodenally utilisable crude protein (94.7 +/- 16.87 g/100 g CP), and a high content of metabolizable energy (9.5 +/- 0.38 MJ ME/kg DM), making this plant most promising for various purposes including anthelmintic action.

ABSTRACT

The effects of tanniferous sainfoin on digestion and metabolism have been investigated in 12 lambs in an incomplete cross-over design (n = 6). Effects of condensed tannins (CT) were evaluated by comparing dehydrated and ensiled sainfoin treated with and without polyethylene glycol (PEG). Dehydrated and ensiled grass-clover mixtures served as controls. The lambs were fed the treatment diets, including a mineral supplement, for 21 d. During the last 7 d excreta, rumen fluid and blood were sampled. The CT of sainfoin decreased rumen fluid ammonia concentration (p < 0.001) and increased the plasma concentration mainly of essential amino acids (p < 0.001). Body retention of phosphorus, calcium and magnesium was lower with sainfoin compared to PEG-treated sainfoin (p < 0.05). Sainfoin without PEG resulted in lower digestibilities of organic matter and neutral detergent fibre than sainfoin with PEG and the grass-clover mixture (p < 0.001). Ensiling of sainfoin led to the lowest N-retention. In conclusion, the reduction in ruminal ammonia and urine-N losses by sainfoin CT did not improve N-retention.
