ABSTRACT

This study explores the effects and possible mechanisms of nuclear factor E2 related factor 2 (NRF2) on ovarian granulosa cells, providing a scientific basis to prevent premature ovarian failure. An ovarian cell injury model was constructed by treating human ovarian granulosa cell (KGN cell) with 4-Vinylcyclohexene dioxide (VCD). Firstly, KGN cells were treated with different concentrations of VCD, and cell counting kit 8 (CCK-8) was used to detect ovarian cell proliferation. After determining IC50 by CCK8, the levels of estradiol and progesterone in the cell supernatant were detected using enzyme-linked immunosorbent assay (ELISA), reactive oxygen species (ROS) assay kit was used to detect the content of ROS in ovarian cells, real-time fluorescence quantitative polymerase chain reaction (qRT PCR) was used to detect the mRNA expression level of NRF2, and Western blot was used to detect the protein expression level of NRF2. Further, NRF2 silence (siNRF2) and overexpression (NRF2-OE) cell models were constructed through lentivirus transfection, and the effects of regulating NRF2 on VCD treated cell models were investigated by detecting hormone levels, oxidative stress indicators (ROS, SOD, GSH-Px), and autophagy (LC3B level). The results showed that VCD intervention inhibited the proliferation of ovarian granulosa cells in a time-dependent and dose-dependent manner (F>100, P<0.05), with an IC50 of 1.2 mmol/L at 24 hours. After VCD treatment, the level of estradiol in the cell supernatant decreased from (56.32±10.18) ng/ml to (24.59±8.75) ng/ml (t=5.78, P<0.05). Progesterone decreased from (50.25±7.03) ng/ml to (25.13±6.67) ng/ml (t=6.54, P<0.05). After VCD treatment, the SOD of cells decreased from (44.47±7.71) ng/ml to (30.92±4.97) ng/ml (t=3.61, P<0.05). GSH-Px decreased from (68.51±10.17) ng/ml to (35.19±6.59) ng/ml (t=5.73, P<0.05). Simultaneously accompanied by an increase in autophagy and a decrease in NRF2. This study successfully constructed KGN cell models that silenced NRF2 and overexpressed NRF2. Subsequently, this study treated each group of cells with VCD and found that the cell proliferation activity of the siNRF2 group was significantly reduced (t=8.37, P<0.05), while NRF2-OE could reverse the cell activity damage caused by VCD (t=3.37, P<0.05). The siNRF2 group had the lowest level of estradiol (t=5.78, P<0.05), while NRF2-OE could reverse the decrease in cellular estradiol levels caused by VCD (t=5.58, P<0.05). The siNRF2 group had the lowest progesterone levels (t=3.02, P<0.05), while NRF2-OE could reverse the decrease in cellular progesterone levels caused by VCD (t=2.41, P<0.05). The ROS level in the siNRF2 group was the highest (t=2.86, P<0.05), NRF2-OE could reverse the increase in ROS caused by VCD (t=3.14, P<0.05), the SOD enzyme content in the siNRF2 group was the lowest (t=2.98, P<0.05), and NRF2-OE could reverse the decrease in SOD enzyme content caused by VCD (t=4.72, P<0.05). The GSH-Px enzyme content in the siNRF2 group was the lowest (t=3.67, P<0.05), and NRF2-OE could reverse the decrease in antioxidant enzyme content caused by VCD (t=2.71, P<0.05). The LC3B level was highest in the siNRF2 group (t=2.45, P<0.05), and NRF2-OE was able to reverse the LC3B elevation caused by VCD (t=9.64, P<0.05). In conclusion, NRF2 inhibits ROS induced autophagy, thereby playing a role in reducing ovarian granulosa cell damage, which may be a potential target for premature ovarian failure.

ABSTRACT

Oral epithelial adhesion to the lamina propria underlies the physiologic function of the oral mucosa and contributes to resisting bacterial invasion, preventing body fluid loss, and maintaining routine chewing; thus, understanding the factors that positively influence oral epithelial adhesion is a research topic of great interest. Rete pegs contribute to oral epithelial adhesion by enlarging the contact areas, whereas integrins are the major molecules that mediate epithelial cell adhesion to the basement membrane. Keratinocyte growth factor (KGF) can promote both rete peg elongation in the skin and the expression of integrins in various cell types. Herein, we tested the effects of submucosal injection of KGF in the ventral surfaces of rat tongues on oral epithelial adhesion. The data confirmed that topical injection of KGF elevated the adhesive forces, elongated the rete pegs, and increased the abundance of integrins, KGF, and KGF receptor on the rat tongue ventral surface. However, HYD-1 (Lys-Ile-Lys-Met-Val-Ile-Ser-Trp-Lys-Gly), an integrin antagonist, inhibited the KGF-enhanced epithelial adhesion and rete peg elongation. Moreover, KGF promoted the expression of integrin subunits α6, ß4, α3, and ß1 in human immortalized oral epithelial cells in 2- and 3-dimensional culture systems. In vitro cell attachment assays demonstrated that HYD-1 inhibited the adhesion of human immortalized oral epithelial cells on Matrigel. Strikingly, the expression of integrins, KGF, and KGFR in human masticatory mucosae with longer rete pegs was more abundant than that in the lining mucosae with shorter rete pegs. In addition, rete peg lengths were positively correlated with the expression levels of integrins, KGF, and KGF receptor. These findings indicate that KGF strengthens oral epithelial adhesion and rete peg elongation via integrins.

ABSTRACT

Reactions of neutral V(n), Nb(n), and Ta(n) metal clusters (n< or =11) with CO+H(2) mixed gases and CH(3)OH in a flow tube reactor (1-50 Torr) are studied by time of flight mass spectroscopy and density functional theory calculations. Metal clusters are generated by laser ablation, and reactants and products are ionized by low fluence (approximately 200 microJ/cm(2)) 193 nm excimer laser light. Nb(n) clusters exhibit strong size dependent reactivity in reactions both with CO+H(2) and CH(3)OH compared with V(n) and Ta(n) clusters. A "magic number" (relatively intense) mass peak at Nb(8)COH(4) is observed in the reaction of Nb(n) clusters with CO+H(2), and CH(3)OH is suggested to be formed. This feature at Nb(8)COH(4) remains the most intense peak independent of the relative concentrations of CO and H(2) in the flow tube reactor. No other Nb(n), Ta(n), or V(n) feature behaves in this manner. In reactions of CH(3)OH with metal clusters M(n) (M=V, Nb, and Ta, n=3-11), nondehydrogenated products M(n)COH(4)/M(n)CH(3)OH are only observed on Nb(8) and Nb(10), whereas dehydrogenated products M(n)CO/CM(n)O are observed for all other clusters. These observations support the suggestion that CH(3)OH can be formed on Nb(8) in the reaction of Nb(n) with CO+H(2). A reaction mechanism is suggested based on the experimental results and theoretical calculations of this work and of those in the literature. Methanol formation from CO+H(2) on Nb(8) is overall barrierless and thermodynamically and kinetically favorable.

ABSTRACT

Neurofibromatosis type 1 (NF1) is an inherited autosomal dominant disorder. Haematoma is an unusual complication of neurofibromatosis and extremely rare in the maxillofacial region. A case of haematoma in NF1 of the left face is presented. MR images of acute haematoma in NF1 and radiographic features of the mandible are described. Stenosis of the internal jugular vein was noted in MR angiography (MRA). Surgical resection of the tumour and evacuation of blood clots were performed. Histological and immunohistochemical examination demonstrated that the neurofibroma tumour cells infiltrated the mural layer of vessels without malignant translation. MRI is a good choice for depicting haematoma in neurofibromatosis. Intratumour haemorrhage may result from the infiltration of vessels into the lesion and minor trauma on the affected area.

ABSTRACT

Neutral niobium and tantalum clusters (Nbn and Tan) are generated by laser ablation and supersonic expansion into a vacuum and are reacted in a pickup cell with various low pressure (approximately 1 mTorr) unsaturated hydrocarbons (acetylene, ethylene, propylene, 1-butene, 1,3-butadiene, benzene, and toluene) under nearly single collision conditions. The bare metal clusters and their reaction products are ionized by a 193 nm laser and detected by a time of flight mass spectrometer. Partially and fully dehydrogenated products are observed for small (nor=m) neutral metal clusters, respectively, with m ranging from 2 to 5 depending on the particular hydrocarbon. In addition to primary, single collision products, sequential addition products that are usually fully dehydrogenated are also observed. With toluene used as the reactant gas, carbon loss products are observed, among which Nb8C12 and Ta8C12 are particularly abundant, indicating that the Met-Car molecule M8C12 can be formed from the neutral metal cluster upon two collisions with toluene molecules. The dehydrogenation results for low pressure reactions are compared with those available from previous studies employing flow tube (high pressure) reactors. Low pressure and high pressure cluster ion reactions are also compared with the present neutral metal cluster reactions. Reactions of unsaturated hydrocarbons and metal surfaces are discussed in terms of the present neutral cluster results.

ABSTRACT

Neutral vanadium, niobium, and tantalum oxide clusters are studied by single photon ionization employing a 26.5 eV/photon soft x-ray laser. During the ionization process the metal oxide clusters are almost free of fragmentation. The most stable neutral clusters of vanadium, niobium, and tantalum oxides are of the general form (MO2)0,1(M2O5)y. M2O5 is identified as a basic building unit for these three neutral metal oxide species. Each cluster family (Mm, m=1,...,9) displays at least one oxygen deficient and/or oxygen rich cluster stoichiometry in addition to the above most stable species. For tantalum and niobium families with even m, oxygen deficient clusters have the general formula (MO2)2(M2O5)y. For vanadium oxide clusters, oxygen deficient clusters are detected for all cluster families Vm (m=1,[ellipsis (horizontal)],9), with stable structures (VO2)x(V2O5)y. Oxygen rich metal oxide clusters with high ionization energies (IE>10.5 eV, 118 nm photon) are detected with general formulas expressed as (MO2)2 (M2O5)y O1,2,3. Oxygen rich clusters, in general, have up to three attached hydrogen atoms, such as VO3H1,2, V2O5H1,2, Nb2O5H1,2, etc.

ABSTRACT

This study evaluates the effects of two different designs of platysma myocutaneous flap, vertical and transverse, used in the reconstruction of defects following the excision of oral and facial tumors. Modified radical neck dissection or selected neck dissection was also performed. Out of the 48 patients, vertical and transverse platysma myocutaneous flaps were used for 41 and 7, respectively. The postoperative outcome for the vertical flaps was 37 cases surviving, two cases of complete necrosis, and two cases of partial necrosis. With the transverse flaps, six survived and there was one case of complete necrosis. The success rate was 90.2% and 85.7% for the vertical and the transverse flap, respectively. The form and function of recipient sites were well recovered. In conclusion, the platysma myocutaneous flap has clinical value in selected patients needing reconstruction of small and medium-sized intraoral or facial defects. It is recommended that the vertical design be used for reconstruction of buccal mucosa defects, and the transverse design for mouth-floor and facial defects.

ABSTRACT

BACKGROUND AND OBJECTIVES: K-ras (Kirsten-ras) point mutation (PM) in codon 12 are suggested to be significantly associated with the tumorigenesis of pancreatic cancer. The incidences of K-ras PMs in human pancreatic cancer are reported to be different between Europeans and Japanese. The present study was designed to compare the incidences and profile of K-ras PMs and ras-p21 expression in primary invasive ductal carcinoma (IDC) of the pancreas between Japanese and Chinese. METHODS: The specimens included 51 Japanese and 34 Chinese patients with the primary IDC of the pancreas. K-ras PMs were tested by allele specific oligonucleotide dot blot hybridization methods and ras-p21 expression was stained by the immunohistochemical method. RESULTS: K-ras PMs were detected in 48 Japanese IDCs (94%) and in 24 Chinese ones (71%). There was a significant difference between the two groups. The GAT mutation was more frequent both in Japanese (61%, 33/54) and in Chinese (60%, 18/30) IDCs. The transitions/transversions ratio in the Japanese group was 2.4 in this study. By contrast, that in the Chinese group was 1.5. The expression of p21 was detected in 24 Japanese IDCs (47%) and in 24 Chinese IDCs (71%). There was a significant difference between the two groups. The expression of p21 and the patterns of K-ras PMs did not show any significant influence on the survival of the patients both in Japanese and Chinese. In the present study, Chinese IDC had a lower frequency of K-ras PMs in codon 12 than Japanese IDC. The pattern of K-ras PMs in Chinese IDC was different from that in Japanese and European IDC, respectively. CONCLUSIONS: Ki-ras PM and p21 expression were frequently seen both in Japanese and Chinese patients with pancreatic cancer. Factors such as lifestyle and environment may have influences on pancreatic carcinogenesis in various populations.

ABSTRACT

Cholelithiasis differs considerably from area to area in the world. Calcium bilirubinate stones or brown pigment stones are said to predominate in the Orient, however, this situation may differ within the Orient. In order to compare cases in China and Japan, 102 consecutive cases of cholelithiasis operated on in Shenyang, China were analyzed for the composition of gallstones and bacterial species isolated from bile in relation to the location and composition of gallstones. In Shenyang, calcium bilirubinate stones predominated, occurring in 49.0 per cent of the cholelithiasis cases. This was much higher than in Japan, which had 17.5 per cent. The incidence of bacteria was also very high, ranging from 20 to 96 per cent, with an average of 66.7 per cent depending on the kind of gallstone present. Bacterial species possessing beta-glucuronidase activity were present in nearly all the cases of calcium bilirubinate stones (92.0 per cent). The incidence of bacteria with beta-glucuronidase activity especially of E. coli was much higher than in Japan (50.8 per cent versus 21.8 per cent) in concordance with the higher incidence of calcium bilirubinate stones in China (49.0 versus 17.5 per cent).
