ABSTRACT

Risk taking is imperative for the survival and fitness of animals since they are constantly facing innumerable threats from various sources. Indeed, the ability of the individual to balance the costs and benefits of various options and adopt a wise decision is critical for the animal well-being. We modified several traditionally used anxiety tests [The modified light-dark box (mLDB), the modified open field test (mOFT) and the modified defensive withdrawal apparatus (mDWA)] by adding a palatable food reward within the anxiogenic zone which granted us to assess the sex differences in risk-taking behavior in Smith's zokors (Eospalax smithii), a typical subterranean rodent species endemic to the Qinghai-Tibetan Plateau. Concomitant with our working hypothesis, female zokors showed strong aversion and avoidance behavioral responses when tested in the mOFT and mDWA while there were no apparent sexually dimorphic behavioral changes when they were tested in the mLDB (Except for the percentage of food consumed and the latency till start feeding). Furthermore, comparison between the three behavioral paradigms revealed that both sexes showed different behavioral responses toward the different behavioral tests. Sex differences in repeatable behaviors were more profound in females than males. This might reflect different degrees of risk perception and emotionality that may differ considerably between the different models of anxiety. Our results highlighted the functional significance of a trade-off between risk and incentives in natural environment that both male and female zokors differ in the processing of risk assessment in the presence of a food reward.

ABSTRACT

The plateau zokor (Eospalax baileyi) is employed as an ideal model for examining the relationships between phenotypic and ecological adaptations to the underground conditions in which the skull morphology evolves to adapt to tunnel environment. We evaluated the influence of environmental factors (altitude, temperature, and precipitation) and geographical distance on the variations in skull morphology of a native subterranean rodent plateau zokor population. Thin-plate spline showed that the trend of morphological changes along the CV1 axis was as follows: the two zygomatic arch and the two postorbital processes moved down, the two mastoid processes and the tooth row moved upward, and the tympanic bulla grew longer. The changes along the CV2 axis were as follows: the nasal bone and the tooth row became longer, the distance between the two anterior tips of zygomatic arch lengthened, the infraorbital foramen became smaller, the whole posterior part of the skull became shorter, the zygomatic bone and the two posterior tips of zygomatic arch moved down, and the foramen magnum became bigger. Thus we found significant differences in the skull shape among the seven populations studied. Along with the reduction in the altitude and increase in the mean annual temperature and mean annual precipitation, the nasal bone became shorter, the distance between the two anterior tips of the zygomatic arch became shorter, the whole posterior part of the skull lengthened, the infraorbital foramen became smaller, the two mastoid processes moved upward, and the occipital bone moved down on the dorsal surface of the skull. On the ventral surface of the skull, with an increase in the altitude, mean annual temperature, and mean annual precipitation, the tympanic bulla became shorter, the tooth row moved down, and the foramen magnum became smaller. The morphological changes in the skull were significantly positively correlated with environmental factors. Finally, there was a significant positive correlation between the Procrustes distance matrix of the skull and the geographic distance matrix, which indicates that the evolution of the plateau zokor follows the distance isolation model, but it needs to be further explored from genetic perspectives.

ABSTRACT

Erwinia persicinus (E. persicina) is a plant pathogenic bacterial species that was previously isolated from a case of human infection. This study aimed to create an experimental infection protocol for E. persicina in laboratory mice. Seventy-two adult mice were divided into four groups (18 animal/group): the control group (G1), the group infected with E. persicina (G2), the group immune-suppressed with cyclophosphamide (G3) and the group immune-suppressed with cyclophosphamide and infected with E. persicina (G4). G2 and G4 were injected with 200 µL of (1 × 1013 cfu/ml) concentration intraperitoneally. Clinical signs, such as diarrhoea, apathy and mortality were observed only in G2 and G4 animals. E. persicina was not detected in blood. Necropsies of the G2 and G4 animals showed lesions in the intestine, liver, kidney and lung tissue. These lesions were characterized by infiltration of inflammatory cells, hyperaemia and focal areas of tissue necrosis in the liver. The results of the pro-inflammatory cytokines analysis revealed a significant increase in the levels of TNF-α and IL1-ß in the liver tissue of the G4 group. E. persicina is an emerging bacterium that can cause pathological lesions into mammalian tissue, which warrants further investigation.

ABSTRACT

Sexual dimorphism is prevalent in most living organisms. The difference in size between sexes of a given species is generally known as sexual size dimorphism (SSD). The magnitude of the SSD is determined by Rensch's rule where size dimorphism increases with increasing body size when the male is the larger sex and decreases with increasing average body size when the female is the larger sex. The unique underground environment that zokors (Eospalax baileyi) live under in the severe habitat of the Qinghai-Tibetan Plateau (QTP) could create SSD selection pressures that may or may not be supported by Rensch's rule, making this scientific question worthy of investigation. In this study, we investigated the individual variation between sexes in body size and SSD of plateau zokors using measurements of 19 morphological traits. We also investigated the evolutionary mechanisms underlying SSD in plateau zokors. Moreover, we applied Rensch's rule to all extant zokor species. Our results showed male-biased SSD in plateau zokors: The body- and head-related measurements were greater in males than in females. Linear regression analysis between body length, body weight, and carcass weight showed significant relationships with some traits such as skull length, lower incisor length, and tympanic bulla width, which might support our prediction that males have faster growth rates than females. Further, the SSD pattern corroborated the assumption of Rensch's rule in plateau zokors but not in the other zokor species. Our findings suggest that the natural underground habitat and behavioral differences between sexes can generate selection pressures on male traits and contribute to the evolution of SSD in plateau zokors.

ABSTRACT

Around the world, climate change has impacted many species. In this study, we used bioclimatic variables and biophysical layers of Central Asia and the Asian Highlands combined with presence data of brown bear (Ursus arctos) to understand their current distribution and predict their future distribution under the current rate of climate change. Our bioclimatic model showed that the current suitable habitat of brown bear encompasses 3,430,493 km2 in the study area, the majority of which (>65%) located in China. Our analyses demonstrated that suitable habitat will be reduced by 11% (378,861.30 km2) across Central Asia and the Asian Highlands by 2,050 due to climate change, predominantly (>90%) due to the changes in temperature and precipitation. The spatially averaged mean annual temperature of brown bear habitat is currently -1.2°C and predicted to increase to 1.6°C by 2,050. Mean annual precipitation in brown bear habitats is predicted to increase by 13% (from 406 to 459 mm) by 2,050. Such changes in two critical climatic variables may significantly affect the brown bear distribution, ethological repertoires, and physiological processes, which may increase their risk of extirpation in some areas. Approximately 32% (1,124,330 km2) of the total suitable habitat falls within protected areas, which was predicted to reduce to 1,103,912 km2 (1.8% loss) by 2,050. Future loss of suitable habitats inside the protected areas may force brown bears to move outside the protected areas thereby increasing their risk of mortality. Therefore, more protected areas should be established in the suitable brown bear habitats in future to sustain populations in this region. Furthermore, development of corridors is needed to connect habitats between protected areas of different countries in Central Asia. Such practices will facilitate climate migration and connectivity among populations and movement between and within countries.

Recognition memory is important for the survival and fitness of subterranean rodents due to the barren underground conditions that require avoiding the burden of higher energy costs or possible conflict with conspecifics. Our study aims to examine the object and object/place recognition memories in plateau zokors (Eospalax baileyi) and test whether their underground life exerts sex-specific differences in memory functions using Novel Object Recognition (NOR) and Object-in-Place (OiP) paradigms. Animals were tested in the NOR with short (10min) and long-term (24h) inter-trial intervals (ITI) and in the OiP for a 30-min ITI between the familiarization and testing sessions. Plateau zokors showed a strong preference for novel objects manifested by a longer exploration time for the novel object after 10min ITI but failed to remember the familiar object when tested after 24h, suggesting a lack of long-term memory. In the OiP test, zokors effectively formed an association between the objects and the place where they were formerly encountered, resulting in a higher duration of exploration to the switched objects. However, both sexes showed equivalent results in exploration time during the NOR and OiP tests, which eliminates the possibility of discovering sex-specific variations in memory performance. Taken together, our study illustrates robust novelty preference and an effective short-term recognition memory without marked sex-specific differences, which might elucidate the dynamics of recognition memory formation and retrieval in plateau zokors.

ABSTRACT

Prey species show specific adaptations that allow recognition, avoidance, and defense against predators. This study was undertaken to investigate the processing of a chronic, life-threatening stimulus to Norway rats (Rattus norvegicus). One hundred forty-four Norway rats were tested by repeated presentation of cat urine for 1 h at different days in a defensive withdrawal apparatus. Rats exposed to urine for short periods showed significantly larger defensive behavioral and medial hypothalamic c-fos messenger RNA (mRNA) responses than other groups. These defensive responses habituated shortly after the presentation of cat urine. Serum levels of adrenocorticotropic hormone and corticosterone increased significantly when animals were repeatedly exposed to cat urine. However, the hormonal responses took longer to habituate than the behavioral and molecular responses did. We conclude that the behavioral and c-fos mRNA responses are "primed" for habituation to repeated exposures to cat urine, while the hormonal responses show "resistance." The results support our hypothesis that the strongest anti-predator responses at three levels would occur during short-term exposure to cat urine and that these responses would subsequently disappear on prolonged exposure. This study assists understanding the way in which the different levels of defensive responses are integrated and react during chronic stress.

ABSTRACT

Predator odors are non-intrusive and naturalistic stressors of high ethological relevance in animals. Upon exposure to a predator or its associated cues, robust physiological and molecular anti-predator defensive strategies are elicited thereby allowing prey species to recognize, avoid and defend against a possible predation threat. In this review, we will discuss the nature of neuroendocrine stress responses upon exposure to predator odors. Predator odors can have a profound effect on the endocrine system, including activation of the hypothalamic-pituitary-adrenal axis, and induction of stress hormones such as corticosterone and adrenocorticotropic hormone. On a neural level, short-term exposure to predator odors leads to induction of the c-fos gene, while induction of ΔFosB in a different brain region is detected under chronic predation stress. Future research should aim to elucidate the relationships between neuroendocrine and behavioral outputs to gage the different levels of anti-predator responses in prey species.

Predator odors are non-intrusive natural stressors of high ethological relevance. Animals are daily challenged with stressors of varying intensity and it is essential for their survival to respond to a wide range of threats. Behavioral and hormonal responses and changes in the level of medial hypothalamic c-fos mRNA were examined in Brandt's voles (Lasiopodomys brandtii) exposed to the feces of a domestic cat (Felis catus) stored for different periods. One hundred voles were tested in the defensive withdrawal apparatus. The voles showed an aversion to freshly collected cat feces, indicated by high levels of flight-related behaviors, increased freezing behavior, and more vigilant rearing compared to old feces. The serum levels of adrenocorticotropic hormone and corticosterone significantly increased when the voles were exposed to fresh cat feces. The level of c-fos mRNA in the medial hypothalamic region was highest in the individuals exposed to fresh cat feces. All of these behavioral, endocrine and c-fos-mRNA responses were lower when voles were subjected to older cat feces. We conclude that these responses depend on volatile chemical constituents of cat feces rather than their physical characteristics and that this accounts for the lower responses to feces stored for longer periods.

Predator odors are non-intrusive natural stressors of high ethological relevance. The objective of this study was to investigate the processing of a chronic, life-threatening stimulus during repeated prolonged presentation to Brandt's voles. One hundred and twenty voles were tested by repeated presentation of cat feces in a defensive withdrawal apparatus. Voles exposed to feces for short periods showed more avoidance, more concealment in the hide box, less contact time with the odor source, more freezing behavior, less grooming, more jumping, and more vigilant rearing than did non-exposed voles, and those exposed for longer periods. Serum levels of adrenocorticotropic hormone and corticosterone increased significantly when animals were repeatedly exposed to cat feces for short periods. The behavioral and endocrine responses habituated during prolonged presentation of cat feces. ΔfosB mRNA expression level was highest in voles exposed to cat feces for 6 and 12 consecutive days, and subsequently declined in animals exposed to cat feces for 24 days. We therefore conclude that the behavioral and endocrine responses to repeated exposure to cat feces undergo a process of habituation, while ΔfosB changes in the medial hypothalamic region exhibit sensitization. We propose that habituation and sensitization are complementary rather than contradictory processes that occur in the same individual upon repeated presentation of the same stressor.
