ABSTRACT

BACKGROUND: The most significant sexually transmissible fungal disease, semen candidiasis, is caused by Candida albicans and impacts male reproductive potential. Actinomycetes are a group of microorganisms that could be isolated from various habitats and used for the biosynthesis of various nanoparticles with biomedical applications. OBJECTIVE: Testing antifungal activity of biosynthesized Ag nanoparticles versus isolated C. albicans from semen as well as its anticancer activity versus the Caco-2 cell line. METHODS: Screening 17 isolated actinomycetes for the biosynthesis of Ag nanoparticle biosynthesis. Characterization of biosynthesized nanoparticles, testing its anti-Candida albicans, and antitumor activity. RESULTS: Streptomyces griseus was the isolate that identified silver nanoparticles using UV, FTIR, XRD and TEM. Biosynthesized nanoparticles have promising anti-Candida albicans with MIC (125 ± 0.8) µg/ml and accelerate apoptotic rate versus Caco-2 cells (IC50 = 7.30 ± 0.54 µg/ml) with minimal toxicity (CC50 = 142.74 ± 4.71 µg/ml) versus Vero cells. CONCLUSION: Certain actinomycetes could be used for the biosynthesis of nanoparticles with successive antifungal and anticancer activity to be verified by in vivo studies.

ABSTRACT

Objectives: Tellurium has received substantial attention for its remarkable properties. This study performed in vitro and in vivo testing of the antibacterial action of tellurium nanoparticles biosynthesized in actinomycetes against methicillin-resistant Staphylococcus aureus (MRSA), a common blood bacterial pathogen. Methods: Nine actinomycete isolates were tested for their potential to reduce potassium tellurite (K2TeO3) and form tellurium nanoparticles (TeNPs). The most efficient actinomycete isolate in producing Tellerium nanoparticles was identified through molecular protocols. The generated TeNPs were characterized using UV, TEM, EDX, XRD and FTIR. The bacterial species implicated in bloodstream infections were detected at El Hussein Hospital. Bacterial identification and antibiotic susceptibility testing were performed using Vitek 2. An animal infection model was used to test the efficacy of the produced TeNPs against the most commonly isolated methicillin-resistant S. aureus using survival assays, colony counting, cytokine assessment and biochemical testing. Results: The most efficient actinomycete isolate was identified as Streptomyces graminisoli and given the accession number (OL773539). The mean particle size of the produced TeNPs was 21.4 nm, and rods and rosette forms were observed. Methicillin-resistant S. aureus (MRSA) was the main bacterium (60%) causing blood stream infections, and was followed by Escherichia coli (25%) and Klebsiella pneumoniae (15%). The produced TeNPs were tested against MRSA, the bacterium most frequently isolated from blood, and showed a promising action inhibition zone of 24 ± 0.7 mm and an MIC of 50 µg/ml. An animal infection model indicated the promise of TeNPs alone or in combination with standard drugs to combat MRSA in a rat intravenous infection model. Conclusion: TeNPs combined with vancomycin have successive impact to combat bacteremia for further verification of results.

ABSTRACT

Background: Because of the growing incidence of obesity, the use of synthetic antiobesity medicines as weight-loss agents has grown in popularity, although their usefulness has yet to be established. Two of such medicines are Aplex and Venera. This study is designed to determine the potential dangers of Aplex and Venera on certain biochemical and physiological indicators in obese adult male rats. Methods: Twenty-one obese male albino rats (9 weeks old and having a body mass of 220 ± 20 g) were divided into three equal groups: the control group (vehicle treatment), the Aplex group (0.1 mg/kg/day) for 30 days, and the Venera group (0.1 mg/kg/day) for 30 days. Results: The values of serum glucose, insulin, homeostatic model assessment of insulin resistance (HOMA-IR), total protein, total cholesterol (TC), high-density lipoprotein (HDL), very-low-density lipoprotein (VLDL), TC/HDL ratio, testosterone, thyroxine (T4), and leptin did not differ significantly between the treated and control groups. In contrast, the treated groups had substantial changes in bodyweight, serum alanine aminotransaminase (ALT), aspartate aminotransaminase (AST), albumin, globulin, albumin/globulin ratio (A/G ratio), triglycerides (TG), low-density lipoproteins (LDL), LDL/HDL ratio, urea, creatinine, and triiodothyronine (T3) levels. Conclusion: The findings indicate that Aplex and Venera have negative impacts on crucial biochemical and physiological indicators, particularly liver and kidney functioning.

ABSTRACT

OBJECTIVES: To study the hemodynamic changes of hepatic & renal vessels in systemic bacterial infection with fever in HCV related cirrhosis with possible complications. METHODS: Three groups of patients with systemic bacterial infection with fever were included in the study; group І included 15 patients with decompensated cirrhosis, group ІІ included 15 patients with compensated cirrhosis and group ІІІ included 10 patients without liver affection. Laboratory parameters and Doppler US of hepatic and renal vessels were evaluated during and after subsidence of fever in all patients. RESULTS: Forty patients were enrolled in this prospective study. There were 22 male and 18 female patients. We found that the direction of blood flow in the portal and splenic veins was hepatopetal and the veins were non pulsatile in all cases with no change during and after subsidence of infection. There was no significant difference in portal or splenic vein diameters during and after subsidence of infection in the three studied groups. However, the mean values of portal and splenic veins peak velocities were significantly lower during infection in cirrhotic groups. The mean value of hepatic artery resistive index during fever was significantly higher than after fever in cirrhotic groups. Renal resistive and pulsatility indices were significantly higher during fever in cirrhotic groups. CONCLUSION: Systemic bacterial infection with fever can affect hepatic haemodynamics leading to aggravation of portal hypertension and increasing the risk of complications as variceal bleeding and hepatic encephalopathy and can also affect renal haemodynamics with increased risk of renal impairment.

ABSTRACT

PURPOSE: The aim of this study was to evaluate available bone quality and quantity after performing sinus augmentation using simvastatin/ß-TCP combination versus ß-TCP alone. MATERIALS AND METHODS: This study included eight sinus lift procedures conducted on six patients. The sinuses were divided into two equal groups. The patients were recalled one, two weeks two, five, nine months post-operatively for post-operative evaluation. Radiographic evaluation involved cone beam computed tomography (CBCT) radiographs taken for every patient one week and nine months post-operatively to evaluate the changes in bone height, while histomorphometric evaluation involved transcortical bone biopsies taken after nine months during the second-stage surgery for implant placement. RESULTS: The histomorphometric results showed that the amount of newly formed bone was higher in the simvastatin group when compared to the ß-TCP group nine months after the surgery; the difference between the two groups was statistically significant. On the other hand, the radiographic evaluation showed that the rate of resorption of the simvastatin group was found to be higher than the control group; however, the difference between both groups was statistically insignificant. CONCLUSION: These results showed that Simvastatin is safe to be used in sinus lift with promising osteoinductive capacity, yet further studies using larger sample size is needed.

ABSTRACT

AIM: This study was established to evaluate the amount of bone height changes in the posterior mandibular area of edentulous patients receiving screw-retained prostheses versus removable telescopic implant overdentures. MATERIAL AND METHODS: Every patient received four inter-foraminal implants regarding the mandibular arch and four anterior implants for the maxillary arch, computer guided surgical guides were planned for the insertion of the implants accurately with a flapless technique. Panoramic radiographs were made immediately, six months and twelve months after the prostheses` use proportional area and vertical measurements were applied to determine changes in the bone height of the posterior mandibular edentulous area. RESULTS: After twelve months, a statistically non-significant amount of bone resorption was reported for both groups. CONCLUSION: Up to the limitations of this study both treatment options the screw retained and telescopic overdenture can be used for rehabilitation of completely edentulous patients. These cases must be followed for a longer period to have a definite answer regarding their efficiency in the long run.

Some snails of Biomphalaria alexandrina can resist the infection of Schistosoma mansoni so this study aimed to clearly this mechanism by using light and electron microscopy (EM) and determine the role of Nitric oxide in this mechanism. B. alexandrina snails used in this study were exposed individually to S. mansoni infection according to their response they were classified into susceptible group (shed cercariae) and resistant group (failed to shed cercariae). Snails not exposed to infection were included in this study as control group. Nitric oxide (NO) level was assayed directly in the soluble fraction of B. alexandrina haemolymph supernatants collected from each group of B. alexandrina snails were subjected to NO assay by the Greiss reaction. The level of NO in haemolymph of infected snails was significantly increased (p < 0.001) than both control and non infected snails groups, however, in non infected snails group had significantly (p < 0.05) compared to control group. This study when correlated the changes recognized by EM with NO level the pro apoptotic effect of high level of NO on the haemocytes. Characterization and identification of cell shape of haemocytes in both haemolymph and tissue were examined by light and electron microscopy. Examination of B. alexandrina snail's haemocytes revealed three types of different cells classified according to their shape and granular contents. These cells are granulocytes, amoebocytes and hyalineocytes. Electron microscope study also revealed the important role of granulocytes and amoebocytes as defense mechanism against snail infection. NO is considered an important anti parasite molecule; intra-molluscan stages of parasites switch off host NO defense response.
