ABSTRACT

Campylobacter is a virulent Gram-negative bacterial genus mainly found in the intestines of poultry. The indiscriminate use of traditional antibiotics has led to drug resistance in these pathogens, necessitating the development of more efficient and less toxic therapies. Despite their complex biologically active structures, the clinical applications of essential oils (EOs) remain limited. Therefore, this study aimed to increase the bioavailability, stability, and biocompatibility and decrease the photodegradation and toxicity of EO using nanotechnology. The diffusion disk test revealed the potent anti-Campylobacter activity of cinnamon, lemongrass, clove, geranium, and oregano EOs (>50 mm). These were subsequently used to prepare nanostructured lipid carriers (NLCs). Formulations containing these EOs inhibited Campylobacter spp. growth at low concentrations (0.2 mg/mL). The particle size, polydispersity index, and zeta potential of these systems were monitored, confirming its physicochemical stability for 210 days at 25 °C. FTIR-ATR and DSC analyses confirmed excellent miscibility among the excipients, and FE-SEM elucidated a spherical shape with well-delimited contours of nanoparticles. The best NLCs were tested regarding nanotoxicity in a chicken embryo model. These results indicate that the NLC-based geranium EO is the most promising and safe system for the control and treatment of multidrug-resistant strains of Campylobacter spp.

ABSTRACT

Background: There is no consensus on the ideal strategy to treat posttransplant focal segmental glomerulosclerosis. The multiple-target therapy, which consisted of high-dose intravenous cyclosporine, prednisone, and plasmapheresis, showed favorable results. Methods: This single-center, prospective study sought to evaluate the multiple-target therapy in an independent cohort of patients. Results: Thirteen patients with posttransplant focal segmental glomerulosclerosis received multiple-target therapy. Complete remission was achieved in 2 patients (15.4%), and partial remission in another 2 patients (15.4%). Four patients (30.7%) did not show remission, and 5 patients (38%) lost the graft because of posttransplant focal segmental glomerulosclerosis during the 12-mo follow-up. Premature discontinuation of treatment occurred in 10 patients (77%), all associated with infectious adverse events. Cytomegalovirus was the most common complication, and preemptive therapy was used instead of prophylaxis. Conclusions: In this cohort of patients, the efficacy of the multiple-target therapy was poor and limited by the high incidence of infectious adverse events.

ABSTRACT

An environmental disaster caused by the rupture of a mining tailings dam has impacted a large area of the Rio Doce watershed in the Brazilian Atlantic Forest, resulting in unprecedented damage at spatial and temporal scales. The Atlantic Forest is one of the world's most important biodiversity hotspots. A long history of land use conversion has resulted in a highly fragmented landscape. Despite numerous restoration initiatives, these efforts have often biased criteria and use limited species assemblages. We conducted a comprehensive synthesis of the plant community in riparian forests along the Rio Doce watershed. Our work detailed vegetation composition (tree and sapling strata) and examined its relationship with edaphic and landscape factors, aiming to inform restoration projects with scientifically robust knowledge. A total of 4906 individuals from the tree strata and 4565 individuals from the sapling strata were recorded, representing a total of 1192 species from 75 families. Only 0.8% of the tree species and 0.5% of the sapling species occurred in all sampled sectors, with over 84% of the species occurring in a single watershed sector for both strata. We observed a high species heterogeneity modulated by turnover (92.3% in the tree, and 92.7% in the sapling strata) among sites. Overall, our research revealed a gradient of soil fertility influencing species composition across different strata. Additionally, we discovered that preserved landscapes had a positive impact on species diversity within both strata. The species exclusivity in the sampled sites and the high turnover rate imply the need to consider multiple reference ecosystems when restoring the watershed to reduce the risk of biotic homogenization. Finally, the reference ecosystems defined here serve as a basis for the selection of locally particular species in the implementation of restoration projects that aim to improve biodiversity, ecosystem services, and water security.

ABSTRACT

Lately, the bacterial multidrug resistance has been a reason to public health concerning around world. The development of new pharmacology therapies against infections caused by multidrug-resistant bacteria is urgent. In this work, we developed 10 NLC formulations composed of essential oils (EO), vegetable butter and surfactant. The formulations were evaluated for long-term and thermal cycling stability studies in terms of (particle size, polydispersion index and Zeta potential). In vitro antimicrobial assays were performed using disk diffusion test and by the determination of the minimum inhibitory concentration (MIC) performed with fresh and a year-old NLC. The most promising system and its excipients were structurally characterized through experimental methodologies (FTIR-ATR, DSC and FE-SEM). Finally, this same formulation was studied through nanotoxicity assays on the chicken embryo model, analyzing different parameters, as viability and weight changes of embryos and annexes. All the developed formulations presented long-term physicochemical and thermal stability. The formulation based on cinnamon EO presented in vitro activity against strains of Acinetobacter baumannii, Klebsiella pneumoniae and Pseudomonas aeruginosa isolated from humans and in vivo biocompatibility. Considering these promising results, such system is able to be further tested on in vivo efficacy assays.

ABSTRACT

BACKGROUND: Recently, anosmia and ageusia (and their variations) have been reported as frequent symptoms of COVID-19. Olfactory and gustatory stimuli are essential in the perception and pleasure of eating. Disorders in sensory perception may influence appetite and the intake of necessary nutrients when recovering from COVID-19. In this short commentary, taste and smell disorders were reported and correlated for the first time with food science. SCOPE AND APPROACH: The objective of this short commentary is to report that taste and smell disorders resulted from COVID-19 may impact eating pleasure and nutrition. It also points out important technologies and trends that can be considered and improved in future studies. KEY FINDINGS AND CONCLUSIONS: Firmer food textures can stimulate the trigeminal nerve, and more vibrant colors are able to increase the modulation of brain metabolism, stimulating pleasure. Allied to this, encapsulation technology enables the production of new food formulations, producing agonist and antagonist agents to trigger or block specific sensations. Therefore, opportunities and innovations in the food industry are wide and multidisciplinary discussions are needed.

ABSTRACT

Inselbergs are granitic or gneissic rocky outcrops prevalent in the landscape of southeastern Brazil. These ecosystems represent islands of isolated habitats that harbor a peculiar flora with high richness and endemism. The present study lists the species of vascular plants occurring in the Pedra da Andorinha Complex, located in the municipality of Jerônimo Monteiro/Cachoeiro de Itapemirim, southern Espírito Santo state, aiming to generate subsidies for the creation of a protected area. The survey was performed between July 2017 and October 2018, resulting in a record of 121 species, 96 genera, and 40 families. Bromeliaceae (17), Orchidaceae (12) and Fabaceae (10) were the richest families. The phytophysiognomy of exposed rock vegetation comprises a greater number of species (79 species) compared to the woody rupicolous communities (42). Eighteen of the collected species are threatened by extinction; a new species was discovered; and five were described based on materials previously collected in the studied location — Alcantarea patriae, Anthurium martinellii, Coleocephalocereus uebelmanniorum, Stigmatodon attenuatoides and Pitcairnia azouryi, the first four being endemic to the region. We also found Tabebuia reticulata, a rare species among Brazilian flora. Our results highlight the biological importance of the Pedra das Andorinhas Complex and reinforce the need to create a protected area to preserve biodiversity and the regional natural heritage.

ABSTRACT

Abstract Regional floristic lists are essential for defining biodiversity conservation strategies and are key to assist in filling knowledge gaps. They aim to provide a data source for applying tools to reduce extinction rates and to conserve ecosystems. Herein we present the results of an inventory of vascular plants in a rainforest in the Caparaó National Park (CNP) and approach their implications for conservation and management of this protected area and the surrounding communities. We conducted botanical expeditions between the years 2012 and 2017 in a montane and upper-montane forest of the CNP. We found 361 species distributed in 78 families and 181 genera. The study area is home to new species for science that have recently been described in other publications outside that location, and 4 new records for Espírito Santo State; also 43 species listed in different extinction threat categories (VU, EN and CR) and another 190 categorized with lesser concerns (LC and NT). The families with the highest species richness were: Melastomataceae (41 spp.), Lauraceae and Myrtaceae (30), Orchidaceae (26), Rubiaceae (24), and Asteraceae (20). Our results contribute to greater knowledge of the CNP flora, of the montane environments in Brazil and the vegetation of Espírito Santo state, in addition to demonstrate the importance of this protected area to the conservation Atlantic Forest biodiversity.

ABSTRACT

Vitamins are organic compounds essential for normal physiological functioning and they need to be provided in adequate amounts by the diet. They are nutrients mainly associated to fruit consumption, playing an important role in the cellular function, growth and development of individuals. The present study aimed to analyze levels of vitamins B, C and carotenoids of fruits from the agrobiodiversity of Northeastern Brazil, among them cajuí (Anacardium spp), murici (Byrsonima crassifolia (L.) Kunth), pequi (Caryocar coriaceum Wittm.), jenipapo (Genipa americana L.), mangaba (Hancornia speciosa Gomes), bacuri (Platonia insignis Mart.), cajá (Spondias mombin L.), umbu-cajá (Spondias bahiensis P. Carvalho, Van den Berg & M. Machado), umbu (Spondias tuberosa Arruda), pitanga (Eugenia uniflora L.), araçá (Psidium sobralianum Landrum & Proença). The vitamins were quantified using the analytical method High Performance Liquid Chromatography (HPLC). Vitamin B complex levels varied from 0.003 ± 0.01 mg/100 g to 6.107 ± 0.06 mg/100 g. Vitamin C ranged from 0.36 ± 0.06 mg/100 g to 253.92 ± 9.02 mg/100 g. Carotenoid values ranged from 0.12 ± 0.02 µg/100 g to 395.63 ± 113.69 µg/100 g. Thus, the profile of water-soluble vitamins and carotenoids of the fruits analyzed was quantified. Therefore, these fruits can provide varied amounts of vitamins important to human health. However, it is interesting for the individual to consume fruits in a diversified manner, avoiding monotony and thus guaranteeing the daily intake of more nutrients.

ABSTRACT

Plant co-products currently represent an attractive alternative to the food industry, especially to the growing market of development low-fat products. Among the co-products resulting from tropical fruits' processing, the cashew apple's fibre presents unusual nutritional and sensory characteristics. In several food preparations could use it as an ingredient. In this work, the bioactive compounds of both artisanal and industrialized cashew apple fibre were studied and the influence of the different cooking methods on their bioactive content, and the acceptance and sensory preference of products new plants-based products formulated. It was observed that both artisanal and industrial cashew apple fibres presented a rich composition in the bioactive compounds, especially regarding the content of ascorbic acid found in artisanal (147.8 mg.100g-1) and carotenoids in industrialized fibre (1.87 mg 100 g-1), which resulted in a higher antioxidant activity for both samples in each method evaluated. Frying (180 °C/3 min) and cooking in a combination oven (98 °C/10 min) exhibited higher averages regarding the retention of the bioactive compounds in the fibres, resulting in a higher antioxidant activity for the products processed by these methods. In contrast, it was boiling processing (100 °C/18min) leads to leach of water-soluble biocompounds and, consequently, their products presented a reduced antioxidant activity. The cashew apple "paçoca" and "meatballs" were judged in terms of their attributes (appearance, aroma, taste, overall impression) and buy intention. In general, the average of these results indicated a high sensorial acceptance and a partial possibility in their purchase of these products. The cashew apple fibres are a source of nutrients. Its incorporation in culinary preparations can be a friendly way to avoid waste and promote new food products.

ABSTRACT

This study reflects on the preservation of digital archival records from a systemic perspective, based on standards recognized by scientific literature. In such a way, it uses a holistic view to emphasize the relevance of preservation to be considered throughout the life cycle of records. The methodology is based on a bibliographic survey composed of articles, books, and technical publications, to obtain a narrative review. It is noteworthy that digital preservation has evolved to new heights, and requires the use of standards to implement reliable computer systems. With this, the entire life cycle can be involved in an uninterrupted chain of custody capable of ensuring the authenticity of digital records. Finally, a systemic-holistic approach is advocated, in which records are planned and produced with a view of the preservation and access in the long-term.

ABSTRACT

In this work, cashew gum (CG) and gelatin (GE) complexation was explored to encapsulate green coffee oil (GCO), rich in cafestol and kahweol, for use as ingredient in fruit juice. The microcapsules were loaded with 25, 50 and 75% (w/w) GCO and characterized by scanning electron microscopy, encapsulation efficiency and accelerated oxidation by Rancimat. Gas chromatography coupled to the mass detector was used to cafestol quantification in simulated gastrointestinal digestion and during fruit juice storage. Particles with 25% GCO (14.56 ± 6.36 µm) presented good encapsulation efficiency (85.57 ± 1.41%), reduced the GCO oxidation by six-fold and were resisted in the pasteurization conditions. The beverage added of capsules showed good sensory quality when compared to the control formulation. For the first time, the incorporation of GCO capsules into fruit juice has been reported, promoting a diterpene-rich drink with good rheological and sensory properties.

ABSTRACT

Several Passiflora species are known for their sedative and anxiolytic properties. However, the functional properties of Passiflora tenuifila Killip are still unexplored. The objective of this work was to evaluate the phenolic composition and acute toxicity, anxiolytic, sedative, and anticonvulsant effects using in vivo assays. The whole fruit (peel, pulp, and seed) was lyophilized and used for all assays. LC-MS showed 19 phenolic compounds, tentatively identified as flavonoids and phenolic acids. Acute treatment with single doses of up to 2000 mg kg-1 in Wistar rats showed no signs of mortality or toxicity over 14 days. The assay of functional effects was performed with Swiss mice, four groups, received by gavage, doses of P. tenuifila (200 or 400 mg kg-1 body weight), water, and diazepam (as negative and positive control), and behavior tests were performed after 60 min of the treatments. The animals treated with P. tenuifila fruit showed a significant decrease in locomotor activity, indicating a sedative and anxiolytic activity. No significant changes were observed in the rotarod apparatus, suggesting that the P. tenuifila fruit did not cause muscle relaxation. The 400 mg kg-1 dose of P. tenuifila exerted a protective effect against pentylenetetrazole-induced seizures, decreasing the severity and not causing the death of the animals. In conclusion, P. tenuifila showed no acute toxicity and had a promising effect as an anxiolytic agent, hypnotic-sedative and anticonvulsant, which could be related to its composition of flavonoids and phenolic acids.

ABSTRACT

INTRODUCTION: Thrombotic microangiopathy (TMA) in post-transplant setting has heterogeneous clinical manifestations. METHODS: We retrospectively studied data of 89 patients with post-transplant TMA, which was characterized by thrombi in at least one glomerulus and/or arteriole. Systemic TMA was defined by thrombocytopenia and microangiopathic anemia and early onset TMA, when occurred less than 90 days post transplant. RESULTS: The cumulative incidence was 0.93%. The majority of the recipients were young (mean age 39 years), female (52%) and Caucasian (48%) with primary kidney disease of unknown etiology (37%). Early TMA occurred in 51% of the patients and systemic TMA, in 25%. Underlying precipitating factors were: infection (54%), acute rejection (34%), calcineurin inhibitor toxicity (13%) and pregnancy (3%). 18% of the patients had several triggers. Glomerular TMA was observed in 50% of the biopsies and endothelial cell activation, in 61%. The 1-year patient survival was 97% and corresponding graft survival, 66%. Allograft survival was inferior when acute antibody mediated rejection (ABMR) occurred (with 41%; without 70%, p = 0.01), however no differences were determined by hemolysis, time of onset, thrombi location or endothelial cell activation. CONCLUSIONS: Our results suggest that post-transplant TMA is a rare but severe condition, regardless of its clinical and histological presentation, mainly when associated to ABMR.

ABSTRACT

Studies involving foods associated with pain reversal and anti-inflammatory effects using zebrafish are rarely reported in the literature. This study aimed to evaluate the effect of graviola (Annona muricata L.) fruit bar (GFB) and GFB added with acerola (Malpighia glabra L) seed extract (ASE) on acute nociception and abdominal inflammation in adult zebrafish (Danio rerio). Acute nociception was induced by formalin, capsaicin, cinnamaldehyde, acidic saline, glutamate (cutaneous models), and hypertonic saline (corneal model), and inflammation was induced by carrageenan. Both GFB and ASE exhibited antinociceptive effect modulated by the nitrergic system, guanylate cyclase, and transient receptor potential ankyrin 1 and acid-sensing ion channels. The antinociceptive effect of GFB also appears to be modulated by the opioid system and glutamatergic receptors (N-methyl-D-aspartate receptor). Only ASE presented corneal antinociceptive effect. Both samples showed anti-inflammatory effect, being more significant the effect of GFB. The addition of acerola by-product extract in GFB results in a product with greater biological potential.

ABSTRACT

Breast cancer is considered a public health problem with an increasing incidence worldwide. Several factors contribute to late diagnosis and hinder the initiation of the treatment, resulting in a worse prognosis. Objectives:To analyze the time interval between diagnosis and the beginning of the first oncologic treatment in the Brazilian population, in addition to assessing the factors associated with the longest time intervals. Method: Retrospective cohort study with 540,529 patients registered in the Hospital Cancer Registry System (SisRHC) from 2000 to 2017. The outcome was the time between diagnosis and the beginning of the first oncologic treatment, considering the interval greater than 60 days as delay. Descriptive analysis and simple logistic regression were performed (95% CI; p <0.05) to analyze the variables. Results: 204,130 cases were analyzed, mean age of 55.8 years (± 13.24), predominantly females (99.1%), 55.1% were from the southeast region and 71% lived in non-capital cities. The median of the time interval between diagnosis and the beginning of the first oncologic treatment was 63 days (interquartile range = 36-109). Sociodemographic, clinical and treatment-related variables affect the time interval, except the gender variable. Conclusion: The time between diagnosis and the beginning of the first oncologic treatment was high. Sociodemographic, clinical and treatment-related factors influence time intervals. Their early identification can contribute to guide the actions toward these most vulnerable groups to delay.

ABSTRACT

Carnauba wax is widely used in food, due to its physico-chemical characteristics with a predominance of esters and inert and stable components. Even with so many possibilities for the use of carnauba wax in food, there are still a large number of researchers around the world searching for new applications and a demand for new products with new technologies to improve existing ones. Recently, many parts of research which focus on the use of this wax in conservation and food processing have been carried out, some of which highlight the role of this wax in the microencapsulation of flavours, in preparing edible films and super hydrophobic and biodegradable packaging. This paper discusses the use of carnauba wax in food, including the extraction process of the wax, its chemical and physical characteristics, safety aspects, national and international law and permitted uses, along with the presentation of the main scientific research conducted.

ABSTRACT

This study aimed to evaluate the potential for the extraction of non-timber forest products (NTFPs) and environmental valuation by the willingness to receive from the residents of the damping zone of the National Forest (FLONA) of Pacotuba, state of Espírito Santo. Forms were applied in a sample of 78 families, in the community of Monte Alegre, located in the damping zone of the FLONA de Pacotuba, for the NTFPs ethnobotanical survey and the willingness to receive to maintain the forest cover. The mapping use of the soil Pacotuba FLONA and the damping zone were obtained through a Landsat image. The contingent valuation method was used using the forms to estimate the willingness to receive to maintain the forest cover. At FLONA de Pacotuba there is potential to produce NTFPs, with an offer mainly for medicinal and ecological purposes. The use of the soil demonstrates that the FLONA of Pacotuba presents a high percentage of forest cover, however the damping zone is with a high percentage of pasture and use of the soil Anthropic, with the payment for environmental services (PES) an alternative to increase the forest cover in the cushioning zone. The willingness to receive to maintain the forest cover of the owners in the damping zone and in the FLONA was greater than the PES of the state of Espírito Santo, which hinders the adoption of a payment program for environmental services. The PES of Espírito Santo, through the program Reflorestar, associated with the management of NTFPs, mainly medicinal and ecological, would be alternatives for income generation and preservation of the forest cover.
