ABSTRACT

ABSTRACT

ABSTRACT

800 samples of pharyngeal exudate were taken from healthy children and 28 healthy intrafamilial contacts of children with meningitis caused by H. influenzae type b. H. influenzae type be was isolated in 8.7 per cent of nursery children, H. influenzae type e in 0.12 per cent and non typifiable H. influenzae in 5.8 per cent. On intrafamilial contacts, 32 per cent had H. influenzae type b and non typifiable in 3.5 per cent. Percent age of ampicillin resistant H. influenzae type b was 14 per cent. No strains were found resistant to chloramphenicol. S. pneumoniae was isolated in 37 per cent of pharyngeal exudate samples. 25.5 per cent of strains tested were penicillin sensitive; 74.5 per cent has a decrease sensitivity. Based on these results we consider chloramphenicol to the the first drug of choice for treatment of severe H. influenzae infections and penicillin for S. pneumoniae infections recommending in the latter higher doses when dealing with central nervous system infections.

ABSTRACT

The sera of 25 patients with bacteriological diagnosis of typhoid fever were analyzed to identify and measure the antibodies to S. typhi somatic antigens using the enzyme-linked immunosorbent assay technique (ELISA). The results were positive in all the serologic tests, with titers varying from 1:600 to 1:2500. Development and standardization of the technique, results of the intra-assay and inter-assay tests and their correlation to Widal's reaction are described in detail.

ABSTRACT

50 sera from patients with a bacteriological diagnosis of typhoid fever and 325 sera from healthy individuals were studied to quantify antibodies against S. typhi somatic antigen by means of Widal's technique and the enzyme linked inmmunosorbent assay ELISA and by means of the latter, determine the minimum diagnostic titer. Only 2.3 per cent of the healthy population had titers up to 1:300 while sera from patients varied from 1:150 (2 per cent) to 1:2500. A titer was established by ELISA--1:3000 as suggestive of typhoid fever. Widal's agglutination reaction was positive in 1:160 titers in 50 per cent of patients while ELISA technique was positive in 98 per cent of cases (p < 0.001). ELISA technique was more efficient, rapid and sensitive than Widal's test and its utilization is proposed for the clincial laboratory as the method of choice in the serological diagnosis of typhoid fever.

ABSTRACT

In the serum of patients with typhoid fever counterimmunoelectrophoretic techniques were used for the detection of antibodies to Salmonella "O" antigen. Lipopolysacharides (LPS) obtained with phenol and water from Salmonella typhi (antigens 0, 9 and 12) were used. Positive results were obtained in those patients with typhoid fever (20). The lower and higher titration levels were 1:8 and 1:32 respectively; the geometric mean was 1:16. The variation coefficient during the intra assay tests was 0.19, and remained stable throughout the inter-assay tests. Reproducibility, as well as a rapid technique, make this test a valuable tool for the serologic diagnosis of typhoid fever.

ABSTRACT

Two different population groups were studied. In one, 50 patients with a confirmed diagnosis of typhoid fever. Serum determinations were made for the detection of antibodies to S. typhi somatic antigen using Widal technique, surface fixation test and counterimmunoelectrophoresis (CIE). In the other group, 350 healthy subjects were studied to determine the minimum diagnostic titer by means of CIE. It was possible to establish that a 1:16 titer was suggestive of typhoid fever when CIE techniques were used. Surface fixation test showed the highest sensitivity levels. CIE with sensitivity levels similar to those found in Widal's reaction exceeds the other test because of its standardization and greater reproducibility.

ABSTRACT

A microanalytical reversed-phase chromatographic system for the separation of aminoacyl-tRNA's on microbore (0.3cm X 25cm) columns is described. Chromatography of lysyl-tRNA from polyoma virus transformed mouse fibroblasts revealed at least seven identifiable isoacceptors. The system has the advantages of speed, sensitivity and excellent resolution of isoaccepting tRNA's.
