ABSTRACT

Autism Spectrum Disorder (ASD) is characterized by differences in social communication and interaction, as well as areas of focused interests and/or repetitive behaviors. Recent studies have highlighted a higher prevalence of endocrine and reproductive disturbances among females on the autism spectrum, hinting at potential disruptions within the hypothalamus-pituitary-ovary (HPO) axis. This research aims to explore the reproductive health disparities in ASD using an animal model of autism, the C58/J inbred mouse strain, with a focus on reproductive performance and hormonal profiles compared to the C57BL/6J control strain. Our findings revealed that the estrous cycle in C58/J females is disrupted, as evidenced by a lower frequency of complete cycles and a lack of cyclical release of estradiol and progesterone compared to control mice. C58/J females also exhibited poor performance in several reproductive parameters, including reproductive lifespan and fertility index. Furthermore, estrogen receptor alpha content showed a marked decrease in the hypothalamus of C58/J mice. These alterations in the estrous cycle, hormonal imbalances, and reduced reproductive function imply dysregulation in the HPO axis. Additionally, our in-silico study identified a group of genes involved in infertility carrying single-nucleotide polymorphisms (SNPs) in the C58/J strain, which also have human orthologs associated with autism. These findings could offer valuable insights into the molecular underpinnings of neuroendocrine axis disruption and reproductive issues observed in ASD.

ABSTRACT

Background: End-stage renal disease (ESRD) patients experience immune compromise characterized by complex alterations of both innate and adaptive immunity, and results in higher susceptibility to infection and lower response to vaccination. This immune compromise, coupled with greater risk of exposure to infectious disease at hemodialysis (HD) centers, underscores the need for examination of the immune response to the COVID-19 mRNA-based vaccines. Methods: The immune response to the COVID-19 BNT162b2 mRNA vaccine was assessed in 20 HD patients and cohort-matched controls. RNA sequencing of peripheral blood mononuclear cells was performed longitudinally before and after each vaccination dose for a total of six time points per subject. Anti-spike antibody levels were quantified prior to the first vaccination dose (V1D0) and 7 d after the second dose (V2D7) using anti-spike IgG titers and antibody neutralization assays. Anti-spike IgG titers were additionally quantified 6 mo after initial vaccination. Clinical history and lab values in HD patients were obtained to identify predictors of vaccination response. Results: Transcriptomic analyses demonstrated differing time courses of immune responses, with prolonged myeloid cell activity in HD at 1 wk after the first vaccination dose. HD also demonstrated decreased metabolic activity and decreased antigen presentation compared to controls after the second vaccination dose. Anti-spike IgG titers and neutralizing function were substantially elevated in both controls and HD at V2D7, with a small but significant reduction in titers in HD groups (p<0.05). Anti-spike IgG remained elevated above baseline at 6 mo in both subject groups. Anti-spike IgG titers at V2D7 were highly predictive of 6-month titer levels. Transcriptomic biomarkers after the second vaccination dose and clinical biomarkers including ferritin levels were found to be predictive of antibody development. Conclusions: Overall, we demonstrate differing time courses of immune responses to the BTN162b2 mRNA COVID-19 vaccination in maintenance HD subjects comparable to healthy controls and identify transcriptomic and clinical predictors of anti-spike IgG titers in HD. Analyzing vaccination as an in vivo perturbation, our results warrant further characterization of the immune dysregulation of ESRD. Funding: F30HD102093, F30HL151182, T32HL144909, R01HL138628. ABSTRACT

Individuals with autism spectrum disorder (ASD) often exhibit atypical hippocampal anatomy and connectivity throughout their lifespan, potentially linked to alterations in the neurogenic process within the hippocampus. In this study, we performed an in-silico analysis to identify single-nucleotide polymorphisms (SNPs) in genes relevant to adult neurogenesis in the C58/J model of idiopathic autism. We found coding non-synonymous (Cn) SNPs in 33 genes involved in the adult neurogenic process, as well as in 142 genes associated with the signature genetic profile of neural stem cells (NSC) and neural progenitors. Based on the potential alterations in adult neurogenesis predicted by the in-silico analysis, we evaluated the number and distribution of newborn neurons in the dentate gyrus (DG) of young adult C58/J mice. We found a reduced number of newborn cells in the whole DG, a higher proportion of early neuroblasts in the subgranular layer (SGZ), and a lower proportion of neuroblasts with morphological maturation signs in the granule cell layer (GCL) of the DG compared to C57BL/6J mice. The observed changes may be associated with a delay in the maturation trajectory of newborn neurons in the C58/J strain, linked to the Cn SNPs in genes involved in adult hippocampal neurogenesis.

ABSTRACT

Introduction: Vigour at work is characterized by high levels of energy and high desire to make an effort at work. This article is the result of a research carried out with university teachers in Spain whose main objective is to show what type and frequency of physical exercise and diet influence Vigour. Methods: The sample consisted of 121 subjects, 62% of whom were women and 37.2% men. A questionnaire was administered to collect information on sociodemographic data, physical exercise habits, eating habits and Vigour at work. Cross-tabulations of the dimensions of Vigour with sex, age and type of contract offered were performed. Spearman correlations and Correspondence Analysis are also carried out to provide information on the intensity and type of relationships between the Vigour dimensions. Finally, the influence of the frequency of physical exercise and diet on Total Vigour is investigated. Results: The results show that the relationships between the dimensions are very strong (sig = 0.001). In addition, the practice of moderate-high intensity physical exercise and maintaining a good adherence to the Mediterranean Diet is related to high levels of Total Vigour (F = 7.955; sig = 0.006). As for the influence of the sociodemographic variables used, significant differences were only observed in the Physical Strength dimension for sex (X2 = 6.173; p = 0.046) and age (X2 = 9.449; p = 0.051) and, with respect to the type of contract, in Emotional Energy (X2 = 19.487; p < 0.001). Discusión and conclusions: The main conclusions of our study show that practicing physical exercise of medium-high intensity four hours or more per week and a high adherence to the MD is more related to high Vigour levels than just eating well or just practicing exercise. And more studies are needed on the influence of sociodemographic variables on Vigour and its different dimensions.

BACKGROUND AND AIM: There is no consensus regarding the most appropriate treatment of scaphoid nonunion. This systematic review aimed to investigate whether wrist arthroscopy exerts a positive influence on bone union and clinical outcomes. METHODS: We searched the literature on Medline (PubMed), Web of Science, Embase and Scopus databases using the combined keywords "scaphoid" AND "arthroscopy" AND "pseudoarthrosis" OR "nonunion". Eighteen studies were finally included in our review. The quality of the studies was assessed using the Coleman Methodological Score. RESULTS: Our systematic review has shown that arthroscopic management of scaphoid nonunion achieves a high rate of union and satisfactory clinical outcomes with minimal complications. CONCLUSIONS: There is need to perform randomized controlled trials reporting on the use of arthroscopy. In addition, the different pattern of pseudoarthrosis should be better classified to manage the patients who will benefit after the management.

ABSTRACT

The university experience can cause academic stress that, in turn, can lead to comorbidities. Students increasingly face demands and challenges that require a large amount of physical and psychological resources. These circumstances can make them experience physical and mental fatigue, lower their interest in studying, and even lead them to lose control over their academic performance and health. The objective of this work is to determine the relationship between the practice of physical exercise, eating patterns, and academic stress among university students. A questionnaire was administered to 742 students using non-probabilistic sampling. The mean age was 21.24 (DT = 3.8), and 20.1% were men and 79.4% were women. To identify academic stress, the Stress Manifestation Scale of the Students Stress Inventory (SSI) subscale was used; the Mediterranean Diet Score was applied for eating patterns, and the practice of exercise was measured by weekly hours of exercise. The results show that there is a relationship between academic stress and physical exercise, but not with adherence to the Mediterranean Diet. However, there is a relationship between the consumption of "unhealthy" foods exceeding the recommendations for the Spanish population and academic stress. In short, physical activity and diet are variables that are related to psychological well-being. Therefore, they should constitute the backbone of actions designed by university managers to eliminate or reduce stress suffered by students. Finally, the work demonstrates the need to create new scales that consider not only the foods that help alleviate stress but also their portions.

ABSTRACT

The Mediterranean Diet (MedD), which UNESCO recognizes as an Intangible Cultural Heritage, constitutes a healthy eating pattern that helps prevent illness. The aim of this work is to know how well the university community of Almeria (Spain) adheres to MedD as a healthy lifestyle standard. For this purpose, the authors administered a survey to students, teachers, and administrative and service personnel at the University of Almeria. The sample for the survey comprised 610 people. Of whom, 64.7% were women; 23% were Teaching, and Research Staff (PDI); 17.3% were Administration and Services Staff (PAS); and 59.7% were students. The average age was 32 years. Results show an average level of MedD adherence overall in the university community, although 40.9% have a low adherence level. The most representative MedD adherent can be profiled as a young Spanish female, who values sustainability, reads the labels of the products she consumes, exercises regularly, cooks healthy food, and recycles waste. We suggest [to the University authorities] to advertise the benefits of the Mediterranean Diet among the university community and offer menus based on the MedD in the university canteen.

Eating disorders are a growing societal problem, especially among young people. This study aims to determine the individual and social factors that support and perpetuate the risk of eating disorders (ED) and their possible consequences on an individual's athletic and academic performances. The sample consisted of 395 athletes between 12 and 16 years of age (M = 14.07; SD = 1.35), of whom 142 (35.9%) were female, and 253 (64.1%) were male. A questionnaire was administered to collect information on sociodemographic data, body image, use of social networks, social relationships, sports practice, risk of developing ED, and academic and sports performance. In the resulting analysis, an initial cross-tabulation was carried out to observe the body distortion of the respondents as a function of BMI, followed by a linear regression to analyze the factors influencing the risk of suffering from ED. In addition, correlations were made to determine the relationship between the risk of manifesting ED and academic and sports performance. The main results show that 77.7% of the young athletes present a risk of ED (M = 13.3; DT = 3.33) due to a high body image distortion, which becomes the determining factor. In addition, relationships with family and friends have a significant influence on this. On the other hand, behaviors related with eating disorders affect concentration (r = -0.122; p = 0.01) and fatigue (r = -0.376; p < 0.01). For all these reasons, generating and promoting prevention and early detection guidelines during adolescence is necessary.

ABSTRACT

Eating disorders (ED) in adolescents represent a significant problem in today's society, with multiple factors influencing them, such as predisposing factors, precipitating factors and perpetuating factors. OBJECTIVE: This paper aimed to determine the relationships between some of the factors considered to be predisposing and precipitating in terms of the development of ED in adolescents and to relate them to the SCOFF index. PARTICIPANTS: The sample was made up of 264 subjects aged between 15 and 19 (48.8% females and 51.1% males). METHODS: This study was conducted in two phases. In the first study phase, a descriptive analysis of the sample was performed, encompassing the frequencies of the independent variables and dependent variable (ED). In the second phase of study, we created several linear regression models. RESULTS: A total of 11.7% of adolescents are at high risk for ED, and the variables that predict the variability of manifesting the danger of ED are the following: physical self-conception and family relationships. CONCLUSIONS: This work shows the need to approach eating disorders in a multidisciplinary way (biological and social), since this will enable the disease to be better conceptualized and prevention guidelines to be more effective.

ABSTRACT

An Ugi-Zhu three-component reaction (UZ-3CR) coupled in one pot manner to a cascade process (N-acylation/aza Diels-Alder cycloaddition/decarboxylation/dehydration) was performed to synthesize a series of bis-furyl-pyrrolo[3,4-b]pyridin-5-ones in 45 to 82% overall yields using ytterbium triflate as a catalyst, toluene as a solvent, and microwaves as a heat source. The synthesized molecules were evaluated in vitro against human SARS-CoV-2 through a time-of-addition approach, finding that compound 1e, at a concentration of 10.0 µM, exhibited a significant reduction at the initial infection stages, thus showing prophylactic potential. On the other hand, it was found that compound 1d, at the same concentration, was significantly active when applied post-infection, thus exhibiting a therapeutic profile. Moreover, compound 1f showed both, prophylactic and therapeutic activity. Then, to understand interactions between synthesized compounds and the main proteins related to the virus, docking studies were performed on spike-glycoprotein, main-protease, and Nsp3 protein, finding moderate to strong binding energies, matching accurately with the in vitro results. Additionally, a pharmacophore model was computed behind further rational drug design.

Current studies show an increase in the risk of eating disorders in runners. Since it is known that abusive exercise can be both a cause and a consequence of such developments, the main objective of the present study was to examine the risk and possible relationships between negative running addiction (NRA), as measured by the reduced and validated SAS-40 scale, and the tendency to be a compulsive eater (measured by YFAS 2.0), anorexia nervosa (AN), and/or bulimia nervosa (BN) (measured by EAT-40). This study highlights the novelty of researching the level of influence of NRA on each defined eating disorder. METHOD: A total of 167 Spanish-speaking federated runners in cross-country and track running (42% women and 58% men), with an average age of 24 years and an average BMI of 21 kg/m2, responded to an online questionnaire that asked about sociodemographic data and the Spanish versions of the SAS-40, YFAS 2, YFAS 3, and YFAS 4. Through a quantitative methodology using logistic regressions-the coefficient of determination and Pearson's correlation coefficient-we created a sample analysis that related the significant items of the DSM-V to the results of the questionnaires administered, as well as their relationship with the practice of the sport in question and various variables of the environment. RESULTS: The rates of CE, AN, and BN were 65, 11.4, and 16.2%, respectively. The tendency towards CE increased with a lower weight (r = 0.156, p < 0.05), not having been overweight in childhood (r = 0.151, p < 0.05), and being a long-distance runner (r = 0.123 p < 0.05). The risk of AN increased with the absence of menstruation for more than 3 months (r = 0.271 p < 0.01), having suffered from childhood obesity (r = 0.213 p < 0.05), and being underweight (r = 0.064 p < 0.05). The risk of BN increased with having suffered from childhood obesity (r = 0.194 p < 0.05), having a higher weight (r = 0.140, p < 0.05), and practicing athletics, especially the relay modality (r = 0.044 p < 0.05). CONCLUSIONS: A considerable number of runners are at risk of suffering from some type of eating disorder. A significant relationship was observed between long-distance runners and the risk of eating disorders (AN, BN, and CE), and the association is stronger for CE than for AN and BN. Lastly, childhood experiences (such as being obese/a healthy weight) were notorious for increasing the risk of eating disorders. Further studies are needed to research each particular parameter and the relationships between the possible levels of dependence on exercise. LEVEL OF EVIDENCE: Level III, cohort analytic study.

The benefits of emotional intelligence and their alleviating impact on work-related stress among teachers have been gaining salience in this research field. The aim of this study was to analyze the relationship between emotional abilities and the burnout syndrome and whether the affective balance mediates this interplay. Using a structural equation modeling framework with self-report data from 200 schoolteachers aged between 22 to 64 years (M = 44.97, SD = 9.31, 73.50% women), the study design was a cross-sectional one. Results showed significant direct and indirect effects of mood clarity and emotional repair on all four burnout dimensions, while attention did not predict teacher burntout. Positive affect mediated the relationship between emotional intelligence and enthusiasm toward the job, while negative affect mediated the association between emotional abilities and negative burnout symptoms. Our findings suggest that enhanced emotional abilities are directly associated with lower levels of work-related stress, however, negative affect restrains this positive influence. (AU)


La utilidad de la inteligencia emocional y su efecto tranquilizador sobre el estrés laboral de los profesores han ido cobrando importancia en este campo de investigación. El objetivo de este estudio ha sido analizar la relación entre las habilidades emocionales y el síndrome de burnout y si el equilibrio afectivo media esta asociación. Se han utilizado modelos de ecuaciones estructurales con datos de autoinforme de 200 profesores cuya edad estaba comprendida entre los 22 y los 64 años (M = 44.97, SD = 9.31, 73.50% mujeres). Se utilizó un diseño transversal para el estudio. Los resultados mostraron un efecto significativo directo e indirecto de la claridad y de la reparación emocional en las cuatro dimensiones del burnout, mientras que la atención no predijo el burnout de los profesores. El afecto positivo medió la relación entre la inteligencia emocional y el entusiasmo hacia el trabajo, mientras que el afecto negativo medió la asociación entre las habilidades emocionales y los síntomas negativos del burnout. Dichos resultados indican que el desarrollo de las habilidades emocionales está directamente asociado a un nivel más bajo de estrés laboral. Sin embargo, el afecto negativo restringe esta influencia positiva. (AU)

ABSTRACT

Swine enteric viral infections are responsible for substantial economic losses in the pork industry worldwide. Porcine epidemic diarrhea (PEDV) is one of the main causative agents of diarrhea in lactating pigs, and reports of PEDV coinfection with other enteric viruses highlight the importance of viral interactions for disease presentation and outcomes. Using next-generation sequencing (NGS) and sequence analyses from samples taken from piglets with acute diarrhea, we explored the possible interactions between PEDV and other less reported pathogens. PEDV coinfection with porcine kobuvirus (PKV) was detected in 36.4% (27/74) of samples. Full genomes from porcine coronavirus and kobuvirus were obtained, as was a partial porcine sapovirus genome (PSaV). The phylogenetic results show the clustering of these strains corresponding to the geographical relationship. To our knowledge, this is the first full genome and isolation report for porcine kobuvirus in México, as well as the first phylogenetic analysis for porcine sapovirus in the country. The NGS approach provides a better perspective of circulating viruses and other pathogens in affected production units.

ABSTRACT

Adolescents' problematic use of the internet and the risk of sexual online victimization are an increasing concern among families, researchers, professionals and society. This study aimed to analyze the interplay between adolescents' addiction to social networks and internet, body self-esteem and sexual-erotic risk behavior online: sexting, sextortion and grooming. While sexting refers to the voluntary engagement in texting sexual-erotic messages, sextortion and grooming are means of sexual-erotic victimization through the use of the internet. Participants were 1763 adolescents (51% girls), aged 12 to 16 years (M = 14.56; SD = 1.16), from public (n = 1068; 60.60%) and private (n = 695; 39.40%) high schools in the Basque Country (Spain). We carried out structural equation modeling (SEM) using Mplus to assess the mediating effects of body self-esteem in the relationship between addiction to social media and internet and sexual-erotic risk behavior. The results showed that internet addiction predicts online sexual victimization; specifically, the best predictors of sexting, sextortion and grooming victimization were symptoms of internet addiction and geek behavior. Body self-esteem and sexting mediated the relationship between internet addiction and sexual online victimization in adolescents. These results highlight the importance of attending to adolescents' mental health regarding their online behavior, considering the risk and protective factors involved, due to its close association with online sexual victimization.

ABSTRACT

La alta transmisibilidad del SARS-CoV-2 antes y poco después de la aparición de los síntomas sugiere que sólo diagnosticar y aislar a pacientes sintomáticos puede no ser suficiente para interrumpir la propagación de la infección; por ello son también necesarias medidas de salud pública como el distanciamiento social. Adicionalmente será importante detectar a los nuevos infectados que permanecen asintomáticos, que pueden ascender al 50% o más de los casos. Las técnicas moleculares son el patrón de referencia para el diagnóstico de infección por SARS-CoV-2. Sin embargo, el uso masivo de estas técnicas ha generado algunos problemas. Por un lado, la escasez de los recursos (analizadores, fungibles y reactivos), y por otro el retraso en la notificación de resultados. Estos dos hechos se traducen en un retraso en la aplicación de las medidas de aislamiento entre casos y contactos, lo que favorece la expansión de la infección. Las pruebas de detección de antígenos son también métodos de diagnóstico directo, con la ventaja de obtener el resultado en pocos minutos y en el mismo lugar de atención. Además, la sencillez y el bajo coste de estas pruebas permiten repetirlas en días sucesivos en determinados contextos clínicos. La sensibilidad de las pruebas de antígenos es generalmente menor que la de las que detectan ácidos nucleicos, si bien su especificidad es comparable. Se ha comprobado que las pruebas antigénicas tienen más validez en los días alrededor del inicio de síntomas, cuando la carga viral en nasofaringe es mayor. Disponer de un análisis de detección viral rápido y en tiempo real como la prueba de antígenos se ha demostrado más útil para controlar la expansión de la infección que pruebas más sensibles, pero de mayor coste y tiempo de respuesta, como son las pruebas moleculares. Las principales instituciones sanitarias como la OMS, los CDC y el propio Ministerio de Sanidad del Gobierno de España plantean el uso de las pruebas antigénicas en una amplia variedad de estrategias para responder a la pandemia. El presente documento pretende servir de apoyo a los médicos implicados en la atención de pacientes con sospecha de infección por SC2, en el contexto de una incidencia creciente en España desde septiembre de 2020 que representa ya la segunda onda pandémica de COVID-19


The high transmissibility of SARS-CoV-2 before and shortly after the onset of symptoms suggests that only diagnosing and isolating symptomatic patients may not be sufficient to interrupt the spread of infection; therefore, public health measures such as personal distancing are also necessary. Additionally, it will be important to detect the newly infected individuals who remain asymptomatic, which may account for 50% or more of the cases. Molecular techniques are the "gold standard" for the diagnosis of SARS-CoV-2 infection. However, the massive use of these techniques has generated some problems. On the one hand, the scarcity of resources (analyzers, fungibles and reagents), and on the other the delay in the notification of results. These two facts translate into a lag in the application of isolation measures among cases and contacts, which favors the spread of the infection. Antigen detection tests are also direct diagnostic methods, with the advantage of obtaining the result in a few minutes and at the very "pointof-care". Furthermore, the simplicity and low cost of these tests allow them to be repeated on successive days in certain clinical settings. The sensitivity of antigen tests is generally lower than that of nucleic acid tests, although their specificity is comparable. Antigenic tests have been shown to be more valid in the days around the onset of symptoms, when the viral load in the nasopharynx is higher. Having a rapid and real-time viral detection assay such as the antigen test has been shown to be more useful to control the spread of the infection than more sensitive tests, but with greater cost and response time, such as in case of molecular tests. The main health institutions such as the WHO, the CDC and the Ministry of Health of the Government of Spain propose the use of antigenic tests in a wide variety of strategies to respond to the pandemic. This document aims to support physicians involved in the care of patients with suspected SC2 infection, in the context of a growing incidence in Spain since September 2020, which already represents the second pandemic wave of COVID-19

ANTECEDENTES Y OBJETIVO: La anatomía vascular de la mano ya ha sido ampliamente descrita a nivel macroscópico. Sin embargo, existen muy pocos trabajos que estudien el patrón de normalidad de la vascularización in vivo y describan y analicen las arterias principales de la mano. El objetivo de este trabajo es realizar un estudio que sirva de referencia para los valores normales de tamaño y flujo de la arteria radial y ulnar a nivel de la muñeca y de las arterias digitales radiales y ulnares a nivel de los dedos. MATERIAL Y MÉTODO: Estudio descriptivo observacional de corte trasversal sobre 200 manos en 100 voluntarios sanos entre 20-30 años. Se realizó ecografía Doppler-color de las arterias ulnar y radial en la muñeca, así como de las arterias digitales radial y ulnar de cada dedo. Una vez tomadas las medidas se llevó a cabo un análisis comparativo de forma general y también teniendo en cuenta la lateralidad, la dominancia y el género. RESULTADOS: Se observó que existe un mayor tamaño de la arteria radial sobre la ulnar a nivel de la muñeca; sin embargo, es la arteria ulnar la que presentó dominancia de flujo a este nivel. A nivel de los dedos, en los tres primeros fue la arteria digital ulnar la que presentó un mayor tamaño y mayor flujo. No obstante, en el cuarto y quinto dedos fue la arteria digital radial la que presentó un mayor tamaño y dominancia de flujo. CONCLUSIÓN: Ha quedado confirmado que la dominancia de flujo, pero no de tamaño, a nivel de la muñeca es de la arteria ulnar. A nivel de los dedos, existe un mayor tamaño y flujo de las arterias en las zonas de los dedos más protegidas de las lesiones (arteria digital ulnar en los tres primeros y radial en el cuarto y quinto)
