ABSTRACT

The ovicidal and larvicidal effect of a full aqueous extract (FE) and two phases: an aqueous (Aq-Ph) and an ethyl acetate (EtOAc-Ph) from Ruta chalepensis (Rc) stems and leaves against Haemonchus contortus (Hc) were assessed. The egg hatching inhibition (EHI) assay and larval mortality (LM) test were performed by triplicate in 96-well micro-titration plates (n=4 wells). The FE against Hc eggs and larvae was assessed at 1.25, 2.5, 5, 10 and 20 mg/mL; and 30, 60, 90, 120,150 and 200 mg/ mL, respectively. The ovicidal effect of Aq-F and EtOAc-F was assessed at 1.25, 2.5, 5, 10 and 20 mg/mL. Plates were incubated at 28 °C for 48 (ovicidal assay) and 72 h (larvicidal assay). The EHI results were considered based on the mean number of eggs hatching failure after 48 h exposure. The LM was recorded after 72 h exposure to the Rc phases and expressed as mortality percentage. The Rc FE caused 96 and 100% EHI at 10 and 20 mg/mL, respectively; meanwhile, 74% LM was recorded at 200 mg/mL (p<0.05). The Aq-Ph showed 78.5% EHI at 2.5 mg/mL. Likewise, the EtOAc-Ph caused 100% EHI in almost all concentrations. Eighteen compounds including alkaloids, cumarins, triterpens, flavonoids, tannins, saponins and sterols were identified by GC-MS analysis. The results indicate that the aqueous extract from Rc possesses bioactive compounds with in vitro nematocidal activity against Hc; mainly in the EtOAc-Ph. Further studies should be performed to elucidate those compounds searching for alternative methods of control of the sheep haemonchosis.

ABSTRACT

Haemonchus contortus is a nematode parasite that causes anaemia and affects the health of sheep. The mean corpuscular haemoglobin concentration (MCHC) is an excellent indicator to detect anaemia that could help to characterize resistant or susceptible lambs to gastrointestinal nematodes. The aim of this study was to evaluate the predictive value of MCHC in detecting changes in red blood cells and their relation to anaemia in lambs re-infected with H. contortus. An analysis of information was performed using 24 Pelibuey lambs previously infected in grazing, dewormed and experimentally re-infected with H. contortus. At the first haematological sampling (admission) the lambs were classified based on MCHC quartiles (Q). Subsequently, the lambs were housed for 56 days. Blood samples were taken every seven days to determine the haematological parameters using an impedance haematological instrument. Confidence limits were constructed with the records of the lambs that recovered their haematological parameters. Each quartile was analysed as a treatment in a repeated measures design over time. To know the optimal combination of sensitivity and specificity of MCHC to detect anaemia a curve of receiver operating characteristic (ROC) curve and the cut-off values were evaluated. In quartile 4 (Q4), lambs showed the highest faecal egg count (FEC, 764 eggs/g of faeces), mean corpuscular haemoglobin (17.0 pg) and MCHC (54.6 g/dL). This group also presented the lowest RBC values (5.8 × 106/mL), haematocrit (HCT, 18.3%), total plasma protein (5.7 g/dL), and HGB (9.7 g/dL). The optimal point of MCHC with ROC curve was 42.4 (sensitivity 88.2% and specificity 86.5%); the area under the curve was 0.91 (CI 95%, 0.86-0.96). These results are related to the haematological effects caused by H. contortus in susceptible lambs. In conclusion, the highest FEC and lower HCT in Q4 are important elements of the haematological damage caused by H. contortus and could identify susceptible lambs.

ABSTRACT

The objective of this study was to evaluate the reduction in nematode faecal egg count (FEC) in Pelibuey lambs segregated as resistant (RES), susceptible (SUS) and intermediate (INT) to gastrointestinal nematodes. Twenty-nine weaned Pelibuey lambs, aged five months old, free of nematode infection, were used. Nine lambs were RES, six were SUS and 14 were INT lambs. The study consisted of two phases: in Phase 1 the lambs were infected experimentally with Haemonchus contortus. In Phase 2, the lambs were naturally infected by grazing. Faecal and blood samples were taken every week. The packed cell volume and total protein were quantified. The FEC value (FECmax) per lamb was recorded together with a natural reduction in FEC in the two phases. The data were analysed with a model of measures repeated over time. During Phase 1, the RES lambs showed the lowest FEC (1061 ± 1053) compared to the other groups (INT: 2385 ± 1794 eggs per gram of faeces (EPG); and SUS: 3958 ± 3037 EPG). However, in Phase 2 no significant differences (p > 0.05) were observed between the groups of lambs (RES: 275 ± 498 EPG; SUS: 504 ± 1036 EPG; and INT: 603 ± 1061 EPG). At the end of Phase 1, the FEC of RES lambs was naturally reduced by 75.5% in respect to FECmax (p < 0.05), and at the end of Phase 2 the reduction in FEC was 90% in respect to FECmax (p > 0.05); the same behaviour was observed in RES and SUS lambs. It is concluded that the artificial infection in the lambs induced a more rapid immune response in RES than SUS lambs, and all lambs developed high acquired resistance by continuous infection.

ABSTRACT

Linear or nonlinear genetic relationships between productive life and functional productive life at 72 mo, with final score (SCO), stature, strength, dairyness (DAI), teat diameter, rear legs (side view), rump angle, rump width (RUW), fore udder attachment (FUA), rear udder height, rear udder arch, udder depth (UDD), suspensory ligament (SUS), and teat placement, as well as heritabilities and correlations were estimated from multibreed US dairy goat records. Productive life was defined as the total days in production until 72 mo of age (PL72) for goats having the opportunity to express the trait. Functional productive life (FPL72) was analyzed by incorporating first lactation milk yield, fat yield, protein yield, and SCO in the statistical model. Heritabilities and correlations were estimated using linear mixed models with pedigree additive genetic relationships and ASReml software. Nonlinearity of genetic relationships was assessed based on second-degree polynomial (quadratic) regression models, with the breeding values of PL72 or FPL72 as responses and the breeding values for each type trait (linear and quadratic) as predictor variables. Heritability estimates were 0.19, 0.14, 0.18, 0.20, 0.14, 0.07, 0.28, 0.20, 0.15, 0.13, 0.25, 0.18, 0.20, 0.21, 0.21, and 0.32 for PL72, FPL72, SCO, stature, strength, DAI, teat diameter, rear legs, rump angle, RUW, FUA, rear udder height, rear udder arch, UDD, SUS, and teat placement, respectively. The type traits SCO, RUW, and FUA were the most correlated with PL72 and FPL72, so these may be used as selection criteria to increase longevity in dairy goats. An increase in the coefficient of determination >1% for the second degree, compared with that for the linear model for either PL72 or FPL72, was taken as evidence of a nonlinear genetic relationship. Using this criterion, PL72 showed maximum values at intermediate scores in DAI, UDD, and RUW, and maximum values at extreme scores in FUA and SUS, whereas FPL72 showed maximum values at intermediate scores in DAI and UDD, and maximum values at extreme scores in FUA, RUW, and SUS. Selecting for increased SCO, RUW, and FUA will lead to an increase of FPL72 in goats. Consideration of nonlinear relationships between DAI, FUA, RUW, SUS, and UDD may help in the design of more efficient breeding programs for dairy goats using conformation traits.

Objectives of this study were: (a) to compare live weight (LW) and zoometric measures (ZM) of local goats in two locations, (b) to fit the best regression equation for goat LW prediction using ZM. LW, body length (BL), trunk length (TL), withers height (WH), hearth girth (HG), rump width (RW), rump length (RL), head length (HL), head width (HW), and ear length (EL) were measured in 318 Local does in Amatepec and Tejupilco, State of Mexico. Statistical methods included student's "t" tests for comparison of means, and correlation, principal components (PC), and multiple linear regression analyses. To evaluate the goodness of fit for LW prediction models the R(2) value was used as a criterion. Differences (P ≤ 0.05) were found between does of Amatepec and Tejupilco in LW, BL, TL, HG, RL, HL, HW, and EL. In Amatepec, LW was correlated with HG, BL, and HW (P ≤ 0.01), whereas in Tejupilco LW was correlated with HG, BL, TL, and HW (P ≤ 0.01). From the Amatepec measures 5 PC were extracted, and which in a multiple regression analysis explained 83.3 % of the total variance, whereas from Tejupilco 4 PC were extracted, and which in a multiple regression analysis explained 82.4 % of the total variance. The best regression model to predict doe LW in Amatepec included TL, HG, RW, and HW, whereas for Tejupilco the best model included BL, HG, HW, and EL. It is concluded that: (1) Amatepec does surpass those of Tejupilco in LW and most ZM, (2) there are reliable ZM for predicting LW of local does in both locations, HG, and HW being common measures for both populations.

ABSTRACT

Heritabilities and correlations for milk yield (MY), fat yield (FY), protein yield (PY), combined fat and protein yield (FPY), fat percentage (F%), protein percentage (P%), age at first kidding (AFK), interval between the first and second kidding (KI), and real and functional productive life at 72mo (FPL72) of 33,725 US dairy goats, were estimated using animal models. Productive life was defined as the total days in production until 72mo of age (PL72) for goats having the opportunity to express the trait. Functional productive life was obtained by correcting PL72 for MY, FY, PY, and final type score (FS). Six selection indexes were used, including or excluding PL72, with 6 groups of different economic weights, to estimate the responses to selection considering MY, FY, PY, and PL72 as selection criteria. The main criteria that determined the culling of a goat from the herd were low FS, MY, and FY per lactation. Heritability estimates were 0.22, 0.17, 0.37, 0.37, 0.38, 0.39, 0.54, 0.64, 0.09, and 0.16 for PL72, FPL72, MY, FY, PY, FPY, F%, P%, KI, and AFK, respectively. Most genetic correlations between the evaluated traits and PL72 or FPL72 were positive, except for F% (-0.04 and -0.06, respectively), P% (-0.002 and -0.03, respectively), and AFK (-0.03 and -0.01, respectively). The highest genetic correlations were between FPL72 and MY (0.39) and between PL72 and MY (0.33). Most phenotypic correlations between the traits evaluated and FPL72 and PL72 were positive (>0.23 and >0.26, respectively), except for F% (-0.004 and -0.02, respectively), P% (-0.05 and -0.02), KI (-0.01 and -0.07), and AFK (-0.08 and -0.08). The direct selection for PL72 increased it by 102.28d per generation. The use of MY, FY, PY, KI, or AFK as selection criteria increased PL72 by 39.21, 27.33, 35.90, -8.28, or 2.77d per generation, respectively. The inclusion of PL72 as selection criterion increased the expected response per generation from 0.15 to 17.35% in all selection indices studied.

ABSTRACT

The preference of lactating dairy cows for grazed herbage or maize silage (MS), simultaneously offered ad libitum in the field, was examined at two sward heights (SH 4-6 and 8-10cm) and two concentrate levels (CL 0 and 6kg day-1) in a 2x2 factorial arrangement within a completely randomised experimental design. The experiment lasted 35 days and was carried out in spring using 24 multiparous Holstein Friesian cows. On average, the cows proportionately spent more time grazing than eating MS (0.85:0.15) and even though the higher rate of intake (RI) of dry matter (DM) of MS compared with grazed herbage (76 versus 26g DM min-1), the proportion of total DM intake as herbage was higher compared to that of MS (0.56:0.44). The higher crude protein and low fibre content of grazed herbage appeared to have a higher priority of choice than RI, as the cows chose to graze for longer (grazing time 385 min, MS feeding time 67min) despite the lower RI of herbage. The low proportion MS intake indicated that RI was a secondary factor of choice. Concentrate supplementation had a greater depressing effect on herbage intake than on MS intake. These results suggest that the animals reduce the intake of feed with lower RI when the labor associated to eat is decreased. The factors influencing the choice for herbage over maize silage remain unclear.


A preferência de vacas leiteiras em lactação por pasto ou silagem de milho (SM) oferecidas ad libitum simultaneamente no pasto foi avaliada quanto a duas alturas de pastagem (4-6 e 8-10cm) e dois níveis de concentrado (0 e 6kg dia-1), em um arranjo fatorial 2x2 dentro de um delineamento inteiramente ao acaso. O experimento, com duração de 35 dias, foi executado na primavera utilizando 24 vacas multíparas da raça Holandesa. As vacas passaram, em média, proporcionalmente mais tempo pastando do que comendo SM (0,85:0,15) e, mesmo considerando a maior taxa de consumo (TC) de matéria seca (MS) de SM comparada com a da pastagem (76 versus 26g MS min-1), a proporção do total do consumo de MS como pastagem foi mais alta comparada com aquela da SM (0,56:0,44). A proteína crua mais alta e o menor conteúdo de fibra da pastagem pareceram ter prioridade na escolha pelos animais do que a TC, pois as vacas preferiram pastar por mais tempo (tempo de pastejo = 385min, tempo para comer SM = 67min), não obstante ter havido menor TC na pastagem. A baixa proporção de consumo de SM indicou que a TC foi um fator secundário na preferência dos animais. A suplementação concentrada teve maior efeito depressor no consumo de pastagem que o consumo de SM. Estes resultados sugerem que a prioridade dos animais, quando o trabalho associado com o comer é reduzido, é o de diminuir o consumo de alimento com menor TC. Os fatores que influenciaram a preferência por pastagem sobre silagem de milho ainda não são totalmente claros.

ABSTRACT

Twenty hair (Pelibuey x Katadin x BlackBelly) and 20 wool (Rambouillet x Criollo) lambs were fattened in an intensive feeding system to reach 40kg body weight at ten months of age. The yield of lamb sides and back fat (51.9 percent, 2.5mm vs. 47.0 percent, 2.3mm) were greater in wool lambs. Meat pH was not different between phenotypes but meat temperature was higher during the first hours postmortem in wool lamb sides (27, 23, and 20, vs. 24, 20, and 17ºC). Protein content in meat from the neck (braquicephalic and esternocephalic muscles) and loin (Longissimus lumborum muscle) were higher in hair lambs (21.1 and 20.4 vs. 19.7 and 19.3 percent), while moisture of the same muscles was higher in wool lambs (75.6 and 75.9 vs. 73.6 and 74.3 percent). Ash content was similar in both phenotypes, but palmitoleic acid was greater in hair lambs (2.9 and 2.7 vs. 2.7 and 2.5 percent). In fresh meat, pH, meat toughness, and water activity (Wa) of loin did not differ; but water holding capacity (WHC) was higher in hair lambs, and the same response was observed for cooked meat toughness from the neck, and in raw and cooked meat from loin. There were differences in the colour of meat from the neck, having values for L*, b* and tonalities greater in wool lambs, and a* greater for hair lambs. Sensorial parameters were similar in both phenotypes.


Vinte cordeiros deslanados (Pelibuey x Katadin x BlackBelly) e 20 cordeiros lanados (Rambouillet x Criollo) foram mantidos em um sistema intensivo de alimentação até atingirem 40kg, aos 10 meses de idade. O rendimento de carcaça e a espessura da gordura foram maiores nos cordeiros lanados (51,9 por cento, 2,5mm vs. 47,0 por cento, 2,3mm). Não houve diferenças entre os dois grupos de cordeiros para pH, mas a temperatura da carne foi mais alta durante as primeiras horas pós-morte, nas meias-carcaças dos cordeiros lanados (27, 23, 20 vs. 24, 20, 17ºC). O conteúdo proteico da carne do pescoço (músculos braquicefálico e esternocefálico) e do lombo (músculo Longissimus lumborum) foi maior nos cordeiros deslanados (21,1 e 20,4 vs. 19,7 e 19,3 por cento) enquanto o teor de umidade foi maior nas lanados (75,6 e 75,9 vs 73,6 e 74,3 por cento). O conteúdo de cinzas não diferiu entre os dois grupos de cordeiros, mas o ácido palmitoleico foi maior nos cordeiros deslanados (2,9 e 2,7 vs 2,7 e 2,5 por cento). Na carne fresca, o pH, a maciez da carne e a atividade da água (Wa) do lombo não diferiram entre os grupos, mas a capacidade de retenção de água (WHc) foi maior nos cordeiros deslanados, e a mesma resposta foi observada para a maciez da carne cozida do pescoço e do lombo fresco e cozido. Houve diferenças para cor da carne do pescoço com valores de L*, b* e maior tonalidade entre os cordeiros lanados, e maior a* para os deslanados. Os parâmetros sensoriais foram semelhantes nos dois grupos.

This study tested whether providing cows a 4-wk period on pasture would improve gait and change lying behavior. Eighteen groups, each of 4 lactating Holstein cows initially housed in a freestall barn, were assigned to either continued housing in the same freestall barn, or moved to pasture to provide changes in both physical environment and diet. To assess lameness, gait scores (1 to 5) were recorded weekly for 4 wk. Gait improved by an average of 0.22 units/wk for those cows kept on pasture. We also recorded 4 specific gait attributes (head bob, back arch, tracking up, and reluctance to bear weight evenly on all 4 hooves), and found that the latter 2 attributes also improved during the pasture period. Improved gait for cows on pasture was not because of increased lying times. Cows on pasture actually spent less time lying down than cows kept indoors (10.9 vs. 12.3 h/d), although this lying time was spread over a larger number of bouts (15.3 vs. 12.2 bouts). Cows housed on pasture also lost more weight and produced less milk relative to cows in freestalls, likely because of reduced nutrient intake. These results indicate that a period on pasture can be used to help lame cattle recover probably because pasture provides a more comfortable surface upon which cows stand, helping them to recover from hoof and leg injuries.
