ABSTRACT

Background: Parkinson's disease (PD) is the second most common neurodegeneration disease worldwide. Necroptosis, which is a new form of programmed cell death with high relationship with inflammation, plays a vital role in the progression of PD. However, the key necroptosis related genes in PD are not fully elucidated. Purpose: Identification of key necroptosis-related genes in PD. Method: The PD associated datasets and necroptosis related genes were downloaded from the GEO Database and GeneCards platform, respectively. The DEGs associated with necroptosis in PD were obtained by gap analysis, and followed by cluster analysis, enrichment analysis and WGCNA analysis. Moreover, the key necroptosis related genes were generated by PPI network analysis and their relationship by spearman correlation analysis. Immune infiltration analysis was used for explore the immune state of PD brain accompanied with the expression levels of these genes in various types of immune cells. Finally, the gene expression levels of these key necroptosis related genes were validated by an external dataset, blood samples from PD patients and toxin-induced PD cell model using real-time PCR analysis. Result: Twelve key necroptosis-related genes including ASGR2, CCNA1, FGF10, FGF19, HJURP, NTF3, OIP5, RRM2, SLC22A1, SLC28A3, WNT1 and WNT10B were identified by integrated bioinformatics analysis of PD related dataset GSE7621. According to the correlation analysis of these genes, RRM2 and WNT1 were positively and negatively correlated with SLC22A1 respectively, while WNT10B was positively correlated with both OIF5 and FGF19. As the results from immune infiltration analysis, M2 macrophage was the highest population of immune cell in analyzed PD brain samples. Moreover, we found that 3 genes (CCNA1, OIP5 and WNT10B) and 9 genes (ASGR2, FGF10, FGF19, HJURP, NTF3, RRM2, SLC22A1, SLC28A3 and WNT1) were down- and up- regulated in an external dataset GSE20141, respectively. All the mRNA expression levels of these 12 genes were obviously upregulated in 6-OHDA-induced SH-SY5Y cell PD model while CCNA1 and OIP5 were up- and down- regulated, respectively, in peripheral blood lymphocytes of PD patients. Conclusion: Necroptosis and its associated inflammation play fundamental roles in the progression of PD and these identified 12 key genes might be served as new diagnostic markers and therapeutic targets for PD.

This article reviewed the structure and forms of TREM-1, mechanism on regulation of inflammation and relationship with immune-related intestinal diseases, thereby to investigate the role and potential value of TREM-1 on diagnosis, prognosis prediction and treatment target in immune-related intestinal diseases.

ABSTRACT

The effects of VEGFA isoforms on the vascular permeability and structure are still unclear. In this study, we found that VEGFA121 and VEGFA165, 2 isoforms of VEGFA, exerted the opposing effects of antiangiogenesis and proangiogenesis on regulating vascular endothelia cells proliferation and tube formation. The 2 isoforms affected the protein expression of Ras-related protein 1-GTPase-activating protein 1 (Rap1GAP) and thrombospondin 1, 2 important signal molecules of Rap1GAP/thrombospondin 1 signal pathway in primary human umbilical vein endothelial cells by regulating 2 different phosphorylating sites of VEGFR2, Tyr(1175) and Tyr(1214). We also found that VEGFA121 and VEGFA165 regulating angiogenesis was related to their regulating VEGFR2 and Rap1GAP/thrombospondin 1 signal pathway with the technology of RNA intervening the gene expression of VEGFR2 and Rap1GAP. Meanwhile, 2 inhibitors of VEGFR2, cabozantinib malate and ZM 323881 HCl (ZM), were used to investigate the relationship among VEGFA(121 and 165), VEGFR2, and angiogenesis. It was demonstrated that cabozantinib malate blocked VEGFA121 and VEGFA165 binding to VEGFR2 and inhibited angiogenesis by specifically binding to VEGFR2 rather than changing VEGFR2 phosphorylation or regulating the expression of VEGFR2. However, ZM antagonized the effect of VEGFA on angiogenesis by specifically reversing the phosphorylation induced by VEGFA121 and VEGFA165. The experiments in vivo also demonstrated that obvious abnormality of VEGFA121 and VEGFA165 presented in the serum of ulcerative colitis (UC) rats compared with that of the normal rats. ZM could promote the repairation of the injuries of the vessels and tissues of colonic mucosa of UC rats and caused mild inflammation in colonic mucosa of normal rats. On the contrary, cabozantinib malate caused injury of vessels and inflammation in the colonic mucosa of normal rats and aggravated the injuries of the vessels and inflammation in the colonic mucosa of UC rats. Hence, our data indicated that the activation of different phosphorylation sites of VEGFR2 leaded to VEGFA121 and VEGFA165 exerting opposing effects on angiogenesis, and it might be an underlying pathogenesis of UC and a potential target for UC treatment.

ABSTRACT

Objective To observe the clinical effect of brachial plexus anesthesia shoulder manipulation solution combined with individualized treatment in treatment of periarthritis of shoulder.Methods 80 patients with periarthritis of shoulder were randomly divided into control group and observation group,40 cases in each group.The control group was given brachial plexus anesthesia shoulder manipulation solution,and the observation group was given brachial plexus anesthesia shoulder manipulation solution combined with individualized treatment.Joint function score and cure rate were compared before and after treatment in the two groups.Results The joint function score [(93.68 ±3.74)] and cure rate (62.50%) of the observation group were significantly higher than those in the control group[(79.35 ±3.21),35.00%] after 2 months treatment (t =1.57,3.86,χ2 =6.05,all P <0.05). Conclusion Brachial plexus anesthesia shoulder manipulation solution with individualized treatment has obvious effect in treatment of periarthritis of shoulder,and can effectively restore joint function in patients.

ABSTRACT

Hedgehog (Hh) signaling pathway is present in many animals and plays an important role in regulating embryonic development and differentiation. Aberrant activation of Hh signaling contributes to the pathogenesis of many malignancies. Recent studies have shown that dysregulated Hh signaling pathway participates in the tumorigenesis, tumor invasion, and metastasis of hepatocellular carcinoma (HCC). Investigation of the relationship between Hh signaling pathway and HCC will help elucidate the molecular mechanism of pathogenesis of HCC and provide a new insight into the development of novel anticancer therapy and therapeutic target.

ABSTRACT

Objective To explore the correlation between polymorphism of interleukin-28B(IL-28B) rs12979860 T/ C and susceptibility of hepatocellular carcinoma( HCC). Methods All eligible case-control studies published up to 2014-09-30 were identified by searching PubMed,EMBase,CNKI,CBM,VIP and WanFang databases. Two reviews independently identified the literature according to inclusion and exclusion criteria. Meta-analysis was performed using Rev Man 5. 2 and Stata 12. 0 software. Results A total of 6 stud-ies comprising 1 138 cases and 955 controls were finally included. Meta-analysis showed that IL-28B rs12979860 T/ C polymorphism was associated with the susceptibility of HCC. Compared with the genotype CC and CT + CC,genotype TT increased the risk of suffering from HCC(TT vs CC:OR = 2. 26,95% CI:1. 40-3. 64,Z = 3. 33,P = 0. 000 9;TT vs CT + CC:OR = 1. 90,95% CI:1. 23-2. 93,Z = 2. 89,P = 0. 004). In stratification analysis by ethnicity,we observed that the polymorphism of IL-28B rs12979860 T/ C was associat-ed with the susceptibility of HCC among Caucasian populations(TT vs CC:OR = 2. 06,95% CI:1. 22-3. 47, Z = 2. 70,P = 0. 007;TT vs CT + CC:OR = 1. 71,95% CI:1. 07-2. 72,Z = 2. 23,P = 0. 03). Conclusion The polymorphism of IL-28B rs12979860 T/ C is associated with the susceptibility of HCC,genotype TT may increase the susceptibility to HCC.

ABSTRACT

Objective To evaluate the effectiveness of nurse-led discharge planning for hospital inpatients with chronic disease.Methods The Cochrane Library,PubMed,Medline,EMBASE,Chinese Biomedical Literature Database Database(CBM),China National Kmowledge Infrastructure (CNKI),VIP Chinese full text database (VIP) and Wanfang Data were searched for randomized controlled trials assessing nurse-directed discharge planning for hospital inpatients with chronic disease.Two reviewers independently extracted data and assessed risk of bias.Meta-analysis was conducted for the eligible studies by RevMan 5.3.4.Results Seventeen randomized controlled trials and 5 331 participants were included.Meta-analysis demonstrated that,compared to standard care,nurse-led discharge planning were effective in reducing hospital readmission [risk radio (RR)=0.73,95% confidence interval (CI) 0.64-0.84,P<0.01],duration of inpatient readmissions [mean difference (MD)=-2.48,95%CI-3.68--1.29,P<0.01],and all-cause mortality (RR=0.78,95%CI 0.62-0.97,P< 0.05).However,no reduction in the length of stay of the index admission (MD=-0.18,95%CI-0.64-0.27,P > 0.05) was demonstrated.Conclusions Nurse-led discharge planning has a positive impact on several aspects of care for hospital inpatients with chronic disease,including reducing readmission,readmission length of stay,and all-cause mortality.It is worth being popularized.

ABSTRACT

OBJECTIVE@#To filtrate and prove the different microRNAs (miRs) profiles in nasopharyngeal carcinoma.@*METHOD@#Screening the different expressions of miRs between nasopharyngeal carcinoma and the inflammatory tissues by the application of expression profiling of chip high-throughput and large-scale microarray analysis. Then we used RT-QPCR technology to prove the accuracy of screening results.@*RESULT@#There were significant expression differences of miRs between nasopharyngeal carcinoma and the control tissues, 144 human miRs had 2 or more fold the difference ratio. Compared with the inflammatory tissues, we have found that miRs-34b, miRs-449b and miRs-7-1 significantly low expressed in nasopharyngeal carcinoma, yet miRs-125b, miRs-184, miRs-196b, miRs-205 and miRs-24-1 expressed high. The results were consistent with the microarray analysis.@*CONCLUSION@#The difference expressed miRs might be closely related to the process of nasopharyngeal carcinoma, and the research on miRs profiles maybe provide a powerful target basis for early diagnosis and therapy of nasopharyngeal carcinoma.

OBJECTIVE@#To investigate the expression profile and the clinical significance of CDK3 in the tissues of nasopharyngeal carcinoma.@*METHOD@#Immunohistochemistry was utilized to detect the expression of CDK3 in 94 cases of NPC and 40 cases of nasopharyngeal inflammation.@*RESULT@#CDK3 was highly expressed in NPC cell lines. Immunohistochemistry showed that CDK3 was mostly expressed in the cytoplasm. The positive expression rate of NPC was 67% and that of nasopharyngeal inflammation was only 12. 5%. The difference between these two groups was highly statistically significant. The CDK3 expression in NPC was related to the TNM clinical staging of NPC (P < 0.01).@*CONCLUSION@#The expression levels of CDK3 were obviously higher in NPC tissues. The CDK3 expression in NPC was related to the TNM clinical staging of NPC. It suggests that CDK3 expression may play an important role in the occurrence and development of nasopharyngeal carcinoma.

ABSTRACT

Observe and study the morphological changes and effects on yoleperoxidase (MPO) of traditional Chinese medical complex Intestine-clearing Suppository (ICS) in preventing and treating ulcerative colitis (UC).Method.. Divide the experimental rats randomly into 6 groups, i.e., blank, salicylazosulfapyridine (SASP), model, high, middle and low dosage of ICS, except the blank group, makeother rats experimental UC model with albumen immunologic method of rabbit colon mucosa, further divide them randomly into 2 experimental groups, i. e. , prevention and treatment, afterwards at 2nd~21st day, give prevention group the medicine, 22nd~35th day, treatment group, then decapitate them and take the colon tissue, and observe it morphologically under optical microscope, measure active MPO, analyse statastic results. Result: Through general morphological observation ,we found there were serious congestion ,edema, mucosa erosion, many large ulcers in the rats of model group; less serious mucosa erosion and quantity of ulcers in SASP and low-dosage ICS groups: small ulcers, not serious mucosa erosion, low degree of congestion and edema in high-and middle-dosage ICS groups; the scores of inflammatory injury of ICS groups were markedly lower than that of model (P<0. 01), meanwhile, the preventive and reating effectiveness of high-and middle-dosage groups was better than that of SASP (P<0.05). Under the optical microscope, there were large area of mucosa coloboma in the rats of model group, the ulcer penetrated to muscular layer and serous coat; in SASP group and low-dosage ICS group, there were erosive mucosa and inflammatory cell infiltration, granulation on the base; in middle-and high-dosage groups, large part of colon mucosa recovered, granulation formed on the base, ulcers were obviously less than that in model group (P <<0.01); there had marked difference compared with that in SASP group (P<0.05). The MPO active values of high-and middle-dosage groups were markedly lower than that of model group (P < 0.01). Conclusion: Traditional Chinese complex ICS had good prevention and treatment to rats UC, itseffectiveness was positively correlated with dosage and better than that of SASP suppository.
