ABSTRACT
This study aimed to assess the leaf aqueous extract composition of Erythrina crista-galli and the effects of its inclusion on the diet of red-eye tetra (Moenkhausia forestii), concerning enzyme content of digestive, hepatic, and oxidation metabolism and pigmentation. Fish (1.78 ± 0.54 g) were divided into groups: fasting (without feeding), control (commercial feed), and treatment (commercial feed with leaf aqueous extract of E. crista-galli) and feeding apparent satiety for 21 days, in the extracted analysis by mass spectrometry, phenolic compost, and flavonoids. The agitation degree and number of dashes, in all supplemented treatments, were lower than those in the control diet when fed for 7 or 15 days. In the digestive enzymatic activity did not differ (p > 0.05). The hepatoprotective treatment group showed lower alanine aminotransferase (ALT) and higher levels of antioxidant catalase (CAT). The results indicated that the aqueous leaf extract of Erythrina crista-galli assists in function maintenance of the liver; and stimulates CAT in red-eye tetra, suggesting that the identified compounds act on the liver and skin, showing hepatoprotective effects and stimulating tranquility.
ABSTRACT
Live food in the initial life stages of neotropical fish is essential for their development and health; however, it can significantly increase production costs. This study uses complete diets with varying moisture contents (47%, 35%, 24%, and 8%) as a cost-effective alternative in the co-feeding phase of surubim larvae, assessing their influence on development, digestive enzymes, and liver metabolism. In a completely randomized design, 3200 three-day-old Pseudoplatystoma sp. larvae (0.001 g) were distributed evenly among 16 aquariums (20 L), with 200 individuals per aquarium. For the first five days, all larvae were fed Artemia exclusively, after which they were fed experimental diets with varying levels of humidity (47%, 35%, 24%, and 8%) six times a day across four treatments and four replicates. The 21-day feeding trial demonstrated that larvae fed with 24% and 8% moisture diets exhibited increased (p < 0.05) weight gain, final length, and protein efficiency rate. The remaining growth parameters (i.e., specific growth rate and condition factor) did not show significant differences (p > 0.05) among the dietary treatments. Enzymatic analysis revealed that the 47% moisture diet enhanced the amylase and alkaline phosphatase activities, whereas the 24% and 35% moisture diets elevated the lipase and protease activities. The 47% moisture diet also resulted in increased alanine aminotransferase, aspartate aminotransferase, and albumin levels, along with visible hepatic histopathologies in samples, such as visible lipid vacuoles, displacement of the nucleus of the hepatocyte, and increased sinusoid spaces. No significant liver changes were observed in fish fed with other diets. Principal component analysis showed that diets with 8-24% moisture content were the most beneficial during the co-feeding phase of surubim larviculture.

Nile Tilapia (Oreochromis niloticus) management procedures are directly linked to the final quality of the product. The aim of this study was to evaluate the effect of pre-slaughter density and different stunning methods on biochemical, respiratory and muscle injury parameters associated with quality and sensory characteristics of Nile tilapia fillets. Fish with an average weight of 762±105 g were used, first collected called the control group. The experiment was conducted in a 2 × 2 factorial scheme, with two densities (50 and 300 kg of live weight m-3) and two stunning methods thus totaling four treatments, with 15 repetitions per treatment totaling 75 fish sampled. Blood gas analysis, evaluation of biochemical parameters, analysis of meat quality and sensory analysis were carried out. For blood gas, biochemical and enzymatic parameters, the highest values were obtained for the density of 300 kg m-3 and asphyxia method: partial pressures of CO2; glucose and lactate, the highest values presented were 268.98 and 11.33 mg dL-1 respectively. As well as enzymatic activities, Creatinine kinase (CPK); Creatinine kinase isoenzyme (CKMB) showed higher values (768.93 and 1078.98 mg dL-1 respectively) in the higher density and asphyxia method. Conversely, when evaluating the quality parameters, the highest values were observed for lower density and thermonarcosis. High depuration density (300 kg m-3), combined with the asphyxiation stunning method, promotes changes in respiratory dynamics and provides greater stress, less firm fillet texture and greater weight loss due to cooking, as well as changes in creatine kinase (CK) and its CK-MB isoenzyme, demonstrating greater muscle damage. On the other hand, the density of 50 kg m-3 during pre-slaughter, combined with the method of stunning by thermonarcosis, provide a longer period of permanence in pre rigor mortis, which will result in fillets with a better sensory profile.

Mycotoxins, produced by fungi, can contaminate fish food and harm their health. Probiotics enhance immune balance and primarily function in the animal intestine. This study aimed to assess aflatoxin's impact on Piaractus mesopotamicus and explore probiotic-based additive (PBA) benefits in mitigating these effects, focusing on antioxidant activity, biochemical indices, and hepatic histopathology. Two experiments were conducted using P. mesopotamicus fry. The first experimental assay tested various levels of aflatoxin B1 (0.0, 25.0, 50.0, 100.0, 200.0, and 400.0 µg kg-1) over a 10-day period. The second experimental assay examined the efficacy of the probiotic (supplemented at 0.20%) in diets with different levels of aflatoxin B1 (0.0, 25.0, and 400.0 µg kg-1) for 15 days. At the end of each assay, the fish underwent a 24-hour fasting period, and the survival rate was recorded. Six liver specimens from each treatment group were randomly selected for metabolic indicator assays, including superoxide dismutase, catalase, alanine aminotransferase, aspartate aminotransferase, and albumin. Additionally, histopathological analysis was performed on six specimens. The initial study discovered that inclusion rates above 25.0 µg kg-1 resulted in decreased activity of AST (aspartate aminotransferase), ALT (alanine aminotransferase), ALB (albumin), CAT (catalase), and SOD (superoxide dismutase), accompanied by liver histopathological lesions. In the second study, the inclusion of PBA in diets contaminated with AFB1 improved the activity of AST and ALT up to 25.0 µg kg-1 of AFB1, with no histopathological lesions observed. The study demonstrated the hepatoprotective effects of PBA in diets contaminated with AFB1. The enzyme activity and hepatic histopathology were maintained, indicating a reduction in damage caused by high concentrations of AFB1 (400.0 µg kg-1 of AFB1). The adverse effects of AFB1 on biochemical and histopathological parameters were observed from 25.0 µg kg-1 onwards. Notably, PBA supplementation enhanced enzymatic activity at a concentration of 25 µg kg-1 of AFB1 and mitigated the effects at 400.0 µg kg-1 of AFB1. The use of PBAs in pacu diets is highly recommended as they effectively neutralize the toxic effects of AFB1 when added to diets containing 25.0 µg kg-1 AFB1. Dietary inclusion of aflatoxin B1 at a concentration of 25.0 µg kg-1 adversely affects the liver of Piaractus mesopotamicus (Pacu). However, the addition of a probiotic-based additive (PBA) to the diets containing this concentration of aflatoxin neutralized its toxic effects. Therefore, the study recommends the use of PBAs in Pacu diets to mitigate the adverse effects of aflatoxin contamination.

This study aims to investigate the effect of different periods of fasting and re-feeding on compensatory responses in Nile tilapia fingerlings and the frequency of muscle fiber distribution. A total of 108 Nile tilapia fingerlings with initial weight of 1.64 ± 0.41 g and mean initial length of 3.60 ± 0.39 cm were used for 55 days. The fish were distributed in a water recirculation system in a completely randomized design with three treatments and four replications: Control - CO - (fish fed until apparent satiation throughout the experimental period); fasting 10 - J10 - (fish fed to apparent satiation for 15 days, followed by 10 days of fasting and re-feeding to satiation for 30 days); and fasting 15 - J15 - (fish fed to apparent satiation for 15 days, followed by 15 days of fasting and re-feeding to satiation for 25 days). Fish from the J15 treatment showed unsatisfactory results in terms of productive performance (p<0.05), such as lower final weight, apparent feed conversion, protein efficiency ratio, and survival, while fish from the J10 treatment achieved the same results as those animals kept in the CO treatment with the exception of the variables of relative weight gain and feed intake. Furthermore, food restriction directly influenced the growth of muscle fibers with a diameter smaller than 20 µm (p<0.05), and fish from the J15 treatment had the lowest frequency of fibers in this diameter class. Therefore, this study concludes that food restriction in short periods (10 days) and at low temperatures can present a compensatory growth, as they alter the process of hyperplasia and hypertrophy of muscle fibers without affecting the morphology of the fibers; however, 15 days of fasting under low temperatures do not compensate for growth and delays the hypertrophic growth of muscle fibers.(AU)

The objective of this study was to evaluate the effects of jabuticaba (Plinia cauliflora) peel extract on the diets of female blue Betta splendens concerning the level of digestive enzymes, liver metabolism, and antioxidant activity. The sample constituted 150 individuals subdivided into five groups in tripli-cate, totaling 10 fish per 20-liter experimental unit. Commercial diets (40.88% crude protein (CP) and 4374.8 Kcalâ¢kg-1v) were added to hydroalcoholic jabuticaba peel extract (EHJ) at concentrations of 0 (control), 0.5, 1.0, 1.5, and 2.0 gâ¢kg-1. The diets were provided twice a day, until apparent satiety, for 21 days. Fish mortality was not observed during the study. Growth indices did not show any significant differences, apart from feed conversion. The use of the extract promoted an increase in the luminosity of the fish; however, there was no statistical difference in chromaticity a* and b*. A decrease in the activity of SOD on the skin of fish fed with diets of 1.5 and 2 g kg-1 EHJ was observed in comparison to the other diets. There was no change in catalase (CAT) activity among the experimental treatments. It is concluded that the use of 2 gâ¢kg-1 EHJ has an antioxidant effect that reflects the greater luminosity of blue female Betta splendens.(AU)

The aim of this study was to evaluate the chronic salinity tolerance of Carassius auratus and the effects on blood parameters, gill morphology, and survival. In the first test, nine different concentrations (0.0, 0.5, 1.0, 2.5, 5.0, 10, 15, 20, and 25 g L-¹) of NaCl were used with nine repetitions for 96 h. The survival of fish subjected to 15 g L-¹ NaCl was 4 h, and 5 min at a concentration of 25 g L-¹. The mortality of fish with 15g L-¹ NaCl was 100%. Morphological analyses of the gills showed hyperplasia of the coated cells in the interlamellar space and hypersecretion of mucus in fish exposed to 10 g L-¹ of NaCl. At concentrations of 20 and 25 g L-¹, necrosis of the support collagen caused the cells to detach from the lamellar structure mucosa. In the chronic test, two concentrations were used, with four replications containing nine fish in each aquarium for a period of 21 days. Blood samples and gills from the fish were collected, and it was observed that the fish showed a decrease in the concentration of bicarbonate (NaHCO3) in the blood, indicating hypernatremia. Acute exposure of C. auratus to sodium chloride (NaCl) should be at a maximum of 10 g L-¹ of NaCl, after which level there would be a loss in animal performance and/or mortality. Chronic exposure to 5 g L-¹ of NaCl promotes acidemia, ionic imbalance, and pathological changes in the gills; therefore, it is not recommended.(AU)

Symphysodon aequifasciatus is a fish with a disk-shaped body and bright colors, important characteristics of ornamental fish. We evaluated amino acid supplementation strategies to reduce crude protein in the diet for evaluation of performance, the content of digestive enzymes, liver metabolism, and intestinal histopathology. A total of 180 fish were randomly distributed in 12 separate 50 L glass aquariums, consisting of a completely randomized design with four treatments (DC - Control diet with 34.4% crude protein; DL - Control diet plus 1% of lysine; DEAA - Control diet plus 1% free essential amino acids (threonine, phenylalanine, leucine, valine, arginine, and tryptophan); and DHP - Diet with a high level of crude protein 48.4%), three repetitions, lasting 60 days. The use of DL and DEAA diets resulted in higher intestinal villus height and higher zootechnical performance. The use of DL diet increased alkaline phosphatase and digestive amylase activity. The use of DHP diets promotes severe liver changes due to increased activity of Alanine aminotraserase. Therefore, it was possible to observe that the use of amino acids can supply the nutritional need of blue discus. Supplementation of diets with AAs allows the reduction of dietary protein, which is a strategy for feeding management.

This study assesses the respiratory dynamics related to stress parameters and resting time before slaughter, in the quality of surubim (Pseudopatystoma spp.) fillets. A completely randomized design was conducted using five treatments: resting time before slaughter of 0, 2, 4, 8 and 24 hours, with 15 fish sampled per treatment. Time 0 corresponded to the treatment without resting time, where the fish were slaughtered immediately after arriving at the processing plant. The resting time did not affect the electrolyte balance, hemoglobin, plasma, hepatic glycogen, myofibrillar fragmentation index (MFI) and water holding capacity (WHC) of surubins. However, with increased resting time, there was a significant decrease in muscle glycogen and an increase in blood pH and blood bicarbonate levels. Additionally, respiratory parameters showed an increase in pO2 and, consequently, in O2 saturation and a decrease in pCO2.The hematocrit and MCV values of the surubins after 24 hours of resting decreased significantly. In the first hours of resting, the highest values of erythrocytes and CHCM were observed. The lowest level of stress was observed for fish having 24 hours of resting. Fish having longer resting periods (8 and 24 hours) presented fillets with a higher pH (P <0.05) and the rigor mortis establishment time was shorter for the first 2 hours and 24 hours of resting time. There was a linear decrease in fillet lightness and an increase in the intensity of red (CIE a*) color up to 24 hours when resting was increased. In CIE b*, a linear decrease (P <0.05) of the yellow intensity of the fillets was observed as the surubim resting time increased. A resting time of 4 to 8 hours before slaughter is effective in reestablishing homeostasis after transporting surubim, providing fillets with higher quality and a greater length of the pre-rigor mortis period.

Phytotherapy has been used in fish breeding to promote stability in the cultivation environment and in the proflatic management reflecting on animal heath. Erythrina crista-galli has a sedative effect and antioxidant and antimicrobial activity, besides functioning as a natural tranquilizer. In the absence of formulations for use in cultivated fish, therapeutic protocols emerge without studies that reveal their characteristics and actions in the animal organism. Therefore, this study evaluated the effects of acute exposure of Carassius auratus to Erythrina crista-galli extract evaluating the physiological blood parameters. In the acute tolerance assay of Erythrina crista-galli, specimens of Carassius auratus were used, exposed to control concentrations (zero), 50, 100 and 200 mg L-1, with three replications, for 96 hours. Survival of 100% was observed in all treatments. These results reveal that the extract of Erythrina crista-galli does not present toxicity to this species of fish. The plasmatic electrolytic balance showed no changes. The use of up to 100 mg L-1 did not promote changes in gaseous exchanges. The pH and the concentration of bicarbonate and glucose were increasing until the concentration of 100 mg L-1. It is concluded that the use of Erythrina crista-galli for acute exposure of Carassius auratus does not promote mortality, but causes undesirable alterations in blood physiological parameters when doses above 100 mg L-1 are used.(AU)

The aim of this study was to assess the leaf aqueous extract composition of Serjania marginata and the effects of its inclusion on the diet of Nile tilapia (Oreochromis niloticus), with respect to the activity of digestive enzymes and enzymes associated with the metabolism of the liver and intestine and liver histopathology. Fish (initial mean weight, 54.36 ± 17.04 g) were divided into groups: fasting (without feeding), control (commercial feed), and treatment (commercial feed with leaf aqueous extract of Serjania marginata), and in each aquarium, there were five individuals and the fish fed ad libitum for 15 days. Treatment fish had ingested on average 224.3 mg of extract/kg of fish/day. In the extract analysis by mass spectrometry, quercitrin, isoquercitrin, A-type proanthocyanidin trimer, and quinic acid were identified. In the enzymatic activity, fish from the treatment group showed higher level of alkaline phosphatase, while the hepatotoxic markers (AST and ALT) and levels of lipase, amylase, and nonspecific protease did not differ (p > 0.05). In liver histopathological analysis, it was observed that fish from the treatment showed normal structure, while abnormalities were associated with control (fibrosis, loss of cordonal architecture, vacuolated hepatocytes with nucleus displaced to the periphery) and fasting (reduction in hepatocyte size and sinusoidal space). The intestine histopathology evidenced that the extract favored the development of goblet cells and intestinal fold height. The results indicated that the leaf aqueous extract of S. marginata assists in the structural maintenance of the liver and intestine and stimulates intestinal alkaline phosphatase production in Nile tilapia, suggesting that the identified compounds act on the liver and intestine, showing hepatoprotective effects and stimulating intestinal digestion.

It is concluded that the use of Erythrina crista-galli for acute exposure of Carassius auratus does not promote mortality, but causes undesirable alterations in blood physiological parameters when doses above 100 mg L-1 are used.

Jatropha has been highlighted as an oleaginous potential for the production of biofuel. The cake, produced by oil extraction, could be used in animal feed. However, some varieties of jatropha are toxic by limiting their incorporation into animal diets. The objective of this study was to evaluate the hepatotoxicity of diets added with jatropha cake JC (Jatropha curcas) in rats. Thirty-five (35) male Wistar adults rats (Rattus norvegicus) with initial weight of 352.1 ± 26.8 g were used. The animals were fed for 21 days with the diets: control, 10, 25, 40 and 50% JC. In the feeding with 50% JC the animals presented themselves prostrate and with piloerection. Development and survival decreased, since the inclusion of JC in diets increased. In rats submitted to 10 and 25% JC there was an increase of 17.52% in the hepatosomatic index in relation to the control group. Increase of JC in the rat diet promoted an increase in the activity of ALT and AST enzymes. Anatomic-histopathological evaluation demonstrated that, regardless of the levels tested, JC in rat diet causes hypertrophy of the hepatocytes, with a reduction in energy reserves. This study demonstrated that the use of JC resulted in decreased food intake, associated with weight loss due to the clinical pattern of toxicity, demonstrated by biochemical and histopathological changes in the liver. It was concluded that the inclusion of jatropha cake in rat feeding presents high hepatotoxic potential leading to lesions in the liver parenchyma.(AU)

This study aimed to evaluate the effect of diets plus pie of Jatropha (Jatropha curcas l.) in performance and metabolism of male Wistar rats. 24 rats were used, albino males (Rattus norvegicus) in growth phase and initial average weight 109.38 ± 6, 16 g. The animals were weighed and randomly divided into two groups with 12 animals, constituting the treatments: control standard diet and diets containing 4, 281TPM. Food consumption, body weight gain and feed conversion ratio of rats has been deprecated by the inclusion of Jatropha bran. The liver enzymes ALT and AST in rats fed diets increased Jatropha. The values of hemoglobin and hematocrit were within this range considered adequate for rodents. Not been reported differences in the electrolytic balance and respiratory metabolism. It is concluded that the inclusion of Jatropha pie in the feeding of rats depresses growth and development promotes hepatic deleterious changes.(AU)

ABSTRACT: Angelfish (Pterophyllum scalare) is a very important species in Brazil's domestic market, however feeding managements differ from fish breeders to aquarists, so the cost of feed and labor become relevant items when cultivating the species. The objective of this research was to assess feeding frequency and feed deprivation based on growth performance, parasite infestation and cost-benefit in farming this species. The experiment was conducted in a completely randomized design in a 3x2 factorial scheme with 3 daily feeding levels, 4 meals, 2 meals and 1 meal; with and without feed deprivation and two repetitions. Feed deprivation consisted of offering feed 5 days a week only. Feeding twice a day without deprivation and four times a day with or without feed deprivation resulted in higher growth performance than feeding once a day. Monogenean and nematode parasitic loads were not influenced by feeding management. The cost-benefit analysis enabled us to observe that the treatment with the best benefit was the one involving two feedings a day with no deprivation. Thus, considering the parameters mentioned above, we concluded that the two daily feedings with no feed deprivation is the most adequate for farming this specie.
