ABSTRACT

Proprioceptive accuracy (PAc), i.e., the acuity of perception of the state of different parts of the motor system, shows substantial intraindividual differences, and is often considered a general ability. However, it is questionable whether there is an association between accuracies measured with different tests at different body sites. With the participation of 87 young individuals with regular physical activity, PAc with respect to both knee and elbow joints (joint position reproduction) and the flexors of the upper arms (weight discrimination) was measured with multiple indices. Expected and perceived performance was also assessed for each behavioral task. Frequentist and Bayesian analysis largely supported the idea that PAc with respect to various parts of the motor system are unrelated. No dominant-subdominant differences for actual performance were found; however, PAc for the dominant and subdominant limb were associated in many cases. Finally, perceived performance was related to expected but not to actual performance for all three proprioceptive modalities. In conclusion, actual accuracy of perception of the actual state of a part (i.e., joint, muscle) of the motor system cannot be generalized to other parts. Perceived accuracy, dominantly shaped by expectations, is independent from actual accuracy.

ABSTRACT

Empirical studies on yoga have shown that regular practice can have a beneficial effect on risk factors of cardiovascular diseases; also, it can decrease body weight, body fat and increase muscle mass. Positive effects on balance and flexibility were also reported. This study evaluated the impact of a 3-month complex yoga-based program, including physical exercises, education, and social support, with a quasi-randomized design in a middle-aged community sample. The intervention group consisted of 46 participants while the passive control group was 29 participants. The intervention group showed a significant increase in core muscle strength and hamstring flexibility and a decrease in body  fat. No significant changes were found concerning risk factors of cardiovascular diseases, body weight, muscle mass, balancing ability, and dietary habits. A 3-month yoga intervention cannot substantially impact the cardiovascular system and body weight control in middle-aged adults.

ABSTRACT

Genetically distinct groups of Erysiphe necator, the fungus causing grapevine powdery mildew infect grapevine in Europe, yet the processes sustaining stable genetic differences between those groups are less understood. Genotyping of over 2000 field samples from six wine regions in Hungary collected between 2017 and 2019 was conducted to reveal E. necator genotypes and their possible differentiation. The demethylase inhibitor (DMI) fungicide resistance marker A495T was detected in all wine regions, in 16% of the samples. Its occurrence differed significantly among wine regions and grape cultivars, and sampling years, but it did not differ between DMI-treated and untreated fields. Multilocus sequence analyses of field samples and 59 in vitro maintained isolates revealed significant genetic differences among populations from distinct wine regions. We identified 14 E. necator genotypes, of which eight were previously unknown. In contrast to the previous concept of A and B groups, European E. necator populations should be considered genetically more complex. Isolation by geographic distance, growing season, and host variety influence the genetic structuring of E. necator, which should be considered both during diagnoses and when effective treatments are planned.

ABSTRACT

Current approaches to somatic symptom perception conceptualize somatic symptoms partly as somato-visceral or body illusions evoked by an interaction between bottom-up (sensory) and top-down (expectations, attention) processes. Similar processes of multisensory integration are assumed to contribute to the rubber hand illusion (RHI). Findings concerning the strength and direction of associations between these two phenomena, symptom perception and the RHI, are equivocal. Individuals of a non-clinical sample (N = 63; 56% females; Mage = 20.4; SD = 1.6) completed the Patient Health Questionnaire Somatic Symptom Scale (PHQ-15) and participated in an experiment that evoked the RHI. In repeated measures analyses of variance with the PHQ-15 score as covariate, no significant interaction effects between the PHQ-15 score and indicators of the RHI, i.e., proprioceptive drift (F(1,61) < 0.001 p = 0.993, partial η2 < 0.001; BF10 = 0.307), felt body ownership(F(1,59) = 0.043, p = 0.836, partial η2 = 0,001; BF10 = 0.501), and felt body disownership (F(1,59) = 0.148, p = 0.702, partial η2 = 0.002; BF10 = 1.972) were found. Overall, frequentist and Bayesian analysis indicated that the support for a possible association between the PHQ-15 and indicators of the RHI remains inconclusive, i.e., neither the null nor the alternative hypotheses were sufficiently supported. At least in this non-clinical sample, the association between somatic symptom distress and the strength of the RHI appears so weak (perhaps non-existing), that both phenomena (somatic symptom distress and the RHI) appear distinct and largely unrelated.

ABSTRACT

Placebo and nocebo effects could influence the perceived, actual, or both postural stabilities. Therefore, this experiment examined whether postural stability is susceptible to placebo and nocebo effects. Driven by expectations, these cognitions could influence the motor stability of people in physical rehabilitation and those with motion instability. We randomly assigned 78 participants to a placebo, nocebo, or control group. Then, we applied a sham sports cream with positive, negative, or neutral instructions about its impact on balance. Next, we tested postural stability with a modified version of the Modified Clinical Test of Sensory Interaction in Balance, including standard, proprioceptive, visual, and vestibular tests before and after the intervention. Further, we measured expected and perceived performance with visual analog scales and assessed trait anxiety, change in state anxiety, optimism, holistic thinking, persistence, and cooperation with questionnaires. The intervention did not affect actual test performances; similarly, trait and state variables and expectations did not have an impact. Furthermore, the experimental manipulation and trait and state variables did not significantly affect perceived performance. However, the association between expectation and perceived performance was strong (ϱ = 0.627, p < 0.001). These findings suggest that postural stability is not susceptible to placebo and nocebo influences. Still, there is a dissociation between objective and subjective performance, showing that expectations impact perceived but not actual performance, which could fuel motivation in rehabilitation settings.

ABSTRACT

BACKGROUND: Proprioceptive accuracy refers to the individual's ability to perceive proprioceptive information, that is, the information referring to the actual state of the locomotor system, which originates from mechanoreceptors located in various parts of the locomotor system and from tactile receptors located in the skin. Proprioceptive accuracy appears to be an important aspect in the evaluation of sensorimotor functioning; however, no widely accepted standard assessment exists. In this systematic review, our goal was to identify and categorize different methods that are used to assess different aspects of proprioceptive accuracy. METHODS: A literature search was conducted in 5 different databases (PubMed, SPORTDiscus, PsycINFO, ScienceDirect, and SpringerLink). RESULTS: Overall, 1139 scientific papers reporting 1346 methods were included in this review. The methods assess 8 different aspects of proprioception: (a) the perception of joint position, (b) movement and movement extent, (c) trajectory, (d) velocity, and the sense of (e) force, (f) muscle tension, (g) weight, and (h) size. They apply various paradigms of psychophysics (i.e., the method of adjustment, constant stimuli, and limits). CONCLUSION: As the outcomes of different tasks with respect to various body parts show no associations (i.e., proprioceptive accuracy is characterized by site-specificity and method-specificity), the appropriate measurement method for the task needs to be chosen based on theoretical considerations and/or ecological validity.

ABSTRACT

Proprioceptive information makes us able to perceive the position of our joints from an internal point of view. In the certain cases, proprioceptive information has to be stored in short-term memory, for example, during the learning of new motor skills or the assessment of proprioceptive accuracy. However, there are contradictory findings about the modality-specific storage of proprioceptive information in working memory. In this preregistered study, we applied the interference paradigm, assessing proprioceptive memory capacity in the subdominant elbow joint for 35 young individuals in five different experimental conditions: (a) without competing task/interference (baseline condition), (b) with motor interference, (c) with spatial interference, (d) with visual interference, and (e) with verbal interference. Proprioceptive span was lower in the verbal and spatial interference condition than in the baseline condition, whereas no significant differences were found for the motor and visual conditions. These results indicate that individuals use verbal and spatial strategies to encode proprioceptive information in short-term memory, and, in contrast to our expectation, the motor subsystem of working memory is not substantially involved in this process.

ABSTRACT

Activation of thermogenic brown and beige adipocytes is considered as a strategy to improve metabolic control. Here, we identify GPR180 as a receptor regulating brown and beige adipocyte function and whole-body glucose homeostasis, whose expression in humans is associated with improved metabolic control. We demonstrate that GPR180 is not a GPCR but a component of the TGFß signalling pathway and regulates the activity of the TGFß receptor complex through SMAD3 phosphorylation. In addition, using genetic and pharmacological tools, we provide evidence that GPR180 is required to manifest Collagen triple helix repeat containing 1 (CTHRC1) action to regulate brown and beige adipocyte activity and glucose homeostasis. In this work, we show that CTHRC1/GPR180 signalling integrates into the TGFß signalling as an alternative axis to fine-tune and achieve low-grade activation of the pathway to prevent pathophysiological response while contributing to control of glucose and energy metabolism.

ABSTRACT

Directionally opposite to placebo effects are the nocebo effects that negatively impact people's thoughts, feelings, and actions. An important but scarcely studied aspect of everyday functioning is motor performance, in which nocebo effects might impair athletic skills and the much-needed purposeful daily movements and motor actions. The aim of this literature review is to unveil the nocebo effects on motor performance. Searched databases were PubMed, PsycINFO, and SPORTDiscus. Twenty-one articles, reporting 23 studies, met the eligibility criteria for inclusion in the current review. All reports exhibited "some" risk of bias. Of the 23 studies, 14 found a nocebo effect on motor performance, equivocal results emerged from two studies, and negative findings were reported in seven studies. Most (10/12) studies using a between-subjects design have reported a nocebo effect. The mean effect size was 0.60, suggesting a medium-to-large effect of nocebo intervention on motor performance. Based on this review, we conclude that nocebo effects do influence motor performance and can be evoked with negative verbal information. This effect may be more robust than the placebo effect but also depends on the type of motor performance, on the examined sample, and on the nocebo agent. Hence, nocebo effects should be recognized and controlled in empirical research on motor performance, and they should be prevented or extinguished in practical and therapeutic settings. More extensive examination of the nocebo effect on motor performance is warranted, especially using between-subjects research design and a "no agent" control condition.

ABSTRACT

Athletic performance relies on tendons, which enable movement by transferring forces from muscles to the skeleton. Yet, how load-bearing structures in tendons sense and adapt to physical demands is not understood. Here, by performing calcium (Ca2+) imaging in mechanically loaded tendon explants from rats and in primary tendon cells from rats and humans, we show that tenocytes detect mechanical forces through the mechanosensitive ion channel PIEZO1, which senses shear stresses induced by collagen-fibre sliding. Through tenocyte-targeted loss-of-function and gain-of-function experiments in rodents, we show that reduced PIEZO1 activity decreased tendon stiffness and that elevated PIEZO1 mechanosignalling increased tendon stiffness and strength, seemingly through upregulated collagen cross-linking. We also show that humans carrying the PIEZO1 E756del gain-of-function mutation display a 13.2% average increase in normalized jumping height, presumably due to a higher rate of force generation or to the release of a larger amount of stored elastic energy. Further understanding of the PIEZO1-mediated mechanoregulation of tendon stiffness should aid research on musculoskeletal medicine and on sports performance.

ABSTRACT

Among the stimuli to which cells are exposed in vivo, it has been shown that tensile deformations induce specific cellular responses in musculoskeletal, cardiovascular, and stromal tissues. However, the early response of cells to sustained substrate-based stretch has remained elusive because of the short timescale at which it occurs. To measure the tensile mechanical properties of adherent cells immediately after the application of substrate deformations, we have developed a dynamic traction force microscopy method that enables subsecond temporal resolution imaging of transient subcellular events. The system employs a novel, to our knowledge, tracking approach with minimal computational overhead to compensate substrate-based, stretch-induced motion/drift of stretched single cells in real time, allowing capture of biophysical phenomena on multiple channels by fluorescent multichannel imaging on a single camera, thus avoiding the need for beam splitting with the associated loss of light. Using this tool, we have characterized the transient subcellular forces and nuclear deformations of single cells immediately after the application of equibiaxial strain. Our experiments reveal significant differences in the cell relaxation dynamics and in the intracellular propagation of force to the nuclear compartment in cells stretched at different strain rates and exposes the need for time control for the correct interpretation of dynamic cell mechanics experiments.

ABSTRACT

The rubber hand illusion (RHI) is a widely used tool in the study of multisensory integration. It develops as the interaction of temporally consistent visual and tactile input, which can overwrite proprioceptive information. Theoretically, the accuracy of proprioception may influence the proneness to the RHI but this has received little research attention to date. Concerning the role of cardioceptive information, the available empirical evidence is equivocal. The current study aimed to test the impact of proprioceptive and cardioceptive input on the RHI. 60 undergraduate students (32 females) completed sensory tasks assessing proprioceptive accuracy with respect to the angle of the elbow joint, a heartbeat tracking task assessing cardioceptive accuracy (the Schandry-task) and the RHI. We found that those with more consistent joint position judgements (i.e., less variable error) in the proprioceptive task were less prone to the illusion, particularly with respect to disembodiment ratings in the asynchronous condition. Systematic error, indicating a systematic distortion in position judgements influenced the illusion in the synchronous condition. Participants with more proprioceptive bias toward the direction of the rubber hand in the proprioceptive test reported a stronger felt embodiment. The results are in accordance with Bayesian causal inference models of multisensory integration. Cardioceptive accuracy, however, was not associated with the strength of the illusion. We concluded that individual differences in proprioceptive processing impact the RHI, while cardioceptive accuracy is unrelated to it. Theoretical and practical relevance of the findings are discussed.

The Joint Position Reproduction test (JPR), one of the most widely used measurements to estimate proprioceptive accuracy, requires the short term storage of proprioceptive information. It has been suggested that visuospatial sketchpad plays a fundamental role in the memorization of proprioceptive information. The current study aimed to investigate this assumption. To do so, we developed and used a novel JPR protocol to measure the retention capacity with respect to sequences of different positions. Our goal was to develop the original task further to make it comparable with other widely used short-term memory measurements, in which the memory capacity was determined by the number of the items participants retain (memory span). We compared participants' (N=39) performance in this task to that of results of Corsi block-tapping task (capacity of the visuospatial sketchpad) and Digit span task (capacity of the phonological loop). Proprioceptive memory capacity did not correlate either with spatial or verbal memory capacity. The exploratory analysis revealed that proprioceptive span correlated positively with the performance if 5 joint positions had to be retained. Further associations with verbal span for 6 or 7 positions, and spatial span for 5 positions were found. Our findings do not support the idea that visuospatial sketchpad plays a fundamental role in the storage of proprioceptive information. The independence of span measures indicates that proprioceptive information might be stored in a subsystem independent of the visuospatial sketchpad or phonological loop.

ABSTRACT

Interoception in the broader sense refers to the perception of internal states, including the perception of the actual state of the internal organs (visceroception) and the motor system (proprioception). Dimensions of interoception include (1) interoceptive accuracy, i.e., the ability to sense internal changes assessed with behavioral tests, (2) confidence rating with respect to perceived performance in an actual behavioral test, and (3) interoceptive sensibility, i.e., the self-reported generalized ability to perceive body changes. The relationship between dimension of cardioceptive and proprioceptive modalities and their association with affect are scarcely studied. In the present study, undergraduate students (N = 105, 53 males, age: 21.0 ± 1.87 years) filled out questionnaires assessing positive and negative affect (Positive and Negative Affect Schedule), interoceptive sensibility (Body Awareness Questionnaire), and body competence (Body Competence Scale of the Body Consciousness Questionnaire). Following this, they completed a behavioral task assessing cardioceptive accuracy (the mental heartbeat tracking task by Schandry) and two tasks assessing proprioceptive accuracy with respect to the tension of arm flexor muscles (weight discrimination task) and the angular position of the elbow joint (joint position reproduction task). Confidence ratings were measured with visual analog scales after the tasks. With the exception of a weak association between cardioceptive accuracy and the respective confidence rating, no associations between and within modalities were found with respect to various dimensions of interoception. Further, the interoceptive dimensions were not associated with state and trait positive and negative affect and perceived body competence. In summary, interoceptive accuracy scores do not substantially contribute to conscious representations of cardioceptive and proprioceptive ability. Within our data, non-pathological affective states (PANAS) are not associated with the major dimensions of interoception for the cardiac and proprioceptive modalities.

ABSTRACT

Understanding cell-material interactions requires accurate characterization of the substrate mechanics, which are generally measured by indentation-type atomic force microscopy. To facilitate cell-substrate interaction, model extracellular matrix coatings are used although their tensile mechanical properties are generally unknown. In this study, beyond standard compressive stiffness estimation, we performed a novel tensile mechanical characterization of collagen- and fibronectin-micropatterned polyacrylamide hydrogels. We further demonstrate the impact of the protein mat on the tensile stiffness characterization of adherent cells. To our knowledge, our study is the first to uncover direction-dependent mechanical behavior of the protein coatings and to demonstrate that it affects cellular response relative to substrate mechanics.

ABSTRACT

Powdery mildews, ubiquitous obligate biotrophic plant pathogens, are often attacked in the field by mycoparasitic fungi belonging to the genus Ampelomyces. Some Ampelomyces strains are commercialized biocontrol agents of crop pathogenic powdery mildews. Using Agrobacterium tumefaciens-mediated transformation (ATMT), we produced stable Ampelomyces transformants that constitutively expressed green fluorescent protein (GFP) to (i) improve the visualization of the mildew-Ampelomyces interaction and (ii) decipher the environmental fate of Ampelomyces fungi before and after acting as a mycoparasite. Detection of Ampelomyces structures, and especially hyphae, was greatly enhanced when diverse powdery mildew, leaf, and soil samples containing GFP transformants were examined with fluorescence microscopy compared with brightfield and differential interference contrast optics. We showed for the first time, to our knowledge, that Ampelomyces strains can persist up to 21 days on mildew-free host plant surfaces, where they can attack powdery mildew structures as soon as these appear after this period. As saprobes in decomposing, powdery mildew-infected leaves on the ground and also in autoclaved soil, Ampelomyces strains developed new hyphae but did not sporulate. These results indicate that Ampelomyces strains occupy a niche in the phyllosphere where they act primarily as mycoparasites of powdery mildews. Our work has established a framework for a molecular genetic toolbox for the genus Ampelomyces using ATMT.

ABSTRACT

Osteosarcoma is the most frequent primary tumor of bone and is characterized by its high tendency to metastasize in lungs. Although treatment in cases of early diagnosis results in a 5-yr survival rate of nearly 60%, the prognosis for patients with secondary lesions at diagnosis is poor, and their 5-yr survival rate remains below 30%. In the present work, we have used a number of analytical methods to investigate the impact of increased metastatic potential on the biophysical properties and force generation of osteosarcoma cells. With that aim, we used two paired osteosarcoma cell lines, with each one comprising a parental line with low metastatic potential and its experimentally selected, highly metastatic form. Mechanical characterization was performed by means of atomic force microscopy, tensile biaxial deformation, and real-time deformability, and cell traction was measured using two-dimensional and micropost-based traction force microscopy. Our results reveal that the low metastatic osteosarcoma cells display larger spreading sizes and generate higher forces than the experimentally selected, highly malignant variants. In turn, the outcome of cell stiffness measurements strongly depends on the method used and the state of the probed cell, indicating that only a set of phenotyping methods provides the full picture of cell mechanics.

According to the proposition of several theoretical accounts, the perception of the bodily cues, interoceptive accuracy and interoceptive sensibility, has a significant positive impact on subjective well-being. Others assume a negative association; however, empirical evidence is scarce. In this study, 142 young adults completed questionnaires assessing subjective well-being, interoceptive sensibility, and subjective somatic symptoms and participated in measurements of proprioceptive accuracy (reproduction of the angle of the elbow joint), gastric sensitivity (water load test), and heartbeat tracking ability (Schandry task). Subjective well-being showed weak to medium positive associations with interoceptive sensibility and weak negative associations with symptom reports. No associations with measures of interoceptive accuracy were found. Gastric sensitivity as opposed to heartbeat perception and proprioceptive accuracy moderated the association between interoceptive sensibility and well-being. Thus, subjective well-being is associated only with the self-reported (perceived) aspect of interoception but not related to the sensory measures of interoceptive accuracy.

ABSTRACT

Objective: The term interoception refers to the perception of bodily cues. In empirical studies, it is assessed using heartbeat detection or tracking tasks, often with the implicit assumption that cardioception reflects general interoceptive ability. Studies that applied a multichannel approach measured only a limited number of modalities. In the current study, six modalities were assessed to gain a deeper understanding of the relationship between the different sensory channels of interoception. Methods: For 118 university students (53% male) gastric perception (water load test), heartbeat perception (Schandry task), proprioception (elbow joint), ischemic pain (tourniquet technique), balancing ability (one leg stand), and perception of bitter taste were measured. Pair-wise correlation analysis and exploratory factor analyses (principal component analysis (PCA) and maximum likelihood (ML) extraction with oblimin rotation) were then carried out with a three-factor solution to investigate the underlying associations. Results: Correlation analysis only revealed significant associations between variables belonging to the same sensory modality (gastric perception, pain, bitter taste). Similarly, the three factors that consistently emerged in the factor analyses represented the three aforementioned modalities. Discussion: Interoceptive sensitivity assessed by using one channel only cannot be generalized. Interoceptive modalities carrying crucial information for survival are not integrated with other channels.

ABSTRACT

PURPOSE: The purpose of this study is to provide an overview of primary modulation based scatter estimation techniques and evaluate their performance as well as to propose improvements. There have been published two distinct approaches to estimate scatter with primary modulators; however they have not been compared, so far. This work seeks to quantify their performance (focusing on how accurately they can estimate a smooth scatter from primary modulated images with real textures and signal to noise ratios) along with that of proposed improved variants. The influence of the parameters of the described algorithms on the estimation error is also investigated. METHODS: Simulations were carried out using primary components derived from real chest DTS projections. After modulation, artificial scatter distributions were added to these projections, and the difference between the estimated and the artificial scatter distributions was quantified. Two existing and four proposed methods were assessed. The performance of the scatter estimators was examined under noisy and noiseless circumstances and with scatter distributions of different bandwidths. The proposed methods (SSE, SSENP, ADASE) solve the problem of varying effective transmission factors in the case of cell-image based methods, and they include an improved gradient based method (LSGSE) as well, which estimates scatter distribution more accurately. RESULTS: It was found that the gradient based algorithms outperformed the cell-image based algorithms under noiseless conditions, while the cell-image based solutions proved to be insensitive to noise. The proposed improved methods reached considerably lower scatter estimation errors than the existing ones. Modulator cell sampling windows of different sizes and shapes were examined. Although much lower effective transmission factors (higher contrast between the attenuated and unattenuated cells) can be achieved using narrow windows, in our case the best estimation results are provided by windows that cover about half of the pixels of a modulator cell. The effect of detector backscatter on scatter estimation was also studied, and a solution was proposed to incorporate this type of scatter into the primary modulation framework. CONCLUSIONS: Considering all the above results, the authors suggest the use of LSGE for scatter estimation with primary modulation.
