ABSTRACT

Herpesviruses have been reported in several Australian marsupial species, with an overt, sometimes fatal disease described in macropods. Our study identifies a gammaherpesvirus in northern brown bandicoots (Isoodon macrourus) and provides virus prevalence data for bandicoots in southeast Queensland, Australia. Herpesvirus DNA was detected using pan-Herpesviridae family primers in a nested PCR format. Samples from 35 northern brown bandicoots were screened, including whole blood (n=29), oropharyngeal swabs (n=34), urine (n=22), and feces (n=23). Combining all sample types, herpesvirus DNA was detected at a total prevalence of 51% (18/35). Whole blood and oropharyngeal swabs proved to be the optimal samples for detection of this virus, with prevalences of 34% and 38%, respectively. Herpesvirus DNA was detected in 4.5% (1/22) of urine samples and not at all in fecal samples. Detection of herpesvirus was more likely in males than females. Animals were trapped at eight different locations, and at all but one location at least one herpesvirus positive animal was detected. This study indicates a high prevalence of the virus within northern brown bandicoot populations in southeast Queensland. Further research is required to understand the clinical manifestations, if any, of herpesvirus infection in this species and how this may affect populations in the face of stressors such as land clearing and habitat fragmentation.

ABSTRACT

The impetus of this study was the imperative to establish blood biomarker values for clinically healthy mahogany gliders (Petaurus gracilis) in order to monitor the health status of eight captive individuals during their movement to a new facility. The study established ranges for 18 hematologic and 21 biochemical blood biomarkers for healthy individuals in a captive environment. The reported values are consistent with those published for other Australian glider and possum species. No statistically significant differences were found between the sexes, but significant age effects were observed. Specifically, subadult animals reported significantly higher total white cell counts, lymphocyte counts, alkaline phosphatase, creatine kinase, and glucose and chloride levels, compared to adult animals. Although there were no clinically significant changes in blood biomarkers associated with the relocation, many of the hematologic and biochemical biomarkers demonstrated the expected changes associated with the physiological stress of relocation. Specifically, triglycerides, glucose, globulins, creatinine kinase, aspartate transferase (AST), total protein, urea, phosphorous, potassium, sodium, chloride, neutrophils, and hematocrit showed changes with the large environmental change. The majority of the blood biomarkers returned to baseline levels 5 wk postrelocation, with all but one aged animal showing no signs of chronic health derangements following the relocation. The abnormal blood biomarker profiles of two geriatric individuals, one male diagnosed with pericloacal and adrenal gland tumors at the beginning of the study, and one female diagnosed with chronic urinary tract infections and suspected bone marrow disease following the relocation, are presented. The findings of this study inform the health monitoring of native gliders in captivity, rehabilitation, and clinical research scenarios. These findings also provide useful baseline data to aid in the health assessment of captive-bred individuals during their reintroduction into free-living populations.

ABSTRACT

Australia's wildlife is being considerably impacted by introduced mammalian predators such as cats (Felis catus), dogs (Canis lupus familiaris), and foxes (Vulpes vulpes). This is often attributed to native wildlife being naïve to these introduced predators. A systematic review of the literature reveals that native metatherians (body mass range 0.02-25 kg) do not recognise, and show relaxed antipredator behaviours towards, native and some introduced mammalian predators. Native eutherians (all with body mass < 2 kg), however, do appear to recognise and exhibit antipredator behaviours towards both native and introduced predators. Based on our findings, we propose a novel theory, the 'Relaxed Predation Theory'. Our new theory is based on the absence of large mammalian predators leading to reduced predation pressure in Australia during the past 40000-50000 years, and on three key differences between Australian metatherians and eutherians: size, sex, and brains. In light of this Relaxed Predation Theory, we make a number of recommendations for the conservation of Australian wildlife: (i) predator avoidance training of suitable species; (ii) exclusion fencing to exclude some, but not all, predators to facilitate the development of antipredator behaviours; (iii) captive breeding programs to prevent the extinction of some species; and (iv) reintroduction of Australia's larger predators, potentially to compete with and displace introduced predators. A more detailed understanding of the responses of Australian mammals to predators will hopefully contribute to the improved conservation of susceptible species.

ABSTRACT

Cats (Felis catus) are significant predators of mammals, birds, frogs and reptiles and are implicated in mammal species extinctions in Australia. Current controls fail to eradicate entire populations allowing survivors to re-establish. The use of the Mata Hari Judas (MHJ) technique, i.e., inducing prolonged oestrus using hormone implants, can enhance the eradication of remnant animals and would greatly improve conservation efforts. The hypotheses tested were that hormone implants could induce prolonged oestrus in queens (adult female cats), and that prolonging oestrus would result in sustained attractiveness to toms (adult male cats). Queens (n = 14) were randomly allocated to five treatments including a control and four treatments using hormone implants. Queens were observed daily; alone and during indirect contact with a tom for 30 consecutive days. There were significant increases (p < 0.001) in oestrus duration (19 to 27 days) for entire and ovariohysterectomised queens given Compudose100™ implants (1/8 or 1/4 implant). This study shows that it is possible to induce and prolong oestrus in queens using Compudose100™ implants where these queens are attractive to toms. The MHJ queen is a new tool with the potential to enhance the detection and thus the control of feral cats in remnant populations.

ABSTRACT

Commercially available microchip-automated devices for companion animals also have potential application with captive wildlife. To explore this potential, a captive bridled nailtail wallaby was trained to use a SureFlap Microchip Pet Door. Throughout the 62 day study the wallaby's interactions with the door increased in frequency and intensity, culminating in the repeated use of the microchip-automated door. This was the first record of any captive macropod being trained to use a microchip-automated device and demonstrates proof-of-concept that captive macropods can successfully utilize commercially available microchip-automated devices. Further research is recommended to develop methodology and evaluation techniques for training captive macropods while also exploring intra- and inter-species variations in responses.

ABSTRACT

The greatest limitation involved with providing enrichment and evaluating its effects on captive mammals is a lack of available staff time [Hoy et al., 2009]. One method to overcome this is through the use of automated husbandry systems to dispense enrichment to individual animals by using implantable radio frequency microchips. An international multi-institutional survey was conducted to determine whether the personnel involved with management, research, and husbandry of captive mammals would support the development of such automated husbandry systems. The survey was also designed to establish which applications of the system would be considered to be most beneficial. Overall, there was a very high level of interest by survey respondents in adopting automated husbandry systems and all potential applications were considered beneficial to varying degrees. These results, together with the relatively low cost of installation and operation, demonstrate that in the future automated enrichment could be an important and effective means of enhancing captive animal management.

ABSTRACT

Environmental enrichment of captive mammals has been steadily evolving over the past thirty years. For this process to continue, it is first necessary to define current enrichment practices and then identify the factors that limit enhancing the quality and quantity of enrichment, as well as the evaluation of its effectiveness. With the objective of obtaining this information, an international multi-institutional questionnaire survey was conducted with individuals working with zoo-housed mammals. Results of the survey showed that regardless of how important different types of enrichment were perceived to be, if providing them was particularly time-consuming, they were not made available to captive mammals as frequently as those requiring less staff time and effort. The groups of mammals provided with enrichment most frequently received it on average fewer than four times per day, resulting in less than two hours per day spent by each animal care staff member on tasks related to enrichment. The time required for staff to complete other husbandry tasks was the factor most limiting the implementation and evaluation of enrichment. The majority of survey respondents agreed that they would provide more enrichment and carry out more evaluation of enrichment if it was manageable to do so. The results of this study support the need for greater quantity, variety, frequency, and evaluation of enrichment provided to captive mammals housed in zoos without impinging on available staff time.

ABSTRACT

It is becoming increasingly evident that the urocortins (Ucns) and their receptors are involved in the initiation and development of inflammation in the gastrointestinal (GI) tract. There has not been a systematic study of the basal expression of Ucns or their receptors in the GI tract. Here, we examined basal expression of Ucn 2 and its high-affinity receptor, CRF-R2 in the rat GI tract. Ucn 2 mRNA was expressed throughout the small and large intestine. Surprisingly, CRF-R2 mRNA expression was detected in only a subset of GI regions that expressed Ucn 2. Immunohistochemical study showed that both Ucn 2 immuno-reactivity (Ucn 2-IR) and CRF-R2-IR were consistently seen in the neurons of the myenteric plexus and the nerve fibers innervating the circular muscle. By and large, Ucn 2-IR was detected in all layers, including the mucosal and the submucosal layers throughout the GI regions. In contrast, CRF-R2-IR was very low or undetectable in the mucosal layers of all regions examined. The role of Ucn 2 and CRF-R2 was then examined in a rat model of chemically-induced colitis. In the early phase of colitis, Ucn 2 mRNA levels peaked, whereas, in striking contrast, CRF-R2 mRNA expression decreased approximately 2.5-fold below control levels. At the peptide level, Ucn 2-IR was specifically induced in a large population of immune cells that infiltrated the lamina propria and submucosa of the distal colon, whereas CRFR2-IR was detected in only a small fraction of infiltrated immune cells. CRF-R2-IR was dramatically reduced in the neurons of the myenteric plexus. Thus, we show, for the first time, that in the acute phase of inflammation, Ucn 2 levels are increased whereas expression levels of its only identified receptor, CRF-R2, are decreased. This suggests that Ucn 2 exerts its effects only in part via CRF-R2.

ABSTRACT

Calcitonin gene-related peptide (CGRP) mediates neurogenic inflammation and modulates intestinal motility. The CGRP receptor is a heterodimer of calcitonin receptor-like receptor (CLR) and receptor-associated modifying protein 1. We used RNA interference to elucidate the specific role of CLR in colonic motility and inflammation. Intramural injection of double-stranded RNA (dsRNA) against CLR (dsCLR) into the colonic wall at two sites caused the spatial and temporal downregulation of CLR in the colon within 1 day of dsRNA injection. Knockdown of CLR persisted for 7-9 days, and the effect of knockdown spread to approximately 2 cm proximal and distal to the injection sites, whereas control dsRNA injection did not affect CLR expression. Measurement of isometric contractions of isolated colonic muscle segments revealed that in control dsRNA-injected rats, CGRP abrogated contractions entirely and decreased resting muscular tone, whereas in dsCLR-injected rats, CGRP decreased muscle tone but slow-wave contractions of varying amplitude persisted. In trinitrobenzene sulfonic acid-induced colitis, rats with knockdown of CLR displayed a significantly greater degree of edema and necrosis than saline- or control dsRNA-injected rats. Levels of the proinflammatory cytokines TNF-alpha and IL-6 were markedly upregulated by trinitrobenzene sulfonic acid treatment. TNF-alpha mRNA levels were further increased in CLR knockdown rats, whereas levels of IL-6 were unaltered. Thus this study demonstrates that CLR is a functional receptor for CGRP.
