ABSTRACT

To investigate the physicochemical conditions necessary to stably remove antibiotic-resistant bacteria (ARB) via contact with activated sludge (AS), the adhesion of ciprofloxacin (CIP)-resistant and -susceptible Escherichia coli to AS was simulated by contact tests in the laboratory. The CIP-resistant E. coli and susceptible E. coli were removed by a 3 log smaller concentration by a 5 h contact test at maximum. Considering the hydraulic retention time of a reaction tank (∼5 h) and step-feeding operation, we considered the removal rate of E. coli in the current simulated contact test to be in agreement with the actual situation where 1-2 log concentrations of E. coli were reported to be removed from an AS reaction tank. With the increase in the AS concentration and/or dissolved oxygen, the removal rate of E. coli increased. The removal rate of CIP-resistant E. coli was greater than that of susceptible E. coli under all experimental conditions. Although the mechanism by which CIP-resistant E. coli preferably adhered to AS was not clearly understood in detail, finding optimum conditions under which bacteria, including ARB, were efficiently removed by the AS process may be possible.

ABSTRACT

We speculated that stroke volume would be higher and heart rate would be lower when the head and upper trunk were mainly upright in the Fowler's position. We therefore analyzed the effects of three trunk postures in Fowler's position on heart rate, blood pressure and circulatory volume. Heart rate (HR), blood pressure (BP), stroke volume (SV), cardiac output (Q), systemic vascular resistance (SVR), ejection time (ET) and pre-ejection period (PEP) were measured in 10 healthy male volunteers (mean age ± SEM, 20.7 ± 0.5 y; range, 19-23 y) while in three trunk postures in Fowler's position. Stroke volume and Q were measured using impedance cardiography. The three trunk postures were 30° of lower and upper trunk inclination (WT30°), 30° and 60° of lower and upper trunk inclination (UT 60°), respectively and 60° of upper and lower trunk inclination (WT60°). Both SV and ET were significantly higher and HR and PEP were lower at UT60° than at WT60° (p < 0.01) whereas these values did not significantly differ between WT30° and UT60° (p > 0.05). None of Q, SVR and BP significantly differed among the three conditions (p > 0.05). These findings suggested that SV and preload are higher when the upper trunk is upright (UT60°) than when the entire trunk is upright (WT60°) while in Fowler's position. In addition, Q might be maintained without increasing HR through vagal withdrawal when only the upper trunk is upright in healthy young males in Fowler's position.

ABSTRACT

BACKGROUND: We previously identified a 5.9 kDa peptide fragment of fibrinogen α C-chain (FIC 5.9) as a novel biomarker candidate for heavy drinking. In an effort to improve FIC 5.9 measurement for potential use in clinical diagnostics, we combined the ClinProt System and a stable isotope-labeled peptide standard dilution as a simple and reproducible system for measuring FIC 5.9. METHODS: We analyzed 104 serum samples that were obtained from patients with alcohol dependency, from patients with chronic hepatitis C, and from healthy volunteers. Serum FIC 5.9 levels were measured using the ClinProt system with and without a stable isotope-labeled synthetic FIC 5.9 as an internal standard. RESULTS: The within-day and between-day CVs were significantly smaller with stable isotope dilution mass spectrometry (SID-MS) than with conventional MALDI-TOF MS. Of the two different MALDI-TOF MS platforms, we obtained concordant results with SID-MS. Furthermore, only SID-MS detected a small but significant difference between the serum FIC 5.9 levels in the chronic hepatitis C group and the controls. CONCLUSIONS: MALDI-TOF MS with a stable isotope-labeled peptide spike can determine serum FIC 5.9 levels more precisely than conventional MS. This will make inter-laboratory FIC 5.9 comparisons possible.

ABSTRACT

Continuous measurements using proton transfer reaction mass spectrometry (PTR-MS) can be used to describe the production processes of secondary products during ozone induced oxidation of terpenes. Terpenes are emitted from woody building materials, and ozone is generated from ozone air purifiers and copy machines in indoor environments. Carbonyl compounds (CCs) are emitted by human activities such as smoking and drinking alcohol. Moreover, CCs are generated during ozone oxidation of terpenes. Therefore, coexistent CCs should affect the ozone oxidation. This study has focused on the measurement of secondary products during the ozone oxidation of terpenes based on the use of PTR-MS analysis and effects of coexistent CCs on oxidized products. Experiments were performed in a fluoroplastic bag containing α-pinene or limonene as terpenes, ozone and acetaldehyde or formaldehyde as coexistent CCs adjusted to predetermined concentrations. Continuous measurements by PTR-MS were conducted after mixing of terpenes, ozone and CCs, and time changes of volatile organic compounds (VOCs) concentrations were monitored. Results showed that, high-molecular weight intermediates disappeared gradually with elapsed time, though the production of high-molecular weight intermediates was observed at the beginning. This phenomenon suggested that the ozone oxidation of terpenes generated ultrafine particles. Coexistent CCs affected the ozone oxidation of α-pinene more than limonene.
