ABSTRACT

For generations, individuals have been incorporating wild edible fungi into their diets. Precisely identifying mushrooms is essential for harnessing their potential benefits for human use. Specifically, the taxonomical identification of Pleurotus species is known for its complexity. In modern times, it is always necessary to use molecular analysis as a valuable approach for gaining a deeper understanding of the systematics and the delineation of species. Thus, this study confirmed the identity of two wild isolates of Pleurotus djamor (pink oyster mushroom) using the internal transcribed spacer sequences. The amplicons underwent sequencing, and the resulting data was assembled and analyzed. The nucleotide sequences of the two wild isolates blasted against sequences from GenBank database revealed that two wild isolates matched 96.80 and 95.27% P. djamor with accession number KF280324.1 and KT273366.1. Commercially cultivated species took less time for spawn run (17.75 days), primordial initiation (22.25 days) and recoded highest biological efficiency (73.92%) among wild isolates. Nutritional analysis revealed that wild isolates showcase an abundance of nutrients, surpassing commercially cultivated species in terms of moisture, protein, and crude fiber content. This distinctive nutritional profile positions wild isolates as an exceptional superfood, fostering overall health enhancement.

ABSTRACT

This paper evaluated the effect of different substrate disinfection methods viz. hot water treatment, autoclaving and steam pasteurization at 100°C, 121°C, and 65°C, respectively, on yield of Hypsizygus ulmarius and effects of sun drying, oven drying and cabinet drying techniques at 25 ± 2°C, 40 ± 2°C, and 50 ± 2°C, respectively, on nutritional qualities of H. ulmarius. Hot water treated substrate gave higher yield (215.36 ± 1.49 g) and biological efficiency (71.78%) compared with autoclaved (194.45 ± 3.36 g and 64.81%) and steam pasteurized substrate (194.45 ± 3.36 g and 65.28%). The different drying methods used for drying the fruit bodies significantly influenced the nutrient profile of H. ulmarius. Highest moisture (6.95%), fiber (16.94%) and protein content (22.49%) was retained in sun dried fruit bodies whereas ash content (5.52%) was highest for oven dried fruit bodies. Cabinet dried fruit bodies showed significantly higher percentage of fats (1.82%) and carbohydrates (53.58%). In the DPPH assay the cabinet dried fruit bodies showed the highest antioxidant activity followed by sun dried and oven dried fruit bodies with respective IC50 values as 104.21 ± 0.14, 117.17 ± 0.07 and 163.57 ± 0.02. The drying methods proved to be effective in controlling the bacterial growth for a period of two months after which significant bacterial growth was noted.

ABSTRACT

Combination therapy of antibiotics and nanoparticles can be used against multi drug resistant microorganisms. Nanoparticles (NPs) have been reported to show antimicrobial activity. The antimicrobial activities of doped ZnO nanoparticles (ZnO NPs) were studied against fungi, gram-positive and gram-negative bacteria using the standard microdilution method. The interaction between the nanoparticle and the antibiotic was estimated by calculating the fractional inhibitory concentration (FIC index) of the combination through checkerboard assay. Experimental results demonstrated that 10% doped zinc oxide nanoparticles (ZnO NPs) exhibited the maximum antimicrobial effect in contrast with that of the 1% loading and pure ZnO nanoparticles. The enhancement in antimicrobial effect was seen when combined with antibiotic. Synergistic and additive effects were found. No antagonistic effect was found. More synergistic effect was observed when combined with ciprofloxacin than ampicillin. Fungus showed only additive effect. The results are quite in terms with MIC clearly depicting that high doping agent is most suitable for combined therapy. 100% synergistic interaction was observed in higher doping with both ciprofloxacin and ampicillin. This study provides a preliminary report of the synergistic activity of nanoparticles with antibiotics against different pathogenic strains. This provides groundwork for further studies on the combination therapy of nanoparticles with antibiotics.

ABSTRACT

To find a cost-effective alternative substrate, the medicinal mushroom Ganoderma lucidum was grown on sawdusts of sheesham, mango, and poplar. Optimum spawn level was determined by spawning in substrates at various levels (1, 2, 3, and 4%). To determine the effect of supplementation, substrates were supplemented with wheat bran, rice bran and corn flour at different concentrations (10, 20, and 30%). Duration of growth cycle, mushroom yield, and biological efficiency data were recorded. Among substrates, mango sawdust was superior, with 1.5-fold higher yields than poplar sawdust, which was the least suitable. However with respect to fructification, mango sawdust produced the first primordia earlier (21±1 days) compared with the other investigated substrates. 3% spawn level was found to be optimal irrespective of the substrate. Yield and biological efficiency (BE) were maximally enhanced by supplementation with wheat bran, whereas rice bran was the least suitable supplement among those tested. Growth cycle shortened and mushroom yield increased to a maximum at the 20% level of supplements. Mango sawdust in combination with 20% wheat bran, if spawned at the 3% level, resulted in a high yield (BE = 58.57%).

ABSTRACT

The present study investigated the effects of four forestry byproducts (sawdust of oak, mango, khair, and tuni) and three agricultural residues (paddy straw, wheat straw, and soybean waste) along with four supplements (wheat bran, rice bran, corn flour, and gram powder) on growth characteristics (spawn run and primordial formation) and yield of Ganoderma lucidum. There were significant differences (P=0.05) in yield regardless of substrates and supplements used in experimentation. Among substrates, agriculture residues supported better yield and biological efficiency of G. lucidum compared to forestry byproducts irrespective of the supplements. The highest yield (82.5 g) and biological efficiency (27.5%) were recorded from paddy straw supplemented with wheat bran, which invariably resulted in significantly higher yield compared to the unsupplemented check(s) or other supplements used in this study.
