ABSTRACT

Natural history collections are essential to a wide variety of studies in biology because they maintain large collections of specimens and associated data, including genetic material (e.g., tissues) for DNA sequence data, yet they are currently under-funded and collection staff have high workloads. With the advent of aggregate databases and advances in sequencing technologies, there is an increased demand on collection staff for access to tissue samples and associated data. Scientists are rapidly developing large DNA barcode libraries, DNA sequences of specific genes for species across the tree of life, in order to document and conserve biodiversity. In doing so, mistakes are made. For instance, inconsistent taxonomic information is commonly taken from different lending institutions and deposited in data repositories, such as the Barcode of Life Database (BOLD) and GenBank, despite explicit disclaimers regarding the need for taxonomic verification by the lending institutions. Such errors can have profound effects on subsequent research based on these mis-labelled sequences in data repositories. Here, we present the production of a large DNA barcode library of reptiles from the National Museum of Natural History tissue holdings. The library contains 2,758 sequences (2,205 COI and 553 16S) from 2260 specimens (four crocodilians, 37 turtles, and 2,219 lizards, including snakes), representing 583 named species, from 52 countries. In generating this library, we noticed several common mistakes made by scientists depositing DNA barcode data in public repositories (e.g., BOLD and GenBank). Our goal is to raise awareness of these concerns and offer advice to avoid such mistakes in the future to maintain accurate DNA barcode libraries to properly document Earth's biodiversity.

ABSTRACT

Dendrobatid frogs are among the best known anurans in the world, mainly due to their toxicity and associated bright colors. A recently described dendrobatid genus, Andinobates, comprises frogs distributed among the Colombian Andes and Panama. During field work in the Distrito de Donoso, Colón province, Panama, we found a poison frog that we here describe as a new species. The new species belongs to the A. minutus species group and is described herein as Andinobates geminisae sp. nov. This new species differs from all other members of the group by having uniformly orange smooth skin over the entire body and a distinctive male advertisement call. The new species is smaller than other colorful dendrobatids present in the area, such as Oophaga pumilio and O. vicentei. We also provide molecular phylogenetic analyses of mitochondrial DNA sequences of dendrobatids and summarize genetic distances among Andinobates species. Andinobates geminisae occurs in Caribbean versant rainforest on the westernmost edge of the known distribution of A. minutus, and represents the fourth species within this genus in Panama. This is vulnerable to habitat loss and excessive harvesting and requires immediate conservation plans to preserve this species with a restricted geographic range.

ABSTRACT

We describe morphological variation (scalation and coloration) observed among eight individuals of the Panamanian lizard species Anolis casildae. This variation was not observed in the holotype and aids in identification of this recently described species (originally described on the basis of a single, male specimen). This species occurs only in the Reserva Forestal Fortuna (ChiriquÝ Province) and the adjacent Bosque Protector Palo Seco (Bocas del Toro Province) in western Panama. Anolis casildae can be distinguished from all other Panamanian anole species via six features: (1) two enlarged superciliary scales (the first larger than the second); (2) an anterior nasal scale in contact with the rostral scale or separated from the rostral by one scale; (3) 6-8 sublabial scales to the center of the eye; (4) 3-4 scales between the supraobital semicircles; (5) unique coloration (4-6 oblique brown bands interspersed by blue-outlined yellow patches; dewlap is a dirty cream color with broad yellow scale rows irregularly interspersed with smaller emerald green scales) and (6) A. casildae occurs from 1,050 to 1,400 m in the Cordillera Central. We also compare our natural history observations of A. casildae to a similar large anole, A. frenatus, a species which we believe A. casildae to be closely related.
