ABSTRACT

A series of TiO2 - based photocatalysts have been prepared by the incorporation of 10 wt% of various carbon-based nanomaterials as modifying agents to titania. More specifically, commercial TiO2 P25 was modified through a wet impregnation approach with methanol with four different carbon nanostructures: single-walled carbon nanotubes (SWCNTs), partially reduced graphene oxide (prGO), graphite (GI), and graphitic carbon nitride (gCN). Characterization results (XPS and Raman) anticipate the occurrence of important interfacial phenomena, preferentially for samples TiO2/SWCNT and TiO2/prGO, with a binding energy displacement in the Ti 2p contribution of 1.35 eV and 1.54 eV, respectively. These findings could be associated with an improved electron-hole mobility at the carbon/oxide interface. Importantly, these two samples constitute the most promising photocatalysts for Rhodamine B (RhB) photodegradation, with nearly 100% conversion in less than 2 h. These promising results must be associated with intrinsic physicochemical changes at the formed heterojunction structure and the potential dual-role of the composites able to adsorb and degrade RhB simultaneously. Cyclability tests confirm the improved performance of the composites (e.g., TiO2/SWCNT, 100% degradation in 1 h) due to the combined adsorption/degradation ability, although the regeneration after several cycles is not complete due to partial blocking of the inner cavities in the carbon nanotubes by non-reacted RhB. Under these reaction conditions, Rhodamine-B xanthene dye degrades via the de-ethylation route.

ABSTRACT

De novo mutations are known to play a prominent role in sporadic disorders with reduced fitness. We hypothesize that de novo mutations play an important role in severe male infertility and explain a portion of the genetic causes of this understudied disorder. To test this hypothesis, we utilize trio-based exome sequencing in a cohort of 185 infertile males and their unaffected parents. Following a systematic analysis, 29 of 145 rare (MAF < 0.1%) protein-altering de novo mutations are classified as possibly causative of the male infertility phenotype. We observed a significant enrichment of loss-of-function de novo mutations in loss-of-function-intolerant genes (p-value = 1.00 × 10-5) in infertile men compared to controls. Additionally, we detected a significant increase in predicted pathogenic de novo missense mutations affecting missense-intolerant genes (p-value = 5.01 × 10-4) in contrast to predicted benign de novo mutations. One gene we identify, RBM5, is an essential regulator of male germ cell pre-mRNA splicing and has been previously implicated in male infertility in mice. In a follow-up study, 6 rare pathogenic missense mutations affecting this gene are observed in a cohort of 2,506 infertile patients, whilst we find no such mutations in a cohort of 5,784 fertile men (p-value = 0.03). Our results provide evidence for the role of de novo mutations in severe male infertility and point to new candidate genes affecting fertility.

ABSTRACT

Feingold syndrome 1 (FGLDS1) is an autosomal dominant malformation syndrome, characterized by skeletal anomalies, microcephaly, facial dysmorphism, gastrointestinal atresias and learning disabilities. Mutations in the MYCN gene are known to be the cause of this syndrome. Congenital absence of the flexor pollicis longus (CAFPL) tendon is a rare hand anomaly. Most cases are sporadic and no genetic variants have been described associated with this abnormality. We describe here a pedigree combining familial CAFPL tendon as a feature of FGLDS1. Molecular analyses of whole exome sequence data in five affected family members spanning three generations of this family revealed a novel mutation in the MYCN gene (c.1171C>T; p.Arg391Cys). Variants in MYCN have not been published in association with isolated or syndromic CAFPL tendon, nor has this been described as a skeletal feature of Feingold syndrome. This report expands on the clinical and molecular spectrum of MYCN-related disorders and highlights the importance of MYCN protein in normal human thumb and foramen development.

ABSTRACT

This paper describes the relationship between the landscape and the socio-economic and political characteristics of a highly biodiverse Andean region of Colombia, which is now recovering from the socio-ecological impact of protracted armed conflict. We quantify the current spatial relationship between nature and society, and we include legacy effects from the most recent period of armed conflict and its consequences of forced displacement and land use disruption. The procedure followed provides a quantitative model where a minimum number of socio-economic and political variables explain the variation in land cover. The results represent the relationship between land use intensity and the main socio-economic and political indicators, highlighting a close interaction between landscape configuration, socio-economic structure of local populations, coercive conservation and armed conflict. A simulated post-conflict landscape shows a clear transition gradient towards agrarian expansion and intensification, also in systems where naturalness is a relevant feature. The peace process in Colombia offers opportunities for new schemes of land planning and management, including natural resource governance and policy reforms to improve welfare and resilience of local communities. The results allow to define options for future planning given the possible consequences of socio-political legacy effects yet to fully play out across Colombia.

ABSTRACT

In this Letter, we report on the generation of 1060 W average power from an ultrafast thulium-doped fiber chirped pulse amplification system. After compression, the pulse energy of 13.2 µJ with a pulse duration of 265 fs at an 80 MHz pulse repetition rate results in a peak power of 50 MW spectrally centered at 1960 nm. Even though the average heat-load in the fiber core is as high as 98 W/m, we confirm the diffraction-limited beam quality of the compressed output. Furthermore, the evolution of the relative intensity noise with increasing average output power has been measured to verify the absence of transversal mode instabilities. This system represents a new average power record for thulium-doped fiber lasers (1150 W uncompressed) and ultrashort pulse fiber lasers with diffraction-limited beam quality, in general, even considering single-channel ytterbium-doped fiber amplifiers.

ABSTRACT

Socio-ecological systems maintain reciprocal interactions between biophysical and socioeconomic structures. As a result of these interactions key essential services for society emerge. Urban expansion is a direct driver of land change and cause serious shifts in socio-ecological relationships and the associated lifestyles. The framework of rural-urban gradients has proved to be a powerful tool for ecological research about urban influences on ecosystems and on sociological issues related to social welfare. However, to date there has not been an attempt to achieve a classification of municipalities in rural-urban gradients based on socio-ecological interactions. In this paper, we developed a methodological approach that allows identifying and classifying a set of socio-ecological network configurations in the Region of Madrid, a highly dynamic cultural landscape considered one of the European hotspots in urban development. According to their socio-ecological links, the integrated model detects four groups of municipalities, ordered along a rural-urban gradient, characterized by their degree of biophysical and socioeconomic coupling and different indicators of landscape structure and social welfare. We propose the developed model as a useful tool to improve environmental management schemes and land planning from a socio-ecological perspective, especially in territories subject to intense urban transformations and loss of rurality.

ABSTRACT

High-average power laser sources delivering intense few-cycle pulses in wavelength regions beyond the near infrared are promising tools for driving the next generation of high-flux strong-field experiments. In this work, we report on nonlinear pulse compression to 34.4 µJ-, 2.1-cycle pulses with 1.4 GW peak power at a central wavelength of 1.82 µm and an average power of 43 W. This performance level was enabled by the combination of a high-repetition-rate ultrafast thulium-doped fiber laser system and a gas-filled antiresonant hollow-core fiber.

ABSTRACT

We investigate the average power scaling of two diode-pumped Yb-doped fiber amplifiers emitting a diffraction-limited beam. The first fiber under investigation with a core diameter of 30 µm was able to amplify a 10 W narrow linewidth seed laser up to 2.8 kW average output power before the onset of transverse mode instabilities (TMI). A further power scaling was achieved using a second fiber with a smaller core size (23µm), which allowed for a narrow linewidth output power of 3.5 kW limited by stimulated Brillouin scattering (SBS). We mitigated SBS using a spectral broadening mechanism, which allowed us to further increase the output power to 4.3 kW only limited by the available pump power. Up to this power level, a high slope efficiency of 90% with diffraction-limited beam quality and without any sign of TMI or stimulated Raman scattering for a spectral dynamic range of higher than -80 dB was obtained.

ABSTRACT

The combination of high-repetition-rate ultrafast thulium-doped fiber laser systems and gas-based nonlinear pulse compression in waveguides offers promising opportunities for the development of high-performance few-cycle laser sources at 2 µm wavelength. In this Letter, we report on a nonlinear pulse compression stage delivering 252 µJ, sub-50 fs-pulses at 15.4 W of average power. This performance level was enabled by actively mitigating ultrashort pulse propagation effects induced by the presence of water vapor absorptions.

ABSTRACT

Thulium-doped fibers with ultra large mode-field areas offer new opportunities for the power scaling of mid-IR ultrashort-pulse laser sources. Here, we present a laser system delivering a pulse-peak power of 2 GW and a nearly transform-limited pulse duration of 200 fs in combination with 28.7 W of average power. This performance level has been achieved by optimizing the pulse shape, reducing the overlap with atmospheric absorption lines, and incorporating a climate chamber to reduce the humidity of the atmospheric environment.

ABSTRACT

We report on the experimental realization of a compact, fiber-based, ultrashort-pulse laser system in the 2 µm wavelength region delivering 24 fs pulse duration with 24 MW pulse peak power and 24.6 W average power. This performance level has been enabled by the favorable quadratic wavelength-dependence of the self-focusing limit, which has been experimentally verified to be at approximately 24 MW for circular polarization in a solid-core fused-silica fiber operated at a wavelength around 2 µm. The anomalous dispersion in this wavelength region allows for a simultaneous nonlinear spectral broadening and temporal pulse compression. This makes an additional compression stage redundant and facilitates a very simple and power-scalable approach. Simulations that include both the nonlinear pulse evolution and the transverse optical Kerr effect support the experimental results.

ABSTRACT

The efficient coherent combination of two ultrafast Tm-doped fiber amplifiers in the 2-µm wavelength region is demonstrated. The performance of the combined amplifiers is compared to the output characteristics of a single amplifier being limited by the onset of detrimental nonlinear effects. Nearly transform-limited pulses with 830-fs duration, 22-µJ pulse energy, and 25-MW peak power have been achieved with a combining efficiency greater than 90%. Based on this result, it can be expected that 2-µm-ultrafast-fiber-laser systems will enter new performance realms in the near future.

ABSTRACT

Nonlinear pulse compression of ultrashort pulses is an established method for reducing the pulse duration and increasing the pulse peak power beyond the intrinsic limits of a given laser architecture. In this proof-of-principle experiment, we demonstrate nonlinear compression of the pulses emitted by a high-repetition-rate thulium-based fiber CPA system. The initial pulse duration of about 400 fs has been shortened to <70 fs with 19.7 µJ of pulse energy, which corresponds to about 200 MW of pulse peak power.

ABSTRACT

In this Letter, we demonstrate a single-mode continuous-wave fiber laser amplifier emitting 146 W of average output power at a wavelength of 1009 nm. The wavelength and bandwidth of the seed oscillator are defined by a pair of fiber Bragg gratings. The seed is amplified in a two-stage ytterbium-doped rod-type amplifier to 146 W with a high slope efficiency of 64%, showing excellent beam quality and stability throughout the experiment. The ASE suppression is as high as 63 dB.

ABSTRACT

We present an enhanced technique for dispersion-free pulse shortening, which exploits the interplay of different third-order nonlinear effects in a waveguide structure. When exceeding a certain value of the pulse energy coupled into the waveguide, the typical oscillations of self-phase modulation (SPM)-broadened spectra vanish during pulse propagation. Such smoothed spectra ensure a high pulse quality of the spectrally filtered and, therefore, temporally shortened pulses independently of the filtering position. A reduction of the pulse duration from 138 to 24 ps has been achieved while preserving a high temporal quality. To the best of our knowledge, the nonlinear smoothing of SPM-broadened spectra is used in the context of dispersion-free pulse duration reduction for the first time.

BACKGROUND AND OBJECTIVE: Gingival keratinocytes are used in model systems to investigate the interaction between periodontal bacteria and the epithelium in the initial stages of the periodontal disease process. Primary gingival epithelial cells (GECs) have a finite lifespan in culture before they enter senescence and cease to replicate, while epithelial cells immortalized with viral proteins can exhibit chromosomal rearrangements. The aim of this study was to generate a telomerase immortalized human gingival epithelial cell line and compare its in vitro behaviour to that of human GECs. MATERIAL AND METHODS: Human primary GECs were immortalized with a bmi1/hTERT combination to prevent cell cycle triggers of senescence and telomere shortening. The resultant cell-line, telomerase immortalized gingival keratinocytes (TIGKs), were compared to GECs for cell morphology, karyotype, growth and cytokeratin expression, and further characterized for replicative lifespan, expression of toll-like receptors and invasion by P. gingivalis. RESULTS: TIGKs showed morphologies, karyotype, proliferation rates and expression of characteristic cytokeratin proteins comparable to GECs. TIGKs underwent 36 passages without signs of senescence and expressed transcripts for toll-like receptors 1-6, 8 and 9. A subpopulation of cells underwent stratification after extended time in culture. The cytokeratin profiles of TIGK monolayers were consistent with basal cells. When allowed to stratify, cytokeratin profiles of TIGKs were consistent with suprabasal cells of the junctional epithelium. Further, TIGKs were comparable to GECs in previously reported levels and kinetics of invasion by wild-type P. gingivalis and an invasion defective ΔserB mutant. CONCLUSION: Results confirm bmi1/hTERT immortalization of primary GECs generated a robust cell line with similar characteristics to the parental cell type. TIGKs represent a valuable model system for the study of oral bacteria interactions with host gingival cells.

ABSTRACT

The impedance of normal osteoblast function by microorganisms is at least in part responsible for the failure of dental or orthopedic implants. Staphylococcus aureus is a major pathogen of bone, and exhibits high levels of adhesion and invasion of osteoblasts. In this article we show that the commensal oral bacterium Streptococcus gordonii also adheres to and is internalized by osteoblasts. Entry of S. gordonii cells had typical features of phagocytosis, similar to S. aureus, with membrane protrusions characterizing initial uptake, and closure of the osteoblast membrane leading to engulfment. The sensitivities of S. gordonii internalization to inhibitors cytochalasin D, colchicine and monensin indicated uptake through endocytosis, with requirement for actin accumulation. Internalization levels of S. gordonii were enhanced by expression of S. aureus fibronectin-binding protein A (FnBPA) on the S. gordonii cell surface. Lysosomal-associated membrane protein-1 phagosomal membrane marker accumulated with intracellular S. aureus and S. gordonii FnBPA, indicating trafficking of bacteria into the late endosomal/lysosomal compartment. Streptococcus gordonii cells did not survive intracellularly for more than 12 h, unless expressing FnBPA, whereas S. aureus showed extended survival times (>48 h). Both S. aureus and S. gordonii DL-1 elicited a rapid interleukin-8 response by osteoblasts, whereas S. gordonii FnBPA was slower. Only S. aureus elicited an interleukin-6 response. Hence, S. gordonii invades osteoblasts by a mechanism similar to that exhibited by S. aureus, and elicits a proinflammatory response that may promote bone resorption.

ABSTRACT

We present a dispersion-free method for the pulse duration reduction of passively Q-switched microchip laser (MCL) seed sources. This technique comprises two stages: one that carries out the self-phase modulation induced spectral broadening in a waveguide structure and a subsequent spectral filtering stage in order to shorten the pulses in time domain. The setup of a proof-of-principle experiment consists of a fiber-amplified passively Q-switched MCL, a passive single-mode fiber used as nonlinear element in which the spectrum is broadened, and a reflective volume-Bragg-grating acting as bandpass filter. A reduction of the pulse duration from 118 to 32 ps with high temporal quality has been achieved with this setup.

ABSTRACT

We report on the suppression of stimulated Raman scattering (SRS) in a double-clad fiber amplifier using long-period gratings (LPGs). The LPGs, fabricated with a CO(2) laser, achieve SRS suppression by coupling the Stokes wavelength from the active core into the cladding. With only three LPGs inserted into a fiber pulse amplifier, the extractable Raman-free output power was nearly doubled. A numerical simulation of the setup shows good agreement with the experimental results.

ABSTRACT

An efficient and simple approach for converting pulsed near-IR laser radiation into visible and mid-IR light by exploiting degenerate four-wave-mixing in an endlessly single-mode, large-mode-area photonic-crystal fiber is presented. Coupling a 1 MHz, 200 ps, 8 W average power pulsed source emitting at 1064 nm into this fiber results in average powers of 3 W at 673 nm signal wavelength and of 450 mW at 2539 nm idler wavelength, respectively. The excellent pulse energy conversion efficiencies of 35% for the signal and 6% for the idler wavelength are due to the unique combination of characteristics of this type of fiber.
