ABSTRACT

A series of amphiphilic poly(amidoamine) dendrimers (PAMAM, G2-G5) composed of a hydrophilic core and a hydrophobic shell of aniline pentamer (AP) were synthesized and characterized. The modified dendrimers self-assembled to vesicular aggregates in water with the critical aggregation concentration (CAC) decreased in the order of G2 > G3 > G4 > G5. It was found that the modified dendrimers self-organized into spherical aggregates with a bilayer vesicular structures and that the dendrimers in higher generation have more order structure, which may be attributed to the crystallization induced by the compacted effect of AP segments. In addition, larger spherical vesicles were observed under acidic and alkaline conditions, as compared with sizes of aggregates in neutral medium. At low pH, the tertiary amine groups of PAMAM-AP were transformed to ammonium salts; the polarons were formed from AP units by doping with strong acids, thereby leading to the stability of vesicular aggregates being better than that in double distilled water. Nevertheless, in high pH environment, the deprotonation of PAMAM-AP caused the enhancement of π-π interactions, resulting in generation of twins or multilayered vesicles.

Color schemes: A mechanochromic material composed of two types of peptides bearing a pyrene group and rhodamine B moieties, respectively, displays multiluminescent colors, such as blue, green, and reddish in one sample (see picture). The mechanochromic behavior is based on a combined switching of molecular assemblies and chemical structure.

ABSTRACT

A smart luminescent material whose emission color and emission intensity can be separately modulated by external force is demonstrated. The rational manipulation of rich noncovalent interactions and fluorophore packing style promotes an in-depth understanding between supramolecular structure and photophysical property and offers an effective strategy to modulate the light-emitting property in a predicative way.

ABSTRACT

There is a delicate balance for a low-weight molecule to behave as a gelator or crystal. The synthesis of two novel amino acid-based naphthalene-dendrons, Nap-G1 and Nap-G2 is described. Both dendrons display polymorphic properties in organic solvents. Nap-G1 developed a fibrous network with ß-sheet architecture in cyclohexane but exhibited a spherulitic network in mixed solvents (chloroform/petroleum ether 1:5, v/v). On the other hand, Nap-G2 acted as an efficient organogelator in chloroform but formed crystalline fibers in relatively high polarity solvents (such as acetone and methanol). Combinations of characterizations have been employed to study the polymorphism.

ABSTRACT

A dual-targeting drug carrier (PAMAM-PEG-WGA-Tf) based on the PEGylated fourth generation (G = 4.0) PAMAM dendrimer with transferrin (Tf) and wheat germ agglutinin (WGA) on the periphery and doxorubicin (DOX) loaded in the interior was synthesized and its BBB penetration and tumor targeting properties were explored. DLS and TEM measurements revealed the size of PAMAM-PEG-WGA-Tf was in the range of 14-20 nm. It reduced the cytotoxicity of DOX to the normal cells greatly, while efficiently inhibited the growth rate of the C6 glioma cells. The assay of transport across the BBB showed that PAMAM-PEG-WGA-Tf delivered 13.5% of DOX in a period of 2 h, demonstrating an enhanced transport ratio as compared to the ratio of 8% for PAMAM-PEG-WGA, 7% for PAMAM-PEG-Tf and 5% for free DOX in the same period of time. The accumulation of DOX in the tumor site was increased due to the targeting effects of both Tf and WGA, leading to the complete breakage of the avascular C6 glioma spheroids in vitro.

ABSTRACT

Spherical co-assemblies were generated from an equivalent-molar-ratio mixture of amphiphilic poly(amidoamine) dendrons that were modified with focal carbazole and/or pyrene chromophores from the first to the third generation (DnCz I and DnPy, where n=1, 2, 3 denotes the generation of dendrons) in water at a concentration of 3.0 x 10(-6) M, lower than the individual critical aggregation concentrations (CAC) of either DnCz I or DnPy. UV-vis absorption and fluorescence emission spectra indicated that fluorescence resonance energy transfer (FRET) from carbazole to pyrene units occurred efficiently within the co-assemblies. Further study implied that part of the carbazole and/or pyrene units stacked homogeneously in the co-assemblies via pi-pi stacking and the dendritic branches served as connectors due to the hydrogen-bonding interaction.

ABSTRACT

Three novel series of amphiphiles based on poly(amidoamine) dendrons (from G1 to G3) and having different aromatic chromophores (Cz I, Cz II, and Py) at the focal point were synthesized and studied for their self-assembly behavior in aqueous solution by using electronic microscopies (i.e., SEM and TEM), UV-vis, fluorescence, IR, and (1)H NMR spectroscopy. It was found that the generation of dendrons affected significantly the self-assembly of these amphiphiles in aqueous solution and the morphological structures of the resulting assemblies depended greatly on the architecture of the focal chromophores. As a result, the first generation of dendrons assembled readily into vesicles at low concentrations. These vesicular structures subsequently fused to form a stable tubular structure. Similar tubular structures could also be directly obtained through self-assembly of these amphiphilic dendrons at high concentrations. X-ray investigations showed that the resulting tubules possessed a lamellar structure. A head-to-head packing model of amphiphilic dendrons in the assemblies was proposed.

ABSTRACT

OBJECTIVE: To evaluate the possibility of self-assembly oligopeptide(T2) for dental enamel biomimetics, especially for the prism's crystal texture since it could prompt calcium phosphate precipitated in gel carrier. METHODS: SEM (Scanning electron microscope) and TEM (Transmission electron microscope) were used to observe the morphologic presentation and ED(Electron diffraction) to crystal texture comparing with the human molar enamel powder. RESULTS: (a) Flake-like and needle-like octacalcium phosphate precipitated in the gel carrier with self-assemble oligopeptide(T2). They transformed into rod-like hydroxyapatite crystals gradually in the following 2-4 weeks. (b) The rod-like hydroxyapatite may arrange or grow into bundles which are similar to the human enamel prisms in both appearance and size. (c) The rod-like hydroxyapatite showed polycrystal while the enamel prisms showed monocrystal under examination of ED. CONCLUSION: The self-assemble oligopeptide(T2) could regulate the speed of nucleation and crystallization of hydroxyapatite in morphology and crystalline size. Thus, the self-assembly oligopeptide and the gel carrier mineralization system could be primarily applied in biomimetic use for the crystallization of hydroxyapatite in dental prism in vitro.

ABSTRACT

PAMAM dendrimers of the zeroth to fifth generation (G0-5) have been peripherally modified with phenyl, naphthyl, pyrenyl, and dansyl chromophores. Their fluorescence behaviors are strongly affected by the dendritic architectures at different generations. These dendrimers modified with hydrophobic chromphores can self-organize into vesicular aggregates at the low generations G0-3 in water. The size and aggregation number of these vesicles decrease with increasing generation from G0 to G3. Critical aggregation concentration determined by fluorescence spectroscopy reveals that these aggregates can be favorably formed in the order of G3 > G2 > G1. In contrast to the vesicles made from traditional amphiphilic compounds, these dendrimer-based vesicles are very adhesive due to the H-bonding interaction and entanglement of dendritic branches located in the outer layer. A large number of multivesicle assemblies, i.e., "twins" and "quins" consisting of two and five vesicles, were clearly identifiable with transmission electron (TEM) and atomic force microscopy. For the dendrimers with peripheral pyrenyl chromophores, triangle-like vesicles were observed in water. The hydrophobic interphase thickness of the vesicular bilayer is ca. 2.0-3.2 nm determined by fluorescence resonance energy transfer methods, which agrees well with the thickness directly observed with TEM.
