ABSTRACT

Objective To observe the clinical efficacy of umbilical compress with anti-cancer Xiaogu formula in managing malignant ascites. Methods A total of 56 patients with malignant ascites who met the inclusion criteria were randomized into 28 patients in the treatment group and 28 patients in the control group. The control group was treated with one type of diuretic, and the treatment group was given umbilical compress using Anti-Cancer Xiaogu formula and diuretics. Both groups were treated for 14 days. The changes of abdominal girth, 24 h urine volume, ascites efficacy, TCM syndrome scores, quality of life and adverse reactions were observed. Results The decrease in the maximum depth difference of the ascites in the treatment group was significantly greater than that of the control group (1.16 ± 1.29 vs. 0.00 ± 1.34, Z=-2.553). The difference was statistically significant (P<0.05). The decrease in abdominal girth in the treatment group was significantly larger than that in the control group (0.57 ± 0.55 vs. 2.61 ± 0.28, Z=-2.264). The difference was statistically significant (P<0.05). The in 24-hour urine volume in the treatment group after intervention (-800.18 ± 64.12 vs.-683.57 ± 55.38, Z=-1.770) was no statistically significant (P>0.05). The response rate in the treatment group was 92.9% (26/28), while that of the control group was 89.3% (25/28).treatment group was 71.4%, while that of the control group was 35.7%. The difference was statistically significant (P<0.05). The increase in KPS in the treatment group was significantly higher than that of the control group. The difference was statistically significant (P<0.05). Conclusions Anti-cancer Xiaogu umbilical cord combined with diuretic can reduce the degree of malignant ascites, alleviate clinical symptoms, improve quality of life and decrease the occurrence of adverse reactions when used concomitantly with diuretics in the management of malignant ascites.

ABSTRACT

Objective To determine the spectrum of mutations responsible for Phenylalanine hydroxylase (PAH) deficiency on phenylketonuria (PKU) patients of Han Chinese people in the Huaihai region of central China. Methods One hundred and one patients diagnosed with PKU were referred to Xuzhou Maternity and Child Health Care Hospital for genetic counseling/analysis from January 2003 to December 2013.Thirteen exons of PAH gene mutations,as well as their flanking introns, were identified in 202 of chromosomes using polymerase chain reaction-single strand conformation polymorphism(PCR-SSCP) and sequencing. Results (1) The spectrum was composed of 24 different mutation types, which had been submitted to the National Center for Biotechnology Information(NCBI) dbSNP databases under accession number SS#2137543837_SS#C2137543860.(2)The most commonly affected region was exon 7 and its flanking introns. The most prevalent mutations were c.728G > A (p.R243Q),followed by c.721C > T (p.R241C),c.1155G > C(p.L385L),c.1068C > A(p.Y356X),c.-71A > C(-71A > C) and c.60 + 62C > T (IVS1 +62C >T),accounting for 18.317%,8.416%,4.950%,3.960%,3.465% and 2.970% of the mutant chromosomes,respectively.(3)Two novel mutations were identified in PAH gene in PKU patients of Han Chinese people:c.60+62C>T(IVS1 +62C >T) and c.782G >T(p.R261L).Conclusions The vast majority of PAH mutations identified corresponded to those observed for the PKU populations in the other regions in China, whereas a few are considerably different from others. The mutational spectrum of PAH gene found in patients with PKU in the Huaihai region exhibit regional association.

ABSTRACT

Objective To explore the security of propofol used in internal carotid artery anesthesia by observing the effect of internal carotid artery infusion of propofol on neurovascular tissue of cerebral infusion areas in rats. Methods A total of 40 SD rats were randomly divided into a V group and an A group (n=20); rats in the V group were given treatment of right femoral vein infusion of 30 mg/ (kg· h) propofol, and those in the A group were given treatment of right internal carotid artery infusion of 30 mg/(kg· h) propofol for 3 h.Rats were sacrificed 4 h after propofol withdrawal.HE staining and transmission electron microscopy were employed to detect the morphological and ultrastructural changes in the right cerebral cortex tissues; and semi-quantitative real time PCR was used to observe the mRNA expressions of bcl-2 and Bax.Results No obvious acute injury was noted in both groups, and the morphology of cortical neurons, neuralgia cells and vascular structures was normal in both groups. And the neuronal nuclei, organelles and capillary endomembrane of both groups were normal, without obvious changes. No significant difference in mRNA expressions ofbcl-2 and Bax was noted between the two groups (P>0.05).Conclusion It is insignificant in the effects of internal carotid artery infusion of propofol on neurovascular tissues of cerebral infusion areas in rats.
