ABSTRACT

Type 2 diabetes mellitus (T2DM) can cause prostate damage and affect male reproductive function, but the underlying mechanisms are not completely understood. In this study, we used liquid chromatography-mass spectrometry (LC-MS)-based untargeted metabolomics to identify endogenous metabolites in the prostate of a T2DM mouse model. The selected endogenous metabolites were then subjected to bioinformatics analysis and metabolic pathway studies to understand their role in the development of T2DM-induced prostate damage. We used male homozygous BTBR ob/ob mice (n = 12) and BTBR WT mice (n = 11) in this study. We monitored changes in blood glucose, body weight, prostate weight, and prostate index, as well as performed hematoxylin and eosin (H&E) staining and observed that the prostate of the BTBR ob/ob was damaged. We then used ultra-high-performance liquid chromatography coupled to quadrupole time-of-flight mass spectrometry (UPLC-Q-TOF-MS) for metabolomics analysis. The stability of the model was validated using principal component analysis (PCA) and orthogonal partial least squares-discriminant analysis (OPLS-DA). Using variable importance in projection (VIP) > 1, false discovery rate (FDR) < 0.05, and coefficient of variation (CV) < 30 as criteria, a total of 149 differential metabolites (62 upregulated and 87 downregulated) were identified between the prostates of the two groups of mice. Topological pathway analysis showed that these differential metabolites were mainly involved in sphingolipid (SP) and glycerophospholipid (GP) metabolism. In conclusion, our study not only emphasizes the damage caused by T2DM to the prostate but also provides new insights into the potential mechanisms of T2DM-induced male reproductive dysfunction.

ABSTRACT

The bond between a steel reinforcement/rod and glulam plays a crucial role in the resistance and deformation capacity of timbers joints. Existing studies provide different bond-slip models for reinforcements and rods with different anchorage lengths, in which the relationship between local bond stress and global bond behaviour cannot not be established. This study presents a unified analytical method for predicting the bond-slip behaviour of ribbed bars and threaded rods along the grain using a local bond-slip model of reinforcement at the elastic and post-yield stages. In the analytical method, equilibrium, compatibility, and constitutive models for reinforcement and rods are considered. The method is verified using test data of rebars and rods with different anchorage lengths. Comparisons between the experimental and calculated results suggest that the analytical method yields reasonably good predictions of the load-slip relationship and failure mode. Furthermore, the embedment lengths required for yield and the ultimate strengths of the reinforcement and rods along the grain are determined by assuming uniform bond stress distributions over the elastic and post-yield steel segment. The average bond stress over the entire anchorage length is calculated and compared with existing equations. Design recommendations for anchorage lengths are proposed for ribbed bars and threaded rods glued in glulam.

ABSTRACT

Unintentional ingestion of metal-contaminated soils may pose a great threat to human health. To accurately evaluate the health risks of heavy metal(loid)s in soils, their bioaccessibility has been widely determined by in vitro assays and increasingly employed to optimize the assessment parameters. Given that, using meta-analysis, we analyzed the literature on farmland heavy metal(loid)s (As, Cd, Cr, Cu, Hg, Pb, Ni, and Zn) in Chinese main grain production regions, and collected their total and bioaccessibility data to accurately assess their human health risks. Monte Carlo simulation was used to reduce the uncertainty in metal concentration, intake rate, toxicity coefficient, and body weight. We found that the mean concentration (0.47 mg/kg) and geological accumulation index (Igeo, 0-5.24) of Cd were the priority position of controlling metals. Moreover, children are more vulnerable to carcinogenic risks than adults. Soil mineralogy, physicochemical properties, Fe, and the types of in vitro assays are the influencing factors of bioaccessibility discrepancy. Furthermore, appropriate bioaccessibility determination methods can be adapted according to the differences in ecological receptors for the risk assessment, like developing a "personalized assessment" scheme for polluted farmland soil management. Collectively, bioaccessibility-based models may provide an accurate and effective approach to human health risk assessment.

ABSTRACT

Sex-determining region Y-box transcription factor 9(SOX9)is essential for prostate development. The dysregulation of SOX9 not only affects the occurrence of Prostate cancer (PCa), but also plays a key role in castration-resistant prostate cancer (CRPC). However, the mechanism of SOX9 affecting the evolution of PCa is still unclear. This paper mainly reviews the molecular mechanism and signal pathway related to the occurrence and development of SOX9 and PCa. SOX9 gene may be an important new biomarker in the development of PCa,providing new ideas for clinical diagnosis and treatment.

ABSTRACT

The superatomic orbital splitting (SOS) method is developed to understand the electronic structures of coinage metal nanoclusters, in which delocalized electron counts are not magic numbers. Because the symmetry of a metal core can significantly affect the electronic structure of a nanocluster, this method takes the shape of the core into account in determining the order of group orbital levels. By taking nanoclusters as superatoms, a highly positively charged core is established by removing the ligands and staples. The superatomic orbitals split into group orbitals at different energy levels because of the nonspherical shape of the cluster core. Therefore, the electron configuration of the nonmagic-number nanocluster can be qualitatively analyzed without quantum chemical calculations, which is very important for understanding the stability of the cluster.

ABSTRACT

Concrete mix design is one of the most critical issues in concrete technology. This process aims to create a concrete mix which helps deliver concrete with desired features and quality. Contemporary requirements for concrete concern not only its structural properties, but also increasingly its production process and environmental friendliness, forcing concrete producers to use both chemically and technologically complex concrete mixtures. The concrete mix design methods currently used in engineering practice are joint analytical and laboratory procedures derived from the Three Equation Method and do not perform well enough for the needs of modern concrete technology. This often causes difficulties in predicting the final properties of the designed mix and leads to precautionary oversizing of concrete properties for fear of not providing the required parameters. A new approach that would make it possible to predict the newly designed concrete mix properties is highly desirable. The answer to this challenge can be methods based on machine learning, which have been intensively developed in recent years, especially in predicting concrete compressive strength. Machine learning-based methods have been more or less successful in predicting concrete compressive strength, but they do not reflect well the variability that characterises the currently used concrete mixes. A new adaptive solution that allows estimating concrete compressive strength on the basis of the concrete mix main ingredient composition by including two observations for a given batch of concrete is proposed herein. In presented study, a machine learning model was built with a deep neural network architecture, trained on an extensive database of concrete recipes, and translated into a mathematical formula. Testing on four concrete mix recipes was performed, which were calculated according to contemporary design methods (Bolomey and Fuller method), and a comparative analysis was conducted. It was found out that the new algorithm performs significantly better than that without adaptive features trained on the same dataset. The presented algorithm can be used as a concrete strength checking tool for the concrete mix design process.

ABSTRACT

OBJECTIVE: To analyze the polymorphism of the HPA1-5,15 system of the donors in Zhangjiakou area. METHODS: DNA was extracted from the blood samples of the donors, PCR- SSP method was used to divide HPA1-6, 15 genotype. The gene frequency and genotype frequency were calculated, compared with the difference and regiahal specificity of the populations in our country and foregiens was compared other populations. RESULTS: The gene expression in the HPA-1, HPA-2 and HPA-4 systems were all homozygous aa, and the donors who expressed homozygous bb was not exessed. Among them, one heterozygous ab expression was found in both HPA-1 and HPA-4 systems (1%), and 14 cases of heterozygous ab expression were found in HPA-2 system (14%). The gene expression in the HPA-5 system was mainly homozygous aa (98%), and a very few expressed homozygous bb (2%) was found. The degree of heterozygosity of gene expression in the HPA-3 and HPA-15 systems was relatively high. The proprotion of the expression of aa, ab and bb in the HPA-3 system was respectively 46%, 40% and 14%, the proprotion of the expression of aa, ab and bb in the HPA-15 system was respectively 21%, 64% and 15%. CONCLUSION: The gene frequency of platelet-specific antigen HPA1-5,15 system in zhangjiakou region shows local characteristics. The heterozygosity degree of gene expression in the HPA-3 and HPA-15 systems are both high, suggesting that they are more likely to result in alloimmunization and ineffective platelet transfusion, which should be pays attention to.

ABSTRACT

The role of androgen receptor (AR) in breast cancer has been unveiled in succession for the past few years. In this study, we conducted a comprehensive analysis based on four online public databases of data from many previous studies. We found that the expression of AR is significantly related to age, histological grade, and subtype but not to lymph node status. The low expression level of AR is strongly associated with poor recurrence-free survival, especially with poor distance metastasis-free survival in luminal A patients, but inverse in HER2 (human epidermal growth factor receptor-2) enriched patients. AR might be a biomarker of chemosensitivity in the basal subtype. Besides, the expression of melanophilin (MLPH) is distinctly in accordance with that of AR. AR could play diverse roles in different subtypes of breast cancer.

ABSTRACT

We report a case of a fetus with rhizomelic chondrodysplasia punctata type Ⅰ. Ultrasound examination of the pregnant women at 23 weeks of gestation showed multiple fractures in bilateral femurs, thick metaphysis, severely short, thickened and curved bilateral humerus with multiple fracture images, some of which were callu formation after fracture. The pregnancy was terminated at 23 +2 gestational weeks, samples of fetal skin tissue were taken after birth, and parental peripheral blood was collected for whole exome sequencing, which revealed a frameshift mutation c.179delT (p.F61Lfs*13) in the PEX7 gene of chromosome 6, and the heterozygous deletion (141 kb) occurred in the region of chromosome 6 137105182-137245871, covering the pathogenic gene PEX7. The analysis of parental samples suggested that the mutations were compound heterozygous mutations, none of which had been previously reported and were determined to be pathogenic mutations. The severe clinical phenotype of this case may be caused by severe damage of PEX7 gene contributed by the frameshift mutation and large fragment deletion mutations.

ABSTRACT

A robust adaptive recurrent cerebellar model articulation controller (RARC) neural network for non-linear systems using the genetic particle swarm optimization (GPSO) algorithm is presented in this study. The RARC is used as the principal tracking controller and the robust compensation controller is designed to recover the residual of the approximation error. In the RARC neural network, the steepest descent gradient method and the Lyapunov function are used for deriving the adaptive law parameter of the system. Besides, the learning rates play an important role in these adaptive laws and they have a great effect on the functions of control systems. In this paper, the combination of the genetic algorithm with the mutation particle swarm optimization algorithm is applied to seek for the optimal learning rates of the RARC adaptation laws. The numerical simulations about the inverted pendulum system as well as the robot manipulator system are given to confirm the effectiveness and practicability of the GPSO-RARC-based control system. Compared with other control schemes, the proposed control scheme is testified to be reliable and can obtain the optimal parameter about the learning rates and the minimum root mean square error for non-linear systems.

ABSTRACT

Based on the plasma activation and the sensing ability of cataluminescence, a low temperature plasma-assisted cataluminescence sensor was developed for ethylene detection using the low-cost and abundant alkaline-earth oxides of MgO nanomaterials as the sensing materials.Taking advantage of the high activity of the plasma, the working temperature of this method was greatly decreased than that of traditional detection method (300-500℃), and the sensing of ethylene was realized at room temperature without any heating device.This ethylene cataluminescence sensor gave a linear range of 112-4997 ng/mL (90-3998 ppm, R=0.97669) with a detection limit of 37 ng/mL (30 ppm).Besides, the sensor showed good selectivity and stability in ethylene detection.Due to the absence of the heating element, the present sensor was simple, rapid, low-cost, low energy-consumption and stable for ethylene sensing.This study improved the applicability of cataluminescence sensors and might promote the development of cataluminescence sensors.

ABSTRACT

We mined the literature for proteomics data to examine the occurrence and metastasis of prostate cancer (PCa) through a bioinformatics analysis. We divided the differentially expressed proteins (DEPs) into two groups: the group consisting of PCa and benign tissues (P&b) and the group presenting both high and low PCa metastatic tendencies (H&L). In the P&b group, we found 320 DEPs, 20 of which were reported more than three times, and DES was the most commonly reported. Among these DEPs, the expression levels of FGG, GSN, SERPINC1, TPM1, and TUBB4B have not yet been correlated with PCa. In the H&L group, we identified 353 DEPs, 13 of which were reported more than three times. Among these DEPs, MDH2 and MYH9 have not yet been correlated with PCa metastasis. We further confirmed that DES was differentially expressed between 30 cancer and 30 benign tissues. In addition, DEPs associated with protein transport, regulation of actin cytoskeleton, and the extracellular matrix (ECM)-receptor interaction pathway were prevalent in the H&L group and have not yet been studied in detail in this context. Proteins related to homeostasis, the wound-healing response, focal adhesions, and the complement and coagulation pathways were overrepresented in both groups. Our findings suggest that the repeatedly reported DEPs in the two groups may function as potential biomarkers for detecting PCa and predicting its aggressiveness. Furthermore, the implicated biological processes and signaling pathways may help elucidate the molecular mechanisms of PCa carcinogenesis and metastasis and provide new targets for clinical treatment.

ABSTRACT

Currently, using human prostate cancer (PCa) tissue samples to conduct proteomics research has generated a large amount of data; however, only a very small amount has been thoroughly investigated. In this study, we manually carried out the mining of the full text of proteomics literature that involved comparisons between PCa and normal or benign tissue and identified 41 differentially expressed proteins verified or reported more than 2 times from different research studies. We regarded these proteins as seed proteins to construct a protein-protein interaction (PPI) network. The extended network included one giant network, which consisted of 1,264 nodes connected via 1,744 edges, and 3 small separate components. The backbone network was then constructed, which was derived from key nodes and the subnetwork consisting of the shortest path between seed proteins. Topological analyses of these networks were conducted to identify proteins essential for the genesis of PCa. Solute carrier family 2 (facilitated glucose transporter), member 4 (SLC2A4) had the highest closeness centrality located in the center of each network, and the highest betweenness centrality and largest degree in the backbone network. Tubulin, beta 2C (TUBB2C) had the largest degree in the giant network and subnetwork. In addition, using module analysis of the whole PPI network, we obtained a densely connected region. Functional annotation indicated that the Ras protein signal transduction biological process, mitogen-activated protein kinase (MAPK), neurotrophin and the gonadotropin-releasing hormone (GnRH) signaling pathway may play an important role in the genesis and development of PCa. Further investigation of the SLC2A4, TUBB2C proteins, and these biological processes and pathways may therefore provide a potential target for the diagnosis and treatment of PCa.

Objective To investigate the feasibility of using clustered regularly interspaced short palindromic repeats ( CRISPR)-mediated genome editing to downregulate the expression of programmed cell death protein 1 (PD-1) on primary T cells by using a lentivirus delivery system. Methods Lentivirus vec-tors pLentiCRISPR A1-A6 containing different PD-1 genomic DNA sequences as single guide RNA ( sgRNA) for Cas9 targeting were constructed individually. The lentivirus vectors were tranduced into primary CD4 T cells. Flow cytometry analysis was performed to detect the expression of PD-1 for evaluating the knockout ef-ficiency. Results The lentivirus vectors pLentiCRISPR A1-A6 carrying six different target sites were con-structed and respectively tranduced into primary CD4 T cells. The expression of PD-1 accompanied with the activation of T cells. Co-expression of CD25 and PD-1 was observed on activated T cells. All of the six sites could be targeted by Cas9, of which A2 and A6 sites were more efficient in knocking out the gene encoding PD-1 with a rate of 19% and 29%, respectively. Conclusion This study suggests that it is feasible to knock out the expression of PD-1 on primary T cells by using CRISPR.

ABSTRACT

<p><b>BACKGROUND</b>In order to minimize the injury reaction during the surgery and reduce the morbidity rate, hence reducing the mortality rate of esophagectomy, minimally invasive esophagectomy (MIE) was introduced. The aim of this study was to compare the postoperative outcomes in patients with esophageal squamous cell carcinoma undergoing minimally invasive or open esophagectomy (OE).</p><p><b>METHODS</b>The medical records of 176 consecutive patients, who underwent minimally invasive esophagectomy (MIE) between January 2009 and August 2013 in Cancer Institute & Hospital, Chinese Academy of Medical Sciences, were retrospectively reviewed. In the same period, 142 patients who underwent OE, either Ivor Lewis or McKeown approach, were selected randomly as controls. The clinical variables of paired groups were compared, including age, sex, Charlson score, tumor location, duration of surgery, number of harvested lymph nodes, morbidity rate, the rate of leak, pulmonary morbidity rate, mortality rate, and hospital length of stay (LOS).</p><p><b>RESULTS</b>The number of harvested lymph nodes was not significantly different between MIE group and OE group (median 20 vs. 16, P = 0.740). However, patients who underwent MIE had longer operation time than the OE group (375 vs. 300 minutes, P < 0.001). Overall morbidity, pulmonary morbidity, the rate of leak, in-hospital death, and hospital LOS were not significantly different between MIE and OE groups. Morbidities including anastomotic leak and pulmonary morbidity, inhospital death, hospital LOS, and hospital expenses were not significantly different between MIE and OE groups as well.</p><p><b>CONCLUSIONS</b>MIE and OE appear equivalent with regard to early oncological outcomes. There is a trend that hospital LOS and hospital expenses are reduced in the MIE group than the OE group.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the reasons of secondary fracture after percutaneous vertebroplasty (PVP) for osteoporotic vertebral compression fractures (OVCFs) and discuss the measure of prevention and cure.</p><p><b>METHODS</b>From January 2011 to January 2013, the clinical data of 180 patients with primary OVCFs treated by PVP were retrospectively analyzed. There were 75 males and 105 females, aged from 68 to 95 years old with an average of (79.50 ± 5.45) years. The involved vertebrae were identified according to the clinical symptoms and imaging data. PVP were performed in 362 vertebrae and the patients were followed up with an average of 12 months. Subsequent vertebral fractures were found through the pain's reappearance and MRI or bone scan. The patients were divided into secondary fracture group and no-secondary fracture group according to the subsequent fractures or no. Secondary fracture group was divided into two groups according to gender, and the patients with secondary fracture were also categorized into the original surgical vertebral fractures, adjacent vertebral fracture and remote vertebral fractures. The age, gender, the cement volume, the cement leakage, secondary fracture site, the incidence and type of secondary fracture were observed and compared among different groups.</p><p><b>RESULTS</b>Among the 362 vertebrae of PVP, there were 109 vertebrae in male and 253 vertebrae in female. And 27 vertebrae (10 in male and 17 in female) of 22 cases (9 males and 13 females) occurred secondary fracture. The second PVP were performed in 13 cases (16 vertebrae) and the third PVP in 2 cases (4 vertebrae); 7 cases (7 vertebrae) were treated with conservative therapy. There was no statistically significant difference on age, gender, cement volume and leakage between secondary fracture group and no-secondary fracture group (P > 0.05). There was no statistically significant difference on the incidence and type of secondary fracture between male and female (P > 0.05). No significant difference was found on the adjacent and remote vertebral fractures (P > 0.05). Most of secondary fracture occurred in 6 months, and whether the single and double side injection, cement leakage had no obvious relation.</p><p><b>CONCLUSION</b>There is no significant difference in the subsequent fracture after PVP for the OVCFs different gender and fractured site, and also no significant difference in the adjacent and remote vertebral fractures. The report didn't support the biomechanical viewpoint that vertebral body stiffness increasing after PVP would lead to adjacent vertebral stress increasing and result easily in adjacent vertebral fracture. Secondary fracture occurs always in 6 months after operation, which is the natural course of osteoporosis.</p>

ABSTRACT

OBJECTIVE: To explore the efficacy and safety of 980 nm diode laser vaporization in the treatment of benign prostatic hyperplasia (BPH). METHODS: We treated 92 BPH patients by 980 nm diode laser vaporization. The patients were aged 65 - 89 years, with a mean prostate volume of (50.1 +/- 13.0) ml. We analyzed and compared the mean operation time, intra-operative blood loss, postoperative complications, international prostate symptom score (IPSS), quality of life (QOL) score, maximum urine flow rate (Qmax), and post void residual (PVR) before and after surgery. RESULTS: Operations were successful in all the 92 cases, with an average operation time of (70.2 +/- 16.9) min, very little blood loss and no blood transfusion. The transurethral catheter indwelling time was 2 -5 (2.4 +/- 0.3) days. The patients were followed up for 1 to 3 months, which revealed a significant reduction in IPPS (P < 0.01) and improvement in Qmax and PVR (P < 0.01) as compared with preoperation. No severe complications were reported, including urinary incontinence and bladder irritation symptoms. None of the patients complained of impaired erectile function. CONCLUSION: Transurethral 980 nm diode laser vaporization is a safe and effective treatment for BPH.

ABSTRACT

For a long time, the diagnosis and treatment protocol for esophageal cancer has been made either entirely based on the experience of Western countries or on our own experience alone. A suitable standardized guideline for diagnosis and treatment of esophageal cancer in our country has not been established until 2010. Due to lack of opportunities for mutual exchange and learning, the overall level of standardized diagnosis and treatment was relatively low in China. In addition, less advanced technologies were applied and varying treatment protocols were implemented in different districts, the treatment results has been unsatisfactory, and the overall 5-year survival rate after surgical treatment has been maintained from 30% to 40%. Therefore, it is imperative that China needs to conduct its own clinical studies and establish its own suitable standardized treatment and diagnosis guideline for esophageal cancer. This article reviews and also made comments on the process of academic speaking tour and development of standardized diagnosis and treatment guidelines as well as the problems resolved by these activities for esophageal cancer since the beginning of this century.

ABSTRACT

The elasticity and nanomechanical response of Aspergillus niger spores determined using atomic force microscopy (AFM) and nanoindentation are discussed. The force-displacement curve of the spore surfaces shows that the average surface roughness of spores was approximately 33 nm and that the adhesion force ranged from 9 to 28 nN. The Young's modulus of the A. niger spores ranged from 0.1 to 21.4 GPa and the hardness ranged from 0.01 to 0.17 GPa. The critical buckling load of the spore membrane is 290 µN.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the changes in the mRNA expression of adiponectin (Adp), adiponectin receptors(AdpR), and leptin in different adipose tissues of Wannanhua pigs at different stages of development, and their sexual dimorphism.</p><p><b>METHODS</b>Five Wannanhua boars and five Wannanhua gilts were sampled at birth, 30, 45, 90, and 180 days of age respectively. The delta delta Ct relative quantification real-time PCR was used to detect the transcription levels of Adp, AdpR1, AdpR2, and leptin mRNAs in subcutaneous (SC) and perirenal (PR) adipose tissues, and beta-actin were used as internal standards.</p><p><b>RESULTS</b>The expression level of Adp, AdpR1, AdpR2, and leptin mRNA in SC and PR adipose tissue were changed with age significantly (P < 0.01). In general, Adp mRNA expression in SC adipose tissue was significantly lower than that in PR adipose tissue (P < 0.05), while AdpR1, AdpR2, and leptin mRNA expression in SC adipose tissue were significantly higher than those in PR adipose tissue (P < 0.05 or P < 0.01). Although the sexual dimorphism were found in apart genes or apart days of age, Adp, AdpR1, AdpR2, and leptin mRNA expression both in SC adipose tissue and PR adipose tissue had no significant differences between Wannanhua gilts and boars in general. Significant positive correlation was found between Adp and AdpR1, AdpR2 (P < 0.05 or P < 0.01), and significant negative correlation was found between Adp and leptin (P < 0.05) in SC adipose tissue and PR adipose tissue respectively (P < 0.05).</p><p><b>CONCLUSION</b>The expression of Adp, AdpR1, AdpR2, and leptin mRNA in adipose tissue of Wannanhua pigs followed specific developmental patterns and tissue specificity. Adp correlated with its receptors.</p>
