ABSTRACT

Objective:To explore the rules of Professor Zhang Binghou’s experience on treating insomnia. Methods:The formula for insomnia treated by Professor Zhang Binghou in Beijing Hospital of Traditional Chinese Medicine Affiliated to Capital Medical University and Beijing Tongrentang Hospital of Traditional Chinese Medicine from Oct. 1, 2016 to July 31, 2019 were collected and sorted out. By establishing the database of formulasto analyze their frequency, patterns, association rules, core medicines and combination of new formulas by cluster analysis by using Traditional Chinese Medicine Inheritance Support System (TCMISS) software V2.5. Results:Among 83 formulas, there were three main types of TCM syndromes, which were the heart-kidney disjunction, heart spleen deficiency and liver-kidney yin deficiency, and involving 134 Chinese herbs. The high frequently ones were Ziziphi Spinosae Semen, Platycladi Semen, Margaritifera concha, and Mauritia arabica. Forty core Chinese herbs combination. Forty herbal combinations with high frenquency and 12 new formulas were identified. The top 5 herbal combinations with high frenquency were Platycladi Semen- Ziziphi Spinosae Semen, Ziziphi Spinosae Semen- Margaritifera concha, Platycladi Semen- Margaritifera concha, Platycladi Semen- Ziziphi Spinosae Semen- Margaritifera concha and Platycladi Semen- Mauritia arabica. The combined formulas were Cannabis fructus- Paeoniae radix alba- Ophiopogonis radix- Atractylodis macrocephalae rhizoma (baked), Scutellariae radix- Asini corii colla- Margaritifera concha- cinnamomi cortex- Platycladi semen, Glycyrrhizae radix et rhizoma praeparata cum melle- Paeoniae radix alba- Atractylodis macrocephalae rhizome (baked)- Coptidis rhizoma- Glycyrrhizae radix et rhizoma praeparata cum melle- Rehmanniae radix- Coptidis rhizoma, Atractylodis macrocephalae rhizome (baked)- Astragali radix praeparata cum melle and Ginseng radix et rhizoma- Pogostemonis herba- Clematidis armandii Caulis- Nelumbinis semen. Conclusions:Professor Zhang's treatment of insomnia mainly focusing on tranquilizing mind, combined with nourishing yin and relieving the internal heat, communicating heart and kidney, invigorating spleen and nourishing heart, nourishing qi and blood based on syndrome differentiation.

ABSTRACT

Objective:To observe the effect of quercetin on myelin regeneration in desyelinating mice induced by cuprizone (CPZ).Methods:A total of 50 C57BL/6J mice were randomly divided into normal control group, model group, low, medium and high dose quercetin groups, 10 mice in each group. In addition to the normal control group, mice demyelinating model was induced by feeding with 0.2% CPZ rodent feed. Quercetin solution was administered to the low, medium and high dose quercetin groups at 25, 50 and 100 mg/kg, and to the normal control group and the model group at 1 time/d. After 5 weeks of continuous gavage, the weight of mice was recorded every week, and the experiment of rotating bar was carried out. After 5 weeks, the changes of the myelin sheath in the corpus callosum of mice were observed by luxol fast blue (LFB) and transmission electron microscope (TEM). Immunofluorescence method was used to determine the protein expressions of myelin basic protein (MBP) and oligodendrocyte transcription 2actor (Olig2) in mouse brain tissue. The expression of MBP and cyclic nucleotide-3'phosphate hydrolase (CNPase) in mouse corpus callosum was determined by Western blot.Results:After 2-5 weeks, compared with the model group, the body mass of the medium and high dose quercetin groups significantly increased ( P<0.01), and the score of turning rod significantly increased ( P<0.01). The LFB observation showed that the demyelination score of corpus callosum in low, medium and high dose quercetin groups (2.23 ± 0.25, 1.50 ± 0.15, 1.14 ± 0.97 vs. 2.83 ± 0.18) significantly decreased ( P<0.01). TEM observation showed that the G-ratio value in low, medium and high dose quercetin groups (0.75 ± 0.05, 0.75 ± 0.08, 0.73 ± 0.08 vs. 0.87 ± 0.05) significantly reduced ( P<0.01). Immunofluorescence observation showed that the positive expression of MBP (37.40 ± 2.41, 37.40 ± 1.14 vs. 24.40 ± 3.65) and Olig2 (7.40 ± 1.14, 4.60 ± 1.14 vs. 2.80 ± 0.84) in the medium and high dose quercetin groups significantly increased ( P<0.05). WB showed that the expression of MBP protein (1.32 ± 0.12, 0.80 ± 0.34 vs. 0.21 ± 0.07) and CNPase protein (0.72 ± 0.13, 1.06 ± 0.36 vs. 0.36 ± 0.21) in the medium and high dose quercetin groups significantly increased ( P<0.05). Conclusions:Quercetin (50-100 mg/kg) can reduce myelin injury and promote myelin regeneration in CPZ mice, as a neuroprotective effect on CPZ mice.

ABSTRACT

Objective: To explore the value of scrotal ultrasonography for choice of treatment of obstructive azoospermia. Methods: Totally 93 obstructive azoospermia patients were prospectively collected for scrotal ultrasound to evaluate whether the epididymis and vas deferens were intact, dilated or calcified. The diameter of epididymal duct and vas deferens were measured. Then the patients were divided into sperm extraction group (n=27) or vasal reconstruction group (n=66) according to the treatment. The etiology and sonographic appearances were compared between groups. The cut-off values of ultrasonic parameters were evaluated by ROC curve, followed by the sensitivity, specificity, positive predicative value, negative predicative value and accuracy. Results: There were significant differences of epididymal morphology, vas deferens dysplasia, calcification, and the diameter of epididymal duct between 2 groups (all P<0.05). Taken 0.55 mm for the cut-off value of diameter of epididymal duct, the sensitivity, specificity, positive predictive value, negative predictive value and accuracy of correctly identifying the patients suitable for sperm extraction was 87.47%, 86.70%, 77.82%, 92.76% and 87.03%, respectively. Conclusion: Scrotal ultrasonography plays an important role in guiding treatment for patients with obstructive azoospermia.

Objective@#To investigate the efficacy of Linglianhua granule in the treatment of depression after coronary heart disease with syndrome of heart-kidney imbalance and its effect on serum levels of interleukin(IL)-8, IL-17, norepinephrine(NE), and 5-hydroxytryptamine(5-HT).@*Methods@#From December 2016 to December 2018, 260 cases with depression after coronary heart disease were selected in Kangning Hospital Affiliated to Wenzhou Medical University and randomly divided into the control group and the observation group according to the digital table, with 130 cases in each group.Both two groups were given routine therapy.The control group was treated with lorazepam tablets.Based on the control group, the observation group was given Linglianhua granule (1 bag/time and bid). The two groups were treated for 8 weeks.The scores of Hamilton's depression scale(HAMD)-17, self-rating depression scale (SDS), clinical efficacy, and serum levels of IL-8, IL-17, NE, 5-HT were compared between the two groups.@*Results@#After treatment for 8 weeks, the HAMD, SDS score of the observation group were obviously lower than those of the control group(t=7.903, 12.331, all P=0.00). The total effective rate of the observation group(93.85%) was higher than that of the control group(84.62%)(χ2=4.843, P<0.05). After treatment for 8 weeks, the serum levels of IL-8 and IL-17 in the observation group were lower, while the NE and 5-HT levels were higher, compared with those in the control group(t=9.403, 14.439, 19.431, 28.931, all P=0.00).@*Conclusion@#Based on the routine therapy, Linglianhua granule in treating depression after coronary heart disease with syndrome of heart-kidney imbalance can promote improvement of depression symptoms, increase the efficacy.Inhibiting serum levels of IL-8, IL-17 and up-regulation of NE and 5-HT may be related with the efficacy.

ABSTRACT

Annexin A1 (ANXA1) is a kind of endogenous scaffold protein. Previous research showed that ANXA1 could increase markedly with multiple increase of drug resistance in K562/imatinib cell lines in vitro. Here the stable transfection cell strains K562-pEGFP-N1 which was the native control and K562-pEGFP-N1-ANXA1 which can stably express ANXA1 were established using the Lipofectamine 2000 in order to find whether ANXA1 involved in the drug resistance. Cell growth inhibition experiment via MTT and cell proliferation experiment via MTS showed that K562-pEGFP-N1-ANXA1 cell strain was more sensitive to imatinib than the K562-pEGFP-N1 cell strain, and however the ability of proliferation of K562-pEGFP-N1-ANXA1 cell strain did not change compared with the negative control. Western blotting results showed that the expression of proteins in Annexin family did not change; drug resistance proteins, Bcr-Abl/p-Bcr-Abl (Tyr245), Src family kinase for example, did not change; proteins related with cell proliferation and cell cycle, such as ERK1/2MAPK, p-38MAPK, CDK1 and Wee 1, did not change either in the K562-pEGFP-N1-ANXA1 cell strain compared with the negative control. The co-immunoprecipitation result showed that the interaction between ANXA1 and beta-actin in the K562-pEGFP-N1-ANXA1 cell strain increased markedly. The deduction was that ANXA1 may make the K562-pEGFP-N1-ANXA1 cell strain more sensitive to imatinib due to the increased uptake of imatinib via the increase of ANXA1 and the interaction between ANXA1 and beta-actin in the K562-pEGFP-N1-ANXA1 cell strain in vitro.

ABSTRACT

[Objective]To evaluate the methods of operation and clinic effective of thoracolumbar spinal tuberculosis with paraplegia.[Method]Thirty cases of thoracolumbac spinal tuberculosis with paraplegia had been treated with primary anterior debridement,decompression of spinal cord,interbody graft bone,and interbody fixation,anti-tuberculosis drug was taken at 18 monthes ofter the operation.[Result]Thirty cases were followed-up for an overage of 19 months and were healed,in which 2 cases recurrence,29 cases spinal fusion of graft bone,12? of kyphosis correction had been gained,the neuropathic fuction had gained improvement excellently(90%).[Conclusion]It had gained good outcome that operation of the primary anterior debridement decompression of spinal cand interbody graft bone and interbody fixation to treat thoracolumbac spinal tuberculosis with pacaplegia.
