ABSTRACT

Our knowledge of genetic aberrations, that is, variants and copy number variations (CNVs), associated with mantle cell lymphoma (MCL) relapse remains limited. A cohort of 25 patients with MCL at diagnosis and the first relapse after the failure of standard immunochemotherapy was analyzed using whole-exome sequencing. The most frequent variants at diagnosis and at relapse comprised six genes: TP53, ATM, KMT2D, CCND1, SP140, and LRP1B. The most frequent CNVs at diagnosis and at relapse included TP53 and CDKN2A/B deletions, and PIK3CA amplifications. The mean count of mutations per patient significantly increased at relapse (n = 34) compared to diagnosis (n = 27). The most frequent newly detected variants at relapse, LRP1B gene mutations, correlated with a higher mutational burden. Variant allele frequencies of TP53 variants increased from 0.35 to 0.76 at relapse. The frequency and length of predicted CNVs significantly increased at relapse with CDKN2A/B deletions being the most frequent. Our data suggest, that the resistant MCL clones detected at relapse were already present at diagnosis and were selected by therapy. We observed enrichment of genetic aberrations of DNA damage response pathway (TP53 and CDKN2A/B), and a significant increase in MCL heterogeneity. We identified LRP1B inactivation as a new potential driver of MCL relapse.

ABSTRACT

There are many cases where one needs to limit the X-ray dose, or the number of projections, or both, for high frame rate (fast) imaging. Normally, it improves temporal resolution but reduces the spatial resolution of the reconstructed data. Fortunately, the redundancy of information in the temporal domain can be employed to improve spatial resolution. In this paper, we propose a novel regularizer for iterative reconstruction of time-lapse computed tomography. The non-local penalty term is driven by the available prior information and employs all available temporal data to improve the spatial resolution of each individual time frame. A high-resolution prior image from the same or a different imaging modality is used to enhance edges which remain stationary throughout the acquisition time while dynamic features tend to be regularized spatially. Effective computational performance together with robust improvement in spatial and temporal resolution makes the proposed method a competitive tool to state-of-the-art techniques.

ABSTRACT

The authors analyze their experience in the application of mexicor, a Russian cytoprotector, in 50patients with chronic coronary artery disease (CAD) and 51 patients with acute coronary syndrome. In additional to cytoprotective action, the use of mexidor in complex therapy of CAD lowers the functional activity of thrombocytes, eliminates high blood viscosity syndrome, and lowers low density lipoprotein cholesterol level. These favorable changes in hemorheological parameters improves myocardial perfusion, lowers the strength and frequency of coronary pain attacks, retards postinfarction left ventricular remodeling, and increases the quality of life of patients with various CAD forms.

ABSTRACT

Miners of basic professions employed in contemporary metal mining industry are exposed to a range of industrial factors, such as vibration, noise, dustiness, and unfavorable microclimate. Long-term exposure to these factors is hazardous to their health, which is manifested by elevated general and occupational morbidity and a higher rate of biological aging. This is most typical of Far North miners, who are exposed to the combination of the industrial factors and extremal climate and geographic conditions. Research into the health status of miners engaged to underground works and open excavations in regions with contrasting climate and geographic conditions, allowed determination of main industrial risk factors (vibration and noise), leading occupational diseases and groups of high risk of their development (according to the occupation and the length of service). The study found high risk of cardivascular diseases as well as pathology of the musculoskeletal system, which was highest in miners in Norilsk region.

ABSTRACT

A combined system for far- and near-field optical spectroscopy consisting of a compact scanning near-field optical microscope and a dedicated spectrometer was realized. The set-up allows the optical investigation of samples at temperatures from 10 to 300 K. The sample positioning range is as large as 5 x 5 x 5 mm3 and the spatial resolution is in the range of 1.5 micro m in the far-field optical microscopy mode at low temperatures. In the scanning near-field optical microscope mode the resolution is defined by the microfabricated cantilever probe, which is placed in the focus of a double-mirror objective. The tip-to-sample distance in the scanning near-field optical microscope is controlled by a beam deflection system in dynamic scanning force microscopy mode. After a description of the apparatus, scanning force topography images of self-assembled InAs quantum dots on a GaAs substrate with a density of less than one dot per square micrometre are shown, followed by the first spectroscopic investigations of such a sample. The presented results demonstrate the potential of the system.
