ABSTRACT

Extraintestinal pathogenic Escherichia coli (ExPEC) is a major health concern due to emerging antibiotic resistance. Along with O1A, O2, and O6A, E. coli O25B is a major serotype within the ExPEC group, which expresses a unique O-antigen. Clinical studies with a glycoconjugate vaccine of the above-mentioned O-types revealed O25B as the least immunogenic component, inducing relatively weak IgG titers. To evaluate the immunological properties of semisynthetic glycoconjugate vaccine candidates against E. coli O25B, we here report the chemical synthesis of an initial set of five O25B glycan antigens differing in length, from one to three repeat units, and frameshifts of the repeat unit. The oligosaccharide antigens were conjugated to the carrier protein CRM197. The resulting semisynthetic glycoconjugates induced functional IgG antibodies in mice with opsonophagocytic activity against E. coli O25B. Three of the oligosaccharide-CRM197 conjugates elicited functional IgGs in the same order of magnitude as a conventional CRM197 glycoconjugate prepared with native O25B O-antigen and therefore represent promising vaccine candidates for further investigation. Binding studies with two monoclonal antibodies (mAbs) revealed nanomolar anti-O25B IgG responses with nanomolar K D values and with varying binding epitopes. The immunogenicity and mAb binding data now allow for the rational design of additional synthetic antigens for future preclinical studies, with expected further improvements in the functional antibody responses. Moreover, acetylation of a rhamnose residue was shown to be likely dispensable for immunogenicity, as a deacylated antigen was able to elicit strong functional IgG responses. Our findings strongly support the feasibility of a semisynthetic glycoconjugate vaccine against E. coli O25B.

ABSTRACT

The increase of vineyard's water consumption due to the Global Warming Phenomenon (GWP) has forced the winegrowers to strengthen their irrigation and water stewardship efforts, intended for maintaining this resource's long-term sustainable use. Due to water being a limited resource, implementing the Water Footprint (WF) concept in winegrapes production provides helpful information for sustainable water stewardship. Currently, an automated version of the satellite-based METRIC (Mapping Evapotranspiration with Internalized Calibration) model, the Google Earth Engine Evapotranspiration Flux (EEFlux) platform, has been suggested as an alternative to analyzing the spatial variability of an entire field's water consumption throughout the growing season. This work aimed to evaluate the potential application of the EEFlux satellite's actual evapotranspiration (ETa) products and ancillary field data to obtain the WF blue (WFb) and green (WFg) of six commercial vineyards placed in the Chilean central zone. Firstly, the reliability of the daily actual evapotranspiration data from EEFlux (ETa EEFlux) was assessed against measured ETa data, using an available database from previous studies. The results of ETa EEFlux estimations against measured ETa were impressive, presenting a root square error (RMSE) of 0.8 mm day-1. The satellite-derived crop coefficients (Kc Sat) allowed to estimate the total WF of each vineyard, in a range of 200 to 900 m3 t-1, showing an average relative error (RE) of 101%, between the satellite-based WFb (WFb Sat) and those calculated from irrigation records (WFb). These results reflected the particular conditions of each vineyard and can be considered reasonable since they were estimated from ancillary data and EEFlux products. This study provides new insights that may represent opportunities to sustainably managing the irrigation of vineyards.

ABSTRACT

BACKGROUND: Many states have legalized casino gambling, and casinos create increased vehicle traffic, but the strength of the association between casino construction and vehicle crashes is unknown. METHODS: Retrospective analyses of motor vehicle crashes (MVCs) occurring within Anne Arundel County, Maryland (2010-2014) were conducted. The ratio of crashes within one mile of the casino's location after it was opened were compared to the ratio occurring in the same area before it was opened to determine how the incidence of MVCs near the casino changed with time. Logistic regression was used to determine how crash characteristics may have influenced the incidence of MVCs near the casino after it opened. RESULTS: 101,860 persons were involved in 43,328 MVCs in Anne Arundel County during the study period; 29,476 (68.0 %) had an at-fault driver ≥21 years of age and complete data. MVCs proximal to the casino occurred most commonly during the day (N = 421, 76.6 %) and involved drivers <40 years of age (N = 366, 66.6 %) and male (N = 316, 57.4 %). After adjustment for impairment and day of the week, there was a significant association with crashes close to the casino after it opened (ORAdjusted = 1.23, 95 % CI: 1.04-1.46, p = 0.02). Crashes occurring close to the casino, after it opened, involved drivers <40 years of age (OR = 1.74, 95 % CI:1.45-2.08) and occurred on weekends (OR = 1.39, 95 %CI:1.15-1.67). CONCLUSIONS: In this single-site study the opening of a casino was associated with an increase in crashes nearby. The generalizability of this finding should be confirmed with analysis of MVC data near other gambling venues.
