ABSTRACT

The suggested technique allows obtaining large slices of hard tissues such as human skin. Replacement of ethanol for acetone improves quality of histological preparations and decreases their cost.

ABSTRACT

Some improvement has been made of the Belov et al. (1975) method of alkaline dissociation of tissues. No cell loss occurs during cell suspension preparation or other procedures. The improved method was shown to be suited for determination of absolute numbers of cardiomyocytes and hepatocytes. Comparison of the living and dissociated hepatocytes revealed preservation of the total cell dry weight and DNA content (classes of ploidy) in the cells treated with formol and concentrated alkali. The method is recommended for studying the cell number, weight, ploidy and mitoses, and for DNA autoradiography.

ABSTRACT

Biochemical and radioimmunoassay of testosterone content in the blood and urine, epitestosterone and 17-CS level in the urine of 80 boys has shown, that the ovarian androgenic function is reduced in all the forms of cryptorchidism. The potential testis reserves are progressively lowered with the age in patients with bilateral and false retention and remain unchanged in unilateral cryptorchidism. In the absolute majority of patients the change in gonadotropic hormone production is noted, manifesting in the decreased and/or increased LH and FSH content in the blood of different patients, that is seemed to reflect heterogeneity of cryptorchidism pathogenetic forms. The ovarian testosterone production rise after chorionic gonadotropin injection did not inhibit the hypophyseal FSH activity.

ABSTRACT

Study of prostaglandin F2 alpha action, synchronizing the estrus, in experiments on heifers demonstrated that Estrumeit (made by ICI, Great Britain), rostaglandin F2 alpha drug, in a dose of 500 micrograms was an active luteolytic factor, leading to the corpus luteum regress, determined by a fall in the progesteron level in the peripheral blood. During the estrus induction there was a short-term elevation in production and excretion of testosterone, this pointing to the prostoglandins participation in the activation of the androgenic ovarian function. Prolactin hyposecretion was shown to be a necessary condition for the realization of general biological processes for the occurrence of estrus.

ABSTRACT

A method is described for production of occlusive myocardial infarction in mice. The dynamics of development as well as the morphological changes in ischaemized myocardium of mice, large laboratory animals, and humans were identical. Polarization microscopy helped reveal myocardiocytic changes in the ischaemic zone already within the first minutes after coronary artery ligation. The method reported upon has the following advantages: feasibility of all stages of the operation by a single operator, low postoperative mortality of the animals and their modest requirements for postoperative care, and low consumption of isotopes and costly therapeutic remedies.

ABSTRACT

Extraction and quantitative determination of progesterone, testosterone and oestrogens have been made in tissues of cephalopod molluscs and crustaceans. Extraction was made by acetone and ethyl ether. Purification was performed by thin layer chromatography on aluminium oxide and silicagel in different systems. The quantitative assay of progesterone and oestrogens was made by fluorimetric and radioimmunological techniques, that of testosterone--by spectrophotometric determination of hydrozones and radioimmunological procedure. Progesterone and testosterone were obtained from the gonads and liver of cephalopods and from the whole body of crustaceans. During sexual maturation of the animals concentration of these hormones tends to increase. Oestrone and oestradiol were found only in the gonads of cephalopods.

ABSTRACT

A description of three operations on the heart in mice is presented: a supravalvular narrowing of the ascending aorta, banding of the apex of the heart, and ligation of the coronaries. These surgical interventions permit to obtain in mice a functional overloading and hypertrophy of the myocardium, a cardiac aneurysm and occlusive myocardial infarction.

ABSTRACT

In the androgen-sterile rats given 10-250 mug of testosterone-propionate there was observed at the early postnatal period a reduction in the LH-RP level in the hypothalamus, of the LH in the hypophysis and the blood, of the weight of the ovaries and of the corporal luteum count in them, as well as an increase in the dopamine level in the anterior portion of the hypothalamus. Klomiphen administration for 5 days in a dose of 20 mug led to the normalization of these indices and to the appearance of ovulation, but failed to restore the capacity to pregnancy. Prolonged klomiphen administration (20 mug every 3 days for a period of 1 month) also failed to restore this capacity.


PIP: 150 nonparous rats were administered 10, 25, or 250 mcg of testosterone propionate (TP) on the 1st-5th day after birth. Upon reaching puberty, the animals were given 20 mcg of clomiphene over the course of 5 days. The content of luteinizing hormone in the hypophysis was determined by Parloy's method in both the experimental animals and a control group. Postnatal administration of TP had been observed to cause a reduction in the level of LH releasing factor (RF) in the hypothalamus, of LH in the hypophysis and the blood, and in the weight of the ovaries, as well as an increase in the dopamine levels in the anterior part of the hypothalamus. The administration of clomiphene led to the normalization of these conditions and to the appearance of ovulation, but failed to restore the ability to impregnate. Prolonged administration of clomiphene also failed to restore this capacity (20 mcg every 3 days for 30 days).

ABSTRACT

Changes in the absolute number of muscle cells and nuclei in the ventricle during the 1st year of postnatal life were followed in males of the CC57BR mice. The counts were carried out in the cell suspension obtained from the fixed cardiac muscle by means of alkaline dissociation. The number of muscle cells and nuclei reliably increased during the 1st month of life and ceased to increase when the maturity was attained (2-3 months). The number of muscle cells increased twice and that of muscle nuclei almost 4 times. A comparative estimate was made of the role of proliferation and growth of muscle cells in the increase of ventricle weight with the age. The importance of individual variation of the number of muscle cells and nuclei during early postnatal ontogenesis for the study of the myocardium adaptive growth is discussed.
