ABSTRACT

eDNA metabarcoding is an emergent tool to inform aerobiome complexity, but few studies have applied this technology with real-world environmental pollen monitoring samples. Here we apply eDNA metabarcoding to assess seasonal and regional differences in the composition of airborne pollen from routine samples collected across successive years. Airborne pollen concentrations over two sampling periods were determined using a continuous flow volumetric impaction air sampler in sub-tropical (Mutdapilly and Rocklea) and temperate (Macquarie Park and Richmond), sites of Australia. eDNA metabarcoding was applied to daily pollen samples collected once per week using the rbcL amplicon. Composition and redundancy analysis of the sequence read counts were examined. The dominant pollen families were mostly consistent between consecutive years but there was some heterogeneity between sites and years for month of peak pollen release. Many more families were detected by eDNA than counted by light microscopy with 211 to 399 operational taxonomic units assigned to family per site from October to May. There were 216 unique and 119 taxa shared between subtropics (27°S) and temperate (33°S) latitudes, with, for example, Poaceae, Myrtaceae and Causurinaceae being shared, and Manihot, Vigna and Aristida being in subtropical, and Ceratodon and Cerastium being in temperate sites. Certain genera were observed within the same location and season over the two years; Chloris at Rocklea in autumn of 2017-18 (0.625, p ≤ 0.004) and 2018-19 (0.55, p ≤ 0.001), and Pinus and Plantago at Macquarie Park in summer of 2017-18 (0.58, p ≤ 0.001 and 0.53, p ≤ 0.003, respectively), and 2018-19 (0.8, p ≤ 0.003 and 0.8, p ≤ 0.003, respectively). eDNA metabarcoding is a powerful tool to survey the complexity of pollen aerobiology and distinguish spatial and temporal profiles of local pollen to a far deeper level than traditional counting methods. However, further research is required to optimise the metabarcode target to enable reliable detection of pollen to genus and species level.

ABSTRACT

Objectifs: évaluer les résultats thérapeutiques et étudier les facteurs pronostiques de la surdité brusque.Patients et méthodes: Il s'agit d'une étude rétrospective à propos de 61 patients (39 hommes et 22 femmes) pris en charge pour surdité brusque dans le service d'Otorhinolaryngologie et de Chirurgie Cervico-Faciale de l'hôpital Bourguiba de Monastir durant une période de 16 ans (2001-2016). Une analyse statistique a été réalisée afin d'identifier les facteurs influençant le pronostic de la surdité brusque. Le seuil de significativité retenu était de 5%. Résultats:L'âge moyen était de 43 ans [16-80 ans]. Le taux global de récupération auditive était de 45%. Les facteurs pronostiques selon l'analyse uni variée étaient: un antécédent d'hypoacousie controlatérale, la présence d'un vertige associé, la sévérité de la perte auditive initiale, une courbe audiométrique de type E, l'absence du réflexe stapédien et le recours à l'oxygénothérapie hyperbare (OHB). En analyse multivariée, les seuls facteurs retenus étaient une perte auditive initiale supérieure ou égale à 70 dB, la présence d'un vertige et l'absence d'un épisode infectieux précédant la survenue de la surdité. Conclusion: Notre étude a permis de retenir comme facteurs de mauvais pronostic indépendants la perte auditive initiale supérieure ou égale à 70 dB, la présence d'un vertige et l'absence d'un épisode infectieux précédant la survenue de la surdité. Ceci nous incite à une réflexion quant au protocole thérapeutique adopté dans notre service et à indiquer l'OHB, qui est normalement prescrite en cas de non réponse, en première intention, afin d'optimiser la récupération auditive.

ABSTRACT

The importance of grass pollen to the global burden of allergic respiratory disease is well established but exposure to subtropical and temperate pollens is difficult to discern. Current monitoring of airborne pollen relies on light microscopy, limiting identification of taxa to family level. This informs seasonal fluctuations in pollen aerobiology but restricts analysis of aerobiological composition. We aimed to test the utility of DNA metabarcoding to identify specific taxa contributing to the aerobiome of environmental air samples, using routine pollen and spore monitoring equipment, as well as assess temporal variation of Poaceae pollen across an entire season. Airborne pollen concentrations were determined by light microscopy over two pollen seasons in the subtropical city of Brisbane (27°32'S, 153°00E), Australia. Thirty daily pollen samples were subjected to high throughput sequencing of the plastid rbcL amplicon. Amplicons corresponded to plants observed in the local biogeographical region with up to 3238 different operational taxonomic units (OTU) detected. The aerobiome sequencing data frequently identified pollen to genus levels with significant quantitative differences in aerobiome diversity between the months and seasons detected. Moreover, multiple peaks of Chloridoideae and Panicoideae pollen were evident over the collection period confirming these grasses as the dominant Poaceae pollen source across the season. Targeted high throughput sequencing of routinely collected airborne pollen samples appears to offer utility to track temporal changes in the aerobiome and shifts in pollen exposure. Precise identification of the composition and temporal distributions of airborne pollen is important for tracking biodiversity and for management of allergic respiratory disease.

ABSTRACT

The effects of dietary marine-derived n-3 fatty acids (FA) on mare milk and mare and foal plasma FA, postpartum ovarian follicular growth and prostaglandin concentrations were evaluated. Sixty days prior to expected foaling dates, mares were assigned to one of three diets: corn oil (CORN, n = 6), docosahexaenoic acid (DHA) diet (D; 12.64 g/d, n = 7), or eicosapentaenoic acid (EPA) and DHA (ED; 8.84 g EPA, 10.43 g DHA/d, n = 7). Milk and plasma were collected for FA analysis. Follicular data were recorded through the first postpartum ovulation. Post-ovulation serial blood samples were evaluated for prostaglandin F2α metabolite (PGFM). Supplementation with DHA, or DHA plus EPA resulted in lower linoleic acid and greater EPA and DHA in mare plasma (P < 0.05), greater arachidonic acid and DHA, or EPA and DHA in milk (P < 0.05), and greater DHA, or EPA and DHA in foal plasma (P < 0.05). Days to the first postpartum ovulation was greater (P < 0.01) in ED (22.5 ± 2.1) compared to CORN (12.5 ± 2.3) and D (13.3 ± 2.3) groups. Follicular retention (≥35 mm) prior to ovulation was longer (P < 0.05) for ED (12.7 ± 1.9 d) compared to CORN (6.3 ± 2.0 d) or D (6.0 ± 2.0 d) groups. Treatment did not affect PGFM concentrations. Maternal EPA and DHA supplementation beginning in late gestation altered the FA profile of milk and mare and foal plasma, and may result in delayed ovulation in the early postpartum period.

ABSTRACT

The objective of these studies was to investigate the efficacy of an intravaginal triptorelin acetate (TA) gel as an ovulation-inducing agent in mares. In Exp. 1, 24 mares were blocked using a combination of parity and age and randomly assigned to 1 of 3 treatment groups: 5 mL TA gel (TA5), 10 mL TA gel (TA10), or 5 mL vehicle gel only (CON). Following the appearance of a follicle ≥ 25 mm, a blood sample was obtained for measurement of LH from each mare every 24 h until treatment administration. When a follicle ≥ 35 mm was observed, treatments were administered intravaginally. Following treatment, blood samples were collected for measurement of LH and ovaries were scanned via ultrasonography every 12 h until 48 h post-ovulation. Mares in both TA5 and TA10 tended ( = 0.08) to experience a brief surge in LH by 12 h post-treatment. There was a treatment by time interaction ( < 0.005). The interval from treatment to ovulation was not different between groups ( > 0.05). We hypothesized that duration of elevated LH was not sufficient to induce ovulation in most mares. In Exp. 2, 23 mares were blocked by parity/age and randomly assigned to 3 treatment groups: the CON ( = 7) and TA5 ( = 8) treatment groups remained the same, but the TA10 treatment was split into two 5-mL doses administered 24 h apart (TA5×2; = 8). Blood collection and ultrasonography occurred every 12 h on detection of a follicle ≥ 25 mm in diameter. Once a follicle ≥ 35 mm was detected, treatment was administered and ultrasonography and blood collection for measurement of LH occurred every 6 h until 48 h post-ovulation to get a more robust characterization of the effect of TA on LH and a more accurate timeframe in which ovulation was occurring. Mares in both TA5 and TA5×2 had an increase ( < 0.05) in LH by 6 h post-treatment, which was declining by 12 h post-treatment. Following the second dose in TA5×2, another rise in LH occurred, but to a lesser magnitude ( > 0.05) compared with the initial dose. Again there was a treatment by time interaction ( < 0.005) and in Exp. 2 the interval from treatment to ovulation was shorter in TA5 (61.5 ± 8.8 h) and TA5×2 (61.5 ± 9.6 h) compared with CON (123.1 ± 21.7 h; < 0.01). In Exp. 2, administration of TA gel increased LH concentrations and hastened the interval from treatment to ovulation in mares, without an advantage in the timing of ovulation noted between the 5 or 10-mL doses, or administration of two 5-mL doses given 24 h apart.

ABSTRACT

The objective of this study was to retrospectively analyze demographic variables and academic preparation of students to determine how these factors relate to student performance in ASI 521 Horse Science, an upper-level course offered in the Department of Animal Sciences and Industry (ASI) at Kansas State University (KSU). Data were collected for 264 students enrolled in the course from 2008 to 2010. Students who took the course in 2008 received greater final percentages than those who took the course in subsequent years (P = 0.0016). Females tended to receive greater percentages than males (P = 0.096). Location of origin of students did not affect percentages earned (P > 0.26). Although class standing (P = 0.35) did not affect the final percentages that students earned, transfer students received lesser final percentages in the course (P = 0.020). If students were majoring in ASI, they fared better than those in other majors (P = 0.0097), but pre-veterinary medicine students performed similarly to non-pre-veterinary students (P = 0.49). Enrollment in the equine certificate program (which requires students to complete 20 credit hours of equine coursework) did not affect percentages earned (P = 0.89) nor did completion of any individual equine class before enrolling in ASI 521 (P > 0.19). Test scores earned on the American College Testing Program standardized test during high school were not reflective of classroom performance (P = 0.51), but KSU grade point average (GPA) was highly predictive (P < 0.0001), regardless of the term for which GPA was calculated. Students in the course took an identical comprehensive test at the beginning and end of the semester, and those test scores were also predictive of final percentage earned in ASI 521 (P ≤ 0.0002). In general, students with greater GPA performed better in ASI 521, so strategies aimed at improving classroom performance may best be targeted toward students with histories of poor academic performance.

ABSTRACT

This experiment was conducted to evaluate if consumption of endophyte-infected fescue alters digital circulation in the distal thoracic limb of the horse and to assess if soundness of the hooves of horses is affected by consumption of endophyte-infected fescue. Twelve American Quarter Horses (mean initial BW 459 ± 31 kg), 6 mares and 6 geldings, were used in this 90-d study that comprised high-endophyte (E+) and low-endophyte (E-) treatment groups. Fescue seed was integrated into the E+ diet at a rate sufficient to bring total ergovaline to 200 µg/kg, and endophyte-free fescue seed was incorporated into the E- diet from d 0 to 90. From d 30 to 60, native prairie hay was replaced with high- or low-endophyte fescue hay, bringing total dietary ergovaline to 280 µg/kg (E+) and 18 µg/kg (E-). From d 61 to 90, fescue seed was ground to decrease particle size. On d 0, 30, 60, and 90, Doppler ultrasonography and thermographic imaging were used to measure the diam. of the medial palmar artery, velocity of blood flow, and surface temperature of the hoof as indicators of digital circulation. Lameness examinations were conducted on the same days. There were no consistent treatment differences observed when evaluating measurements of digital circulation. On d 60, horses in the E+ treatment group showed increased hoof sensitivity in the left limb (P = 0.02). These horses tended to have increased hoof sensitivity when both thoracic limbs were averaged (P = 0.06), and they demonstrated increased lameness during longeing (P = 0.08). Data indicated that mares may have increased digital circulation, regardless of treatment, compared with geldings (P ≤ 0.05). Heavier horses also had greater arterial diam., velocity of blood flow, and hoof temperature than lighter BW horses (P ≤ 0.05) on d 30, 60, and 90 at time points that ranged from 90 to 180 min after feeding. Although horses consuming the E+ diet demonstrated increased lameness, especially on d 60, compared with horses consuming the E- diet, the measures of digital circulation did not support the hypothesis that digital circulation was reduced. Because of observed lameness issues, limiting the access of horses to endophyte-infected fescue may be prudent.

ABSTRACT

Four of 19 patients with acute renal failure (ARF) requiring dialytic therapy were hypophosphatemic (serum phosphate, 0.8 +/- 0.8 mg/dL) during their illness. The hypophosphatemia could not be attributed to differences in the etiology of renal failure, frequency of dialysis, or residual renal function. No hypophosphatemic patient received phosphate-binding antacids. The hypophosphatemia was related to carbohydrate (CHO) providing a large percentage (greater than 79%) of total calories for a prolonged period. This complication was likely caused by tissue anabolism as well as by CHO-stimulated, insulin-induced intracellular shifts of phosphate. The greater use of parenteral fat as a caloric source and more aggressive phosphate supplementation despite the presence of ARF may help prevent this complication.

ABSTRACT

The ability of retinoic acid (RA), a potent antitumor agent, to stimulate cell-mediated cytotoxicity (CMC) in mice was investigated. Low doses of RA (5-300 micrograms/mouse/day) administered ip into C57BL/6 mice for 5 days daily or for 1--3 months three times a week before immunization in vivo or in vitro with allogeneic BALB/c S194 myeloma cells led to an enhanced cytotoxic activity of their spleen effector cells. Similarly, in a syngeneic situation injection of RA into C57BL/6 or BALB/c mice before in vitro challenge with EL 4 (C57BL/6) or S194 (BALB/c) tumor cells strongly stimulated CMC. The enhanced cytotoxic activity was effected by thymus-derived lymphocytes (T-cells) and specific for the H-2 histocompatibility antigens in the case of the allogeneic sensitization or specific for tumor antigens in the case of the syngeneic sensitization. Because RA had no effect on the effector step of CMC, RA likely enhanced the induction step of T-CMC. The action of RA was antigen-dependent, and it is therefore a true adjuvant rather than a nonspecific stimulator or polyclonal activator of cytotoxic T-cells.
