ABSTRACT

The chemical synthesis of complex oligosaccharides relies on efficient and highly reproducible glycosylation reactions. The outcome of a glycosylation is contingent upon several environmental factors, such as temperature, acidity, the presence of residual moisture, as well as the steric, electronic, and conformational aspects of the reactants. Each glycosylation proceeds rapidly and with a high yield within a rather narrow temperature range. For better control over glycosylations and to ensure fast and reliable reactions, a systematic analysis of 18 glycosyl donors revealed the effect of reagent concentration, water content, protecting groups, and structure of the glycosyl donors on the activation temperature. With these insights, we parametrize the first step of the glycosylation reaction to be executed reliably and efficiently.

ABSTRACT

Chicken is a major source of dietary protein worldwide. The dispersion and movement of chickens constitute vital indicators of their health and status. This is especially evident in Taiwanese native chickens (TNCs), a local variety which is high in physical activity when healthy. Conventionally, the dispersion and movement of chicken flocks are observed in patrols. However, manual patrolling is laborious and time-consuming. Moreover, frequent patrols increase the risk of carrying pathogens into chicken farms. To address these issues, this study proposes an approach to develop an automatic warning system for anomalous dispersion and movement of chicken flocks in commercial chicken farms. Embendded systems were developed to acquire videos of chickens from overhead view in a chicken house, in which approximately 20,000 TNCs were raised for a period of 10 wk. Each video was 5-min in length. The videos were transmitted to a remote cloud server and were converted into images. A You Only Look Once-version 7 tiny (YOLOv7-tiny) object detection model was trained to detect chickens in the images. The dispersion of the chicken flocks in a 5-min long video was calculated using nearest neighbor index (NNI). The movement of the chicken flocks in a 5-min long video was quantified using simple online and real-time tracking algorithm (SORT). The normal ranges (i.e., 95% confidence intervals) of chicken dispersion and movement were established using an autoregressive integrated moving average (ARIMA) model and a seasonal autoregressive integrated moving average with exogenous factors (SARIMAX) model, respectively. The system allows farmers to check up on the chicken farm only when the dispersion or movement values were not in the normal ranges. Thus, labor time can be saved and the risk of carrying pathogens into chicken farms can be reduced. The trained YOLOv7-tiny model achieved an average precision of 98.2% in chicken detection. SORT achieved a multiple object tracking accuracy of 95.3%. The ARIMA and SARIMAX achieved a mean absolute percentage error 3.71% and 13.39%, respectively, in forecasting dispersion and movement. The proposed approach can serve as a solution for automatic monitoring of anomalous chicken dispersion and movement in chicken farming, alerting farmers of potential health risks and environmental hazards in chicken farms.

ABSTRACT

Upper authorities have tightly controlled local budgets, especially at the administrative township level. Taiwan has been facing this particular phenomenon for a long period. This article explains how a township, before general elections, sees the choice of an improved environmental administrative work plan as a political advantage and uses simple and easy-to-use collective decision-making to assist. Then, survey residents' perceptions of implementing the new environmental policy. The research results are discussed not only as suggestions to improve the effectiveness of township public environmental policies and to respond to the needs of civilians appropriately but also to lead future research directions.

ABSTRACT

Enzymatic synthesis that is commenced by the sugar nucleotide regeneration system (SNRS) protocol can minimize 1) the consumption of exorbitant sugar nucleotides, 2) the amount of transferases required, and 3) byproduct feedback inhibition. In this study, LacNAc extensions/modifications of the N-linked mannose core were carried out efficiently with SNRS with high yields and purities on all branches in a uniform manner. In addition, we demonstrate that with SNRS, bacterial glycosyltransferases exhibit a wide acceptor tolerance for bi- and triantennary mannose core structures as substrates for target oligosaccharides. The synthesized small library of mannose core-based glycans and linear O-glycans were screened for their binding affinity against h-Siglecs 2, 4, 7, 9, 14, 15, and m-Siglec-15 to explore their structure-based binding preferences. Microarray data revealed that each Siglec showed few distinct yet overlapping specificities. An increase in branching from mono to di or tri antennary did not necessarily lead to increasing affinity. Glycans with the disialoside sequence α(2,3)α(2,8)/α(2,6)α(2,8) showed high specificity and affinity for Siglec-7, and sLex α(2,3) exhibited a strong affinity for Siglec-9. Explicit recognition of α(2,6)α(2,3)- linear and α(2,3)α(2,6)-branched glycans by Siglecs-2, 4, and 15, respectively, suggests that these structures can act as potential candidates for the further development of high-affinity ligands.

BACKGROUND: Paraquat (1,1-dimethyl-4,4-bipyridinium dichloride) is a toxic herbicide. Accidental ingestion of paraquat in animals and humans causes respiratory failure and death. AIM: To describe the radiographic features of confirmed paraquat intoxication in a group of dogs and determines whether any identified features can facilitate this diagnosis. METHODS: Eleven dogs diagnosed with paraquat intoxication were selected from two institutions between November 2014 and August 2019 comprising five males (all intact) and six females (one intact and five spayed). The mean age was 3.9 ± 2.9 (SD) years and their mean weight was 11.6 ± 5.0 kg. The tentative diagnosis was confirmed through analysis of their urine samples using a colorimetric assay (paraquat concentation 0.39 µg/ml ranging from 0.19-0.65 µg/ml), and their clinical signs were reviewed. Thoracic radiographs were evaluated for the presence of pneumomediastinum, lung patterns (interstitial or alveolar) and their locations (caudodorsal, cranioventral, diffuse, or symmetrical), subcutaneous emphysema, pneumoretroperitoneum, and pneumothorax. RESULTS: The most common clinical signs were dyspnea (11/11, 100%) and anorexia (9/11, 82%). Pneumomediastinum (10/11, 91%) and symmetrically increased lung opacity (7/11, 65%) were the most common radiographic features. Pneumothorax (3/11, 27%), pleural effusion (3/11, 27%), subcutaneous emphysema (2/11, 18%), and pneumoretroperitoneum (1/5, 20%) were the less common findings. None of the dogs survived. CONCLUSION: Pneumomediastinum and diffuse or symmetrical interstitial or alveolar lung patterns are the most common radiographic features in dogs with paraquat intoxication. CLINICAL RELEVANCE: In countries where this herbicide is not banned, paraquat intoxication should be considered if dogs with no history of trauma present with pneumomediastinum.

ABSTRACT

OBJECTIVE: To establish a nicotine intravenous self-administration rat model, and to examine, with this model, the effects of two flavoring additives, menthol and cineole, on nicotine dependence. METHODS: Thirty male Sprague-Dawley (SD) rats were included in the study. After jugular venous catheterization was performed, fixed concentration of nicotine was administered in order to train the rats and establish the rat model of intravenous self-administration groups, receiving intraperitoneal injection of menthol, cineole, and dimethyl sulfoxide (DMSO), the vehicle that was used for the control group. The rats were tested with different fixed-ratio (FR) schedules, including FR1 schedule, in which the rat received one nicotine infusion for every active nose poke, FR2 schedule, in which the rat received one nicotine infusion for every two active nose pokes, and FR5 schedule, in which the rat received one nicotine infusion for every five active nose pokes. The number of active and inactive poke responses and the number of nicotine infusion were documented accordingly. RESULTS: After 10 days of training in nicotine self-administration, the 30 rats demonstrated significant increase in the number of active poke responses and the number of nicotine infusion, which were maintained at a stable and relatively high level. The number of active poke responses was significantly higher that of inactive poke responses ( P< 0.001). The rat model of intravenous nicotine self-administration was successfully established. In the testing phase, under the FR2 schedule, the menthol group showed a reduced number of active poke responses ( P=0.020). Under the FR5 schedule, the groups showed obvious interaction between time and the number of active poke responses ( P<0.011), with the menthol group showing reduced number of active poke responses on day three ( P=0.011) and the cineole group showing rising number of active poke responses on day three ( P=0.003). The DMSO control group did not show any significant change. CONCLUSIONS: Menthol and cineole are shown to have an effect on nicotine dependence. When there is relative difficulty involved in obtaining nicotine, menthol suppresses nicotine dependence, whereas cineole enhances nicotine dependence.

ABSTRACT

Homology is a crucial concept that should be considered while conducting a comparative analysis between organisms. In particular, in the subtribe Ligeriinae, the nectar guide pattern is associated with high diversity in petal shapes and sizes. This largely limits researchers to exclusively examining the interspecific variation in nectar guide patterns on the developmentally homologous region. Thus, to solve this problem, we proposed an approach for defining a homologous region of interest (ROI) that aligns the petal image between specimens based on petal contours and vasculatures. We identified petal contours and vasculatures from the fresh petal image and its histological image through image processing. The homologous ROI was subsequently obtained by applying geometric transformation to the contour and vasculature. The qualification and quantification of nectar guide patterns were subsequently performed based on the homologous ROI. Four patterning modes, namely vascular, random, distal, and proximal, were defined for the qualitative analysis of nectar guide patterns. In the quantitative analysis, principal component (PC) analysis was applied to homologous ROIs, and the PC score of each specimen served as the trait values of nectar guide patterns. The results of the two analyses coincided, and both showed significant associations between nectar guide patterns and pollination types. The proximal mode (corresponding to PC1) and distal mode (corresponding to PC2) together showed the strongest association with pollination types. Species exhibiting the hummingbird and bee pollination types tended to recruit the distal and proximal modes, respectively. Our study conducted a comparative analysis of nectar guide patterns on the developmentally homologous region and provided a comprehensive view of the variation in the nectar guide patterns of Ligeriinae.

ABSTRACT

Objective To investigate the pollution levels of six indicator polychlorinated biphenyls (PCBs) in animal-derived foods in Hebei Province. Methods A total of 96 food samples (including beltfish, snakehead, egg, pork liver, pork and beef meat) were collected from Hebei Province in 2019. The contents of six indicator PCBs (PCB 28, PCB 52, PCB 101, PCB 138, PCB 153 and PCB180) in different food samples were determined using isotope dilution-high resolution gas chromatography-high resolution mass spectrometry (HRGC-HRMS). Results The contents of indicative PCBs in fish samples ranged from 0.12 to 0.32 ngg-1 fresh weight, while the content of PCBs in other samples ranged from 0.11 to 1.10 ng g-1 fat. PCB 138 and PCB 153 were the main compounds detected in fish samples, while PCB 28 and PCB 52 were the main compounds detected in other samples. The average estimated daily dietary intake of PCBs for residents in Hebei Province was 0.14 ng kg-1·bw-1·d-1, and the highest contribution rate was beltfish (61.52%). Conclusion The pollution levels of indicator PCBs in animal-derived foods in Hebei Province are relatively low, and the residents has lower health risks through dietary intake of indicator PCBs.

ABSTRACT

Defining and quantifying corolla traits are essential for studying corolla shape variation. Three-dimensional (3D) images of corollas contain comprehensive information regarding corolla structures and are optimal for studying corolla shapes. Conventionally, corolla traits are identified and quantified manually from 3D images. Manual identification is time consuming and labor intensive. In this study, approaches are proposed to automatically identify first-order veins and corolla contours in 3D corolla images. The first-order veins of the corollas were identified using Hessian of Gaussian and Dijkstra's algorithm. The contours of the corollas were identified using vector harmony and node distance thresholding. A total of 130 3D images of 28 species in the subtribe Ligeriinae were collected and used to test the proposed approaches. The successful detection rate reached 86.54%. Two derived traits, contour-vein ratio and corolla angle, were defined and quantified using the first-order veins and corolla contour results to investigate the relationship between corolla shapes and pollination types of the subtribe Ligeriinae. Analyses revealed that the mean corolla contour, mean absolute corolla angle, and mean contour-vein ratio of the ornithophilic species were significantly smaller compared with the other species. The mean corolla contour, mean corolla angle, and mean contour-vein ratio of the melittophilic species were significantly larger compared with those of the ornithophilic species. The proposed method was also applied to certain Gesneriaceae species in the subtribes Gloxiniinae, Streptocarpinae, and Didymocarpinae. The results revealed that the method could be applied to most fresh sympetalous flowers for identifying first-order veins and corolla contours.

ABSTRACT

The total synthesis of the oligosaccharide moiety of disialosyl globopentaosylceramide (DSGb5 Cer), a dominant ganglioside isolated from malignant renal cell carcinoma tissues, is reported. The synthetic strategy relies on a chemical α(2,6)-sialylation at the internal GalNAc unit of a Gb5 pentasaccharide backbone that furnishes a Neu5Acα(2,6)GalNAc-linked hexasaccharide, suitable for an enzymatic α(2,3)-sialylation of the terminal Gal residue to construct a heptasaccharide glycan. Convergent access to this key α(2,6)-sialylated hexasaccharide was also achieved through a [3+3] glycosylation building upon a Galß(1,3)[Neu5Acα(2,6)]GalNAc-based trisaccharide donor and a Gb3 acceptor. The synthetic DSGb5 glycan bearing a 6-azidohexyl aglycon at the reducing end could undergo further regioselective functionalization. This approach represents a viable chemoenzymatic method for accessing complex ganglioside glycans and should be useful for the synthesis and biological investigation of DSGb5 derivatives.

BACKGROUND: Quantification of corolla shape variations helps biologists to investigate plant diversity and evolution. 3D images capture the genuine structure and provide comprehensive spatial information. RESULTS: This study applied X-ray micro-computed tomography (µCT) to acquire 3D structures of the corollas of clade Corytholoma and extracted a set of 415 3D landmarks from each specimen. By applying the geometric morphometrics (GM) to the landmarks, the first 4 principal components (PCs) in the 3D shape and 3D form analyses, respectively, accounted for 87.86% and 96.34% of the total variance. The centroid sizes of the corollas only accounted for 5.46% of the corolla shape variation, suggesting that the evolutionary allometry was weak. The 4 morphological traits corresponding to the 4 shape PCs were defined as tube curvature, lobe area, tube dilation, and lobe recurvation. Tube curvature and tube dilation were strongly associated with the pollination type and contained phylogenetic signals in clade Corytholoma. The landmarks were further used to reconstruct corolla shapes at the ancestral states. CONCLUSIONS: With the integration of µCT imaging into GM, the proposed approach boosted the precision in quantifying corolla traits and improved the understanding of the morphological traits corresponding to the pollination type, impact of size on shape variation, and evolution of corolla shape in clade Corytholoma.

OBJECTIVE:To provide reference for pharmacists to participate in the management of chronic disease. METHODS:A total of 259 patients with chronic airway disease [included asthma and chronic obstructive pulmonary disease (COPD)] met the inclusion criteria were selected from our hospital and 5 community health care centers of medical consortium. These patients received medication safety assessment management,which was led by clinical pharmacists of our hospital with the participation of community pharmacists,including medication safety comprehensive evaluation and risk classification management, follow-up and medication guidance, integrated prescriptions checking, establishment of shared database. 1 years after the implementation,the effectiveness were evaluated by score the relatived indicators in related groups. RESULTS:After a year of the management mode practice,compared with before intervention,the patients'safety medication cognitive ability score in high-risk and low-risk group increased from(4.49±1.26)and(7.31±1.01)to(5.40±1.56)and(7.44±0.91);medication adherence score increased from(4.96±1.21)and(7.08±1.24)to(6.66±1.08)and(7.38±0.98);ACT score from asthma patients increased from (16.15±2.58)and(21.15±1.03)to(16.80±2.57)and(21.64±1.55);CAT score from COPD patients decreased from(25.51± 4.07) and (14.90 ± 3.95) to (24.20 ± 3.96) and (13.80 ± 4.08);the rate of irrational prescription effective identification and intervention by pharmacists increased from 3.6% and 1.4% to 9.4% and 7.6%,respectively. All the differences above were statistically significant (P<0.05). CONCLUSIONS:The participation of pharmacists in long-term medication safety assessment management for chronic airway disease patients can improve patients'safety medication cognitive ability,medication adherence, disease control and the pharmacists'ability of irrational drug use identification and intervention.

ABSTRACT

Micrometer sized oxidation patterns were created in chemical vapor deposition grown graphene through scanning probe lithography (SPL) and then subsequently reduced by irradiation using a focused x-ray beam. Throughout the process, the films were characterized by lateral force microscopy, micro-Raman and micro-x-ray photoelectron spectroscopy. Firstly, the density of grain boundaries was found to be crucial in determining the maximum possible oxygen coverage with SPL. Secondly, the dominant factor in SPL oxidation was found to be the bias voltage. At low voltages, only structural defects are formed on grain boundaries. Above a distinct threshold voltage, oxygen coverage increased rapidly, with the duration of applied voltage affecting the final oxygen coverage. Finally, we found that, independent of initial conditions, types of defects or the amount of SPL oxidation, the same set of coupled rate equations describes the reduction dynamics with the limiting reduction step being C-C â†’ C=C.

This study used three-dimensional (3D) micro-computed tomography (µCT) imaging to examine petal form variation in a hybrid cross of Sinningia speciosa between a cultivar with actinomorphic flowers and a variety with zygomorphic flowers. The major objectives were to determine the genotype-phenotype associations between the petal form variation and CYCLOIDEA2-like alleles in S. speciosa (SsCYC) and to morphologically investigate the differences in petal types between actinomorphic and zygomorphic flowers. In this study, µCT was used to accurately acquire 3D floral images. Landmark-based geometric morphometrics (GM) was applied to evaluate the major form variations of the petals. Nine morphological traits of the petals were defined according to the form variations quantified through the GM analysis. The results indicated that the outward curvature of dorsal petals, the midrib asymmetry of lateral petals, and the dilation of ventral region of the tube were closely associated with the SsCYC genotype. Multiple analyses of form similarity between the petals suggested that the dorsal and ventral petals of actinomorphic plants resembled the ventral petals of zygomorphic plants. This observation indicated that the transition from zygomorphic to actinomorphic flowers in S. speciosa might be caused by the ventralization of the dorsal petals. We demonstrated that the 3D-GM approach can be used to determine genotype-phenotype associations and to provide morphological evidence for the transition of petal types between actinomorphic and zygomorphic flowers in S. speciosa.

Objective:To investigate the changes of expression levels of TLR9 and IL-6 in the periodontal tissue during the experimental tooth movement of the rats, and the effecst of the MT01 on the expression of TLR9,TRAF6 and IL-6 in periodontal tissue,and to clarify its related mechanisms.Methods:Thirty Wistar rats were randomly divided into MT01 intervention group(n=6) and non-MT01 group(n=24).Force of 0.49 N was applied to move the upper first molars mesially. The rats in Non-MT01 intervention group were sacrificed on the days 3,7,14 and 21, and the rats in MT01 intervention group were all sacrificed on the day 7. RT-qPCR was used to detect the expression levels of TLR9,TRAF6 and IL-6 mRNA in maxillary first molar alveolar bone in each group.Results:The expression levels of IL-6 and TLR9 mRNA in loaded side were higher than those in control side(P<0.05), and reached the maximum levels on the day 7(P<0.01);with the interpose of MT01, the expression levels of TLR9 and TRAF6 mRNA were lower than control side(P<0.01).Conclusion: MT01 could down-regulate the expression levels of TLR9 and TRAF6 during orthodontic tooth movement and eventually resists the inflammation during the tooth movement.

The quantification of floral shape variations is difficult because flower structures are both diverse and complex. Traditionally, floral shape variations are quantified using the qualitative and linear measurements of two-dimensional (2D) images. The 2D images cannot adequately describe flower structures, and thus lead to unsatisfactory discrimination of the flower shape. This study aimed to acquire three-dimensional (3D) images by using microcomputed tomography (µCT) and to examine the floral shape variations by using geometric morphometrics (GM). To demonstrate the advantages of the 3D-µCT-GM approach, we applied the approach to a second-generation population of florist's gloxinia (Sinningia speciosa) crossed from parents of zygomorphic and actinomorphic flowers. The flowers in the population considerably vary in size and shape, thereby served as good materials to test the applicability of the proposed phenotyping approach. Procedures were developed to acquire 3D volumetric flower images using a µCT scanner, to segment the flower regions from the background, and to select homologous characteristic points (i.e., landmarks) from the flower images for the subsequent GM analysis. The procedures identified 95 landmarks for each flower and thus improved the capability of describing and illustrating the flower shapes, compared with typically lower number of landmarks in 2D analyses. The GM analysis demonstrated that flower opening and dorsoventral symmetry were the principal shape variations of the flowers. The degrees of flower opening and corolla asymmetry were then subsequently quantified directly from the 3D flower images. The 3D-µCT-GM approach revealed shape variations that could not be identified using typical 2D approaches and accurately quantified the flower traits that presented a challenge in 2D images. The approach opens new avenues to investigate floral shape variations.

ABSTRACT

This study involved developing a computer-aided diagnosis (CAD) system for discriminating the grades of breast cancer tumors in ultrasound (US) images. Histological tumor grades of breast cancer lesions are standard prognostic indicators. Tumor grade information enables physicians to determine appropriate treatments for their patients. US imaging is a noninvasive approach to breast cancer examination. In this study, 148 3-dimensional US images of malignant breast tumors were obtained. Textural, morphological, ellipsoid fitting, and posterior acoustic features were quantified to characterize the tumor masses. A support vector machine was developed to classify breast tumor grades as either low or high. The proposed CAD system achieved an accuracy of 85.14% (126/148), a sensitivity of 79.31% (23/29), a specificity of 86.55% (103/119), and an A Z of 0.7940.

ABSTRACT

<p><b>BACKGROUND</b>As the only active component in final treatment phase of Tip-Edge Plus technique, the activation of nickel-titanium orthodontic archwires is one of the factors that affect the torque expression. It is necessary to evaluate the mechanical properties of the nickel-titanium wire used in the final treatment phase in simulated oral environments to forecast the treatment outcomes.</p><p><b>METHODS</b>The mechanical properties of 171 thermal nickel-titanium wires of 0.35 mm (0.014-in) in diameters with different deflection of 40 mm in length were investigated with three-point bending test. The samples were divided into 2 groups: as-received and bended groups. In the bended group, samples were divided into 7 subgroups according to the amounts of deflection and named by the canine angulations (-25°, -19°, -13°, -7°, -1°, +5°, +11°). The deflection of wires was made by inserting the wires into the deep tunnel of Tip-Edge Plus brackets positioned in plaster casts with different canine angulations to mimic the use of nickel-titanium wires in the final treatment phase. Immersed the bended group in artificial saliva (pH 6.8) and preserved at 37.0°C. Eight durations of incubation were tested: 1 to 8 weeks. Three analogous samples of each group and subgroups were tested per week. Stiffness (YS:E) and the load-deflection characteristics of unloading plateau section were obtained.</p><p><b>RESULTS</b>Significant changes in specific mechanical properties were observed in long-term immersed and large deflected wires compared with as-received groups. Both immersion time and deflection affected the mechanical properties of wires in the simulated oral environment, and the two factors had synergistic effect. In groups -25°, -19° and -13°, stiffness (YS:E) increased then decreased and average plateau force and ratio of variance decreased then increased correspondingly at specific time.</p><p><b>CONCLUSIONS</b>In the final treatment phase of Tip-Edge Plus technique, the mechanical properties of nickel-titanium wire are associated with the using time and amounts of deflection and it may affect treatment outcomes. As the main reason for wire deflection, canine crown angulation plays an important role in the wire performance. It may be wise to focus on the canine crown angulations and using time in clinic with Tip-Edge Plus technique and make proper adjustment to help to make sure the treatment outcomes.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) and several fluoride products in inhibiting enamel demineralization.</p><p><b>METHODS</b>Seventy healthy premolars extracted before orthodontic treatment were divided into seven groups (A: CPP-ACP; B: CPP-ACP + fluoride mouth rinses; C: fluoride mouth rinses; D: fluoride glass ionomer protection; E: fluoride resin binder; F: fluoride varnish; G: control group). There were ten samples in each group. The teeth were dipped into an artificial caries solution ten minutes at a time, then applied separately with CPP-ACP, CPP-ACP + fluoride mouth rinses, fluoride mouth rinses, fluoride glass ionomer protection, fluoride resin binder and fluoride varnish on labial enamel. The samples were dipped into an artificial saliva solution (37°C). Then all samples were examined with atomic force microscope (AFM) at the end of first, second and third monthes.</p><p><b>RESULTS</b>At the end of the first month, surface roughness in group D [(114 ± 1) nm] was significantly lower than that in group G[(172 ± 9) nm]. At the end of the second month and the third month, significant difference was found in surface roughness between group G and the rest of groups (P < 0.05); between group B and the rest of groups; and between group C and groups (D, E and F).</p><p><b>CONCLUSIONS</b>CPP-ACP and fluoride could both inhibit enamel demineralization in vitro. Solid fluoride (groups D, E and F) had better results than liquid fluoride (group C).</p>

ABSTRACT

ABSTRACT The NSs protein of Watermelon silver mottle virus (WSMoV) was expressed by a Zucchini yellow mosaic virus (ZYMV) vector in squash. The expressed NSs protein with a histidine tag and an additional NIa protease cleavage sequence was isolated by Ni(2+)-NTA resins as a free-form protein and further eluted after sodium dodecyl sulfate-polyacrylamide gel electrophoresis for production of rabbit antiserum and mouse monoclonal antibodies (MAbs). The rabbit antiserum strongly reacted with the NSs crude antigen of WSMoV and weakly reacted with that of a high-temperature-recovered gloxinia isolate (HT-1) of Capsicum chlorosis virus (CaCV), but not with that of Calla lily chlorotic spot virus (CCSV). In contrast, the MAbs reacted strongly with all crude NSs antigens of WSMoV, CaCV, and CCSV. Various deletions of the NSs open reading frame were constructed and expressed by ZYMV vector. Results indicate that all three MAbs target the 89- to 125-amino-acid (aa) region of WSMoV NSs protein. Two indispensable residues of cysteine and lysine were essential for MAbs recognition. Sequence comparison of the deduced MAbs-recognized region with the reported tospoviral NSs proteins revealed the presence of a consensus sequence VRKPGVKNTGCKFTMHNQIFNPN (denoted WNSscon), at the 98- to 120-aa position of NSs proteins, sharing 86 to 100% identities among those of WSMoV, CaCV, CCSV, and Peanut bud necrosis virus. A synthetic WNSscon peptide reacted with the MAbs and verified that the epitopes are present in the 98- to 120-aa region of WSMoV NSs protein. The WSMoV sero-group-specific NSs MAbs provide a means for reliable identification of tospoviruses in this large serogroup.
