ABSTRACT

The ultrastructure of the liver was studied in 3 groups of dogs (5 in each) with acute experimental pancreatitis (AEP) induced by injection of bile-trypsin mixture into the pancreatic duct. Experiments were terminated after 12 h. In the group of untreated animals, severe degeneration of mitochondria, extensive autophagocytosis, hyperactivity of Kupffer cells, depletion of glycogen granules, dilatation of endoplasmic spaces and vacuolisation of cytoplasm were found. In the group treated with prostacyclin PGI2 (20 ng/kg/min) in continuous i.v. infusion for 12 h, ultrastructural alterations were less advanced. Even better results were obtained in the group additionally pretreated for 1 h with the same rate of prostacyclin infusion. The results indicate a protective effect of prostacyclin against damage to the liver in the course of AEP.

ABSTRACT

Possible protective effects of cortisone in experimental post-alcohol liver damage were evaluated in relation to dose and duration of treatment. Model examinations were undertaken in 100 rabbits divided into groups of 10. No protective effect of cortisone could be demonstrated. Treatment did not alter biochemical indices of liver function. Histologic examination revealed somewhat less extensive fatty changes following chronic alcohol exposure in animals receiving 1 mg cortisone per kilogram body weight.

ABSTRACT

The ultrastructure of the liver in acute experimental pancreatitis (AEP) in 18 dogs in three groups was investigated after 6, 12 and 24 h of the pathological process. AEP was induced by injection of an incubated mixture of bile and trypsin into the pancreatic duct. Swelling of mitochondria, depletion of glycogen, formation of autophagosomes more pronounced in later stages of AEP were evident. Complete destruction of some hepatocytes after 24 h was observed. Dilatation of Disse's spaces, activation of Kupffer cells and degranulation of mastocytes especially after 24 h were stated. Ultrastructural changes in the liver were present in 100% of the material. The severity of liver pathology was parallel to the intensity of pancreatic necrosis. The crucial stage of the damage to the liver during AEP seems to be the impairment of mitochondria.

ABSTRACT

Ethanol 48% was applied to the experimental model in 3 examined groups of rabbits in different periods of administration and differentiated daily doses on order to investigate the influence of ethanol upon the formation of morphological and biochemical indices of the liver lesion. Biochemical investigations were carried out on 3 and 7 day of the experiment and repeated at week by intervalls. All the animals were dissected and morphological examinations of the liver were carried out. Total analysis of the results obtained of the biochemical and morphological examinations of the liver in 3 analysed groups of animals proved that the biochemical index of postalcoholic lesion of the liver was a rise in guanase activity, and not in activity aminotransferases. In the investigations of fat balance, however the increase of triglycerides and cholesterol was observed. Morphological examinations showed post-alcoholic lesions of the liver, especially intensified in animals under longlasting alcohol influence, but in reducted doses.

ABSTRACT

The development of a new intima in polyester prostheses Dallon implanted into the abdominal part of dog's aorta was tested in the scanning and transmission electron microscope. It was proved that during the first month of the experiment morphoetic elements of blood appeared on the graft surface and later within 36 months smooth muscle cells, collagen fibres and small amounts of elasticin could be found. The process of endothelialization began in the 4 month and after 12 months the graft was totally covered by endothelium. The picture in the SEM especially in the central part differed from the picture of normal arterial endothelium.
