ABSTRACT

A case of a 16-year-old female with a disseminated tumor was reported six years after surgical treatment. In October 1993 the diagnosis of a bronchial carcinoid tumor was made and a left pneumonectomy was performed. No adjuvant treatment was indicated. In May 1999 a relapse was confirmed by cytology, and a treatment with rec-hIFNalpha (10 MU intramuscular, thrice/week) was indicated. Her clinical condition improved, (ECOG 2 to 0), after three months of stable disease at evaluation, up to March 2002 when she developed a progressive disease as documented by abdominal CT scan. The histological block of the primary tumor was sent to the Centre of Molecular Immunology in order to evaluate the recognition of the ganglioside molecule on the tumor by immunohistochemistry, which was informed as positive. In April 2002 we decided to begin a compassionate treatment with the vaccine NGcGM3/VSSP Montanide ISA 51 in combination with rec-hIFNalpha. Since then up to now (56 months after progression with rec-hIFNalpha alone) the patient still has stable disease. In summary, we observed very encouraging results that could support further studies in this type of patients.

ABSTRACT

Las técnicas de análisis multivariado de datos, ofrecen la posibilidad de evaluar la asociación simultánea de un conjunto de pruebas de laboratorio con determinadas enfermedades y producir funciones discriminantes compuestas de varios resultados de laboratorio, que pueden tener una asociación con la enfermedad en cuestión. Esto es superior a cada una de las pruebas de laboratorio por separado. La característica de la función discriminante cosiste en que es capaz de tomar en cuenta la tendencia del fenómeno, ya que el valor de una prueba, aunque esté dentro del rango normal, si se mueve en el sentido de la alteración, influirá en el resultado que tendrá Y (la variante dependiente)> Los estudios de laboratorio fueron hechos a más de 3 000 pacientes en el momento de su inscripción enel IOR (AU)

Se presenta el montaje de la técnica bioquímica de determinación del receptor de progesterona en útero de rata. Se analizan y discuten los resultados encontrados en la utilización de esta técnica en 87 pacientes con tumores mamarios malignos, a los que se realizó, simultáneamente, la determinación del receptor estrogénico. Se evalúa, además, el control de calidad propuesto por nuestro laboratorio para el uso sistemático de estas dos técnicas, conjuntamente, como mejor criterio bioquímico de hormonodependencia (AU)

ABSTRACT

When BHK21 cells transformed by hamster sarcoma virus are grown in the presence of 5-Bromedeoxyuridine (BUdr), several in vitro properties of the transformed cells such as morphology, adhesiveness, and alignment, revert towards a state close to that of untransformed cells. We have studied plasma membrane changes associated with this phenotypic reversion by several different biochemical methods. Reversion is accompanied by a reappearance of Fibronectin, an increase in a membrane-associated protein of M.W. 100,000 which is increased in transformed cells and a decrease in Con A-agglutinability. On the other hand, several membrane changes associated with malignant transformation namely, the increase in an integral membrane protein M.W. 177,000, the higher rate of hexose uptake, the increase in high molecular weight surface glycopeptides and, to some extent, the increase in the density of intramembranous particles, did not revert under BUdr treatment. Thus, membrane properties of transformed cells may be dissociated into two main groups by BUdr treatment. In addition, the exposure and glycosylation of a growth-regulated membrane protein M.W. 160,000 was highly sensitive to BUdr.

ABSTRACT

We have recently described in BHK cells a plasma membrane protein of molecular weight 177,000, which is significantly increased in Hamster Sarcoma Virus-transformed cells (Lage-Davila, A. and Montagnier, L. (1977) Biochem. Biophys. Res. Commun. 79, 577--584). We present now a study of proteins from purified plasma membrane fractions in the same pair of clones. Solubilization conditions, cross-linking experiments, metabolic labelling and enzymatic radioiodination allow to characterize this 177,000 transformation-increased protein as an integral membrane glycoprotein partially exposed at the outer cell surface. Additional information on other membrane proteins in this system is also given.

ABSTRACT

Some quantitative criteria for spontaneous and virus-induced transformation of BHK21 cells are described. The resistance to acid polysaccharides such as dextran sulphate in agarose medium is a characteristic shared by all tumorigenic clones. The increase in a high molecular weight integral membrane protein seems to be a biochemical feature correlated with this property.
