ABSTRACT

The objective of this study was to evaluate the use of corpus luteum (CL) color Doppler (CD) ultrasonography and pregnancy-associated glycoproteins (PAG) for early pregnancy diagnosis and examine their ability to predict late embryonic/early fetal mortality (LEM) in Bos taurus beef replacement heifers. Beef heifers (n = 178) were exposed to a 7-d CO-Synch + CIDR protocol followed by fixed-time artificial insemination (day 0). On days 20 and 22, B-mode and CD ultrasonography were performed to evaluate CL morphometries and blood perfusion, respectively. Heifers were considered nonpregnant when CL area was <2 cm2 or estimated luteal blood perfusion was ≤30% of the total luteal area. Blood samples were collected on days 25 and 29 to estimate circulating concentrations of PAG. Conventional ultrasonography on days 29 and 94 was utilized to determine pregnancy status and considered the gold standard method for pregnancy diagnosis. Pregnant heifers had greater (P < 0.01) CL diameter, area, volume, and blood perfusion when compared with nonpregnant heifers on days 20 and 22. Accuracy of CD on days 20 and 22, and PAG on days 25 and 29 were 91%, 94%, 96%, and 98%, respectively. No false-negative results were observed for CD on both days 20 and 22 (negative predicted value = 100%) and false-positive results represented 8% and 6% of the diagnoses. Heifers that experienced LEM between days 29 and 94 of gestation had decreased luteal (P = 0.02) volume on day 20 and tended (P = 0.07) to have decreased concentrations of PAG on day 29 compared with heifers that maintained pregnancy. However, both CD and PAG failed to predict embryonic mortality. In conclusion, CD successfully detected most nonpregnant replacement heifers as early as day 20 of gestation, while resulting in no false negative diagnoses. Both CD and PAG failed to predict LEM in the present study.


Identifying nonpregnant females early after breeding allows cattle producers to rapidly rebreed females that failed to conceive after their first artificial insemination and consequently increases reproductive efficiency. Additionally, embryonic and fetal mortality during early gestation are the main drivers of pregnancy failure and represents a significant economic burden to both the beef and dairy industries. This study evaluated the use of color Doppler (CD) ultrasonography of specific ovarian structures and blood concentrations of pregnancy-associated glycoproteins (PAG) as potential methods to diagnose pregnancy earlier than industry-standard techniques. Moreover, the present study investigated the use of CD and blood PAG as predictor of pregnancy loss. Data generated here indicates that CD and blood PAG can accurately detect pregnancy as early as 20 and 25 d after insemination, respectively. In the present study, heifers that experienced pregnancy loss between days 29 and 94 of gestation tended to have decreased plasma concentrations of PAG during early pregnancy. Heifers experiencing pregnancy loss between days 29 and 94 of gestation were also more likely to have a luteal cavity on day 20 of gestation and had decreased luteal volume on the same day. However, results reported herein indicate that both CD and blood concentrations of PAG failed to predict pregnancy loss in replacement heifers.

ABSTRACT

To determine effects of delaying the injection of prostaglandin F2α (PGF) and fixed-time artificial insemination (TAI) in the 14-d CIDR-PG protocol, 1,049 Angus heifers at six locations were enrolled in a completely randomized design. Within location heifers were randomly assigned to one of two treatment groups: 1) PG16 (n = 518), heifers received a controlled internal drug release (CIDR) insert on d 0 for 14 d, a 25-mg injection of PGF 16 d after CIDR removal (d 30), and a 100-µg injection of gonadotropin-releasing hormone concurrent with TAI 66 ± 2 h later; or 2) PG17 (n = 531), heifers were treated the same as PG16, however, PGF was administered 17 d after CIDR removal (d 31), and heifers were TAI 66 ± 2 h later. Estrus detection patches were applied to a subset (n = 482) of heifers at the time of PGF administration and were examined for activation at TAI. Dominant follicle diameter was determined via transrectal ultrasonography at PGF administration and TAI in a subset of heifers (n = 116). Transrectal ultrasonography was performed to determine pregnancy rates to TAI (PR/AI) between 30 and 45 d after TAI. Estrus expression prior to TAI differed by treatment where PG17 heifers had greater (P < 0.01) expression of estrus than PG16 heifers (57.8 ± 6.1% vs. 43.4 ± 6.1%, respectively). Nevertheless, dominant follicle diameters at PGF and at TAI were similar (P ≥ 0.59) between PG16 and PG17 heifers. In addition, PR/AI did not differ (P = 0.29) between PG16 and PG17 treatments (50.5 ± 3.2% vs. 45.7 ± 3.1%, respectively). Results of this experiment indicate that delaying the injection of PGF and TAI in the 14-d CIDR-PG protocol increased estrus expression prior to TAI yet did not improve fertility in beef heifers.

ABSTRACT

An experiment was conducted during the winter of two consecutive years to evaluate the effects of feeding green-chopped cool-season forages on digestibility, ruminal fermentation, and blood parameters in beef steers. Nine ruminally cannulated Angus crossbred steers (year 1: 359 ± 79 kg; year 2: 481 ± 105 kg) received ad libitum green-chopped forages from pastures planted with one of the following mixtures: 1) OAT = Horizon 201 oats (Avena sativa L.)/Prine annual ryegrass (Lolium multiflorum Lam.) at 95 and 17 kg/ha, respectively; 2) RYE = FL401 cereal rye (Secale cereale L.)/Prine annual ryegrass (Lolium multiflorum Lam.) at 78 and 17 kg/ha, respectively; or 3) TRIT = Trical 342 triticale (X Triticosecale spp.)/Prine annual ryegrass (Lolium multiflorum Lam.) at 95 and 17 kg/ha, respectively. Intake was measured using the GrowSafe system and orts were discarded prior to subsequent feeding. After a 14-d adaptation, feed and fecal samples were collected twice daily for 4 d to determine apparent total tract nutrient digestibility using indigestible neutral detergent fiber (NDF) as an internal marker. On day 19, blood and ruminal fluid samples were collected every 3 h during a 24-h period to analyze plasma urea nitrogen (PUN) and glucose, ruminal pH, and concentration of ruminal ammonia nitrogen (NH3-N) and volatile fatty acids (VFA). Data were analyzed as a generalized randomized block design with repeated measures using the PROC MIX of SAS. No effect of treatment (P > 0.05) was observed for intake of dry matter, organic matter (OM), crude protein, NDF, or acid detergent fiber. Apparent total tract digestibility of nutrients was greater (P < 0.05) for OAT and TRIT when compared with RYE, with OM digestibility being 82.7%, 79.6%, and 69.5%, respectively. An effect of time (P < 0.01) was observed for ruminal pH. Plasma concentration of glucose was greater (P < 0.01) in steers consuming OAT, whereas steers fed RYE had greater (P < 0.05) concentrations of ruminal NH3-N and PUN, and the least concentration of total ruminal VFA (P < 0.05), despite having the greatest (P > 0.05) molar proportion of acetate, branched-chain VFA, and acetate:propionate. Increased nutrient digestibility and favorable ruminal fermentation and blood metabolites of OAT and TRIT are potentially conducive to enhanced growth performance when compared with RYE.

ABSTRACT

In Phase 1 (days 0 to 56) of a two-phase experiment, 81 steers (initial BW = 189 ± 22 kg) were arranged in a generalized randomized block design, allocated to nine pens (nine steers per pen) and assigned to one of three dietary treatments. Treatments consisted of different inclusion rates of Fermenten (FER) in a backgrounding diet fed ad libitum, and were as follows: 0% (CON), 2%, or s FER in the diet DM. Steer was considered the experimental unit, and individual DMI was measured using the GrowSafe system. In Phase 2 (days 57 to 112), treatments were discontinued and steers were relocated to a paddock with free access to a common basal diet, to assess potential residual effects of FER. Effect of treatment was measured on performance, concentration of serum urea nitrogen (SUN), NEFA and glucose, and apparent total tract digestibility of nutrients. On day 56, steers fed 4% FER had lesser (P = 0.01) BW than 2% FER steers. Steers fed CON also tended (P = 0.08) to have greater BW than 4% FER steers. ADG was not different (P = 0.96) between CON and 2% FER steers, whereas steers in the 4% FER treatment had lesser (P ≤ 0.01) ADG than CON and 2% FER. Steers in the 4% FER treatment had lesser (P = 0.04) DMI than CON, and were less feed efficient (P ≤ 0.04) than 2% FER and CON steers. However, DMI as a percentage of the BW did not differ (P = 0.07) across treatments. A treatment effect was observed for final BW on day 112 (P = 0.05), where 4% FER tended (P = 0.07) to have lesser BW than CON. In Phase 2, ADG was not different (P = 0.18) among treatments. However, overall ADG (days 0 to 112) was lesser (P = 0.02) for steers fed 4% FER when compared with CON. There was no difference (P ≥ 0.12) in longissimus muscle area gain or 12th rib fat depth gain. There was a treatment × day interaction for serum concentrations of glucose (P < 0.001), NEFA (P < 0.001), and SUN (P < 0.001). Steers fed 4% FER had greater (P < 0.001) DM, OM, NDF, ADF, and CP apparent total tract digestibility than CON. Similarly, 4% FER steers had greater (P ≤ 0.02) DM, OM, NDF, and ADF digestibility than 2% FER. Results show that although inclusion of FER increased apparent total tract digestibility of nutrients, DMI and overall performance of growing beef cattle were negatively affected, mainly when FER was included at 4% of the diet DM.
