ABSTRACT

Increasing antimicrobial resistance requires the development of novel materials and approaches for treatment of various infections. Utilization of photodynamic therapy represents an advanced alternative to antibiotics and metal-based agents. Here, we report the fabrication of electrospun material that possesses benefits of both topical antimicrobial and photodynamic therapies. This material combines chitosan, as a biocompatible polymer, and a second generation photosensitizer. The incorporation of photosensitizer doesn't affect the material morphology and its nearly uniform distribution in fibers structure was observed by confocal Raman microscopy. Owing to photosensitizer the prepared material exhibits the light-induced and spatially limited antimicrobial activity that was demonstrated against Staphylococcus aureus, an important etiological infectious agent. Such material can be potentially used in antibacterial therapy of chronic wounds, infections of diabetic ulcers, and burns, as well as rapidly spreading and intractable soft-tissue infections caused by resistant bacteria.

ABSTRACT

In the present study, Slovenian honey samples produced from different floral sources are evaluated for their antibacterial and antifungal properties. The peroxide contribution to antibacterial activity is also determined. Minimum inhibitory concentration (MIC) of the honeys was assessed against four bacterial species (Escherichia coli, Enterococcus faecalis, Pseudomonas aeruginosa and Staphylococcus aureus) and against eight fungal species (Aspergillus niger, Aureobasidium pullulans, Candida albicans, Candida parapsilosis, Candida tropicalis, Cladosporium cladosporioides, Penicillium chrysogenum and Rhodotorula mucilaginosa). Honey at concentrations between 1% and 50% (v/v) were tested. Although all of the bacterial species were inhibited by the different honey samples, the chestnut and pasture honeys showed the highest antibacterial activities. The antifungal activities were concentration-dependent, with five (Aureobasidium pullulans, Candida parapsilosis, Candida tropicalis, Cladosporium cladosporioides, Rhodotorula mucilaginosa) inhibited only at honey concentrations greater than 50%. The fungi Aspergillus niger, Candida albicans and Penicillium chrysogenum were not inhibited by any of the tested honeys, even at the highest concentrations. The lowest MICs seen were 2.5% (v/v) for the chestnut, fir and forest honeys against Staphylococcus aureus, and 10.0% (v/v) for the chestnut and pasture honeys against Cladosporium cladosporioides. The non-peroxide action of chestnut honey was tested against Escherichia coli. The MIC of the catalase-treated chestnut honey was 50% (v/v). The antibacterial effect of Slovenian honeys is mostly due to peroxide action. These data support the concept that Slovenian honeys are effective antibacterials and antifungals, and can thus be applied for medicinal purposes.

ABSTRACT

The ambient air in nine locations in a pulmonary hospital and a tuberculosis diagnostic laboratory was analysed to determine the hazard posed by Mycobacterium tuberculosis. In four locations, between 6 and 20 M. tuberculosis cell equivalents/m(3) were detected. The results indicated that inadequately ventilated corridors had the highest cell equivalents. In these areas healthcare workers were less aware of infection hazard compared to areas with known sources of M. tuberculosis and the wearing of protective masks and passive ventilation were not in place. Based on these results, further infection prevention and control measures need to be implemented.

ABSTRACT

The aim of our work was to assess the pollution-induced community tolerance (PICT) of isopod gut microbiota and pollution-induced isopod population tolerance (PIPT). Animals collected from a chronically Hg polluted and an unpolluted location were exposed for 14 days to 10microg Hg/g dry food under laboratory conditions. The lysosomal membrane stability, hepatopancreas epithelium thickness, feeding activity and animal bacterial gut microbiota composition were determined. The results confirm the hypothesis that the response to short-term Hg exposure differs for animals from the Hg polluted and the unpolluted field locations. The animals and their gut microbiota from the Hg polluted location were less affected by Hg in a short-term feeding experiment than those from the unpolluted environment. We discuss the pollution-induced population tolerance of isopods and their gut microbiota as a measure of effects of long-term environmental pollution. The ecological consequences of such phenomena are also discussed.

ABSTRACT

Anaerobic bacteria from Porcellio scaber hindgut were identified and, subsequently, isolated using molecular approach. Phylogenetic affiliation of bacteria associated with the hindgut wall was determined by analysis of bacterial 16S rRNA gene sequences which were retrieved directly from washed hindguts of P. scaber. Sequences from bacteria related to obligate anaerobic bacteria from genera Bacteroides and Enterococcus were retrieved, as well as sequences from 'A1 subcluster' of the wall-less mollicutes. Bacteria from the genus Desulfotomaculum were isolated from gut wall and cultivated under anaerobic conditions. In contrast to previous reports which suggested the absence of anaerobic bacteria in the isopod digestive system due to short retention time of the food in the tube-like hindgut, frequent renewal of the gut cuticle during the moulting process, and unsuccessful attempts to isolate anaerobic bacteria from this environment our results indicate the presence of resident anaerobic bacteria in the gut of P. scaber, in spite of apparently unsuitable, i.e. predominantly oxic, conditions.
