ABSTRACT

The therapeutic benefits of phenolic compounds found in plants are well known. The purpose of this study was to determine the phenolic content of ten plant species used as ethnoveterinary treatments in Namibia's Omusati and Kunene regions. The plants of concern were Aloe esculenta, Fockea angustifolia, Boscia albitrunca, Combretum imberbe, Acacia nilotica, Colophospermum mopane, Acacia erioloba, Ziziphus mucronata, Ximenia americana, and Salvadora persica. An LC-MS approach was used to identify the compounds. To analyse high-resolution UPLC-UV/MS, a Waters Acquity ultra-performance liquid chromatograph (UPLC) with a photodiode array detector was connected to a Waters Synapt G2 quadrupole time-of-flight mass spectrometer (MS). The current study identified a total of 29 phenolic compounds. Flavonoids (epicatechin, (-)-Epigallocatechin, and rutin,) were the most abundant, followed by 2R, 3S-Piscidic acid. Methylisocitric acid was found in all species investigated, with the highest quantities in A. esculenta and X. americana leaf extracts. There were differences in composition and quantity of phenolic compounds in aerial and ground sections between species. The overall findings of the present study would act as a standard for subsequent investigations into the pharmacological potentials of plants species utilized as ethnoveterinary remedies. Priority should be given to isolating, purifying, and defining the active compounds responsible for these plants' activity.

ABSTRACT

INTRODUCTION: Traditional healing is often the preferred form of therapy especially in rural and resource-limited communities. The extracts of plants are used to treat many diseases such as arthritis and chronic pain. Four medicinal plant species, namely, Acokanthera oppositifolia, Plantago lanceolata, Conyza canadensis and Artemisia vulgaris used in Southern Africa to treat pain and inflammation-related diseases were selected for evaluation in laboratory-based experiments. METHODS: The selected plant species were evaluated for phytochemical content, antioxidant and anti-inflammatory activities, as well as cytotoxicity effects against mammalian cells in culture. RESULTS: The results indicated that the n-hexane and chloroform extracts of P. lanceolata had the best antioxidant activities with an IC50=0.41 µg/mL. Also, the acetone extracts of P. lanceolata had 93.76% nitric oxide (NO) inhibition. However, the chloroform and n-hexane extracts of C. canadensis produced NO inhibition of 98.53% and 99.2%, respectively, at 100 µg/mL with IC50=17.69 µg/mL. Furthermore, the ethyl acetate extracts also had promising NO inhibitory activity (96.33%), but the cytotoxicity results with cell viabilities of 5.31%, 5.7% and 5.89%, respectively, suggested that the observed activity was due to a cytotoxic effect. Acetone extracts of C. canadensis were also cytotoxic at 30 µg/mL with 6.07-6.67% cell viabilities compared with the acetone extracts of P. lanceolata (99.57%). CONCLUSION: The results partially validate the ethnomedicinal uses of the selected plant species used for inflammation-related conditions. However, because some of the extracts had potential cytotoxic effects, caution is advised in their use, especially those consumed orally.

ABSTRACT

Mycotoxins are toxic secondary metabolites produced by filamentous fungi that may contaminate food and pose a health risk, especially in developing countries, where there is a lack of food security and quality is subsumed by food insufficiency. Aflatoxins are the most toxic known mycotoxins and are a significant risk factor for liver and kidney cancer, teratogenicity, undernutrition, and micronutrient malabsorption in both humans and animals. The main aim of the study was to determine the extent of fungal and aflatoxin contamination in peanuts and peanut butter being sold in both the formal and informal markets in Bulawayo, Zimbabwe. Eighteen peanut samples and 11 peanut butter samples were purchased from retail shops and the informal market. Fungal contamination was determined using standard mycology culture methods, while aflatoxin contamination was determined using high-performance liquid chromatography-fluorescence detection. Four of the six peanut samples tested for fungal contamination were infected with Aspergillus flavus/parasiticus, ranging from 3 to 20% of the kernels examined, while 27% (3 of 11) of the peanut butter samples were infected with A. flavus/parasiticus. Ninety-one percent (10 of 11) of the peanut butter samples were contaminated with aflatoxins (mean, 75.66 ng/g, and range, 6.1 to 247 ng/g), and aflatoxin B1 was the most prevalent (mean, 51.0 ng/g, and range, 3.7 to 191 ng/g). Three of the 18 peanut samples were contaminated with aflatoxins (range, 6.6 to 622 ng/g). The commercial peanut butter samples had very high aflatoxin levels, and manufacturers should be sensitized to the detrimental effects of aflatoxins and measures to reduce contamination.

ABSTRACT

The ultrastructure of the Sertoli cell of the vervet monkey was studied using both scanning and transmission electron microscopic techniques. SEM micrographs revealed perforated sleeve-like processes which encased mature elongated spermatids which are ready for spermiation. TEM micrographs showed a large Sertoli cell nucleus characterized by many lobes (4-5) and consisting of a homogenous nucleoplasm and a distinctive nucleolus. The nucleus occupies a significant portion of the basal region of the cell. The distribution of chromatin clearly shows high activity of these cells. Lipid droplets and free ribosomes are also found scattered throughout the cytoplasm. Well-developed Golgi apparatus is found in the basal region of the cell. There is phagocytic activity in the Sertoli cells as revealed by the presence of numerous phagosomes. Numerous mitochondria with well-developed tubular cristae are found on the basal side of the nucleus, whereas few mitochondria are located on the apical side of the nucleus. Distinct desmosomes are located between cells. A well-developed smooth endoplasmic reticulum and granular endoplasmic reticulum are frequently found in the cytoplasm of the Sertoli cells. The results of this investigation showed that Sertoli cells of the vervet monkey are almost similar to those of humans and show many similarities with other mammalian species.
