ABSTRACT

Significance: Photoacoustic computed tomography (PACT) is a promising non-invasive imaging technique for both life science and clinical implementations. To achieve fast imaging speed, modern PACT systems have equipped arrays that have hundreds to thousands of ultrasound transducer (UST) elements, and the element number continues to increase. However, large number of UST elements with parallel data acquisition could generate a massive data size, making it very challenging to realize fast image reconstruction. Although several research groups have developed GPU-accelerated method for PACT, there lacks an explicit and feasible step-by-step description of GPU-based algorithms for various hardware platforms. Aim: In this study, we propose a comprehensive framework for developing GPU-accelerated PACT image reconstruction (GPU-accelerated photoacoustic computed tomography), to help the research community to grasp this advanced image reconstruction method. Approach: We leverage widely accessible open-source parallel computing tools, including Python multiprocessing-based parallelism, Taichi Lang for Python, CUDA, and possible other backends. We demonstrate that our framework promotes significant performance of PACT reconstruction, enabling faster analysis and real-time applications. Besides, we also described how to realize parallel computing on various hardware configurations, including multicore CPU, single GPU, and multiple GPUs platform. Results: Notably, our framework can achieve an effective rate of ∼ 871 times when reconstructing extremely large-scale three-dimensional PACT images on a dual-GPU platform compared to a 24-core workstation CPU. In this paper, we share example codes via GitHub. Conclusions: Our approach allows for easy adoption and adaptation by the research community, fostering implementations of PACT for both life science and medicine.

ABSTRACT

The dysregulated levels of branched chain amino acids (BCAA) contribute to renal fibrosis in chronic kidney disease (CKD), yet specific analysis of BCAA contents and how they are regulated still remain unclear. It is therefore of great scientific interest to understand BCAA catabolism in CKD and develop a sensitive method for simultaneous determination of individual BCAA and their metabolites branched chain α-ketoacids (BCKA). In this work, the important role of BCAA metabolism that drives renal fibrosis in the process of CKD was first revealed by using transcriptomics. The key target genes controlling BCAA metabolism were then validated, that is, mRNA levels of BCKDHA and BCKDHB, the regulating rate-limiting enzymes during BCAA metabolism were abnormally reduced by quantitative PCR (qPCR), and a similar drop-off trend of protein expression of BCKDH, HIBCH and MCCC2 that are closely related to BCAA metabolism was also confirmed by western blotting. Furthermore, we established a novel strategy that simultaneously determines 6 individual BCAA and BCKA in serum and tissue. The method based on dansylhydrazine derivatization and ultra-high performance liquid chromatography-tandem triple quadrupole mass spectrometry (UHPLC-QQQ-MS) achieved to simultaneously determine the contents of BCAA and BCKA, which is efficient and stable. Compared with normal rats, levels of BCAA including leucine, isoleucine and valine in serum and kidney of CKD rats was decreased, while BCKA including α-ketoisocaproic acid, α-ketomethylvaleric acid and α-ketoisovaleric acid was increased. Together, these findings revealed the abnormality of BCAA metabolism in driving the course of kidney fibrosis and CKD. Our current study sheds new light on changes in BCAA metabolism during CKD, and may facilitate development of drugs to treat CKD and renal fibrosis.

ABSTRACT

Neutrophils, the most abundant immune cells in human blood, play crucial and diverse roles in tumor development. In the tumor microenvironment (TME), cancer cells regulate the recruitment and behaviors of neutrophils, transforming some of them into a pro-tumor phenotype. Pro-tumor neutrophils interact with cancer cells in various ways to promote cancer initiation, growth, and metastasis, while anti-tumor neutrophils interact with cancer cells to induce senescence and death. Neutrophils can also interact with other cells in TME, including T cells, macrophages, stromal cells, etc. to exert anti- or pro-tumor functions. In this review, we will analyze the anti- and pro-tumor intercellular interactions mediated by neutrophils, with a focus on generalizing the mechanisms underlying the interaction of neutrophils with tumor cells and T cells. Furthermore, we will provide an overview of cancer treatment strategies targeting neutrophil-mediated cellular interactions.

BACKGROUND: The temporomandibular joint diseases have been associated with various predisposing factors. Joint spaces, articular eminence height and inclination, and the shapes of the condylar and glenoid fossa have all been shown to vary in temporomandibular joint diseases (TMD) patients. Advanced imaging techniques like cone beam computed tomography (CBCT) have been employed to estimate these parameters. AIMS AND OBJECTIVES: The aim of the current study was to investigate the condylar morphology, condylar and glenoid fossa shapes, and assessment of joint spaces, such as anterior, posterior, superior, lateral, and medial spaces, through CBCT slices in coronal and sagittal planes and compare them between the control group and TMD group. MATERIALS AND METHODS: A cross-sectional study was planned where 80 joints in 40 patients were assessed for the above parameters; group I consisted of healthy patients, and group II included those with temporomandibular joint diseases (TMDs). The articular eminence height and inclination were assessed on the midsagittal section. The condylar changes and shapes of the glenoid fossa and condyles, as well as the joint spaces, were assessed on the selected coronal and sagittal sections. RESULTS: The condylar fossa had a triangular shape in the TMJ group and an oval shape in the control group. The results were highly significant (P = 0.000**). A highly significant difference in morphological parameters, such as AJS, PJS, SJS, MJS, LJS, articular eminence height, and inclination, was found between the two groups (P = 0.000**). The association of morphological parameters, such as AJS, PJS, SJS, MJS, LJS, and articular eminence height and inclination were compared with condylar and glenoid fossa shapes, where the association of superior joint space and articular eminence inclination was observed. A highly significant difference was noted between the two groups with regard to all the parameters with P=0.00*. CONCLUSION: The articular eminence inclination, as well as the superior joint space, were found to be associated with the glenoid and condyle fossa shapes in the TMJ group. These observations would, therefore, help in the early diagnosis of temporomandibular joint diseases.

ABSTRACT

By combining optical absorption contrast and acoustic resolution, photoacoustic imaging (PAI) has broken the barrier in depth for high-resolution optical imaging. Meanwhile, Fluorescence imaging (FLI), owing to advantages of high sensitivity and high specificity with abundant fluorescence agents and proteins, has always been playing a key role in live animal studies. Based on different optical contrast mechanisms, PAI and FLI can provide important complementary information to each other. In this work, we uniquely designed a Photoacoustic-Fluorescence (PA-FL) imaging system that provides real-time dual modality imaging, in which a half-ring ultrasonic array is employed for high quality PA tomography and a specially designed optical window allows simultaneous whole-body fluorescence imaging. The performance of this dual modality system was demonstrated in live animal studies, including real-time monitoring of perfusion and metabolic processes of fluorescent dyes. Our study indicates that the PA-FL imaging system has unique potential for live small animal research.

ABSTRACT

Significance: Various peripheral vascular diseases (PVD) in extremities, such as arterial atherosclerosis or venous occlusion in arm or legs, are a serious global health threat. Noninvasive vascular imaging is of great value for both diagnosis and assessment of PVD. Approach: By scanning a one-dimensional non-focusing linear array, an equivalent large two-dimensional (2D) matrix array with hundreds of thousands or more ultrasound elements is formed, thereby achieving a wide signal reception angle as well as large imaging area for three-dimensional (3D) imaging of peripheral extremities. Aim: To provide a feasible bedside and noninvasive imaging method for vascular imaging in extremities. Results: Our system can achieve high-quality photoacoustic (PA) peripheral vessel imaging. The 3D subcutaneous vascular imaging results of the palms and arms of healthy volunteers demonstrate the superior performance of the system. Conclusions: This work proposes a clinically oriented PA 3D subcutaneous vascular imaging system for human extremities. The system employs a synthetic matrix array via scanning a one-dimensional non-focusing linear probe, providing noninvasive, high-resolution, and high-contrast images of human extremities. It has potential application value in the diagnosis and monitoring of vascular diseases.

ABSTRACT

The integrated analysis of host metabolome and intestinal microbiome is an opportunity to explore the complex therapeutic mechanisms of traditional Chinese medicines. Currently, researchers mainly employ various statistical correlation analytical methods to investigate metabolome-microbiome correlations. However, these conventional correlation techniques often focus on statistical correlations and their biological meanings are always ignored, especially the functional relevance between them. Here, we developed a novel enzyme-based functional correlation (EBFC) algorithm to further improve the interpretability and the identified scope of microbe-metabolite correlations based on the conventional Spearman's analysis. The proposed EBFC algorithm is successfully utilized to reveal the therapeutic mechanisms of Jian-Pi-Yi-Shen (JPYS) formula on the treatment of adenine-induced chronic kidney disease (CKD) rats. JPYS, a TCM formula for treating CKD, has beneficial clinical effects. We tentatively revealed the potential mechanism of JPYS for treating CKD rats from the perspective of the serum metabolome, gut microbiome, and their interactions. Specifically, 11 metabolites and 19 bacterial genera in the CKD rats were significantly regulated to approaching normal status after JPYS treatment, suggesting that JPYS could ameliorate the pathological symptoms of CKD rats by reshaping the disturbed metabolome and gut microbiota. Further correlation analysis between the significantly perturbed metabolites, microbiota, and the related enzymes provided more strong evidence for the study of host metabolism-microbiota interactions and the therapeutic mechanism of JPYS on CKD rats. In conclusion, these findings will help us to deeply understand the pathogenesis of CKD and provide new insights into the therapeutic mechanism of JPYS.

Rubella virus infection can cause vertical transmission to the fetus during pregnancy. In China's Henan province, rubella surveillance needs to be well-established. In this research, a total of 1933 neonates and 2502 pregnant women were enrolled, and their sera for IgG and IgM antibodies against rubella were tested by chemiluminescence assay. Of 1933 neonates' sera tested, the seropositive of rubella IgG was 68.7%. The seroprevalence of rubella IgM in neonates was 0.4%. 30.9% of neonates had negative results for IgG and IgM antibodies. Two thousand five hundred and two pregnant women participated in the serosurvey, and 79.3% were rubella IgG positive. Rubella IgG seropositivity in pregnant women differed by age and number of births. 0.8% of the pregnant women had positive results for IgM against the rubella virus. The seronegative of rubella IgG and IgM antibodies in pregnant women was 19.8%. Due to the negative rubella-specific IgG antibody, many neonates remain at risk of rubella virus infection. Rubella virus continues to spread since some neonates and pregnant women with rubella-specific IgM antibody positive have been detected. Rubella vaccination may be introduced for childbearing-age women to increase immunity levels against rubella with periodic sero-surveillance.

ABSTRACT

Photoacoustic imaging (PAI) uniquely combines optics and ultrasound, presenting a promising role in biomedical imaging as a non-invasive and label-free imaging technology. As the traditional opaque ultrasound (US) transducers could hinder the transportation of the excitation light and limit the performance of PAI system, piezoelectric transparent ultrasonic transducers (TUTs) with indium tin oxide (ITO) electrodes have been developed to allow light transmission through the transducer and illuminate the sample directly. Nevertheless, without having transparent matching materials with appropriate properties, the bandwidth of those TUTs was generally narrow. In this work, we propose to employ polymethyl methacrylate (PMMA) as the matching layer material to improve the bandwidth of lithium niobate (LN)-based TUTs. The effects of PMMA matching layer on the performance of TUTs have been systematically studied. With the optimized PMMA matching layer, the very wide bandwidth of > 50 % could be achieved for the TUTs even with different transducer frequencies, leading to the great enhancement of axial resolution when compared to the similar reported work. In addition, the imaging performance of the developed TUT prototype has been evaluated in a PAI system and demonstrated by both phantom and in vivo small animal imaging.

ABSTRACT

ETHNOPHARMACOLOGICAL RELEVANCE: Jian-Pi-Yi-Shen (JPYS) is a herbal decoction being used to relieve the symptoms of chronic kidney disease (CKD) and its complications, including anemia, for over twenty years. Nonetheless, it is unclear how JPYS influences renal anemia and iron metabolism. AIM OF THE STUDY: An analysis of network pharmacology, chemical profiling, and in vivo experiments was conducted to identify the impact of JPYS on JAK2-STAT3 pathway and iron utilization in renal anemia and CKD. MATERIALS AND METHODS: The chemical properties of JPYS and its exposed ingredients were detected in vivo. And based on the aforesaid chemical compounds, the potential targets and signaling pathways of JPYS for renal anemia treatment were predicted by network pharmacology. Afterward, an adenine-feeding animal model of CKD-related anemia was developed to verify the mechanism by which JPYS modulates iron recycling to treat renal anemia. Renal injury was estimated by serum creatinine (Scr), blood urea nitrogen (BUN), histopathological examinations and fibrosis degree. Western blot, enzyme-linked immunosorbent assay (ELISA), quantitative real-time polymerase chain reaction (qPCR) and immunohistochemistry approaches were utilized to assess the levels of JAK2, STAT3 and iron metabolism-related factors. RESULTS: There were 164 active ingredients identified in JPYS, including prototypes and metabolites in vivo were identified in JPYS, and 21 core targets were found through network pharmacology based on topological characteristics. Combined with the core targets and pathway enrichment analysis, the majority of the candidate targets were associated with the JAK2-STAT3 signaling pathways. Experimental results indicated that JPYS treatment significantly decreased the expression of BUN and Scr, restored renal pathological damage, down-regulated fibrosis degree, and improved hematological parameters such as red blood cell, hemoglobin and hematocrit in CKD rats. Furthermore, JPYS significantly restored iron metabolism from dysregulation by increasing the levels of iron and ferritin in the serum, inhibiting the production of hepcidin in liver and serum, and regulating transferrin receptor 1 in bone marrow. Meanwhile, the expression of JAK2 and STAT3 was suppressed by JPYS treatment. CONCLUSIONS: Based on these results, JPYS reduces hepcidin levels by inhibiting the activation of JAK2-STAT3 signaling, thereby protecting against iron deficiency anemia.

ABSTRACT

Significance: Although several miniature microscope systems have been developed to allow researchers to image brain neuron activities of free moving rodents, they generally require a long cable connecting to the miniature microscope. It not only limits the behavior of the animal, but also makes it challenging to study multiple animals simultaneously. Aim: The aim of this work is to develop a fully wireless miniature microscope that would break constraints from the connecting cables so that the animals could move completely freely, allowing neuroscience researchers to study more of animals' behaviors simultaneously, such as social behavior. Approach: We present a wireless mini-microscope (wScope) that enables simultaneously real-time brain imaging preview from multiple free-moving animals. The wScope has a mass of 2.7 g and a maximum frame rate of 25 Hz at 750 µ m × 450 µ m field of view with 1.8 - µ m resolution. Results: The performance of the wScope is validated via real-time imaging of the cerebral blood flow and the activity of neurons in the primary visual cortex (V1) of different mice. Conclusions: The wScope provides a powerful tool for brain imaging of multiple free moving animals in their much larger spaces and more naturalistic environments.

OBJECTIVE: The purpose of this study was to research the morphological classification and clinical significance of vertebral artery sulcus on atlas based on CT three-dimensional reconstruction. METHODS: Three-dimensional reconstruction images of 300 adult atlases were collected. A total of 600 atlas vertebral artery sulci were selected in this study. The parameters required for placement of C1 pedicle screw, including depth of grinding drilling (ao), width (cd), length ab), height (H), lateral wall thickness (L1), inner wall thickness (L2), medial angle (∠α), and the cephalad angle to the transverse plane of atlas pedicle (∠ß), were measured. RESULTS: CT three-dimensional reconstruction images showed that there were five types of atlas vertebral artery sulci: no process type (n = 494 cases, 82.33%), upper process type (n = 29, 4.83%), lower process type (n = 25, 4.17%), double process type (n = 19, 3.17%), and posterior ring type (33, 5.50%). One-way ANOVA tests showed that the five groups differed significantly in the parameter of ao, L2, H, ∠α and ∠ß. One-way ANOVA with the LSD post hoc tests showed that the parameter ao of the group of no process type was less than that of the group of upper or lower process type (P < 0.05), and ao of the group of lower process or posterior ring type was less than that of the group of the upper type (P < 0.05). The parameter of ao of the male group was larger than that of the female group. CONCLUSION: No process type of the atlas vertebral artery sulcus was the most common, and the medial angle and cephalad angle of the atlas pedicle in this type were the smallest. When pedicle screws are inserted, the above two angles should not be too large. Male's ao was larger than that of female's. All these findings should be considered to avoid the deviation of the nail track.

Pasture biomass and quality are dependent on herbivore grazing and precipitation, but the responses of vegetation to the interactive effects of climate and grazing regimes remain unclear. We conducted an eight-year sheep grazing experiment with 4 stocking rates (0, 3.5, 5.5, and 7.5 sheep/ha) in an alpine meadow of the northeastern Tibetan Plateau. The above-ground net primary productivity (ANPP) and forage nutritional value (FNV) of four dominant species (Poa annua, Kobresia humilis, Astragalus adsurgens and Potentilla fruticosa) were measured during a wet year (360 mm rainfall) and a drought year (216 mm rainfall). The FNV was used as indicator of forage quality and was calculated from the crude protein (CP) content, in vitro true dry matter digestibility (IVTD), metabolic energy (ME) yield, and neutral detergent fiber (NDF) content of the plant. The stocking rate explained a minimum of 76% of the variations of ANPP, and the precipitation sub-additive effect for ANPP ranged from 5% to 12%. The interaction of sheep stocking rate and precipitation affected ANPP of the 4 species, except for P. fruticosa. The FNV of the pasture increased with increasing grazing pressure, but ANPP and forage nutritional yield (FNY) decreased. In calculating FNY, the increase in FNV did not compensate for the decrease in ANPP. In non-grazed plots, the CP yield declined sharply (18%-55%) in response to drought, but there was no effect on ME yield. The interaction between stocking rate and precipitation affected forage quality of the 4 plant species differently. The grassland ANPP and FNY could be maintained at a grazing intensity of 3.5 sheep/ha in wet and dry years. Our results highlight that stocking density affects pasture ANPP and FNV, and is contingent on rainfall.

ABSTRACT

Photoacoustic imaging (PAI) was performed to evaluate oxygen saturation (sO2) of blood-mimicking phantoms, femoral arteries in beagles, and radial arteries in humans at various sO2 plateaus. The accuracy (root mean square error, RMSE) of PAI sO2 compared with reference sO2 was calculated. In blood-mimicking phantoms, PAI achieved an accuracy of 1.49% and a mean absolute error (MAE) of 1.09% within 25 mm depth, and good linearity (R = 0.968; p < 0.001) was obtained between PAI sO2 and reference sO2. In canine femoral arteries, PAI achieved an accuracy of 2.16% and an MAE of 1.58% within 8 mm depth (R = 0.965; p < 0.001). In human radial arteries, PAI achieved an accuracy of 3.97% and an MAE of 3.28% in depth from 4 to 14 mm (R = 0.892; p < 0.001). For PAI sO2 evaluation at different depths in healthy volunteers, the RMSE accuracy of PAI sO2 increased from 2.66% to 24.96% with depth increasing from 4 to 14 mm. Through the multiscale method, we confirmed the feasibility of the hand-held photoacoustic/ultrasound (PA/US) in evaluating sO2. These results demonstrate the potential clinical value of PAI in evaluating blood sO2. Consequently, protocols for verifying the feasibility of medical devices based on PAI may be established.

ABSTRACT: Ischiofemoral impingement is a distinct pathologic finding with abnormal osseous contact between the ischium and the lesser trochanter of the femur. Lesser trochanter excision has been recommended for recalcitrant ischiofemoral impingement through an open or endoscopic approach; however, no study has included ischial tuberosity osteophyte resection and refixation of the hamstring tendon. We report an endoscopic procedure involving ischial tuberosity osteophyte resection with refixation of the partially detached hamstring insertion through a posterior approach in the prone position. Using this technique, it is easier to reach the lesion and less likely to injure the sciatic nerve. The postoperative pain score (visual analogy score) was significantly decreased, the modified Harris hip score increased from 39 preoperatively to 86 postoperatively, and there was no adverse effect on the hamstring tendon.

ABSTRACT

We propose and demonstrate experimentally the strong dissipative acousto-optic interaction between a suspended vibrating microfiber and a whispering-gallery microcavity. On the one hand, the dissipative response driven by an external stimulus of acoustic waves is found to be stronger than the dispersive response by 2 orders of magnitude. On the other hand, dead points emerge with the zero dissipative response at certain parameters, promising the potentials in physical sensing such as precise measurements of magnetic field and temperature. The strong dissipative acousto-optic interaction is then explored for ultrasensitive detection of broadband acoustic waves. A noise equivalent pressure as low as 0.81 Pa at 140 kHz in air is demonstrated experimentally, insensitive to cavity Q factors and does not rely on mechanical resonances.

Large-scale coal mining activities on the QTP have affected the natural grassland ecosystem which will take decades to recover. Therefore, looking for an effective vegetation restoration method is the most effective way to restore the ecosystem. In this study, we explored the effects of three vegetation restoration modes (coal mine spoils planting (CK), organic fertilizer planting (OF), and cover soil planting (CS)) on the five cold-resistance related physiological indices of three native Gramineae plants (Elymus nutans, Poa crymophila and Festuca sinensis) growing in three different habitats (shady slope, sunny slope, and flat slope). The higher contents of soluble sugar (SS) in the Elymus nutans (213.46 mg/g) and Festuca sinensis (202.84 mg/g) were recorded under the CS treatment in the sunny slope habitat. The FP contents of Elymus nutans (421.31 µg/g), Poa crymophila (310.06 µg/g), and Festuca sinens (288.45 µg/g) were higher under the CK treatment in the flat ground habitat. The higher (P < 0.05) content of chlorophyll (Chl) was recorded in the Elymus nutans (2.13 mg/g) under the CS treatment in the sunny slope habitat compared to the other two plants. We observed lower MDA contents in the Elymus nutans (3.19 nmol/g) and Festuca sinensis (3.56 nmol/g) under the CK treatment growing in the shady slope habitat. In the shady slope habitat, the H2O2 contents of Elymus nutans (15.86 µmol/g), Poa crymophila (6.78 µmol/g), and Festuca sinensis (8.76 µmol/g) under the CS treatment were significantly lower compared to other treatments. Based on membership function method, the low-temperature adaptability order of the three restoration plants from strong to weak was Elymus nutans > Festuca sinensis > Poa crymophila. While the effectiveness order of the three vegetation restoration models in alpine mining areas from good to bad was CS > OF > CK. This study will provide a theoretical and guiding significance to evaluate the effectiveness of different restoration methods, formulate optimal restoration strategies and guide the sustainable restoration of the damaged ecosystems of the alpine mining areas.

BACKGROUND: Platelet-rich plasma and concentrated growth factors have been widely utilized in tissue regeneration. However, very few studies have focused on comparing the merits of these two materials in skin repair. AIMS: We aim to compare the wound healing effects of four platelet concentrates. METHODS: Auto-platelet-rich plasma, artificial platelet-rich plasma, concentrated growth factors in a liquid phase, and a gel phase were prepared, and a full-thickness skin defect model of mice was made. The skin defects were treated with normal saline as a control and also the four kinds of platelet concentrates mentioned above, respectively. Wound size was measured and calculated on days 3, 5, 7, and 10, with histological analysis performed. RESULTS: All four platelet concentrates accelerated wound healing in mice. Interleukin-1ß (IL-1ß) and tumor necrosis factor (TNF-α) in concentrated growth factors in a liquid phase, a gel phase, and artificial platelet-rich plasma groups were significantly lower than those in the control group; and vascular endothelial growth factor (VEGF) and CD34 were significantly higher than those in the control group and auto-platelet-rich plasma group. CONCLUSIONS: All four platelet concentrates appear to promote wound healing. Compared with auto-platelet-rich plasma, concentrated growth factors in a liquid phase and in a gel phase, and artificial platelet-rich plasma seem to have more substantial effects in promoting angiogenesis, epithelialization, and reducing inflammation, thus promoting a stronger effect on wound healing.
