ABSTRACT

Ganpu vine tea is a new type of health care citrus fruit tea made from citrus shell, Pu-er tea, and vine tea baked as raw materials. In this study, the in vitro uric acid synthase inhibition system and hyperuric acid cell model were constructed to appraise the uric acid lowering efficacy of Ganpu vine tea, traditional Ganpu tea, and vine tea. Results showed that in the uric acid synthase inhibition system, the aqueous extract can inhibite the puric metabolically related enzymes, such as adenosine deaminase (ADA), purine nucleoside phosphorylase (PNP), and xanthine oxidase (XOD). The ability of the aqueous extract to inhibit the above enzyme was as follows: vine tea > Ganpu vine tea > Ganpu tea; all teas had a strong effect on XOD inhibition. The hyperuric acid cell model test showed that the aqueous extract inhibited uric acid production through accumulating inosine and hypoxanthine and hindering xanthine synthesis. The uric acid reductive ability was as follows: Vine tea > Ganpu vine tea > Ganpu tea. The inhibition of enzymes related to uric acid synthesis and the inhibition of uric acid production were significantly enhanced through adding vine tea to Ganpu tea. It also shows that flavonoids are the main factor driving this ability because they are the main active ingredients in these botanical drinks.

ABSTRACT

BACKGROUND: Angiotensin-converting enzyme 2 (ACE2) has been reported to be the main receptor for SARS-CoV-2 infection of host cells. Understanding the changes in bronchoalveolar epithelial cells after SARS-CoV-2 infection of host cells and the intercellular communication relationship between these epithelial cell changes and immune cells is of great significance for the development of therapeutic methods. METHODS: We explored the single-cell RNA sequence (scRNA-seq) of cells infected with bronchoalveolar lavage fluid (BaLF) of patients with different severities of SARS-CoV-2 and healthy people. RESULTS: We found 11 clusters of epithelial cells in the BaLF, and they were derived from the S group. In the S group, the proportion of cells with positive ACE2 expression was relatively high. ACE2 was relatively more expressed in epithelial cell clusters 1, 3, and 7. Clusters 4 and 5 represented the original state, and there were two differentiation directions: one was cluster 2, and the others were clusters 1, 3, and 6. Cluster 7 was the intermediate state. Clusters 1, 3, 6, and 7 had high similarities (> 0.9), and their main signaling pathways focused on inflammatory activation and immune response. Cluster 2 was relatively specific and was up-regulated in differential genes that were mainly related to apoptosis. The ligand-receptor expression pattern of TNFRSF10D-TNFSF10 showed a special inter-cell regulatory relationship between epithelial cell cluster 2 and macrophages. CONCLUSION: This study revealed the changes in epithelial cells derived from alveolar lavage fluid after SARS-CoV-2 infection and the communication relationship with other immune cells.

ABSTRACT

Flowering Chinese cabbage (Brassica campestris L. ssp. chinensis var. utilis Tsen et Lee) is one of the most popular vegetables in China. However, the loss of the functional ingredients in postharvest flowering Chinese cabbage during storage is still serious, owing to the unclear causes of the metabolic shifts. Herein, benzoic acid, chlorine dioxide, and 1-methylcyclopropene (1-MCP) could maintain the quality of postharvest flowering Chinese cabbage, and 1-MCP showed the best effect. Furthermore, transcript-metabolite profiling of the treatments revealed a transcript-metabolite correlation network of the flavonoid biosynthesis pathways with a range of 3 to 3662 differentially expressed genes (DEGs) and a range of 23 to 37 differentially accumulated metabolites (DAMs). Surprisingly, 1-MCP had the best effect on shelf life among the treatments, although chlorine dioxide could stimulate the expression of four critical differential genes (Bra007142, Bra008792, Bra009358, and Bra027457) involved in delaying flavonoid degradation (hesperetin, chalcone, rutin, baicalein). As a result, our findings will help to improve our understanding of the regulation of flavonoid production in relation to the quality of postharvest flowering Chinese cabbage during storage.

ABSTRACT

OBJECTIVES: To screen and verify differential genes affecting the prognosis of breast cancer. METHODS: Breast cancer gene expression datasets were downloaded from the GEO database, and original data were analyzed in R. The TIMER database was used to analyze the relationship between ANLN and UBE2T and immune cell infiltration. RESULTS: Ten hub-key genes were identified, and survival analysis showed that UBE2T and ANLN were upregulated in breast cancer and their upregulation was associated with a poor prognosis. ANLN and UBE2T upregulation was associated with the prevalence of Th1 and Th2 cells, shifting the Th1/Th2 balance to Th2 in Basal and Luminal-B breast cancers, which indicates a poor prognosis (P < 0.05). CONCLUSION: ANLN and UBE2T are potential biomarkers for predicting the prognosis of breast cancer.

In order to analyze the influence of nitroguanidine (NQ) spheroidization on the corresponding characteristics of slow cook-off molten cast explosives, experiments and simulation calculations were carried out. A calculation method was established, based on a multiphase flow model to simulate the response process of spherical NQ-based molten cast explosives under slow cook-off conditions, to analyze the temperature distribution and liquid phase distribution during the reaction process, and to discuss the reaction temperature, reaction time and reaction location with the change of solid content. The study found that the slow cook-off response level of spherical NQ-based molten cast explosives is deflagration; the phase change cloud diagram can be used to determine the ignition time to obtain more accurate slow cook-off response data; when the solid content is 50%, the ignition temperature of ordinary NQ-based molten cast explosives is 454.3 K, and the ignition time is 50.0 h, while the slow-baking ignition temperature of spherical NQ-based fused-cast explosives is up to 464 K, which is an increase of 2.14%, and the ignition time is 51.8 h, which is a relative increase of 3.55%; it can be seen that the spheroidization of NQ improves the thermal safety of molten-cast explosives has a significant effect.

ABSTRACT

The term "osteoimmunology" was coined to denote the bridge between the immune system and the skeletal system. Osteoimmunology is interdisciplinary, and a full understanding and development of this "bridge" will provide an in-depth understanding of the switch between body health and disease development. B lymphocytes can promote the maturation and differentiation of osteoclasts, and osteoclasts have a negative feedback effect on B lymphocytes. Different subtypes of T lymphocytes regulate osteoclasts in different directions. T lymphocytes have a two-way regulatory effect on osteoblasts, while B lymphocytes have minimal regulatory effects on osteoblasts. In contrast, osteoblasts can promote the differentiation and maturation of T lymphocytes and B lymphocytes. Different immune cells have different effects on chondrocytes; some cooperate with each other, while some antagonize each other. In a healthy adult body, bone resorption and bone formation are in a dynamic balance under the action of multiple mechanisms. In this review, we summarize the interactions and key signaling molecular mechanisms between each type of cell in the immune system and the skeletal system.

ABSTRACT

There is a lack of straightforward methods to prepare high-quality bismuthene nanosheets, or, even more challengingly, to grow their arrays due to the low melting point and high oxophilicity of bismuth. This synthetic obstacle has hindered their potential applications. In this work, it is demonstrated that the galvanic replacement reaction can do the trick. Under well-controlled conditions, large-area vertically aligned bismuthene nanosheet arrays are grown on Cu substrates of various shapes and sizes. The product features small nanosheet thickness of two to three atomic layers, large surface areas, and abundant porosity between nanosheets. Most remarkably, bismuthene nanosheet arrays grown on Cu foam can enable efficient CO2 reduction to formate with high Faradaic efficiency of >90%, large current density of 50 mA cm-2 , and great stability.

In this article, phosphonic acid silver-graphene oxide nanomaterials (Nano-PAS-GO) was synthesized from silver nitrate (AgNO3) solution and phosphoric graphene oxide (PGO) via the convenient ultrasonic-assisted method, and the structure and morphology were characterized, and the photocatalytic activity and recyclability were evaluated through photocatalyzing degradation of Rhodamin B (RhB) aqueous solution, and the possible photocatalytic mechanism was also discussed. Based on those, it was confirmed that Nano-PAS-GO has been synthesized from AgNO3 solution and PGO colloidal suspension under ultrasonic-assisted condition, and Nano-PAS-GO has consisted of phosphoric acid silver nanoparticles and GO with 2D lattice (2D GO lattice) connected in the form of C-P bonds, and the photodegradation rate of Nano-PAS-GO for RhB aqueous solution has reached 93.99%, and Nano-PAS-GO has possessed the nicer recyclability when the photocatalytic time was 50 min. From those results, the strong and stable interface .between PAS nanoparticles and 2D GO lattice connected in the form of the covalent bonds has effectively inhibited the occurrence of the photocorrosion phenomenon.

ABSTRACT

In this article, flake graphite, nitric acid, peroxyacetic acid and phosphoric acid are used to prepare graphene oxide phosphonic and phosphinic acids (GOPAs), and GOPAs and polyvinyl alcohol (PVA) are used to synthesize polyvinyl alcohol graphene oxide phosphonate and phosphinate (PVAGOPs) in the case of faint acidity and ultrasound irradiation, and PVAGOPs are used to fabricate PVAGOPs film, and the structure and morphology of GOPAs, PVAGOPs and PVAGOPs film are characterized, and the thermal stability and mechanical properties of PVAGOPs film are investigated. Based on these, it has been proved that GOPAs consist of graphene oxide phosphonic acid and graphene oxide phosphinic acid, and there are CP covalent bonds between them, and PVAGOPs are composed of GOPAs and PVA, and there are six-member lactone rings between GOPAs and PVA, and the thermal stability and mechanical properties of PVAGOPs film are improved effectively.

In this article, polyvinyl alcohol/phytic acid polymer (PVA/PA polymer) is synthesized from PVA and PA via the esterification reaction of PVA and PA in the case of acidity and ultrasound irradiation, and PVA/PA polymer sponge is prepared via foaming PVA/PA polymer in the presence of n-pentane and ammonium bicarbonate, and the structure of PVA/PA polymer and the structure, morphology and crystallinity of PVA/PA polymer sponge are characterized, and the thermal stability and surface resistivity of PVA/PA polymer sponge are investigated. Based on these, it has been attested that PVA/PA polymer synthesized under the acidity and ultrasound irradiation and PVA/PA polymer sponge are structured by the chain of PVA and the cricoid PA connected in the form of ether bonds and phosphonate bonds, and the thermal stability of PVA/PA polymer sponge attains 416.5 °C, and the surface resistivity of PVA/PA polymer sponge reaches 5.76 × 104 ohms/sq.

In the article, graphene oxide (GO) was prepared by flake graphite, nitric acid and peroxyacetic acid via the sonochemical method and characterized, and polyvinyl alcohol carbonate/GO composite (PVAC/GO composite) was synthesized by polyvinyl alcohol (PVA), dimethyl carbonate (DMC) and GO via the approach of transesterification in the case of ultrasonic-microwave synergistic effects and characterized, and three-dimensional PVAC/GO sponge (3D PVAC/GO sponge) was manufactured by PVAC/GO composite via the foaming approach and characterized, and the thermal stability and surface resistivity of 3D PVAC/GO sponge were investigated. Based on those, it had been attested that PVAC polymer was structured by DMC and PVA and had the six-membered lactone rings and the ether bonds, and PVAC/GO composite was constituted by 2D GO lattice and PVAC polymer, and 3D PVAC/GO sponge was constructed by PVAC/GO composite, and the surface resistivity of 3D PVAC/GO sponge with 0.00, 0.60, 1.20, 1.80 and 2.40 g of GO were 9.07  ×  107, 6.02  ×  107, 4.65  ×  107, 2.47  ×  107 and 1.06  ×  107 O/sq, and the thermal stability of 3D PVAC/GO sponge had improved.

In this paper, flake graphite, nitric acid and acetic anhydride are used to prepare graphene oxide carboxylic acid (GO-COOH) via an ultrasonic-assisted method, and GO-COOH and polyvinyl alcohol polymer (PVA) are used to synthesize graphene oxide carboxylic acid polyvinyl alcohol polymer (GO-COOPVA) via the ultrasonic-assisted method, and GO-COOPVA is used to manufacture graphene oxide carboxylic acid polyvinyl alcohol polymer film (GO-COOPVA film) via a solidification method, and the structure and morphology of GO-COOH, GO-COOPVA and GO-COOPVA film are characterized, and the thermal stability and surface resistivity are measured in the case of the different amount of GO-COOH. Based on the characterization and measurement, it has been successively confirmed and attested that carboxyl groups implant on 2D lattice of GO to form GO-COOH, and GO-COOH and PVA have the esterification reaction to produce GO-COOPVA, and GO-COOPVA consists of 2D lattice of GO-COOH and the chain of PVA connected in the form of carboxylic ester, and GO-COOPVA film is composed of GO-COOPVA, and the thermal stability of GO-COOPVA film obviously improves in comparison with PVA film, and the surface resistivity of GO-COOPVA film clearly decreases.

In this paper, polyvinyl alcohol/phytic acid polymer (PVA/PA polymer) was synthesized through esterification reaction of PVA and PA in the case of acidity and ultrasound irradiation and characterized, and PVA/PA polymer film was prepared by PVA/PA polymer and characterized, and the influence of dosage of PA on the thermal stability, mechanical properties and surface resistivity of PVA/PA polymer film were researched, and the influence of sonication time on the mechanical properties of PVA/PA polymer film was investigated. Based on those, it was concluded that the hydroxyl group on the chain of PVA and the phosphonic group on PA were connected together in the form of phosphonate bond, and the hydroxyl group on the chain of PVA were connected together in the form of ether bond after the intermolecular dehydration; in the meantime, it was also confirmed that PVA/PA polymer film prepared from 1.20mL of PA not only had the high thermal stability and favorable ductility but also the low surface resistivity in comparison with PVA/PA polymer film with 0.00mL of PA, and the ductility of PVA/PA polymer film was very sensitive to the sonication time.

In this paper, phosphorus graphene oxide/poly (vinyl alcohol) polymer (PGO/PVA polymer) was synthesized by PGO and PVA via the esterification in the case of faint acidity and the ultrasound irradiation and characterized; moreover, phosphorus graphene oxide/poly (vinyl alcohol) film (PGO/PVA film) was prepared by PGO/PVA polymer and characterized; also, the surface resistivity of PGO/PVA film was investigated in the case of the different amount of PGO. Based on those, it had been found that PGO reacted with PVA to produce PGO/PVA polymer via the esterification under the ultrasonic-assisted condition, and PGO/PVA polymer was structured by 2D lattice of PGO and the chain of PVA connected in the form of six-member lactone ring and phosphonic ester, and PGO/PVA film was constituted by PGO/PVA polymer, and surface resistivity of 0.00, 0.75, 1.50, 2.25 and 3.00wt% of PGO/PVA film were 6.85×108, 2.98×108, 1.42×106, 7.66×104 and 1.29×105Ω/sq, respectively.

OBJECTIVES: Our study aimed to investigate the impact of fatigue on the severity of stroke and to explore the underlying mechanisms. METHODS: Fatigued male rats underwent middle cerebral artery occlusion and the infarcted brain area was determined. Then, coagulation parameters were assessed in the fatigued group and a control group. In addition, the level of fibrinogen was determined in rats deprived of sleep for various numbers of days. To study whether interleukin-6 was involved in fibrinogen synthesis during fatigue, we also measured levels of interleukin-6 in rats deprived of sleep for various numbers of days. Furthermore, brain injury by middle cerebral artery occlusion was measured in wild-type mice, interleukin-6-/- mice and wild-type mice treated with bezafibrate. RESULTS: More severe cerebral infarction was observed in the fatigued rats, resulting in an infarct ratio of 23.4%. The infarct ratio was significantly increased in the fatigued rats compared with that in the control group (8%, p<0.05). The level of fibrinogen was increased significantly in the fatigued rats compared with that in the control group. In addition, a marked reduction in fibrinogen level was observed in the fatigued interleukin-6-/- mice compared to their wild-type counterparts, whereas no difference was observed between fatigued wild-type mice and interleukin-6-/- rats treated with recombinant human interleukin-6. The reduction in brain injury due to middle cerebral artery occlusion during fatigue was observed in interleukin-6-/- mice and wild-type mice treated with bezafibrate. CONCLUSION: Fatigue could increase stroke severity and was associated with the interleukin-6-induced expression of fibrinogen.
