ABSTRACT

The experiments on rats with a model toxic liver damage (tetrachloromethane hepatitis) showed evidence of a high hepatoprotector activity of liproxol, representing a combination of eplir and lokhein mixed in a 1-12 ratio. Liproxol inhibits lipid peroxidation, stimulates the excretory and detoxicant functions of liver, reduces hyperfermentation, and normalized membrane phospholipid composition.

ABSTRACT

Aggregation of erythrocytes and circulation time of polyhemoglobins in blood plasma was estimated in the model of 50% blood loss compensation in vivo and in vitro using solutions of polyhemoglobins with different content of polymerized form (from 2-3% to 70-73%). Hemoglobin derivatives were characterized by HPLC, sedimentation analysis; the main functional properties of polyhemoglobins were evaluated as well as their influence on erythrocytes aggregation in vitro and in vivo on dogs. The optimal content of polymerized form (25-30%) was estimated which provides sufficient prolongation of circulation time and does not enhance the erythrocytes aggregation. The conclusion was made that influence on erythrocyte aggregation likewise functional properties and circulation time may be considered as criterion of optimal fractional composition and biocompatibility of the polyhemoglobins.

ABSTRACT

The effectiveness of modified polyhemoglobin proposed as an artificial oxygen carrier (AOC) was evaluated in experiments on dogs with acute fatal hemorrhage. It was shown that total oxygen consumption, arterial blood pressure and circulating blood volume were maintained at the normal physiological levels during 4 h after infusion of 10% solution of polyhemoglobin. AOC infusion provided 100% survival of the animals. It has been concluded that hemoglobin polymer modified with pyridoxal-5'-phosphate can be used as the basis for the production of the medicinal form of AOC.

ABSTRACT

The efficacy of the modified polyhemoglobin suggested as an artificial oxygen transporting substitute was estimated in the model of extreme hemodilution in dogs. It was shown that O2 consumption in dogs had been supported on the normal level during 4 hours after total exchange transfusion with the polyhemoglobin solution. Index of hemodynamics testifies that preparation has hemodynamics action. The conclusion was made that this modified polyhemoglobin has been effective O2 carrying blood substitute.

ABSTRACT

Functional activity of the solutions of chemically modified hemoglobins (Hb) with different structure had been investigated during the replacement of acute fatal blood loss in dogs. It was found the correlation between polymerization degree of Hb derivatives and alterations of its oxygen-carrying characteristics in the process of circulation. It was shown that decline of functional activity at the prolonged terms of circulation was more expressed for macromolecular Hb derivatives with heterogeneous structure. Therefore chemically modified Hb free from high molecular weight fractions may be considered as a potential oxygen-carrying fluids, because they are capable of more effective support of the oxygen transport level in the organism.

ABSTRACT

The influence of the plasma substitute oxyamal on shifts in the water and acid-base balance of the blood was studied in experiments on dogs with simulated severe hemorrhagic shock. Oxyamal therapy of the severe hemorrhagic shock led to a stable reduction of the volume of circulating plasma and blood, and to a rapid normalization of blood circulation. No disorders were observed in the fluid distribution in the body, acid-base balance rapidly recovered that has evidenced a full-value transcapillary water-ion exchange.
