ABSTRACT

OBJECTIVES: The aim of this study is to determine whether new European Kidney Function Consortium (EKFC) equation is more applicable than Asian-modified CKD-EPI equation in clinical practice, having a higher accuracy in estimating GFR in our external CKD population. METHODS: We calculated estimated GFREKFC and GFRCKD-EPI independently using the EKFC and Asian-modified CKD-EPI formulas, respectively. The clinical diagnostic performance of the two equations was assessed and compared by median bias, precision, accuracy (P30) and so on, using 99mTc-DTPA dual plasma sample clearance method as a reference method for GFR measurement (mGFR). The equation that met the following targets was superior: (1) median bias within ± 3 mL/min/1.73 m2; (2) P30 > 75%; and (3) better precision and 95% limits of agreement in Bland-Altman analysis. RESULTS: Totally, 160 CKD patients were recruited in our external cohort. GFREKFC was highly related to mGFR, with a regression equation of GFREKFC=mGFR × 0.87 + 5.27. Compared with the Asian-modified CKD-EPI equation, EKFC equation demonstrated a wider median bias (-1.64 vs. 0.84 mL/min/1.73 m2, p < 0.01) that was within 3 mL/min/1.73 m2 and not clinically meaningful. Furthermore, the precision (12.69 vs. 12.72 mL/min/1.73 m2, p = 0.42), 95% limits of agreement in Bland-Altman analysis (42.4 vs. 44.4 mL/min/1.73 m2) and incorrect reclassification index of the two target equations were almost identical. Although, EKFC equation had a slightly better P30 (80.0% vs. 74.4%, p = 0.01). CONCLUSIONS: The overall performance of EKFC equation is acceptable. There is no clinically meaningful difference in the performance of the Asian-modified CKD-EPI and EKFC equations within the limits imposed by the small sample size.

ABSTRACT

In order to determine the effect of different oligosaccharides on growth performance, intestinal health, and antioxidant status of broilers, 240 1-day-old XiangHuang broilers were randomly distributed to 4 treatments with 6 replicates each. Birds were fed corn-soybean-based diets (CON), and birds in xylo-oligosaccharides (XOS), fructo-oligosaccharides (FOS), and iso-maltooligosaccharide (IMO) groups were given the basal diet supplemented with 200 mg/kg XOS, FOS, and IMO, respectively. Result showed that average daily gain (ADG) during the whole 5 weeks in FOS group was greater than that in control group (p < 0.05). Both breast and thigh muscle percentages were higher for birds fed XOS versus CON (p < 0.05). Oligosaccharides supplementation increased jejunal villus height compared with control group (p < 0.05). Malondialdehyde (MDA) concentration in breast muscle was lower for birds fed diet containing FOS versus CON (p < 0.05). Activities of total superoxide dismutase (T-SOD) in serum and thigh muscle were higher in IMO than in control group (p < 0.05). Serum T-SOD and breast muscle's glutathione peroxidase (GSH-Px) activity was higher in XOS compared with control group (p < 0.05). Conclusion, dietary oligosaccharides such as XOS, FOS, and IMO could improve intestinal health and antioxidant ability of muscle without affect growth performance in broilers.

ABSTRACT

Excitatory synapses in the mammalian brain exhibit diverse functional properties in transmission and plasticity. Directly visualizing the structural correlates of such functional heterogeneity is often hindered by the diffraction-limited resolution of conventional optical imaging techniques. Here, we used super-resolution stochastic optical reconstruction microscopy (STORM) to resolve structurally distinct excitatory synapses formed on dendritic shafts and spines. The majority of these shaft synapses contained N-methyl-d-aspartate receptors (NMDARs) but not α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptors (AMPARs), suggesting that they were functionally silent. During development, as more spine synapses formed with increasing sizes and expression of AMPARs and NMDARs, shaft synapses exhibited moderate reduction in density with largely unchanged sizes and receptor expression. Furthermore, upon glycine stimulation to induce chemical long-term potentiation (cLTP), the previously silent shaft synapses became functional shaft synapses by recruiting more AMPARs than did spine synapses. Thus, silent shaft synapse may represent a synaptic state in developing neurons with enhanced capacity of activity-dependent potentiation.
