ABSTRACT

BACKGROUND: Existing studies have found that circular RNAs (circRNAs) act as sponges for micro RNAs (miRNAs) to control downstream genes. However, the specific functionalities and mechanisms of circRNAs in human clear cell renal cell carcinoma (ccRCC) have yet to be thoroughly investigated. METHODS: Patient cohorts from online databases were used to screen candidate circRNAs, while another cohort from our hospital was obtained for validation. CircSOD2 was identified as a potential oncogenic target, and its relevant characteristics were investigated during ccRCC progression through various assays. A positive feedback loop containing downstream miRNA and its target gene were identified using bioinformatics and validated by luciferase reporter assays, RNA pull-down, and high-throughput sequencing. RESULTS: CircSOD2 expression was elevated in tumor samples and significantly correlated with overall survival (OS) and the tumor stage of ccRCC patients, which appeared in the enhanced proliferation, invasion, and migration of tumor cells. Through competitive binding to circSOD2, miR-532-3p can promote the expression of PAX5 and the progression of ccRCC, and such regulation can be salvaged by miR-532-3p inhibitor. CONCLUSION: A novel positive feedback loop, PAX5/circSOD2/miR-532-3p/PAX5 was identified in the study, indicating that the loop may play an important role in the diagnosis and prognostic prediction in ccRCC patients.

ABSTRACT

AIMS: This study aimed to identify gray/white matter volume (GMV/WMV) alterations in Internet Gaming Disorder (IGD), with a special focus on the subregions of the mesocorticolimbic dopaminergic system and their clinical association. RESULTS: Compared with healthy controls, IGDs showed bigger GMV in the bilateral caudate and the left nucleus accumbens (NAc), and bigger WMV in the inferior parietal lobule. The comparison of regions of interest (ROI) confirmed increased GMV in the bilateral caudate (including the dorsal anterior, body, and tail) and the left core of NAc in IGD, but no significant WMV alterations in the mesocorticolimbic dopaminergic system. GMVs in the left lateral orbital gyrus of orbitofrontal cortex (OFC) were associated with craving for games, while GMVs in the left anterior insula, right NAc, right caudate, and right OFC were associated with self-control in IGD. CONCLUSIONS: IGD was accompanied by changed GMV, but not WMV, in the mesocorticolimbic dopaminergic system. GMV in the mesocorticolimbic dopaminergic system may contribute to impaired self-control and craving in IGD.

ABSTRACT

Although innovative technologies for somatic cell reprogramming and transdifferentiation provide new strategies for the research of translational medicine, including disease modeling, drug screening, artificial organ development, and cell therapy, recipient safety remains a concern due to the use of exogenous transcription factors during induction. To resolve this problem, new induction approaches containing clinically applicable small molecules have been explored. Small molecule epigenetic modulators such as DNA methylation writer inhibitors, histone methylation writer inhibitors, histone acylation reader inhibitors, and histone acetylation eraser inhibitors could overcome epigenetic barriers during cell fate conversion. In the past few years, significant progress has been made in reprogramming and transdifferentiation of somatic cells with small molecule approaches. In the present review, we systematically discuss recent achievements of pure chemical reprogramming and transdifferentiation.

ABSTRACT

Background: Evidence indicates that the serum concentration of uric acid (UA) in patients may relate both to the pathophysiology and therapeutics of bipolar disorder (BPD). The purpose of this study was to examine the changes and clinical significance of serum UA concentrations in first-episode manic patients suffering from BPD. Methods: Seventy-six drug-naive patients with first-episode bipolar mania and 76 age- and gender-matched healthy subjects were recruited. Young Mania Rating Scale and Hamilton Depression Rating Scale were used to assess clinical symptoms. We tested serum UA concentrations by sandwich enzyme-linked immunosorbent assay at baseline and at the end of 8-week treatment in BPD patients and in the control group. Results: After 8-week quetiapine and sodium valproate treatment, this study revealed that the serum UA concentrations in remitted patients were significantly lower than nonremitted patients; however, those remitted patients still had higher serum UA than healthy controls. We discovered that the baseline UA concentration was higher in nonremitted than remitted patients after 8 weeks of treatment. Finally, a positive association was found between serum UA and symptom relief in the first episode of manic disorder patients. Conclusion: Patients with first-episode BPD had high levels of serum UA, which responds to treatment mainly in remitted patients. Our results suggest that serum UA concentrations might present potentially a trait marker in bipolar patients.

BACKGROUND: Improving patients' perception of social support is significant not only for their re-adaptation to life but also for alleviating caregivers' burden. AIM: This study aims to examine an integrated model regarding social support, psychotic symptoms and caregiver burden. METHODS: Persons with schizophrenia (N1 = 300) and their family caregivers (N2 = 300) in Xinjin County, Chengdu, China, completed the survey to report their demographics, patients' perception of social support (Duke Social Support Index), psychotic symptoms (Positive and Negative Syndrome Scale) and caregiver burden (Burden Scale for Family Caregivers, Short Version). Structural equation modelling was utilised to test the proposed model. RESULTS: The degree of caregiver burden differed significantly within subgroups of patients' gender and education, as well as caregivers' gender, education and employment. Caregiver burden was negatively related to patients' age and household income. Social interaction partially mediated the relationship between instrumental and subjective social support (total effect = 0.451, p < .01). Subjective social support fully mediated the impact of social interaction on psychotic symptoms (total effect = -0.099, p < .05). In the final model, instrumental social support was positively associated with social interaction (p < .001) and increased subjective social support (p < .05). Increased subjective social support showed correlation with a lower degree of psychotic symptoms (p < .01), which was related to a lower level of caregiver burden (p < .001). CONCLUSION: This study shows the associations of patients' social support with psychotic symptoms and caregiver burden. Culture-specific psychosocial interventions should be provided for both patients and caregivers to enrich external support and reduce psychotic symptoms and caregivers' burden within the health care environment.

ABSTRACT

Drug addiction is a widespread social problem, which not only brings adverse consequences to the human body, but also causes great burden to the society. However, it's still unclear how the long-term and sustained cocaine exposure will affect clock genes' expression in the reward related brain areas. We hypothesize that chronic cocaine exposure causes changes in the circadian rhythmic expression of clock genes in brain regions associated with reward, since previous studies have shown that cocaine use causes circadian disorders. Sprague-Dawley male rats were administrated with cocaine 20 mg/kg at ZT4 through intraperitoneal injection for 21 consecutive days. Twenty-four hours after the last cocaine administration brain samples were collected at 4-h intervals for 24 h (every 4h: ZT 0; ZT 4; ZT 8; ZT 12; ZT 16; ZT 20) to examine expression of rPer1, rPer2, rPer3, rCry, rBmal1 and rClock by quantitative real-time reverse-transcription polymerase chain reaction (RT-PCR). We found that chronic cocaine exposure to rats resulted in significantly disturbances in expression of clock genes in reward related brain areas compared to saline - treated rats. In cocaine-treated rats, rPer1 expression in the suprachiasmatic nucleus (SCN), prefrontal cortex (PFC) and ventral tegmental area (VTA); rPer2 expression in the nucleus accumbens (NAc) shell and hippocampus; rPer3 expression in the NAc core; rCry expression in the SCN and PFC; rBmal1 expression in the SCN and NAc core showed robust circadian rhythms that were essentially identical to those in control rats. However, robust circadian rhythm in rPer1 expression in the SCN and rCry expression in the PFC was nearly completely phase - reversed in cocaine-treated rats. A blunting of circadian oscillations of rPer1 expression occurred in the NAc core and shell and hippocampus; of rPer2 expression occurred in the SCN, PFC, NAc core and hippocampus; of rPer3 expression occurred in the SCN, PFC, NAc shell, hippocampus and VTA; of rCry expression occurred in the NAc core and shell, hippocampus and VTA; of rBmal1 expression occurred in the PFC, NAc shell, hippocampus and VTA in cocaine - treated rats. These rhythm changes accompanied by significant increase in rPer1, rPer2, rPer3 and rBmal1 in the PFC, rPer1, rPer2 and rBmal1 in the hippocampus; significant decrease in rPer2, rPer3 and rCry in the SCN, rPer3, rCry, rBmal1 and rClock in the NAc core compared to control rats. rClock expression in cocaine - treated rats showed no rhythmic change, identical to control rats.These results suggest that chronic cocaine exposure results in disturbances in clock genes' expression in reward related areas.

ABSTRACT

The aim of this study was to evaluate the prognostic value of serum and cerebrospinal fluid (CSF) free fatty acid (FFA) levels in a cohort of patients with an acute ischemic stroke (AIS). In a prospective study, FFA levels were measured using an enzyme cycling method on admission in serum and CSF of 252 consecutive patients with AIS. The prognostic value of FFA to predict the functional outcome and mortality within 90-day was compared with the National Institutes of Health Stroke Scale score and with other known outcome predictors. Serum and CSF levels of FFA increased with increasing severity of stroke as defined by the NIHSS score (all P < 0.001). Patients with an unfavorable outcomes and non-survivors had significantly increased FFA serum and CSF levels on admission (all P < 0.0001). Multivariate logistic regression analysis adjusted for common risk factors showed that serum FFA ≥0.71 mmol/L (third quarters) was an independent predictor of functional outcome (odds ratios (OR) = 4.86; 95 % confidence interval (CI) 2.26-10.48) and mortality (OR = 7.72; 95 % CI 3.01-21.48). The area under the receiver operating characteristic curve of serum FFA was 0.79 (95 % CI, 0.72-0.86) for functional outcome and 0.86 (95 % CI, 0.78-0.94) for mortality. Similarly, CSF FFA level also was an indicator for predicting of functional outcome and mortality. FFA levels in serum and CSF may serve as independent biomarkers in addition of the traditional methods for assessing the functional outcome and mortality of AIS.

ABSTRACT

The aim of this study was to investigate the potential diagnostic and prognostic role of CXC chemokine ligand-12 (CXCL12) in Chinese patients with acute ischemic stroke (AIS). All consecutive patients with first-ever AIS from January 2014 to August 2014 were recruited to participate in the study. CXCL12 and NIH Stroke Scale were measured at the time of admission. Short-term functional outcome was measured by modified Rankin scale 3 months after admission. Multivariate analyses were performed using logistic regression models. Receiver operating characteristic (ROC) curve was used to evaluate the accuracy of serum CXCL12 in diagnosing stroke and prognosing functional outcome. From 375 screened patients, a total of 288 patients with first-ever AIS were included in this study. Based on the ROC curve, the optimal cutoff value of serum CXCL12 levels as an indicator for auxiliary diagnosis of AIS was projected to be 3.5 ng/mL, which yielded a sensitivity of 88.1 % and a specificity of 73.5 %, with the area under the curve at 0.907 (95 % CI 0.882-0.932). In multivariate analysis, there was an increased risk of unfavorable outcome associated with serum CXCL12 levels ≥7.6 ng/mL (OR 4.356, 95 % CI 2.993-7.132, P < 0.0001) after adjusting for possible confounders. Our study demonstrated that elevated serum CXCL12 level at admission was an independent diagnostic and prognostic marker in patients with AIS.

ABSTRACT

OBJECTIVE: To investigate the early psychological effect of the equilibrium psychological intervention on the person who were injured in the disaster incident. METHODS: The equilibrium psychological intervention was used to the injured person in the Lushan earthquake during the early period. The GHQ-12, HAMA and HAMD were used before and after the evaluation. RESULTS: The score of the GHQ-12 decreased from (3.488 +/- 2.900) to (1.610 +/- 0.840), which showed the significant differences (P < 0.001). The total score of the HAMA and the score of the somatic anxiety factor and mental anxiety factor decreased significantly, compared with the base line (P < 0.001 respectively). The total score of the HAMD and the score of the sretardation factor, somatization factor and sleep disorder factor also decreased significantly, compared with that of the base line (P < 0.005 respectively). CONCLUSION: The equilibrium psychological intervention has the positive effect on the persons who were injured in the disaster incident during the early period.

ABSTRACT

OBJECTIVE: To assess the quality of life of children with attention deficit hyperactivity disorders (ADHD). METHODS: The PedsQL 4.0 generic core scales (Chinese Version) were administered to 73 ADHD children and 98 gender and age-matched healthy children. The parents of the children completed the Conners Parent Symptom Questionnaire. RESULTS: A total of 169 out of 171 recruited families completed the questionnaires, with a response rate of 98.8%. The ADHD children had significantly lower scores (72.7 +/- 13.0) of PedsQL 4.0 than that of the healthy children (83.7 +/- 12.0, t = -49.3, P = 0.000). The Parent Proxy-Report total score of the ADHD children (70.0 +/- 12.4) was also lower than the normal controls (82.4 +/- 11.2, t = -57.7, P = 0.000). The psychosocial health functioning of ADHD children (Children Self-Report 68.6 +/- 14.5, Parent Proxy-Report 64.9 +/- 15.4) was consistently poorer than the Physical Functioning (Children Self-Report 81.2 +/- 14.0, Parent Proxy- Report 81.7 +/- 15.6) (P = 0.000). The ADHD children had significantly higher scores in Conners Parent Symptom Questionnaire (44.54 +/- 17.89) than the normal controls (16.09 +/- 9.23, t = 100.08, P = 0.000). The PedsQL 4.0 scores were negatively correlated with school functioning scores, learning problems, hyperactivity index and the total scores of Conners Parent Symptom Questionnaire (r = -0.650, -0.630, and -0.599 respectively, P = 0.000). CONCLUSION: ADHD children suffer from poor quality of life and learning difficulties.

OBJECTIVE: To investigate the prevalence of eating disorders and its correlated factors in female students of universities, senior high schools and junior high schools. METHODS: A total of 1486 female students took part in the study. They were sampled by optimum distributing delaminating grouping method, representing the female students in 6 universities and 8 high schools in the 5 urban areas in Chengdu. Participants completed the Eating Disorder Inventory (EDI), Body Mass Index-based Silhouette Matching Test (BMI-SMT), Self-rating Depression Scale, Beck Anxiety Inventory and a self-administered questionnaire in the class time. We performed the t test and logistic regression to analysis the data. RESULTS: The estimated total prevalence of eating disorders in the female students was 3.32%. 17 years old (7.16%) was the easiest age to develop eating disorders while in senior high school (5.21%) showing the highest prevalence rate. There were differences between the eating disorders tendency group and none eating disorders tendency group in cognitive warp (P = 0.009) and expectation warp (P = 0.000) of BMI-SMT. The related factors of eating disorders were: concerning about the idol at media, non-harmonious parental relationships and being abused in childhood, body dissatisfaction factor of EDI, interoceptive awareness factor of EDI and anxiety. CONCLUSION: The total prevalence of female students' eating disorders in Chengdu was high and more attention should be paid to them on their mental health.
