ABSTRACT

Salmonella is a primary cause of foodborne diseases globally. Despite food contamination and clinical infections garnering substantial attention and research, asymptomatic Salmonella carriers, potential sources of infection, have been comparatively overlooked. In this study, we conducted a comparative analysis of serotype distribution, antimicrobial resistance phenotypes, and genetic profiles of archived Salmonella strains isolated from food (26), asymptomatic carriers (41), and clinical cases (47) in Shiyan City, China. Among the 114 Salmonella strains identified, representing 31 serotypes and 34 Sequence Types (STs), the most prevalent serovars included Typhimurium, Derby, Enteritidis, Thompson, and London, with the most predominant STs being ST11, ST40, ST26, ST34, and ST155. Antimicrobial resistance testing revealed that all strains were only sensitive to meropenem, with 74.6% showing antimicrobial resistance (AMR) and 53.5% demonstrating multidrug resistance (MDR). Strains resistant to five and six classes of antibiotics were the most common. Pearson's chi-square test showed no statistically significant difference in the occurrence of AMR (p = 0.105) or MDR (p = 0.326) among Salmonella isolates from the three sources. Our findings underscore associations and diversities among Salmonella strains isolated from food, asymptomatic carriers, and clinical patients, emphasizing the need for increased vigilance towards asymptomatic Salmonella carriers by authorities.

ABSTRACT

Air pollution in China has becoming increasingly serious in recent years with frequent incidents of smog. Parts of southwest China still experience high incidents of smog, with PM2.5 (particulate matter with diameter ≤2.5 µm) being the main contributor. Establishing the spatial distribution of PM2.5 in Southwest China is important for safeguarding regional human health, environmental quality, and economic development. This study used remote sensing (RS) and geographical information system (GIS) technologies and aerosol optical depth (AOD), digital elevation model (DEM), normalized difference vegetation index (NDVI), population density, and meteorological data from January to December 2018 for southwest China. PM2.5 concentrations were estimated using ordinary least squares regression (OLS), geographic weighted regression (GWR) and geographically and temporally weighted regression (GTWR). The results showed that: (1) Eight influencing factors showed different correlations to PM2.5 concentrations. However, the R2 values of the correlations all exceeded 0.3, indicating a moderate degree of correlation or more; (2) The correlation R2 values between the measured and remote sensed estimated PM2.5 data by OLS, GWR, and GTWR were 0.554, 0.713, and 0.801, respectively; (3) In general, the spatial distribution of PM2.5 in southwest of China decreases from the Northeast to Northwest, with moderate concentrations in the Southeast and Southwest; (4) The seasonal average PM2.5 concentration is high in winter, low in summer, and moderate in spring and autumn, whereas the monthly average shows a "V" -shaped oscillation change.

ABSTRACT

Cronobacter sakazakii (C. sakazakii) is a notorious pathogen responsible for infections in infants and newborns, often transmitted through contaminated infant formula. Despite the use of traditional pasteurization methods, which can reduce microbial contamination, there remains a significant risk of pathogenic C. sakazakii surviving due to its exceptional stress tolerance. In our study, we employed a comparative proteomic approach by comparing wild-type strains with gene knockout strains to identify the essential genes crucial for the successful survival of C. sakazakii during desiccation. Our investigation revealed the significance of envZ-ompR, recA, and flhD gene cassettes in contributing to desiccation tolerance in C. sakazakii. Furthermore, through our comparative proteomic profiling, we identified the maltodextrin-binding protein encoded by ESA_03421 as a potential factor influencing dry tolerance. This protein is regulated by EnvZ-OmpR, RecA, and FlhD. Notably, the knockout of ESA_03421 resulted in a 150% greater reduction in Log CFU compared to the wild-type C. sakazakii. Overall, our findings offer valuable insights into the mechanisms underlying C. sakazakii desiccation tolerance and provide potential targets for the development of new antimicrobial strategies aimed at reducing the risk of infections in infants and newborns.

ABSTRACT

Regional land use change and ecological security have received considerable attention in recent years. The rapid economic development of Kunming and Fuzhou has resulted in environmental damage such as water pollution and urban heat island effect. It is thus important to conduct a comparative analysis of the ecological security response to land use/land cover change (LUCC) in different natural zones. Using the Google Earth Engine (GEE) platform, random forest and support vector machine methods were used to classify land cover types in the study area, after which the ArcGIS platform was used to analyze LUCC. The driving force-pressure-state-impact-response (DPSIR) model and entropy weight method were used to construct an ecological security evaluation system, and gray correlation was used to compare the ecological security responses to LUCC in Kunming and Fuzhou. The findings revealed that: (1) The average dynamic degrees of comprehensive land use in Kunming and Fuzhou from 1995 to 2020 were 1.05% and 0.55%, respectively; (2) From 1995 to 2020, the ecological security index values for Kunming and Fuzhou increased from 0.42 to 0.52 and from 0.36 to 0.68, respectively, indicating that Fuzhou's index is rising more rapidly; and (3)There is a strong correlation between LUCC and ecological security, the correlation between the woodland and the ecological security index is very strong in both places. The expansion of construction land may be an important reason for the reduced ecological security level in Fuzhou City, while water resources have a significant impact on the ecological security level of Kunming City.

ABSTRACT

The process of complex cognition, which includes language processing, is dynamic in nature and involves various network modes or cognitive modes. This dynamic process can be manifested by a set of brain states and transitions between them. Previous neuroimaging studies have shed light on how bilingual brains support native language (L1) and second language (L2) through a shared network. However, the mechanism through which this shared brain network enables L1 and L2 processing remains unknown. This study examined this issue by testing the hypothesis that L1 and L2 processing is associated with distinct brain state dynamics in terms of brain state integration and transition flexibility. A group of late Chinese-English bilinguals was scanned using functional magnetic resonance imaging (fMRI) while listening to eight short narratives in Chinese (L1) and English (L2). Brain state dynamics were modeled using the leading eigenvector dynamic analysis framework. The results show that L1 processing involves more integrated states and frequent transitions between integrated and segregated states, while L2 processing involves more segregated states and fewer transitions. Our work provides insight into the dynamic process of narrative listening comprehension in late bilinguals and sheds new light on the neural representation of language processing and related disorders.

ABSTRACT

Intra-individual variability of neural response has been found to be negatively associated with cognitive proficiency and automaticity. However, whether developmental dyslexia (DD) is marked by greater intra-individual neural variability remains unclear. Using a multivariate approach and dual-control group design, the current study aims to examine whether the pattern similarity of brain activation during a visual spelling task is abnormal in children with DD compared to age control and reading control children. We found that there was reduced intra-subject pattern similarity at the left occipito-temporal regions in children with DD than both control groups, suggesting a neural signature of DD. Furthermore, we found that pattern similarity was positively associated with stability of reaction time and reading fluency in both children with DD and typical control children, suggesting that neural stability supports behavioral stability and automaticity during reading.

ABSTRACT

Recent studies have shown that the brain functional connectome constitutes a unique fingerprint that allows the identification of individuals from a group. However, what information encoded in the brain that makes us unique remains elusive. Here, we addressed this issue by examining how individual identifiability changed along the language hierarchy. Subjects underwent fMRI scanning during rest and when listening to short stories played backward, scrambled at the sentence level, and played forward. Identification for individuals was performed between two scan sessions for each task as well as between the rest and task sessions. We found that individual identifiability tends to increase along the language hierarchy: the more complex the task is, the better subjects can be distinguished from each other based on their whole-brain functional connectivity profiles. A similar principle is found at the functional network level: compared to the low-order network (the auditory network), the high-order network is more individualized (the frontoparietal network). Moreover, in both cases, the increase in individual identifiability is accompanied by the increase in inter-subject variability of functional connectivities. These findings advance the understanding of the source of brain individualization and have potential implications for developing robust connectivity-based biomarkers.

ABSTRACT

Exhibiting deactivation and anticorrelation with task-positive networks, the default mode network (DMN) has been traditionally thought to be suppressed to support externally oriented cognitive processes during spoken language comprehension. In contrast, recent studies examining listener-listener intersubject correlation (ISC) have proposed an active role of DMN in language comprehension. How can we reconcile those seemingly conflicting results? This study adopted a "two-brain" paradigm and combined "within-brain" and "across-brain" analyses to address this issue. We found, despite being deactivated and anticorrelated with the language network (LN) and executive control network (ECN), both the anterior and posterior DMN in the listeners' brains were temporally coupled with the homologous networks in the speaker's brain. Interestingly, the listener-speaker neural couplings persisted even after controlling for listener-listener ISC. Moreover, the coupling strength of posterior DMN positively correlated with the listeners' speech comprehension. Further dynamic causal modeling showed that the LN and ECN, the anterior DMN, and the posterior DMN occupied the bottom, intermediate, and top layers of a hierarchical system, respectively. We suggest the DMN may primarily serve as an internal module that cooperates with the externally oriented modules, potentially supporting the transformation of external acoustic signals into internal mental representations during successful language comprehension.

The engagement of the cerebellum during reading tasks is not unprecedented. However, it is still unclear which regions in the cerebellum are specifically involved in reading and how the cerebellum processes different languages. With functional magnetic resonance imaging, we compared the cerebellar neural activity in Chinese child learners of English between reading and non-reading tasks to identify functionally specialized areas for reading, and between Chinese characters and English words in a passive viewing paradigm to detect regions sensitive to different scripts. Two posterior subregions of right lobule VI, as well as right lobule VIIIA, demonstrated greater activation to viewing Chinese characters and English words compared to the non-reading tasks. However, we did not find any cerebellar regions that were differentially responsive to Chinese versus English print. Instead, we observed that functional connectivity between the two above-mentioned cerebellar regions (lobules VI and VIIIA) and the left inferior parietal lobule was significantly greater in English reading compared to Chinese reading. Overall, these results indicate that the posterior parts of right lobule VI and the right lobule VIIIA could be reading-specific regions, and deepen our understanding of how the cerebellum contributes to reading.

Using graph theory, we examined topological organization of the language network in Chinese children with poor reading during an auditory rhyming task and a visual spelling task, compared to reading-matched controls and age-matched controls. First, poor readers (PR) showed reduced clustering coefficient in the left inferior frontal gyrus (IFG) and higher nodal efficiency in the bilateral superior temporal gyri (STG) during the visual task, indicating a less functionally specialized cluster around the left IFG and stronger functional links between bilateral STGs and other regions. Furthermore, PR adopted additional right-hemispheric hubs in both tasks, which may explain increased global efficiency across both tasks and lower normalized characteristic shortest path length in the visual task for the PR. These results underscore deficits in the left IFG during visual word processing and conform previous findings about compensation in the right hemisphere in children with poor reading.

ABSTRACT

It has been revealed that dyslexic children learning alphabetic languages are characterized by aberrant topological organization of brain networks. However, little is known about the functional organization and the reconfiguration pattern of brain networks in Chinese dyslexic children. Using graph theoretical analysis and functional magnetic resonance images (fMRI), we examined this issue specifically from the perspective of functional integration and segregation. We first compared large-scale topological organizations between dyslexic children and typically developing children during a Chinese phonological rhyming task, and found that dyslexic children showed increased local efficiency and clustering coefficient compared with typically developing children, which were negatively correlated with task performance. Furthermore, dyslexic children and typically developing children could be accurately distinguished at the individual-subject level based on the nodal local efficiency or clustering coefficient. Second, we studied the group difference of network reconfiguration and found that dyslexic children showed more difficulty when shifting from the resting state to the phonological task. Our results suggest an over-segregated brain functional organization and deficits in brain network reconfiguration in Chinese dyslexic children, which helps to advance our knowledge on the neural mechanisms underlying dyslexia.

An increasing number of studies have highlighted the importance of listener-speaker neural coupling in successful verbal communication. Whether the brain-to-brain coupling changes with healthy aging and the possible role of this change in the speech comprehension of older adults remain unexplored. In this study, we scanned with fMRI a young and an older speaker telling real-life stories and then played the audio recordings to a group of young (N = 28, aged 19-27 year) and a group of older adults during scanning (N = 27, aged 53-75 year), respectively. The older listeners understood the speech less well than did the young listeners, and the age of the older listeners was negatively correlated with their level of speech understanding. Compared to the young listener-speaker dyads, the older dyads exhibited reduced neural couplings in both linguistic and extra-linguistic areas. Moreover, within the older group, the listener's age was negatively correlated with the overall strength of interbrain coupling, which in turn was associated with reduced speech understanding. These results reveal the deficits of older adults in achieving neural alignment with other brains, which may underlie the age-related decline in speech understanding.

This study aims to investigate the potential safety hazards and provide reference for improving the medical waste disposal procedure in SARS-CoV-2 testing laboratory. Our SARS-CoV-2 testing group detected the RNA residue on the surface of medical waste with Droplet Digital PCR, and held a meeting to discuss the risks in the laboratory medical waste disposal process. After effective autoclaving, SARS-CoV-2 contaminated on the surface of medical waste bags was killed, but the average concentration of viral RNA residues was still 0.85 copies/cm2. It would not pose a health risk, but might contaminate the laboratory and affect the test results. When the sterilized medical waste bags were transferred directly by the operators without hand disinfection, re-contamination would happen, which might cause the virus to leak out of the laboratory. Furthermore, we found that sterilization effect monitoring and cooperation among operators were also very important. In summary, we investigated and analyzed the potential safety hazards during the medical waste disposal process in SARS-CoV-2 testing laboratory, and provided reasonable suggestions to ensure the safety of medical waste disposal.

BACKGROUND: Aging is a global problem, and the ability assessment of the elderly plays an important role in the formulation of pension policies. It's necessary to evaluate the ability of the elderly in rural China to provide insights into future nursing care and policy making. METHODS: The elderly in 20 rural villages were selected by convenience sampling. We used "Elderly Ability Evaluation Form" issued by the Ministry of Civil Affairs of China as survey tool. The characteristics and score differences of the elderly of different ability level were compared and analyzed. And logistic regression analyses were conducted to identify the potential risk factors for disability in the elderly. RESULTS: A total of 2,878 elders were included, of which there were 1,916 elders with intact ability, 866 elders with mild disability, 42 elders with moderate disability, 54 elders with severe disability. The incidence of disability among respondents was 33.43%. There were significantly statistical differences in the dimensions of activities of daily living, mental state, perception and communication, and social participation among elders with intact ability, mild, moderate and severe disability (all P<0.05). The age, education level, marital status and living situations were all corrected to the scores on the activities of daily living, mental state, perception and communication, and social participation among elders (all P<0.05), and the elderly with age ≥75 years, illiteracy, unmarried and live alone had higher risk for disability (all P<0.05). CONCLUSIONS: The current situation of the ability level of the elderly in rural China seems to be worrying, and it's necessary to establish a long-term nursing care system and aging policy to meet the needs of the elderly with regards to those potential influencing factors.

ABSTRACT

Nursing skill education plays a very important part for the nursing education. It's necessary to analyze the role of the case-based teaching (CBT) in nursing skill education, to provide insights into the nursing education. The sophomore nursing students who received nursing skill teaching from September 2019 to January 2020 were selected. The students either underwent traditional teaching (control group) or CBT (CBT group) intervention. The objective structure clinical examination (OSCE), autonomous learning ability, questionnaire for effects evaluation of CBT, and student's satisfaction level on CBT were analyzed. A total of 146 participants were included. There were no significant differences in the gender, age and the final exam score as freshman between 2 groups (all P > .05). The OSCE scores and autonomous learning ability in CBT group were significantly higher than that of control group (all P < .05). The most students favored the use of CBT, and most students were satisfied with CBT. CBT is beneficial to improve the nursing skill and comprehensive ability of students, and it's conducive to increase the interest of students, which merits application in nursing education.

OBJECTIVE: To identify the relationship between fever and diarrhea 2-week prevalence and Yingyangbao(YYB) effective consumption among infants and young children. METHODS: A total of 2952 infants and young children aged from 6 to 24 month in 10 impoverished counties of Henan province were selected by multi-stage random sampling between June and September 2017. To acquire 2-week prevalence information of infants and young children, their caregivers were investigated by self-made questionnaire. The structural equation model was utilized in multi-factor analysis. RESULTS: After adjusting potential confounders, YYB effective consumption reduced2-week prevalence of fever(ß=-0. 279, P=0. 001) and diarrhea(ß=-0. 182, P=0. 042) among infants and young children. Nutrition knowledge and YYB benefit cognition of caregivers reduced2-week prevalence of fever(γ=-0. 002, 95%CI-0. 004~-0. 001, P=0. 003) and diarrhea(γ=-0. 001, 95%CI-0. 003~0. 000, P=0. 049) indirectly through chain mediation path of "nutrition knowledge-YYB benfit cognition-YYB effective consumption-fever/diarrheal". CONCLUSION: YYB effective consumption can reduce 2-week prevalence of fever and diarrhea among infants and young children. Nutrition knowledge and YYB benefit cognition can improve YYB effective consumption and thus reduce 2-week prevalence of fever and diarrhea indirectly.

Breast cancer is the most common type of malignancy in women, which remains a significant health concern worldwide. Gemcitabine is a frequently applied anticancer pharmacological agent. However, the efficacy of gemcitabine is limited by chemoresistance. In the present study, a combination of reverse transcription quantitative-PCR, cell viability, flow cytometry, luciferase reporter assay and western blot analysis were performed to elucidate the potential effects of miR-187-3p on gemcitabine sensitivity in the breast cancer cell line, MDA-MB-231. The results revealed that miR-187-3p was significantly decreased in the breast cancer tumor tissues. Moreover, the overexpression of miR-187-3p significantly inhibited cell viability and promoted apoptosis in MDA-MB-231 cells. In addition, miR-187-3p overexpression enhanced the anti-proliferative and pro-apoptotic effects of gemcitabine, indicating that miR-187-3p regulated gemcitabine sensitivity in breast cancer cells. Mechanistically, miR-187-3p negatively regulated the expression of fibroblast growth factor 9 (FGF9) by binding to its 3'-untranslated region. Overexpression of FGF9 reversed the aforementioned effects of miR-187-3p overexpression on cell viability and apoptosis in the presence of gemcitabine. In conclusion, the present study indicated that miR-187-3p increased gemcitabine sensitivity in breast cancer cells by targeting FGF9 expression.

Despite substantial research on the brain mechanisms of L1 and L2 processing in bilinguals, it is still unknown whether language modality (i.e., visual vs. auditory) plays a role in determining whether L1 and L2 are processed similarly. Therefore, we examined the neural representational similarity in neural networks between L1 and L2 in spoken and written word processing in Korean-English-Chinese trilinguals. Participants performed both visual and auditory rhyming judgments in the three languages: Korean, English, and Chinese. The results showed greater similarity among the three languages in the auditory modality than in the visual modality, suggesting more differentiated networks for written word processing in the three languages than spoken word processing. In addition, there was less similarity between spoken and written word processing in L1 than the L2s, suggesting a more specialized network for each modality in L1 than L2s. Finally, the similarity between the two L2s (i.e., Chinese and English) was greater than that between each L2 and L1 after task performance was regressed out, especially in the visual modality, suggesting that L2s are processed similarly. These findings provide important insights about spoken and written language processing in the bilingual brain.

ABSTRACT

The rapid development of global COVID-19 pandemic poses an unprecedented challenge to the safety and quality of laboratory diagnostic testing. Little is known about the laboratory surface areas and operation behaviors that may cause potential contamination in SARS-CoV-2 nucleic acid testing. This study aims to provide reference basis for the improvement of laboratory disinfection programs and personal operating protocols. In this study, we compared the qRT-PCR and ddPCR in detecting of residual virus that existed on the object surfaces from sample transportation and reception related facilities, testing related instruments, personal protective equipment and other facilities in nucleic acid testing laboratory. All samples were negative by qRT-PCR, in contrast, 13 of 61 samples were positive for SARS-CoV-2 by ddPCR. The areas with highest density of SARS-CoV-2 nucleic acid were the outer gloves of operator A (37.4 copies/cm2), followed by door handle of 4 °C refrigerator (26.25 copies/cm2), goggles of operator A (22.16 copies/cm2), outer cover of high speed centrifuge (19.95 copies/cm2), inner wall of high speed centrifuge (14.70 copies/cm2) and others. We found that all the positive objects were directly or indirectly contacted by the operator's gloved hands, suggesting that hands contact was the main transmission pathway that led to laboratory environmental contamination. In summary, ddPCR has an advantage over qRT-PCR in tracing laboratory contamination. We evaluated the risk areas and operation behaviors that may easily cause contamination, and provided recommendation for improving the laboratory disinfection programs and personal operating specifications.

ABSTRACT

Where is human language processed in the brain independent of its form? We addressed this issue by analyzing the cortical responses to spoken, written and signed sentences at the level of individual subjects. By applying a novel fingerprinting method based on the distributed pattern of brain activity, we identified a left-lateralized network composed by the superior temporal gyrus/sulcus (STG/STS), inferior frontal gyrus (IFG), precentral gyrus/sulcus (PCG/PCS), and supplementary motor area (SMA). In these regions, the local distributed activity pattern induced by any of the three language modalities can predict the activity pattern induced by the other two modalities, and such cross-modal prediction is individual-specific. The prediction is successful for speech-sign bilinguals across all possible modality pairs, but fails for monolinguals across sign-involved pairs. In comparison, conventional group-mean focused analysis detects shared cortical activations across modalities only in the STG, PCG/PCS and SMA, and the shared activations were found in both groups. This study reveals the core language system in the brain that is shared by spoken, written and signed language, and demonstrates that it is possible and desirable to utilize the information of individual differences for functional brain mapping.
