ABSTRACT

Natural products, while valuable for drug discovery, encounter limitations like uncertainty in targets and toxicity. As an important active ingredient in traditional Chinese medicine, celastrol exhibits a wide range of biological activities, yet its mechanism remains unclear. In this study, they introduced an innovative "Degradation-based protein profiling (DBPP)" strategy, which combined PROteolysis TArgeting Chimeras (PROTAC) technology with quantitative proteomics and Immunoprecipitation-Mass Spectrometry (IP-MS) techniques, to identify multiple targets of natural products using a toolbox of degraders. Taking celastrol as an example, they successfully identified its known targets, including inhibitor of nuclear factor kappa B kinase subunit beta (IKKß), phosphatidylinositol-4,5-bisphosphate 3-kinase catalytic subunit alpha (PI3Kα), and cellular inhibitor of PP2A (CIP2A), as well as potential new targets such as checkpoint kinase 1 (CHK1), O-GlcNAcase (OGA), and DNA excision repair protein ERCC-6-like (ERCC6L). Furthermore, the first glycosidase degrader is developed in this work. Finally, by employing a mixed PROTAC toolbox in quantitative proteomics, they also achieved multi-target identification of celastrol, significantly reducing costs while improving efficiency. Taken together, they believe that the DBPP strategy can complement existing target identification strategies, thereby facilitating the rapid advancement of the pharmaceutical field.

ABSTRACT

The chemical production process typically possesses complexity and high risks. Effective fault diagnosis is a key technology for ensuring the reliability and safety of chemical production processes. In this study, a comprehensive fault diagnosis method based on time-varying filtering empirical mode decomposition (TVF-EMD), kernel principal component analysis (KPCA), and an improved whale optimization algorithm (WOA) to optimize bi-directional long short-term memory (BiLSTM) is proposed. This research utilizes TVF-EMD and KPCA to analyze and preprocess the raw data, eliminating noise and and reducing the dimensions of the fault data. Subsequently, BiLSTM is employed for fault data classification. To address the hyperparameters within BiLSTM, the enhanced WOA is used for optimization. Finally, the efficacy and superiority of this approach are validated through two fault diagnosis examples.

BACKGROUND: Congenital infantile fibrosarcoma (CIF) and congenital hemangioma (CH) have similarities on prenatal ultrasound and are rare. CASE SUMMARY: We report 3 cases of fetuses with superficial hypervascular tumors, confirmed by postnatal pathology as CIF (1 case) and CH (2 cases, including 1 in a twin fetus). In Case 1, a mass with a rich blood supply in the fetal axilla was discovered by prenatal ultrasound at 28+0 wk of gestation. The postpartum pathological diagnosis was CIF, the mass was surgically removed, and the prognosis of the child was good. In Case 2, at 23+1 wk of gestation, a mass was discovered at the base of the fetus's thigh on prenatal ultrasound. The postpartum pathological diagnosis was CH. After conservative treatment, the mass shrank significantly. Case 3 occurred in a twin fetus. At 30+0 wk of gestation, prenatal ultrasound revealed a bulging mass with a rich blood supply on the abdominal wall of one of the fetuses. Three weeks later, the affected fetus died, and the unaffected baby was successfully delivered by emergency cesarean section. The affected fetus was pathologically diagnosed with CH. CONCLUSION: Prenatal ultrasound can provide accurate information, such as the location, size and blood supply of a surface mass in a fetus. We found similarities between CIF and CH in prenatal ultrasound findings. Although it is difficult to distinguish these conditions by prenatal ultrasound alone, for superficial hypervascular tumors that rapidly increase in size in a short period, close ultrasound monitoring of the fetus is required to quickly address possible adverse outcomes.

ABSTRACT

Biomolecular condensates play key roles in various biological processes. However, specific condensation modulators are currently lacking. PROTAC is a new technology that can use small molecules to degrade target proteins specifically. PROTAC molecules are expected to regulate biomolecular condensates dynamically by degrading/recovering key molecules in biomolecular condensates. In this study, we employed a BRD4-targeting PROTAC molecule to regulate the super-enhancer (SE) condensate and monitored the changes of SE condensate under PROTAC treatment using live-cell imaging and high-throughput sequencing technologies. As a result, we found that BRD4-targeting PROTACs can significantly reduce the BRD4 condensates, and we established a quantitative method for tracking BRD4 condensates by PROTAC and cellular imaging. Surprisingly and encouragingly, BRD4 condensates were observed to preferentially form and play specialized roles in biological process regulation for the first time. Additionally, BRD4 PROTAC makes it possible to observe the dynamics of other condensate components under the continued disruption of BRD4 condensates. Together, these results shed new light on research methods for liquid-liquid phase separation (LLPS), and specifically demonstrate that PROTAC presents a powerful and distinctive tool for the study of biomolecular condensates.

BACKGROUND/NEED: Postoperative flap perfusion assessment methods still rely on the evaluation of traditional clinical indicators, which have the disadvantage of being subjective and burdensome. METHODOLOGY: This study describes a self-designed infrared wireless thermometer for flap blood supply monitoring and evaluates its efficacy in the postoperative monitoring of 40 free flaps. DEVICE DESCRIPTION: The device consists of multiple temperature and humidity modules as well as a wireless module, which has the advantages of low cost and continuous remote monitoring. PRELIMINARY RESULTS: The alarm time of the wireless infrared thermometer was 30.5 ± 3.1 hours, and the clinical observation reported 41.7 ± 13.6 hours. CURRENT STATUS: In future studies, the device will be tested on different types of flaps in a porcine model.

ABSTRACT

The application of flap surgery is becoming more and more widespread with the development of microsurgical techniques. Currently, postoperative blood flow monitoring of flaps is still mainly assessed by medical staff for traditional clinical parameters, which has the disadvantage of being subjective and unable to monitor in real-time. This study describes a self-contained infrared wireless infrared thermometry device for flap blood supply monitoring and evaluates its effectiveness on eight porcine flap models. A scapular muscle flap model was established using eight small pigs, and the vessels were ligated at irregular intervals using a lumir ligature to simulate arterial crisis and venous crisis. Laser Doppler flowmetry (LDF), the wireless infrared thermometry device, and traditional clinical observation methods were applied to monitor the blood supply of the flap and evaluate the effect. The time to the determination of blood supply disturbance by wireless infrared thermography (IRT) was 28.75 ± 3.30 min and 96.5 ± 27.09 min for the arterial and venous groups, respectively; by LDF was 6.00 ± 1.41 min and 52.75 ± 15.76 min; by clinical observation was 42.00 ± 8.60 min and 156.50 ± 40.91 min, respectively. Paired t-tests were performed between the wireless IRT device and clinical observations, and the statistical results were significantly different in the arterial group and not significantly different in the venous group. Paired t-testing of the wireless infrared thermometry device with the LDF also showed significant differences in the arterial group and non-significant differences in the venous group. This wireless infrared thermometry device outperforms traditional clinical observation methods in monitoring blood supply in a porcine skin flap model. Because of its low cost, real-time monitoring, simple operation, and non-invasive features, it has the potential to be used in clinical practice as a routine means of postoperative blood supply monitoring in flap surgery.

ABSTRACT

Defect passivation along with promoted charge transport is potentially an effective but seldom exploited strategy for high-performance perovskite solar cells (PSCs). Herein, the in situ defect passivation and carrier transport improvement are simultaneously realized by introducing a conductive polymer (i.e., emerald salt, ES) into the precursor solution of methylammonium (MA)-free perovskites. The interaction between ES and uncoordinated Pb2+ reduces defect density to suppress the non-radiative recombination. Moreover, ES can act as a "carrier driver" to promote the carrier transport due to its conductive feature, resulting in efficient PSC devices with a decent power conversion efficiency (PCE) of 23.0%, which is among the most efficient MA-free PSCs. The ES-based unencapsulated devices show superior stability, retaining 89.1% and 83.8% of their initial PCEs when subjected to 35 ± 5% relative humidity (RH) storage and 85 °C thermal aging for 1000 h, respectively. To further assess the large-area compatibility of our strategy, 5 × 5 cm2 mini modules were also fabricated, delivering an impressive efficiency of 19.3%. This work sheds light on the importance of conductive additives in boosting cell performance by playing multiple roles in passivating defects, retarding the moisture invasion, and enhancing and balancing charge transport.

Background: The systemic immune-inflammation index (SII) is a novel marker of inflammation, and hepatic steatosis and fibrosis are associated with inflammation. This study aimed to investigate the possible relationship between SII and hepatic steatosis and fibrosis. Methods: The datasets from the National Health and Nutrition Examination Survey (NHANES) 2017-2020 were used in a cross-sectional investigation. Multivariate linear regression models were used to examine the linear connection between SII and controlled attenuation parameter (CAP) and liver stiffness measurement (LSM). Fitted smoothing curves and threshold effect analysis were used to describe the nonlinear relationship. Results: This population-based study included a total of 6,792 adults aged 18-80 years. In a multivariate linear regression analysis, a significant positive association between SII and CAP was shown [0.006 (0.001, 0.010)]. This positive association in a subgroup analysis was maintained in men [0.011 (0.004, 0.018)] but not in women. Furthermore, the association between SII and CAP was nonlinear; using a two-segment linear regression model, we found an inverted U-shaped relationship between SII and CAP with an inflection point of 687.059 (1,000 cells/µl). The results of the multiple regression analysis showed that the relationship between SII and LSM was not significant (P = 0.263). Conclusions: Our findings imply that increased SII levels are linked to hepatic steatosis, but SII is not linked to liver fibrosis. To confirm our findings, more large-scale prospective investigations are needed.

ABSTRACT

Kasabach-Merritt phenomenon (KMP) is a life-threatening condition caused by rare vascular tumors. To reduce drug resistance observed in monotherapy of KMP with prednisone, vincristine (VCR) or sirolimus, the present study evaluated the efficacy and safety of triad therapy in the treatment of KMP. A total of 10 KMP infants managed with prednisolone, VCR and sirolimus in The Second Affiliated Hospital of Xi'an Jiaotong University (Xi'an, China) between April 2017 and August 2021 were retrospectively reviewed. The three female and seven male infants with KMP underwent cocktail therapy with prednisone, VCR and sirolimus. At diagnosis, the infants, aged 49.1±41.0 days, showed laboratory test results with platelet counts 22±15.4x109/l, fibrinogen 81.7±26.9 mg/dl and D-dimer 38649±13443.6 ng/ml. The average maximal diameter of the tumors at diagnosis was 84.5±25.1 mm. KMP risk is increased by large tumors with deep lesions infiltrating the muscle. Platelet counts normalized after a median 10 days (range, 5-69 days) of treatment. With combination therapy maintained for 46.8±24.4 days, ultrasound showed that the thickness of the tumors decreased by 51% from 28.9±12.1 to 13.9±6.2 mm. Neutropenia and gastrointestinal disorders were the most common adverse effects. The present study found that the cocktail therapy with prednisolone, VCR and sirolimus has favorable tolerance and efficacy for life-threatening KMP. Once a stable condition has been achieved, cocktail therapy should be replaced by sirolimus monotherapy to reduce potential side effects.

Lymphatic malformation (LM) in the abdomen is uncommon in children, and the standard treatment is surgical excision. The present study aimed to evaluate the safety and efficacy of laparoscopic surgery of intra-abdominal LM in a pediatric population. The medical records of 10 children with intra-abdominal LM treated by laparoscopic resection from March 2017 to June 2021 in The Second Affiliated Hospital of Xi'an Jiaotong University (Xi'an, China) were retrospectively reviewed. Equal numbers of female and male patients were included and underwent surgery at the median age of 55 months (range, 40 days-94 months). Abdominal pain was the most frequent symptom presenting in eight patients (80%). All children were referral patients, and half of them presented to the emergency department with acute abdominal pain. All 10 of the patients benefited from laparoscopic treatment; three patients underwent extraperitoneal segmental intestinal resection and anastomosis via an enlarged umbilical incision, and two patients were converted to laparotomy because of a large retroperitoneal LM with involvement of the colon. No recurrence or complications occurred in the patients during the follow-up period with mean of 35 months (range, 11-60 months) after surgery. Overall, laparoscopic resection of intra-abdominal LM was an effective, minimally invasive therapy in the pediatric population. Segmental intestinal resection is usually required to achieve the complete removal of lesions to lessen the risk for recurrence.

ABSTRACT

MOTIVATION: Disease diagnosis-oriented dialog system models the interactive consultation procedure as the Markov decision process, and reinforcement learning algorithms are used to solve the problem. Existing approaches usually employ a flat policy structure that treat all symptoms and diseases equally for action making. This strategy works well in a simple scenario when the action space is small; however, its efficiency will be challenged in the real environment. Inspired by the offline consultation process, we propose to integrate a hierarchical policy structure of two levels into the dialog system for policy learning. The high-level policy consists of a master model that is responsible for triggering a low-level model, the low-level policy consists of several symptom checkers and a disease classifier. The proposed policy structure is capable to deal with diagnosis problem including large number of diseases and symptoms. RESULTS: Experimental results on three real-world datasets and a synthetic dataset demonstrate that our hierarchical framework achieves higher accuracy and symptom recall in disease diagnosis compared with existing systems. We construct a benchmark including datasets and implementation of existing algorithms to encourage follow-up researches. BACKGROUND: Serum lipids are highly inheritable and play a major role in bone health. However, the relationship between low-density lipoprotein cholesterol (LDL-C) and bone mineral density (BMD) remains uncertain. The goal of this study was to see if there was a link between LDL-C levels and BMD in persons aged 20 to 59. METHODS: Using data from the National Health and Nutrition Examination Survey (NHANES) 2011-2018, multivariate logistic regression models were utilized to investigate the association between LDL-C and lumbar BMD. Fitted smoothing curves and generalized additive models were also used. RESULTS: The analysis included a total of 4909 adults. After controlling for various variables, we discovered that LDL-C was negatively linked with lumbar BMD. The favorable connection of LDL-C with lumbar BMD was maintained in subgroup analyses stratified by gender and race in both males and females, Whites and Mexican Americans, but not in Blacks and other races. The relationship between LDL-C and lumbar BMD in other races was an inverted U-shaped curve with the inflection point: 2.327 (mmol/L). CONCLUSION: In people aged 20 to 59, our research discovered a negative relationship among LDL-C and lumbar BMD. Among races other than Whites, Blacks, Mexican Americans, this relationship followed an inverted U-shaped curve (inflection point: 2.327mmol/L). LDL-C measurement might be used as a responsive biomarker for detecting osteoporosis early and guiding therapy.

ABSTRACT

Microplastic (MP) pollution has been a considerable concern due to its ubiquity in the environment and its potential to harm human health. Unfortunately, the exact levels of MP in various species of seafood species have not been established. It is also unclear whether or not consuming seafood contaminated with MPs directly jeopardizes human health. Here, eight popular species of seafood in Dongshan Bay, China were investigated to determine the presence of MP pollution and its implications on human health. The abundance, color, size, shape, type, surface morphology, danger of the MPs extracted from the seafood were analyzed. Results showed that the average MP abundance in the shellfish and fish was 1.88 ± 1.44 and 1.98 ± 1.98 items individual-1, respectively. The heavy presence of fibers may be attributed to the shellfish and fish's feeding behaviors as well as their habitat and environment. The sizes of MPs found were below 1.0 mm. The main types of MP found in the shellfish were PES and PET, whereas the main types found in the fish were PS and PES. Risk assessment suggested that MPs in the shellfish (risk Level V) posed a greater and more direct threat to human health if the shellfish is eaten whole. The MPs in the gastrointestinal tracts (GITs) of fish (risk Level IV) have a relatively limited effect on human health since GITs are seldom consumed by humans unless the fish is heavily processed (canned or dried). MPs-induced health risk is predicted using a technique called molecular docking. The results of this study not only establish levels of MP pollution in popular seafood species but also help understand the implications of consuming MP-contaminated seafood on human health.

ABSTRACT

Microplastic pollution in fish is a growing concern worldwide due to its implication for human health. Microplastic contaminations and impacts were investigated in 15 wild-caught commercially important dolphinfish (Coryphaena hippurus L.) from the Eastern Pacific Ocean waters. 1741 suspected particles were extracted from gills, esophagus, stomachs, intestinal tracts, and muscle of C. hippurus. Only 139 of them were identified as microplastics by microscopic inspections and micro-Raman spectroscopic analysis. 10, 34, 51, 35, and 9 out of these 139 microplastic particles were extracted from the gill, esophagus, stomach, intestinal tract, and muscle respectively. Overall, microplastics were detected in 15 out of 15 fish (100%), with ~9.3 pieces per individual on average. The prevalence and high incidence of occurrence of microplastics in the C. hippurus suggest that this pelagic species are at high risk of exposure to microplastic pollutions. The chemical composition of microplastics was made of polyester (PES, 46.8%), polyethylene terephthalate (PET, 38.1%), polypropylene (PP, 7.9%), polystyrene (PS, 5.0%), polyethylene-polypropylene copolymer (PE-PP, 1.4%), and polyethylene (PE, 0.7%). 36.7% and 13.7% of microplastics in the fish were 1-2.5 mm and 2.5-5 mm, respectively. Microplastics of 0.1-0.5 mm and 0.5-1 mm roughly shared equally the remaining 50%. Molecular docking results implied that interaction of the four dominant microplastic polymers (PES, PET, PP, and PS) with cytochrome P450 17A1 would lead to impairment of the reproductive function of C. hippurus. The findings provide insights on the harms from microplastic exposure, along with quantitative information of occurrence, abundance, and distribution of microplastics in the fish tissues, which will ultimately improve understanding of bioavailability and hazards of microplastics to the organisms and beyond to human via food chain transfer.

ABSTRACT

Microplastic (MP) pollution has spurred a wide range of concerns due to its ubiquity and potential hazards to humans and ecosystems, yet studies on MP abundance, distribution, and ecological impacts on the small-scale local estuarine systems are insufficient. We conducted the first study of MP pollution in surface water of Dongshan Bay in southern China. A total of six water samples were collected using a Manta trawl (length = 3 m, width = 1 m, height = 0.6 m, and mesh size = âˆ¼330 µm). The abundance, type, shape, color, and size, were measured using light microscopy and micro-Raman spectroscopy. Our results showed that MPs spanned from 0.23 to 4.01 particles m-3 with an average of 1.66 particles m-3. 75% of the MPs were PP, PE, and PS that may be explained by the widespread application of PE, PP, and PS foam in local fishing and aquaculture within the bay. Foam, white, and 1.0-2.5 mm were dominant shape, color, and size of MPs, respectively. Both indices of MPs-induced risk (Hestuary = 13.7) and pollution load (PLIestuary = 14.2) yielded a Hazard Level II for MPs pollution in the Dongshan Bay. The potential ecological risk from combined MPs polymers (RIestuary = 21.5) ended up at a minor risk. Our findings established the first set of baseline data on MPs pollution in Dongshan Bay and provided preliminary quantitative measures on the scale of ecological risk, which would improve the understanding of MP fate, transport, and ecological impacts in the estuarine environment.

Microplastics have pulled the alarm for its far-reaching impacts in the environment and potential risks to human health. Here, we first investigated the microplastic pollution in the Zhangjiang River of southeastern China. Water samples were collected across a river catchment using bulk sampling method. The abundance, distribution, and characteristics of microplastics were analyzed using stereomicroscopy and micro-Raman spectroscopy. The results indicated that microplastic abundance ranged from 50 to 725 items m-3 with an average of 246 items m-3. PP and PE were the major polymers and accounted for ~75%. The dominant shape, color, and size of microplastics are fragment, white, and 0.5-1 mm, respectively. The findings established the baseline information on the microplastic pollution in the Zhangjiang River, which may improve the knowledge of sources of microplastic debris to the local watershed and future evaluation on the role of river catchments in transporting microplastics to the estuary and beyond into the ocean.

ABSTRACT

Surfactin produced by Bacillus subtilis is a powerful biosurfactant in food, cosmetics, and pesticide industries. However, its suitability in wound healing applications is uncertain. In this article, we determined the effects of surfactin A from B. subtilis on wound healing, angiogenesis, cell migration, inflammatory response, and scar formation. The results indicated that 80.65 ± 2.03% of surfactin A-treated wounds were closed, whereas 44.30 ± 4.26% of the vehicle-treated wound areas remained open on day 7 (P < 0.05). In mechanisms, it upregulated the expression of hypoxia-inducible factor-1α (HIF-1α) and vascular endothelial growth factor (VEGF), accelerated keratinocyte migration through mitogen-activated protein kinase (MAPK) and nuclear factor-κB (NF-κB) signaling pathways, and regulated the secretion of proinflammatory cytokines and macrophage phenotypic switch. More attractive, surfactin A showed a seductive capability to inhibit scar tissue formation by affecting the expression of α-smooth muscle actin (α-SMA) and transforming growth factor (TGF-ß). Overall, the study revealed a new function and potential of surfactin A as an affordable and efficient wound healing drug.

ABSTRACT

BACKGROUND: With the development of e-Health, it plays a more and more important role in predicting whether a doctor's answer can be accepted by a patient through online healthcare community. Unlike the previous work which focus mainly on the numerical feature, in our framework, we combine both numerical and textual information to predict the acceptance of answers. The textual information is composed of questions posted by the patients and answers posted by the doctors. To extract the textual features from them, we first trained a sentence encoder to encode a pair of question and answer into a co-dependent representation on a held-out dataset. After that,we can use it to predict the acceptance of answers by doctors. RESULTS: Our experimental results on the real-world dataset demonstrate that by applying our model additional features from text can be extracted and the prediction can be more accurate. That's to say, the model which take both textual features and numerical features as input performs significantly better than model which takes numerical features only on all the four metrics (Accuracy, AUC, F1-score and Recall). CONCLUSIONS: This work proposes a generic framework combining numerical features and textual features for acceptance prediction, where textual features are extracted from text based on deep learning methods firstly and can be used to achieve a better prediction results.

ABSTRACT

Microplastics (MPs) contamination has been recognized as one of major threats to coastal marine environments. Although studies evidenced severe MPs contaminations to the Pacific Ocean, environmental implications of MPs concentrations, distributions, and characteristics have not been evaluated in sufficient detail. Here, we report on the distribution, abundance, and characteristics of MPs at the surface of the Northwestern Pacific Ocean, from which environmental implications are assessed. A manta trawl with a mesh size of ~330 µm and a rectangular net opening of 0.45 × 1 m was used to collect MPs samples on May 11-June 3, 2018. The MPs samples were sequentially isolated, digested, filtered, and characterized using an optical microscope, micro-Raman spectroscopy, and scanning electron microscopy. The results indicate the heterogeneity in abundance, shapes, color, and sizes of MPs. The study provides strong environmental implications such as sources, environmental degradation, residence time, transportation routes, and biological interactions.
