ABSTRACT

Primary Sjögren's syndrome (pSS) is associated with an increased risk of lymphoma, especially non-Hodgkin's lymphoma. The rarest pathological subtype is T-cell lymphoma. We herein report a case of a 52-year-old man with a 17-year history of pSS who was admitted to our hospital with chronic epigastric pain and a positive fecal occult blood test. Colonoscopy revealed multiple colonic ulcers, and histological and immunological studies demonstrated the T-cell origin of this lymphoma. However, the patient rejected all treatments. He developed recurrent intestinal obstruction and infection for 3 years until an intestinal perforation occurred. The right half of the colon was resected and colostomy was performed. However, the patient died of an intestinal fistula and intraperitoneal infection 40 days postoperatively. This case highlights the rarity of the correlation between T-cell lymphoma and pSS.

ABSTRACT

An experiment was conducted to compare effects of emulsified soybean oil and non-emulsified soybean oil on the quality of broiler feed differing in the feed type and the broiler feeding stage in vitro. A 2 × 2 × 3 factorial arrangement was designed with two fat sources (soybean oil and emulsified oil), two feed types (mash and pellet) and three broiler feeding stages (starter, grower and finisher). Four samples of feeds were collected from each combination of factors at the beginning of the experiment and stored at 20°C. Subsamples were taken at 15-day intervals to determine the moisture content, peroxide value (PV), acid value (AV) and the total fungal count over a 45-day period; weight loss percentage was determined by weighting the samples at days 0, 15, 30 and 45; fines percentage in pellets was only determined at day 0. The emulsified oil reduced (P < 0.05) the fines percentage, increased (P < 0.05) the moisture content, decreased (P < 0.05) the weight loss percentage and PV, did not affect (P > 0.05) the AV and the total fungal count. Results showed that the emulsified oil decreased weight loss, increased pelletability, moisture content and oxidation stability without affecting fungal growth.

ABSTRACT

Transcription factors (TFs) play an important role in gene regulation. Computational identification and annotation of TFs at genome scale are the first step toward understanding the mechanism of gene expression and regulation. We started to construct the database of Arabidopsis TFs in 2005 and developed a pipeline for systematic identification of plant TFs from genomic and transcript sequences. In the following years, we built a database of plant TFs (PlantTFDB, http://planttfdb.cbi.pku.edu.cn ) which contains putative TFs identified from 22 species including five model organisms and 17 economically important plants with available EST sequences. To provide comprehensive information for the putative TFs, we made extensive annotation at both the family and gene levels. A brief introduction and key references were presented for each family. Functional domain information and cross-references to various well-known public databases were available for each identified TF. In addition, we predicted putative orthologs of the TFs in other species. PlantTFDB has a simple interface to allow users to make text queries, or BLAST searches, and to download TF sequences for local analysis. We hope that PlantTFDB could provide the user community with a useful resource for studying the function and evolution of transcription factors.

ABSTRACT

Our recent work by Wang et al (2010), together with previous studies published in the last decade, have provided evidence suggesting a link between ethylene signaling and primordial anther specific DNA damage in female cucumber flowers. These findings explained ethylene promotion of female flower by ethylene inhibition of stamen development. However, it is not determined if ethylene promotes carpel development. In addition, an explanation of why the naturally occurring gas ethylene was selected to be involved in such developmental events remains elusive. In this study, we carried out a phylogenetic analysis of cucumber ACS genes, and analyzed the expression levels of some pre-miRNAs in male, female and hermaphrodite flowers. We found the M gene might have evolved prior to, or "co-opted" into unisexual flower development before the F gene, and miRNA might be involved in carpel development regulation. Based on these observations, we propose a new hypothesis to explain why ethylene was selected to be involved in the evolution of the unisexual cucumber flower.

ABSTRACT

Altered glycosylation may be a hallmark of malignant transformation and cancer progression. In the work described, a specific mannose-binding lectin, Pinellia pedatisecta agglutinin (PPA), was genetically fused with the extracellular domain of coxsackie-adenovirus receptor (CAR) to generate the soluble CAR (sCAR)-PPA fusion protein. The adenoviral transduction of acute myeloid leukemia (AML) cell lines Kasumi-1 and HL-60 was increased by sCAR-PPA, indicating that a fraction of AML cells exposing mannose residues was detected by PPA. However, sCAR-PPA did not increase the adenoviral infection of KG-1 cells, suggesting the mannose exposure of AML cells may be cell type specific. Furthermore, the infectious efficiency of Ad-EGFP in chronic myeloid leukemia cell line K562 was significantly increased by sCAR-PPA as well. We, herein, report that PPA recognized a fraction of myeloid leukemia cells showing mannose-exposing phenotype. The sCAR-PPA fusion protein combined with the adenoviral vector system may provide a useful tool for investigating myeloid leukemia cells exposing mannose residues and further elucidating the role of these cells in the leukemia development.

ABSTRACT

A new class of heterogeneous catalysts for asymmetric hydrogenation of enamides was synthesized using molecular imprinting technology. These new catalysts are molecularly imprinted polymers (MIPs) made from rhodium (I) and copper (II) complexes with the bis(oxazoline) chiral ligands. One of the Rh-MIPs showed 87% ee toward L-enantiomeric product while the Cu-MIP showed 82% ee toward D-enantiomeric product. Both MIPs are easy to separate and reusable.

ABSTRACT

Systematic identification and annotation of TFs, followed by construction of TF databases may serve as useful resources for studying the function and evolution of transcription factors. We developed a comprehensive plant transcription factor database PlantTFDB (http://planttfdb.cbi.pku.edu.cn), which contains 26,402 TFs predicted from 22 species, including five model organisms with available whole genome sequence and 17 plants with available EST sequences. Functional domain information and cross-references to various well-known public databases were available for each identified TF. In addition, we predicted putative orthologs of those TFs among the 22 species. PlantTFDB has a simple interface to allow users to search the database by IDs or free texts, to make sequence similarity search against TFs of all or individual species, and to download TF sequences for local analysis.

A review on the principles and applications of boronic acids as affinity ligands for the chromatographic separation of carbohydrates, nucleic acid components, glycoproteins, and other small biomolecules. The mechanisms of interactions between boronate ligands and analytes are described. Various boronate ligands and supports are discussed. Examples of the use of boronate affinity chromatography for separation of each class of analytes are presented.

ABSTRACT

A new type of 2-D separation material was synthesized and studied. The material is suitable for 2-D chromatography utilizing both covalent and noncovalent interactions. The first dimension is boronate affinity chromatography, and the second dimension is RP chromatography (or vice versa). The polymeric media were prepared using p-vinylphenylboronic acid as the functional monomer. This monomer was selected due to the presence of the boronic acid group for the cis-diol/boronate interaction in boronate chromatography. Two crosslinkers were evaluated, namely ethylene glycol dimethacrylate and divinylbenzene. The crosslinker content was varied to maximize the polymer strength and the RP performance of the packed column. Several parameters were evaluated to define the optimum for polymer strength and column performance including crosslinker, porogen, initiator, and column-packing parameters. The polymer-based HPLC columns were successful in separating phenol, catechol, dimethylphthalate, and hydroquinone under RP conditions, and thus can be used as an RP HPLC column. The columns were also successful in separating catechol and adenosine under boronate chromatography conditions, and thus can be used as a boronate affinity column. Moreover, the two types of chromatography can be performed consecutively on the same column during one complete chromatographic run, making it a 2-D chromatography. Under these 2-D conditions, the catechol was separated from a mixture of phenol, catechol, dimethylphthalate, and hydroquinone; the adenosine ribonucleoside was separated from a mixture of adenosine ribonucleoside, adenosine deoxyribonucleoside, and uridine deoxyribonucleoside. This type of single-column 2-D HPLC eliminates the requirement of a complex and expensive multidimensional HPLC instrument and provides increased peak capacity for separation.

ABSTRACT

A novel approach for regioselective synthesis is described. The approach uses the unique binding properties of protein as a guide to achieve regioselectivity. Specifically, steroids, e.g., 4-androstene-3,17-dione, were reduced selectively at carbon 17 using bovine serum albumin (BSA) as a regio auxiliary. The approach has resulted in a high yield (ca. 91%) in an environmental friendly aqueous medium. Both (1)H NMR and (13)C NMR data show that the BSA-assisted reduction of the steroid render the reaction regioselective compared to the control reaction. Various control reactions were performed to verify that the unique binding of BSA to steroids was the origin of regioselectivity. After one cycle of the reduction reaction, BSA was reconditioned such that it could be reused five or more times. This novel approach could be used to develop practical and "green" methods for regioselective synthesis.

Telomere is an important DNA-protein structure. It caps the ends of linear eukaryotic chromosomes. Telomeric DNA consists of tandemly repeated simple sequences. Telomere is synthesized with the action of telomerase, a ribonucleoprotein with reverse transcriptase activity. Telomere plays an important role in maintaining the stability of intact chromosome,genome and cell. This paper is a review of telomere, telomerase, telomere-binding protein and developmental control of telomere change, telomerase activity in higher plant.
