ABSTRACT

It is indicated that the exposure to radon originating from the building materials is not negligible and the radon exhalation measurement should get more attention in the future. The experience with four different methods of the exhalation measurement, established in our laboratory for the purpose of the intercomparison is reported. Additionally, a comparison of advantages and disadvantages of used methods is discussed.

ABSTRACT

In the era of the energy saving policy (i.e. more air tight doors and windows), the radon exhaled from building materials tends to increase its concentration in indoor air, which increases the importance of the measurement of radon exhalation from building materials. This manuscript presents a novel method of the radon exhalation measurement using only a HPGe detector or any other gamma spectrometer. Comparing it with the already used methods of radon exhalation measurements, this method provides the measurement of the emanation coefficient, the radon diffusion length and the radon exhalation rate, all within the same measurement, which additionally defines material's radon protective properties. Furthermore it does not necessitate additional equipment for radon or radon exhalation measurement, which simplifies measurement technique, and thus potentially facilitates introduction of legal obligation for radon exhalation determination in building materials.

ABSTRACT

Rb1 plays an important role in cell cycle progression and therefore may be involved in malignant transformation of colonic cells. The aim of our research was to define the potential role of Rb1 as a prognostic biomarker in tumorigenesis of sporadic colorectal cancer, and to examine the role of miR-106a in Rb1 regulation as it functionally binds to 3'UTR of transcribed mRNA. We examined LOH and promoter methylation status. Real-time PCR was used for Rb1 mRNA and miR-106a, and immunohistochemistry for protein expression analysis. All the results obtained from patients' samples were correlated with the clinicopathological parameters in order to determine its influence on the sporadic colorectal carcinogenesis. LOH showed no correlation with mRNA and pRb expression. 51.5% of tumor samples were scored negative for pRb staining. Despite this finding, we detected overexpression of Rb1 mRNA in tumor samples in comparison to the adjacent normal tissue (p=0.023). mRNA overexpression was consistent with Rb1 promoter methylation analysis results, which showed no methylation in the investigated samples. Expression analysis of miR-106a in the patients samples showed its overexpression in colorectal cancer (p<10(-4)). Negative pRb score was expected according to the definition of tumor suppressor genes and their proposed role in the malignant transformation of the cells. The observed discrepancy between mRNA and protein expression can be explained by a regulatory mechanism that inhibits translation, such as microRNA silencing. Our results suggest that miR-106a might have a regulatory role for Rb1 in sporadic colorectal cancer.

ABSTRACT

OBJECTIVE: The aim of this study was to evaluate the role of cytology in providing a reliable diagnosis upon which the clinician can base further investigative or treatment strategies in patients with laryngeal and pharyngeal tumours. METHODS: Imprint cytology diagnoses from 174 patients were correlated with the histological result of a corresponding biopsy. RESULTS: We found that the imprint cytology proved to be a useful, quick and reliable method with complete diagnostic accuracy, sensitivity, specificity, positive predictive value and negative predictive values of 97%, 96%, 100%, 100% and 92% respectively. CONCLUSION: Imprint cytology allows diagnostic statements in a shorter time than is possible with histological sections and proves a useful adjunct in evaluating laryngeal and pharyngeal lesions. The validity of the method depends on the care with which the specimen is sampled and on the experience of the investigator.

ABSTRACT

OBJECTIVE: Previous studies have pointed to the changes of serum angiotensin-converting enzyme (SACE) values in patients with liver disease and cancer located in different sites. The aim of this study was to determine the changes in SACE values in patients with hepatocellular carcinoma (HCC) and liver cirrhosis. METHODS: The study comprised 30 patients with HCC (22 men and eight women) of average age 48.6 +/- 9.0 years and 30 patients with liver cirrhosis (21 men, nine women) of average age 49.1 +/- 9.5 years. The control group consisted of 30 healthy volunteers with matching anthropometric characteristics. SACE activity was determined by a spectrophotometric method using synthetic hippuryl-glycyl-glycine as substrate. RESULTS: The mean SACE value was considerably lower in patients with HCC, 22.8 U/ml (95% CI, 22.5-23.9), both those in whom HCC developed in cirrhotic liver (n = 23), 23.7 (22.9-24.5) as well as those with HCC without cirrhosis (n = 7), 21.8 (21.0-22.6), with regard to patients with liver cirrhosis, 37.2 (36.6-37.8) (P < 0.001). There was also a statistically significant difference between healthy, 29.9 (29.4-30.3), and both groups of HCC patients (P < 0.001). No significant differences could be found between the group of HCC patients with and without liver cirrhosis (P < 0.05). In patients with liver cirrhosis SACE value was increased in accordance with the severity of the disease expressed by Child's classification; however, at each stage SACE values were considerably lower in patients with HCC in cirrhotic liver (Child A, 35.8 vs 22.1; Child B, 38.7 vs 24.2; Child C, 40.0 vs 28.3) (P < 0.001). Alfa-fetoprotein (AFP) values did not correlate with the SACE activity. The SACE value was also significantly decreased in patients with HCC whose AFP were not altered. CONCLUSION: The study has shown that SACE values are low in patients with advanced HCC. It may be helpful in detecting HCC in patients with cirrhosis, where it can be difficult to differentiate between small HCC tumours and regeneration nodules.
