ABSTRACT

Hypoxia-ischemia (HI) is among the most frequent causes of death and disability in neonates. We aimed here to examine the neuroprotective effects of Remifentanil (RE) and the underlying mechanisms in a rat model of hypoxic-ischemic brain damage (HIBD). We found that RE improved the learning memory ability, reduced neuronal cell damage and apoptosis, reduced inflammation induced by suppressing the expression of BTB domain and CNC homolog 1 (BACH1) in rats with HIBD. BACH1 attenuated the alleviating effect of RE on cognitive impairment in HIBD rats. Moreover, RE inhibited TRAF3 expression by downregulating BACH1, and TRAF3 attenuated the therapeutic effect of RE on cognitive impairment by activating the NF-κB signaling. In conclusion, our findings demonstrated that RE inhibits the expression of BACH1, which in turn inhibits the NF-κB signaling pathway by suppressing TRAF3. RE may be a promising therapeutic agent to attenuate HIBD-induced cognitive impairment.

ABSTRACT

In order to expand the application range of the model for a single kind of fruits in the portable near infrared instrument, this paper comes up with a new method for the soluble solid content (SSC) model transfer between different kinds of fruits. This method is focusing on the idea of model transfer between different instruments. Based on the similar physical and chemical properties of apples, peaches and pears, such as the range of SSC content, fruit size and the thickness of peel, a simple Slope/Bias algorithm is applied to the transfer of apple SSC partial least square (PLS) model. After that, it can be used to predict pear & peach SSC value with very little extra samples. It's more convenient and costs less by using this method. For pear samples, by using extra 35 standard samples to transfer apple SSC model, RMSEP reduced from 1.009 °Brix to 0.565 °Brix. For peaches, extra 40 standard samples led to a significant reduce of RMSEP from 1.726 °Brix to 0.677 °Brix after model transfer. To validate the feasibility of this model transfer method, both pear and peach SSC models were tested using the same Slope/Bias algorithm model transfer respectively. A pear SSC model was firstly set up and then transferred with Slope/Bias method. Taking 30 standard apples as samples, RMSEP value reached 0.597°Brix, while taking 40 standard peaches as samples, RMSEP value reached 0.689°Brix. The peach SSC model was transferred in the same way. For apples, using 35 standard samples, RMSEP value reached 0.654°Brix, and for pears, using 30 standard samples, RMSEP value reached 0.439°Brix. These results show that slope/bias algorithm can be used to transfer model between similar kinds of fruits such as apples, pears and peaches. The paper provides innovative ideas for the model transfer among similar kinds of materials, so that the portable near infrared instruments can be used more conveniently and widely.

ABSTRACT

OBJECTIVE: To investigate the effect of houttuyfonate sodium (HS) on eliminating adhesion of Psedomonas aeruginosa (Pa) and forming biofilms. METHOD: Pa biofilms were established in 96-hold plates. MTT assay was used to evaluate the changes in metabolism of biofilms and assess the minimum eliminating concentration and minimum biofilm inhibitory concentration for adherent Pa. The colony counting method was used to observe the effect of HS on Pa adhesion and biomass in biofilms. SEM was employed to examine the effect of HS on adhesion of tested Pa and morphology of biofilms. RESULT: MEC80 and MEC50 of HS for adherent Pa was 500 mg x L(-1) and 125 mg x L(-1), respectively. Meanwhile, its SMIC80 for either early or mature biofilms of Pa was 500 mg x L(-1), and SMIC50 for early and mature biofilms of Pa were 31.25, 1.95 mg x L(-1), respectively. At the concentration of 250 mg x L(-1), the number of viable bacteria in the state of adhesion and in initial and mature biofilms decreased significantly, compared with the control group (P < 0.05). The number of bacteria on adherent carriers notably reduced under SEM. Following the continuous administration, there were no visible biofilms on carriers in the mature biofilm phase, with the biomass remarkably shrinking and the bacterial morphology changing from bacillus into coccobacillus. CONCLUSION: HS displayed powerful effect on eliminating adherent Pa, and can inhibit Pa biofilm from being formed through continuous administration.

ABSTRACT

OBJECTIVE: To research the application of the single time detection of HPV E6/E7 mRNA and HC2-HPV-DNA in cervical screening project. METHODS: We detected both HPV E6/E7 mRNA and HC2-HPV-DNA of each sample which collected from 130 cervical disease patients' cervix during Jan. 2008 and July. 2009. TCT results were taken as standard to evaluate the diagnostic accuracy of the above two test methods in detecting high-grade cervical disease. RESULTS: 82.3% (107/130)women were confirmed to infect HPV by HC2-HPV-DNA detection, and 40.0% (52/130) women were confirmed to infect HPV by HPV E6/E7 mRNA detection, there was no significant difference between the two results (chi2 = 24.5, P < 0.05). The sensitivity, specificity, positive predictive value, negative predictive value of HC2-HPV-DNA detection were 90.1%, 22.1%, 37.4% and 82.6%, respectively. The sensitivity, specificity, positive predictive value, negative predictive value of HPV E6/E7 mRNA detection were 65.9%, 73.3%, 55.8% and 80.8%, respectively. CONCLUSION: In clinical cervical screening project of single time, the combination of HC2-HPV-DNA detection and HPV E6/E7 mRNA detection wick take on more potential value than applying each of them alone. ABSTRACT

The Human Serum Albumin (HSA) can emit fluorescence (lamda(em) = 350 nm) under irradiation of ultraviolet light (lamda(ex) = 296 nm). After the vitamin B6 (B6) was added into HSA solution the fluorescence of HSA was quenched partially. This quenching was static quenching owing to the fact that ro (fluorescence life time (life time of the molecule excitation state) of HSA without B6) equaled to tau(i) (fluorescence life time of HSA with B6). The formation constant K between HSA and B6 was observed from the experiment, and then the binding distance R0 was determined (R0 = 1.872 nm). The CD spectra of samples (HSA, [HSA] = 1 x 10(-5) mol x L(-1), HSA +B6, [B6] = 5 x 10(-8), 1.5 x 10(-7), 2.5 x 10(-7), 3.5x 10(-7), 4.5x 10(-7) mol x L(-1)) were measured, and all CD spectra were almost the same. From [theta] values, the contents (percent content) of four structure(alpha-Helix, beta-Pleated, Beta-corner, and Randon wind) of samples can be calculated, and we find that all samples contain almost the same structure contents.

ABSTRACT

Fluorescence spectroscopy has achieved great successes in the study on the interaction of drugs and DNA in both quantitative analysis and qualitative analysis. While the fluorescence probe technique can be used to measure the intensity of drugs-DNA interaction, because the injected drugs can change the fluorescence intensity of the probe. In this paper, the authors use berberine as a probe to measure the interaction intensity between some anticancer drugs and DNA, calculate the constant D and determine the effects of those drugs. In addition, the authors point out that this method can not be efficient for certain drug solution.

ABSTRACT

Spectral characteristics of an organic material in acid with different concentrations were investigated in the solution of a proper acid concentration. The results showed that at 252 nm the transmittance T = 56.5%, but at 290 nm the material had a strong absorption peak (epsilon > 10(4) L. (mol.cm)-1). The absorbance A280-298 nm > 8, i.e. T280-298 nm < 10(-8). The band width was about 16 nm. Therefore, this material has conspicuous characteristics such as high transmittance and deep cutoff in the background in ultraviolet-c band. The transmission peak can be shifted in the range of 251-260 nm by changing the pH value of the solution and selecting solution with a different polarity. By analyzing the spectra of the material in acid we found that the two original absorption peaks disappeared, and a new strong absorption peak appeared at 290 nm. Moreover, it seems to be some relation between the two absorption peak intensity and the new peak, which needs to be further studied. Using this result, we can produce high quality UV-filters of this material with traditional coating and colour glass technology.
