ABSTRACT

Objective To investigate the effects of cetirizine on Janus kinase 2/signal transducer and activator of transcription 3 (JAK2/STAT3) pathway and mast cell activation in asthmatic mice by establishing a mouse model of asthma. Methods Sixty SPF BALB/c mice (10 in each group) were divided into control group, model group, low-dose cetirizine group (2 mg/kg), high-dose cetirizine group (5 mg/kg), cetirizine combined with AG490 group (5 mg/kg cetirizine combined with 0.4 mg/kg AG490). The asthma model was established by ovalbumin(OVA) sensitization and eliciting. Each treatment group was given certain amount of cetirizine by gavage every day, and the cetirizine combined with AG490 group was given intraperitoneal injection of 0.4 mg/kg AG490 once a week; the control group was given the same dose of normal saline until the end of modeling. The level of histamine in serum was detected by ELISA; the total number of cells in bronchoalveolar lavage fluid (BALF) was counted; the pathological changes of lung tissue were detected by HE staining; the expression of tryptase in lung tissue was detected by immunohistochemical staining; the expressions of pathway related proteins were detected by Western blot analysis. Results Compared with those in the control group, the level of histamine in serum, the total number of BALF cell, the eosinophilia count, the degree of lung pathological injury, and the expressions of tryptase, p-JAK2/JAK2, and p-STAT3/STAT3 in the model group were significantly higher; compared with those in the model group, the level of histamine in serum, the total number of BALF cell, the eosinophilia count, the degree of lung pathological injury, and the expressions of tryptase, p-JAK2/JAK2, and p-STAT3/STAT3 in low-dose group, high-dose group, and cetirizine combined with AG490 group were significantly lower; compared with those in the high-dose group, the level of histamine in serum, the total number of BALF cell, the eosinophilia count, the degree of lung pathological injury, and the expressions of tryptase and p-STAT3/STAT3 in cetirizine combined with AG490 group were significantly lower, however, the expression of p-JAK2/JAK2 had no significant change. Conclusion Cetirizine inhibits JAK2-STAT3 pathway activation and mast cell activation in lung tissue of asthmatic mice.

ABSTRACT

A technique to improve the accuracy of the retrieval of aerosol size-distribution is introduced. This technique includes a wavelength-selecting method and a "function model." The wavelength-selecting method is used to select the suitable wavelengths for the retrieval of aerosol size-distribution, and the function model is used to express the aerosol size-distributions with a fewer parameters. By using this technique, the accuracy of the retrieval of aerosol size-distribution can be improved.

ABSTRACT

The fluorescence property of DCM in water solution with added different concentration of SDS is reported. With increasing the concentration of SDS in water, the solubility of DCM and the fluorescence intensity increase. When the concentration of SDS increases from 0.025 to 0.5 mol x L(-1), the maximum fluorescence relative intensity is enhanced by a factor of 95. A powerful dye laser at wavelength of 648 nm was generated in the solution of DCM in water with SDS exited by a 532 nm laser. It shifts 13 nm compared with the dye laser wavelength of the solution of DCM in ethanol. And enhancement of SRS of C6H6 was achieved both in the solution of DCM in water with SDS and in the solution of DCM in ethanol. The result is that the second, the third and the fourth order Stokes of C6H6 are all enhanced. But the former mainly enhances the fourth order Stokes (632 nm) and the enhancement gain coefficient is 8.5, while the latter mainly enhances the third order Stokes (595 nm) and the enhancement gain coefficient is 2.5. The mechanism of solubility, fluorescence enhancement and its possible application are discussed.

ABSTRACT

The truncated second-order moments and generalized M2 factor (M(G)2 factor) of two-dimensional beams in the Cartesian coordinate system are extended to the case of three-dimensional rotationally symmetric hard-edged diffracted beams in the cylindrical coordinate system. It is shown that the propagation equations of truncated second-order moments and the M(G)2 factor take forms similar to those for the nontruncated case. The closed-form expression for the M(G)2 factor of rotationally symmetric hard-edged diffracted flattened Gaussian beams is derived that depends on the truncation parameter beta and beam order N. For N --> infinity, the M(G)2 factor equals 4/square root of 3 corresponding to the value of truncated plane waves, which guarantees consistency of the formalism.
