ABSTRACT

PURPOSE: To investigate the effect of lingual wedge shaped coronectomy in extraction of horizontally impacted mandibular third molar(HIMTM). METHODS: A total of 172 patients with HIMTM were randomly divided into experimental group and routine group with 86 cases in each group . Lingual wedge-shaped coronectomy was applied in the experimental group and T-shaped coronectomy was applied in routine group. Operation time, intraoperative and postoperative complications were compared between the two groups.The data were analyzed by using SPSS 26.0 software package. RESULTS: The operation time of the experimental group was significantly shorter than that of the control group (P<0.05). The rate of swelling, pain, lingual bone plate injury and broken root of the experimental group were separately lower than that of the control group, and the difference was statistically significant(P<0.05). CONCLUSIONS: Lingual wedge shaped coronectomy of HIMTM has significant advantages in the extraction of HIMTM. It can reduce operation time,broken roots,fracture of lingual plate, postoperative swelling, pain and other surgical complications.

ABSTRACT

OBJECTIVES: Ferroptosis is a newly discovered cell death mode that has been confirmed to occur in the intestinal epithelial cells in ulcerative colitis (UC). In this study we aimed to elucidate the mechanism of ferroptosis and its association with adenosine monophosphate-activated protein kinase (AMPK) in UC. METHODS: Gene expression profiles of colonic mucosa (GSE87473) were downloaded. Both human colonic samples and dextran sodium sulfate (DSS)-induced colitis murine model were used. The molecular markers of ferroptosis were detected using western blot and immunohistochemistry. Symptoms, iron abundance, and lipid peroxidation level of the mouse model were measured to evaluate the role of AMPK activation in ferroptosis. RESULTS: Both gene and protein expressions of GPX4 and FTH1 were decreased in UC patients compared with the healthy controls. An increased iron abundance and lipid peroxidation level in colon tissues and damaged mitochondria were found in DSS-induced colitis. AMPK expression was decreased in UC patients and correlated with FTH1 and GPX4. Activation of AMPK with metformin inhibited ferroptosis in the colon, improved symptoms, and prolonged the lifespan in DSS-induced colitis mice. CONCLUSIONS: Ferroptosis can be observed in colonic tissues in UC. AMPK activation inhibits ferroptosis in murine colitis model, which may act as a potential target for the treatment of colitis.

ABSTRACT

PURPOSE: To measure the cortical bone thickness of zygomatic alveolar ridge in adolescents and explore the correlation between cortical bone thickness and cervical vertebral bone age. METHODS: Cone-beam CT data of 80 adolescents were collected, including 20 adolescents with cervical vertebral bone ages of Cvs3, Cvs4, Cvs5 and Cvs6, respectively. CBCT images were reconstructed with the maxillary occlusal plane as the reference plane. Cortical bone thickness of different slices in the left maxillary zygomatic alveolar ridge area was measured in the direction parallel to and 60° from the reference plane, and the measured data were statistically analyzed by SPSS 21.0 software package. RESULTS: When the measurement direction was parallel to the reference plane and at 60°, the cortical bone thickness in the zygomatic alveolar ridge area of Cvs3-Cvs6 adolescents was (0.90±0.09) -(1.72±0.21) mm and (1.35±0.44)-(3.98±1.48) mm, respectively. There was significant difference in cortical bone thickness between Cvs4 and Cvs5 group(P<0.05). Spearman correlation analysis showed a strong positive correlation(P<0.01) between cortical bone thickness of zygomatic alveolar ridge and cervical vertebral bone age in adolescents. CONCLUSIONS: The cortical bone thickness of zygomatic alveolar ridge in adolescents increases with the increase of cervical vertebral bone age, and the cortical bone thickness may increase significantly during Cvs4-Cvs5. In terms of cortical bone thickness, all slices of zygomatic alveolar ridge of CVS3-CVS6 adolescents are suitable for implanting miniscrews, and the anterior slices should be selected for implantation as far as possible for Cvs3 and Cvs4 adolescents.

ABSTRACT

Empathy plays an essential role in determining how we understand and respond to others' emotional experiences. Previous studies have mainly focused on the influencing factors of physical pain and monetary loss empathy, as well as on their effects on subsequent feelings. However, little is known about social pain empathy and its influence on subsequent feelings, and the role of personal trait empathy. In this study, we performed Cyberball Games and gambling tasks to explore the effect of social exclusion empathy on subsequent feelings for gambling outcomes (others' losses and gains) by analyzing the brain activity of excluded and non-excluded players at the gambling outcome feedback stage. It was found that both feedback-related negativity (FRN) and theta-band oscillatory are more negative for loss outcome than gain for the excluded players, while these effects cannot be found for the non-excluded players. Furthermore, we only observed a larger P300 amplitude in the gain outcome than that of loss, regardless of the players' role. More interestingly, we found state social pain empathy is negatively correlated with the d-FRN amplitude under non-excluded conditions, while trait empathy is positively correlated with P300 amplitude regardless of the players' role. These results provide insight into the different roles of state social pain empathy and trait empathy on subsequent gambling outcomes. Furthermore, different from previous studies, the current results show that d-FRN could reflect the state empathy discrepancy rather than the biomarker of state empathy.

ABSTRACT

Grounded on the cognitive consistency theory, this paper adopts the prime-probe paradigm and Electroencephalography (EEG) experiment to examine the impact of country-of-origin (COO) stereotypes-brand positioning congruence on consumer behavior, the boundary effect of brand positioning strategy, as well as the underlying cognitive mechanism. Behaviorally, consumers show a higher purchase intention in the congruence condition. Moreover, this congruence effect of purchase intention can be found for competence brand positioning strategies rather than warmth brand positioning strategies. At the brain level, we found that compared with the congruence condition, the incongruence condition enhances consumers' cognitive conflict, reflected in enhanced frontal theta-band oscillation. Furthermore, the cognitive conflict effect is accentuated in the competence positioning strategy condition rather than the warmth strategy positioning condition, confirming the boundary effect of brand positioning strategy from the brain level. These findings provide neural evidence that the congruence between COO stereotypes and brand positioning influences consumer purchase behavior, reveals a boundary effect in the COO stereotype-brand positioning congruence, and highlights the importance of the competence dimension. Finally, the theoretical and practical implications are discussed.

ABSTRACT

Objective: To investigate the effects of Zhongfeng capsule on the autophagy-related proteins expression in rats with cerebral ischemia/reperfusion injury (CI/ RI), and to explore its neural protection mechanisms of the decoction. Methods: Rat middle cerebral artery ischemia/reperfusion injury model (ischemia for 2 h, reperfusion for 24 h) was prepared by the improved line plug method. Sixty male SD rats were randomly divided into sham operation group, model group, butylphthalide group(0.054 g/kg), Zhongfeng capsule high-dose groups (1.08 g/kg), Zhongfeng capsule middle-dose groups (0.54 g/kg), Zhongfeng capsule low-dose groups (0.27 g/kg), with 10 rats in each group. Rats were treated with Zhongfeng capsule by gavage once a day for 10 days. The rats were sacrificed and the brain tissue was obtained after the experiment in each group. Score neurological deficit was evaluated after 24 h of the last intervention in rat of each group. The pathological changes of brain tissue were observed by HE staining. The serum levels of estradiol (E2) and follicle stimulating hormone (FSH) were determined by ELISA. The expressions of key genes and proteins of PI3K/Akt/Beclin1 signaling pathway in brain tissue were detected by qRT-PCR and Western blot respectively. Results: Compared with the sham operation group, the body weight and protein expressions of p-PI3k and p-Akt in brain tissue of rats were decreased significantly in the model group, while the brain index, neurological deficit score, gene and protein expressions of Beclin1 and LC3 were increased markedly in the model group(P<0.05 or P<0.01). In the model group, nerve cells of brain tissue were loosely packed, interstitial edema, triangular in shape, nuclear pyknosis and dark-blue staining were observed. Compared with the model group, the body weight of rats was increased obviously, the neurological deficit score was decreased significantly and the pathological injury of brain tissue was alleviated evidently in high-dose of Zhongfeng capsule group (P<0.05). The brain index, the gene and protein expressions of Beclin1 and LC3 were decreased apparently in Zhongfeng capsule treatment groups(P<0.05 or P<0.01), while the expressions of p-PI3k and p-Akt in brain tissue were increased evidently in Zhongfeng capsule treatment groups(P<0.05 or P<0.01). Conclusion: Zhongfeng capsule can inhibit autophagy and improve brain neurons lesion of CIRI rats, the mechanism may be related to regulate the expression of Beclin1 and LC3 in PI3K/Akt/Beclin1 signaling pathway.

ABSTRACT

There is ample experimental evidence showing that the proposers' social role is related to individuals' fairness perception in the Ultimatum Game (UG). However, various social roles, e.g., degree of economic neediness, have different influences on fairness perception, yet it has not been well studied. In this study, we adapted the UG paradigm and recorded electroencephalography (EEG) to probe the neural signatures of whether and how the degree of neediness influences fairness perception. Behavioral results showed that responders are prone to accept unfair offers from proposers in need more than those who are not in need. At the brain level, MFN (medial frontal negativity) was more negative-going in response to unfair than fair offers for not-in-need proposers. In contrast, we found a reversed MFN difference response to unfair and fair offers for in-need proposers, showing a strongly pure altruistic phenomenon. Moreover, we found smaller P300 amplitude was induced in the proposer-in-need condition, compared with its counterpart, while a negative correlation between empathy rating and P300 amplitude in the proposer-in-need condition regardless of the offers' fairness. The current results indicate that the degree of neediness might reduce fairness perception by promoting the empathic concern toward the in-need proposers rather than decreasing the empathic concern for the not-in-need proposers.

ABSTRACT

This study aimed to explore the main active ingredients and potential targets of Solanum nigrum(SN), so as to reveal the potential molecular mechanism of SN in the treatment of hepatocellular carcinoma(HCC) based on network pharmacology and molecular docking. First,the main active ingredients and predictive targets of SN were collected in the traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP). Then,the targets relating to HCC were collected through retrieval of integrated bio-pharmacological network database for traditional Korean medicine(PharmDB-K), oncogenomic database of hepatocellular carcinoma(OncoDB.hcc). The common targets of disease-drug component were selected through intersection between predictive targets and disease targets. Next, based on the String platform, protein-protein interaction network(PPI) model of the potential anti-HCC targets was constructed using the software Cytoscape 3.7.1. ClueGO and CluePedia APP in Cytoscape were used to analyze the gene function of SN in the treatment of HCC, and construct the main active ingredients-potential targets-signal pathways topology network of SN. Finally,DISCOVERY STUDIO software was applied in verifying the molecular docking between the key active ingredient and potential protein target. The results showed that there were 4 main active ingredients of SN, involving 22 potential targets relating to HCC and 7 signal pathways relating to potential anti-HCC targets of SN. Network analysis showed that SN may play a therapeutic role in HCC by acting on key targets, such as EGFR, TP53, MYC, CCND1 and CTNNB1. Molecular docking results showed that quercetin and EGFR could bind stably and interact through amino acid residues LEU718, LYS745 and GLN791. This study revealed the potential active ingredients and the possible molecular mechanism of SN for treatment of HCC, providing scientific basis for follow-up exploration of the molecular mechanism of SN against HCC.

ABSTRACT

Previous studies have shown that generosity is driven by empathy and that both generosity in economic sharing behavior and monetary loss empathy decay as the social distance increases. However, it is still unclear whether this decay in economic sharing generosity can be influenced by the decay in monetary loss empathy. In the current study, we carried out two experiments to investigate this issue to deepen our understanding of the relationship between monetary loss empathy and generosity in economic sharing behavior. Our results show that in the observation group (observers watch their friend, and a stranger plays a gambling game), a negative correlation between log-transformed k value (ln(k)) and the distinction of d-FRN (feedback-related negativity difference between gain and loss) between friends and strangers was observed. However, in the execution group (executors play a gambling game themselves and watch a stranger play the same gambling game), there was no significant correlation between ln(k) and the distinction of d-FRN between self and strangers. Current results indicate that the decayed generosity across different social distances in economic sharing behavior can be modulated by the decayed monetary loss empathy. The study adds weight to the relationship between decayed monetary loss empathy and decayed generosity in sharing economic behavior at the level of social distance and provides electrophysiological evidence.

ABSTRACT

Periodontal diagnosis requires discovery of the relations among teeth, gingiva (i.e., gums), and alveolar bones, but alveolar bones are inside gingiva and not visible for inspection. Traditional probe examination causes pain, and X-ray based examination is not suited for frequent inspection. This work develops an automatic non-invasive periodontal inspection framework based on gum penetrative Optical Coherence Tomography (OCT), which can be frequently applied without high radiation. We sum up interference responses of all penetration depths for all shooting directions respectively to form the shooting amplitude projection. Because the reaching interference strength decays exponentially with tissues' penetration depth, this projection mainly reveals the responses of the top most gingiva or teeth. Since gingiva and teeth have different air-tissue responses, the gumline, revealing itself as an obvious boundary between teeth and gingiva, is the basis line for periodontal inspection. Our system can also automatically identify regions of gingiva, teeth, and alveolar bones from slices of the cross-sectional volume. Although deep networks can successfully and possibly segment noisy maps, reducing the number of manually labeled maps for training is critical for our framework. In order to enhance the effectiveness and efficiency of training and classification, we adjust Snake segmentation to consider neighboring slices in order to locate those regions possibly containing gingiva-teeth and gingiva-alveolar boundaries. Additionally, we also adapt a truncated direct logarithm based on the Snake-segmented region for intensity quantization to emphasize these boundaries for easier identification. Later, the alveolar-gingiva boundary point directly under the gumline is the desired alveolar sample, and we can measure the distance between the gumline and alveolar line for visualization and direct periodontal inspection. At the end, we experimentally verify our choice in intensity quantization and boundary identification against several other algorithms while applying the framework to locate gumline and alveolar line in vivo data successfully.

ABSTRACT

Digital dental reconstruction can be a more efficient and effective mechanism for artificial crown construction and period inspection. However, optical methods cannot reconstruct those portions under gums, and X-ray-based methods have high radiation to limit their applied frequency. Optical coherence tomography (OCT) can harmlessly penetrate gums using low-coherence infrared rays, and thus, this work designs an OCT-based framework for dental reconstruction using optical rectification, fast Fourier transform, volumetric boundary detection, and Poisson surface reconstruction to overcome noisy imaging. Additionally, in order to operate in a patient's mouth, the caliber of the injector is small along with its short penetration depth and effective operation range, and thus, reconstruction requires multiple scans from various directions along with proper alignment. However, flat regions, such as the mesial side of front teeth, may not have enough features for alignment. As a result, we design a scanning order for different types of teeth starting from an area of abundant features for easier alignment while using gyros to track scanned postures for better initial orientations. It is important to provide immediate feedback for each scan, and thus, we accelerate the entire signal processing, boundary detection, and point-cloud alignment using Graphics Processing Units (GPUs) while streamlining the data transfer and GPU computations. Finally, our framework can successfully reconstruct three isolated teeth and a side of one living tooth with comparable precisions against the state-of-art method. Moreover, a user study also verifies the effectiveness of our interactive feedback for efficient and fast clinic scanning.

Aggressive red imported fire ants (RIFAs) are expanding their habitat due to active international trade and global warming. To prevent infestation and settlement, RIFAs must be removed during the quarantine process. Because RIFAs are social insects and have different morphological characteristics depending on their castes, non-ant taxonomists have difficulty confirming RIFAs based on their morphological characteristics alone. The disadvantages of previously reported RIFA molecular diagnostics are that they require additional steps, such as restriction enzyme digestion followed by agarose gel electrophoresis separation or DNA sequence verification for polymerase chain reaction (PCR)-amplified products. To overcome these drawbacks, two RIFA-specific genes were selected and used to develop diverse PCR-based RIFA molecular diagnostic techniques. We found that RIFAs could be confirmed by conventional PCR targeting of two RIFA-specific genes followed by agarose electrophoresis separation. In addition, TaqMan probe real-time PCR methods had the advantage of confirming RIFAs immediately after the reactions were completed by observing fluorescence indexes. Finally, multiplex PCRs enhanced RIFA specificity and sensitivity. The new molecular diagnostic methods developed in this study had the advantages of reducing false positive and negative results together with high specificity and sensitivity for RIFAs.

Previous studies have documented that people tend to respond faster and memorize better to the in-group traits. It may be particularly manifest for ethnic minorities, due to their salient ethnic identity. However, few studies have explored how the valence of traits modulates the in-group preference effect. The present study examined the impacts of ethnic identity salience and the valence of traits on the group-preference effect among 33 Han Chinese in a Tibetan-dominant area and 32 Tibetan participants in a Han-dominant area. Two weeks before the experiment, we measured the ethnic identity salience of participants in both groups. In the formal experiment, we used the group-reference effect (GRE) paradigm with three encoding tasks. The results showed that, regardless of whether ethnic identity was salient, both groups responded faster to positive traits than to negative traits when evaluating their own group, whereas there were no significant difference between the processing of positive traits and negative traits in the out-group evaluation and font judgment tasks. This suggested a pervasive processing advantage of the in-group positive characteristics. The results imply that self-enhancement motivation had a moderation effect on the GRE, as well as the ethnic identity salience may not be necessary for a GRE.

ABSTRACT

OBJECTIVE: To explore the diagnostic value of MRI in the transient dislocation of the patella. METHODS: The DR and MRI data of 35 patients with acute patellar transient dislocation from January 2015 to December 2017 were retrospectively analyzed, including 12 males and 23 females, ranging in age from 10 to 23 years old, with an average of 17 years old. The differences between DR and MRI diagnosis and surgical or discharge diagnosis were compared, and the difference between them in diagnosis of knee anatomical position, bone injury, signs and soft tissue injury were compared. The R statistical software WiLcoxon signed rank test (WiLcoxon signed rank test) was used for statistical analysis. RESULTS: All 35 MRI findings were completely consistent with postoperative or discharge diagnosis. According to MRI, 7 cases of high patella and 7 cases of patella subluxation were diagnosed. According to DR, 7 cases of high patella and 0 cases of patella subluxation were diagnosed. Fourteen dislocations were diagnosed by MRI and 7 dislocations were diagnosed by DR. There was statistically significant difference between the two methods in the diagnosis of abnormal position of patella(v=10, P<0.05). Total 112 cases of bone injuries were diagnosed by MRI, and 0 was diagnosed by DR. There was statistically significant difference between the two methods in the diagnosis of bone injuries(v=16, P<0.05). Total 43 cases of soft tissue injuries were diagnosed by MRI, and 0 was diagnosed by DR. There was statistically significant difference between the two methods in the diagnosis of soft tissue injuries (v=21, P<0.05). Total 35 cases of hydrops of the knee joint were diagnosed by MRI, and 30 were diagnosed by DR. There was statistically significant difference between the two methods in the diagnosis of hydrops of the knee joint(v=32, P>0.05). CONCLUSIONS: MRI is superior to DR in the diagnosis of acute temporal bone dislocation. MRI should be used as the first choice for imaging diagnosis of acute temporal bone dislocation. It can avoid missed diagnosis and misdiagnosis.

At the beginning of the ancient "Silk Road", traditional Chinese medicine resources (TCM resources) have long been integrated into it, and it was once the "important part" of the ancient "Silk Road" in Chinese history, benefited from this, the political connections was strengthened, the economic and trade was developed, and Chinese medicine culture was spread. Before Qing Dynasty, people took out the "silk" and brought back "herbs" on the "Silk Road", which enriched China's medicinal resources. While in the later Qing Dynasty and the period of the Republic of China, more scientific and technological methods were brought back from abroad, and this in turn enriched the medical research methods. With more than 2 000 years' development, China's boundaries have changed countless times, therefore the word "Foreign" has different meanings in different historical periods. The import, development and utilization of foreign TCM resources is different in different historical periods, at present, with the continuous enhancement of China's comprehensiv national strength,especially the "Belt And Road" strategy, the communication and import of TCM resources with countries that participating in the "Belt and Road" initiativ became much easier. In order to actively respond to the "Belt and Road" initiative promote the development and utilization of foreign medicinal resources; serve people from the countries participating in the "Belt and Road" initiative we sorted out the foreign TCM resources from different periods in ancient and modern China, and based on this, we proposed the strategy of import of foreign TCM resources under the new situation, those are: "three levels, four steps, one support and one key". And "three levels" refers to intelligence collection level, field investigation level, scientific research level, "Four steps" refers to the original identification research, safety evaluation research, functional positioning research and medicinal properties research; "One support" refers to relying on overseas Chinese or Chinese enterprises with working foundation and network resources, "One key" refers to the protection of the intellectual property rights of foreign TCM resources at the policy level and to mobilize the initiative of all parties.

Orientation direction dependency of whitening activated at large strains was studied using four pre-oriented isotactic polypropylene (iPP) samples with different molecular weights stretched along different directions with respect to the pre-orientation (0°, 45°, and 90°) by means of in situ wide-, small-, and ultra-small-angle X-ray scattering techniques. A macroscopic fracture of iPP materials was also observed following the stress whitening at large strains. These two associated processes in pre-oriented iPP samples at elevated temperatures were found to be governed by not only the molecular weight of iPP but also the pre-orientation direction. For a certain pre-orientation direction of iPP, both the critical stress of cavitation induced-whitening and failure stress increased with increasing molecular weight. For one given molecular weight, the pre-oriented iPP showed the smallest critical stress for whitening and failure stress along the pre-orientation direction (0°) while the samples displayed larger values for the same behaviors when stretched at 45° or 90° with respect to the pre-orientation direction. Such behavior suggested that oriented amorphous networks, with different mechanical strengths, can be generated during the second deformation processes in these pre-oriented iPP samples. The evolution of inter-fibrillar tie chains in highly oriented amorphous networks was considered as the main factor controlling the response of the inner network to the external stress since the cavitation-induced whitening activated at large strains was caused by the failure of load bearing inter-fibrillar tie chains in the oriented amorphous network.

ABSTRACT

BACKGROUND: Parkinson's disease (PD) patients with long-term levodopa (L-DOPA) treatment are suffering from severe circadian dysfunction. However, it is hard to distinguish that the circadian disturbance in patients is due to the disease progression itself, or is affected by L-DOPA replacement therapy. This study was to investigate the role of L-DOPA on the circadian dysfunction in a rat model of PD. METHODS: The rat model of PD was constructed by a bilateral striatal injection with 6-hydroxydopamine (6-OHDA), followed by administration of saline or 25 mg/kg L-DOPA for 21 consecutive days. Rotarod test, footprint test, and open-field test were carried out to evaluate the motor function. Striatum, suprachiasmatic nucleus (SCN), liver, and plasma were collected at 6:00, 12:00, 18:00, and 24:00. Quantitative real-time polymerase chain reaction was used to examine the expression of clock genes. Enzyme-linked immunosorbent assay was used to determine the secretion level of cortisol and melatonin. High-performance liquid chromatography was used to measure the neurotransmitters. Analysis of variance was used for data analysis. RESULTS: L-DOPA alleviated the motor deficits induced by 6-OHDA lesions in the footprint and open-field test ( P < 0.01, P < 0.001, respectively). After L-DOPA treatment, Bmal1 decreased in the SCN compared with 6-OHDA group at 12:00 ( P < 0.01) and 24:00 ( P < 0.001). In the striatum, the expression of Bmal1, Rorα was lower than that in the 6-OHDA group at 18:00 (P < 0.05) and L-DOPA seemed to delay the peak of Per2 to 24:00. In liver, L-DOPA did not affect the rhythmicity and expression of these clock genes (P > 0.05). In addition, the cortisol secretion was increased (P > 0.05), but melatonin was further inhibited after L-DOPA treatment at 6:00 (P < 0.01). CONCLUSIONS: In the circadian system of advanced PD rat models, circadian dysfunction is not only contributed by the degeneration of the disease itself but also long-term L-DOPA therapy may further aggravate it.

ABSTRACT

Composite materials based on expanded graphite (EG) and metal oxide (MO) particles was prepared by an explosive combustion and blending method. The objective of the study was to develop EG impregnated with metal oxide particulates (Ag2O, CuO and ZnO) and evaluate the level of protection the materials conferred against biological agents. The physical properties of the EG/MO composites were examined using SEM, EDX and XRD spectroscopy, and the results indicated that the MO particles were incorporated into the EG matrix after impregnation. The antimicrobial activities of the EG/MO composites against Gram-positive bacteria, Gram-negative bacteria and Bacillus anthracis were investigated using zone of inhibition, minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC) and plate-counting methods. EG/Ag2O exhibited a stronger antibacterial activity than EG/CuO and EG/ZnO, with a MIC of 0.3mg/mL and a MBC of 0.5mg/mL. To the best of our knowledge, few studies have demonstrated that EG/MO composites can inhibit the growth of Bacillus anthracis-adhered cells, thus preventing the process of biofilm formation. Nanoscale metal oxides display enhanced reactive properties toward bacteria due to their high surface area, large number of highly reactive edges, corner defect sites and high surface to volume ratio.

ABSTRACT

BACKGROUND: Increasing evidence suggests that microRNAs are functionally involved in the initiation and maintenance of pain hypersensitivity, including chronic morphine analgesic tolerance, through the posttranscriptional regulation of pain-related genes. We have previously demonstrated that miR-219 regulates inflammatory pain in the spinal cord by targeting calcium/calmodulin-dependent protein kinase II gamma (CaMKIIγ). However, whether miR-219 regulates CaMKIIγ expression in the dorsal root ganglia to mediate morphine tolerance remains unclear. RESULTS: MiR-219 expression was downregulated and CaMKIIγ expression was upregulated in mouse dorsal root ganglia following chronic morphine treatment. The changes in miR-219 and CaMKIIγ expression closely correlated with the development of morphine tolerance, which was measured using the reduction of percentage of maximum potential efficiency to thermal stimuli. Morphine tolerance was markedly delayed by upregulating miR-219 expression using miR-219 mimics or downregulating CaMKIIγ expression using CaMKIIγ small interfering RNA. The protein and mRNA expression of brain-derived neurotrophic factor were also induced in dorsal root ganglia by prolonged morphine exposure in a time-dependent manner, which were transcriptionally regulated by miR-219 and CaMKIIγ. Scavenging brain-derived neurotrophic factor via tyrosine receptor kinase B-Fc partially attenuated morphine tolerance. Moreover, functional inhibition of miR-219 via miR-219-sponge in naive mice elicited thermal hyperalgesia and spinal neuronal sensitization, which were both suppressed by CaMKIIγ small interfering RNA or tyrosine receptor kinase B-Fc. CONCLUSIONS: These results demonstrate that miR-219 contributes to the development of chronic tolerance to morphine analgesia in mouse dorsal root ganglia by targeting CaMKIIγ and enhancing CaMKIIγ-dependent brain-derived neurotrophic factor expression.
