ABSTRACT

Parturition and the early neonatal period are critical life history stages in ungulates with considerable implications for population growth and persistence. Understanding the changes in behaviour induced by ungulate parturition is important for supporting effective population management, but reliably identifying birth sites and dates presents a challenge for managers. Rocky Mountain bighorn sheep (Ovis canadensis canadensis) are one such highly valued and ecologically important species in montane and subalpine ecosystems of Western North America. In the face of changing patterns of anthropogenic land use, wildlife managers increasingly require site-specific knowledge of the movement and habitat selection characteristics of periparturient sheep to better inform land use planning initiatives and ensure adequate protections for lambing habitat. We used movement data from GPS collared parturient (n = 13) and non-parturient (n = 8) bighorn sheep in Banff National Park, Canada to (1) identify lambing events based on changes in key movement metrics, and (2) investigate how resource selection and responses to human use change during the periparturient period. We fit a hidden Markov model (HMM) to a multivariate characterization of sheep movement (step length, daily home range area, residence time) to predict realistic lambing dates for the animals in our study system. Leave-one-out cross validation of our model resulted in a 93% success rate for parturient females. Our model, which we parameterized using data from known parturient females, also predicted lambing events in 25% of known non-parturient ewes in a test dataset. Using a latent selection difference function and resource selection functions, we tested for postpartum changes in habitat use, as well as seasonal differences in habitat selection. Immediately following lambing, ewes preferentially selected high-elevation sites on solar aspects that were more rugged, closer to escape terrain, and further from roads. Within-home range habitat selection was similar between individuals in different reproductive states, but parturient ewes had stronger selection for low snow depth, sites closer to barren ground, and sites further from trails. We propose that movement-based approaches such as HMMs are a valuable tool for identifying critical parturition habitat in species with complex movement patterns and may have particular utility in study areas without access to extensive field observations or vaginal implant transmitters. Furthermore, our results suggest that managers should minimize human disturbance in lambing areas to avoid interfering with maternal behaviour and ensure access to a broad range of suitable habitat in the periparturient period.

ABSTRACT

From February to May 2021, four nonmigratory rams from the Radium-Stoddart bighorn sheep (BHS; Ovis canadensis) herd in the Rocky Mountains of southeastern British Columbia, Canada, died from infection with the giant liver fluke, Fascioloides magna. Affected animals were emaciated, weak, and lethargic or were found dead. Gross lesions, histopathology, and parasite burdens were consistent with those reported in experimentally infected BHS, domestic sheep, and other aberrant hosts. Although BHS range does not typically overlap with fluke-contaminated aquatic habitats, the change in migratory behavior recently observed in some Radium-Stoddart rams may have exposed the affected animals to F. magna. We describe clinical signs and gross and histopathologic findings of hepatobiliary trematodiasis associated with F. magna in free-ranging BHS. From experimental data and our findings, giant liver fluke is pathogenic and is a threat to the conservation of the Radium-Stoddart BHS herd and other BHS herds in endemic F. magna regions.

ABSTRACT

An adult female Rocky Mountain bighorn sheep (Ovis canadensis canadensis) was euthanized because of a large mandibular mass. Histopathology revealed an ameloblastic fibro-odontoma, a rare odontogenic neoplasia.

ABSTRACT

The complete genome sequence of a novel circovirus (elk circovirus (ElkCV) Banff/2019) was determined via high throughput sequencing of liver tissue from a euthanized Rocky Mountain elk (Cervus canadensis nelsoni) from Alberta, Canada. The genome is circular and 1,787 nucleotides long, with two major ORFs encoding predicted proteins. Comparative genomic analysis to 4,164 publicly available complete and near complete circovirus genomes showed that ElkCV shares approximately 65% pairwise genome-wide nucleotide identity with the most closely related circovirus species, porcine circoviruses (PCV) 1 and 2 and bat-associated circovirus (BatACV) 11. ElkCV features a stem-loop within the origin of replication region characteristic of circoviruses. However, it differs from those found in PCV1, PCV2 and BatACV11 since it has a longer stem and contains hexamer repeats that overlap the stem in opposing orientations. Interestingly, stem-loop structures of similar length featuring repeats in a similar position and orientation are also seen in some avian circoviruses. Based on the demarcation threshold established by the International Committee on Taxonomy of Viruses (ICTV) for members of Circoviridae (80% pairwise genome-wide nucleotide identity), ElkCV represents a novel species and is the first complete circovirus genome reported from a cervid host.

ABSTRACT

We used molecular analyses to confirm Mycobacterium avium spp. hominissuis infection in lung granulomas and pyogranulomas in the tracheobronchial lymph node in a wild mule deer in Banff, Canada. These lesions are similar to those found in M. bovis-infected animals, emphasizing the critical need for disease surveillance in wildlife populations.

ABSTRACT

The genus Macavirus of the subfamily Gammaherpesvirinae comprises two genetically distinct lineages of lymphotropic viruses. One of these lineages includes viruses that can cause malignant catarrhal fever (MCF), which are known as MCF viruses (MCFV). All MCFVs are genetically and antigenically related but carried by different hosts. In this study, we report the recognition of new MCFV carried by bighorn sheep. The virus was first identified in a bighorn sheep from Banff National Park, Alberta, Canada. Analysis of a conserved region of the viral DNA polymerase gene of the virus carried by this bighorn sheep showed 85.88% nucleotide identity to the MCFV carried by domestic sheep, ovine herpesvirus 2 (OvHV-2). Further investigation of bighorn samples obtained from animals in the US and Canada showed 98.87-100% identity to the DNA polymerase sequence of the first bighorn in the study. Phylogenetic analysis indicated that the MCFV carried by bighorn sheep is closely related but distinct from OvHV-2. Epidemiological and virulence features of the newly recognized MCFV are still unknown and warrant further investigation. Considering the current nomenclature for MCFVs, we suggest a tentative designation of ovine herpesvirus-3 (OvHV-3) for this newly identified bighorn sheep MCFV.

ABSTRACT

Boreal woodland caribou (Rangifer tarandus caribou) are listed as threatened across Canada, and a basic understanding of their health status is lacking. From December 2012 to April 2013, we investigated multiple health indices for adult female boreal caribou (n=163) captured from seven herds in NE British Columbia, Canada. Health indices included physical characteristics, physiologic and trace mineral status, exposure to or infection with selected pathogens, and measures of chronic stress and inflammation, including serum amyloid A, haptoglobin, and hair cortisol concentration. Key findings were exposure to the bacterium Erysipelothrix rhusiopathiae in 14% of individuals, mild to severe hair loss associated with winter tick (Dermacentor albipictus) infestations in 76% of caribou from December to early February and 81% from late February to early April, and evidence of trace mineral deficiencies with 99% and 34% of individuals deficient in copper and selenium, respectively. Seroprevalence for exposure to selected pathogens was: alphaherpesvirus (63%), pestivirus (1%), Besnoitia spp. (60%), and Neospora caninum (2%). All animals were seronegative to Brucella spp. and Toxoplasma gondii. Mycobacterium avium ssp. paratuberculosis was not detected in any fecal samples. Parasite eggs or larvae, including Parelaphostrongylus andersoni (36%), Skrjabinema spp. (1%), Strongyle-type eggs (11%), Moniezia-type eggs (8%), and nematodirines (3%), were detected on fecal examination, but at low intensity. Blood biochemistry values and hair cortisol concentrations were within ranges previously reported in Rangifer tarandus sspp. Some significant differences among herds were noted, including antler morphology, exposure to Besnoitia spp., and concentrations of serum amyloid A, copper, cobalt, manganese, and iron.

Methodological differences among laboratories are recognized as significant sources of variation in quantification of hair cortisol concentration (HCC). An important step in processing hair, particularly when collected from wildlife, is the choice of solvent used to remove or "wash" external hair shaft cortisol prior to quantification of HCC. The present study systematically compared methanol and isopropanol as wash solvents for their efficiency at removing external cortisol without extracting internal hair shaft cortisol in samples collected from free-ranging grizzly bears and polar bears. Cortisol concentrations in solvents and hair were determined in each of one to eight washes of hair with each solvent independently. •There were no significant decreases in internal hair shaft cortisol among all eight washes for either solvent, although methanol removed detectable hair surface cortisol after one wash in grizzly bear hair whereas hair surface cortisol was detected in all eight isopropanol washes.•There were no significant differences in polar bear HCC washed one to eight times with either solvent, but grizzly bear HCC was significantly greater in hair washed with isopropanol compared to methanol.•There were significant differences in HCC quantified using different commercial ELISA kits commonly used for HCC determinations.

ABSTRACT

Malignant catarrhal fever-like clinical disease was diagnosed in a free-ranging bighorn sheep ( Ovis canadensis ) from Alberta, Canada, in June 2015. Antemortem and gross pathology findings included muscle atrophy, marked weight loss, and bilaterally symmetric alopecia with hyperpigmentation and crusting over the face, medial surfaces of the pinnae, dorsal trunk, distal limbs, perineal area, and tail. Histologically, the skin lesions were characterized by granulomatous mural folliculitis with numerous multinucleated giant cells and fewer lymphocytes and eosinophils consistent with previous reports of chronic ovine herpesvirus-2 (OvHV-2) infection. Multiple skin samples were positive for OvHV-2 DNA on PCR, and on partial sequencing of the viral DNA, there was 94% homology with reference GenBank OvHV-2. Quantitative PCR confirmed an increased level of OvHV-2 DNA in the lesional skin tissues. Based on exclusion of other disease processes, gross and histological lesions, PCR, and viral DNA sequencing results, a diagnosis of OvHV-2-mediated malignant catarrhal fever-like dermatitis was made.

Northern ecosystems are currently experiencing unprecedented ecological change, largely driven by a rapidly changing climate. Pathogen range expansion, and emergence and altered patterns of infectious disease, are increasingly reported in wildlife at high latitudes. Understanding the causes and consequences of shifting pathogen diversity and host-pathogen interactions in these ecosystems is important for wildlife conservation, and for indigenous populations that depend on wildlife. Among the key questions are whether disease events are associated with endemic or recently introduced pathogens, and whether emerging strains are spreading throughout the region. In this study, we used a phylogenomic approach to address these questions of pathogen endemicity and spread for Erysipelothrix rhusiopathiae, an opportunistic multi-host bacterial pathogen associated with recent mortalities in arctic and boreal ungulate populations in North America. We isolated E. rhusiopathiae from carcasses associated with large-scale die-offs of muskoxen in the Canadian Arctic Archipelago, and from contemporaneous mortality events and/or population declines among muskoxen in northwestern Alaska and caribou and moose in western Canada. Bacterial genomic diversity differed markedly among these locations; minimal divergence was present among isolates from muskoxen in the Canadian Arctic, while in caribou and moose populations, strains from highly divergent clades were isolated from the same location, or even from within a single carcass. These results indicate that mortalities among northern ungulates are not associated with a single emerging strain of E. rhusiopathiae, and that alternate hypotheses need to be explored. Our study illustrates the value and limitations of bacterial genomic data for discriminating between ecological hypotheses of disease emergence, and highlights the importance of studying emerging pathogens within the broader context of environmental and host factors.

The measurement of cortisol in hair is becoming important in studying the role of stress in the life history, health and ecology of wild mammals. The hair cortisol concentration (HCC) is generally believed to be a reliable indicator of long-term stress that can reflect frequent or prolonged activation of the hypothalamic-pituitary-adrenal axis over weeks to months through passive diffusion from the blood supply to the follicular cells that produce the hair. Diffusion of cortisol from tissues surrounding the follicle and glandular secretions (sebum and sweat) that coat the growing hair may also affect the HCC, but the extent of these effects is thought to be minimal. In this study, we report on a range of factors that are associated with, and possibly influence, cortisol concentrations in the hair of free-ranging brown bears (Ursus arctos). Through two levels of analyses that differed in sample sizes and availability of predictor variables, we identified the presence or absence of capture, restraint and handling, as well as different methods of capture, as significant factors that appeared to influence HCC in a time frame that was too short (minutes to hours) to be explained by passive diffusion from the blood supply alone. Furthermore, our results suggest that HCC was altered after hair growth had ceased and blood supply to the hair follicle was terminated. However, we also confirmed that HCC was inversely associated with brown bear body condition and was, therefore, responsive to diminished food availability/quality and possibly other long-term stressors that affect body condition. Collectively, our findings emphasize the importance of further elucidating the mechanisms of cortisol accumulation in hair and the influence of long- and short-term stressors on these mechanisms.

ABSTRACT

A 12-year-old Clydesdale gelding was presented for colic and dysuria. Obstructive urolithiasis and chronic renal disease were diagnosed via transurethral endoscopy and percutaneous ultrasonography. Nephroliths, hydronephrosis, and peri-ureteral fibrosis were present. Surgical intervention was declined and the gelding was managed medically with antibiotics and dietary modification.
